The physiological and phytosanitary qualities of common bean seeds used on a small farming system depend on the cultural practices, drying system and storage conditions employed. The objective of this work was to evaluate eight lots of common bean seeds using germination, vigor andphytosanitary tests. The lots were produced in 2005 and 2006 in “Zona da Mata”, Minas Gerais State, Brazil, and analyzed for moisture content, germination quality, first germination count, seedling vigor, seedling emergence, electric conductivity, seedling dry weight, seedling length, hypochlorite test and blotter test. The experimental design was completely randomized with four replications. The mean seed moisture content from all lots was 8.4 %. Four lots were classified as high vigor, one as intermediate vigor and three lots as low vigor. The first germination count and electrical conductivity tests were the best for evaluating bean seed physiological quality. All seed lots were infested by fungi, mainly Alternaria, Cladosporium and Fusarium.

ABSTRACT: Macauba palm stands out for having favorable features to biodiesel production such as the high oil content of its fruit. Considering the great potential of the species and their applicability in the renewable energy field, it becomes indispensable to establish the right conditions for storing the seeds for propagation purpose. The aim of this research was to evaluate the effect of seed moisture content, packaging, and storage conditions such as temperature and relative humidity on the quality of seeds from Minas Gerais State, during a 12-month storage period. The research had two independent assays: (I) the seeds were stored with three moisture contents/ranges 4.0≤6.0%; 6.0≤8.0% and 8.0≤10.0% in impermeable packages, under room temperature and at 10ºC; (II) seeds with approximately 5.9% of moisture content were stored in three different types of packages: a) permeable, b) semi-permeable and c) impermeable. Three storing conditions were tested: a) room temperature and RH under laboratory conditions; b) 15ºC and 45% RH; c) 20ºC and 55% RH. Water content, germination rate and germination speed index were evaluated at 0, 4, 8 and 12 months of storing. The best germination results were obtained with the moisture range of 6.0≤8.0%, with seeds kept at room temperature; while the seeds stored at 10ºC, regardless the moisture range, did not survive. The stored seeds with 5.9% moisture content and at both 15ºC/45%RH and 20ºC/55% RH conditions, independently of the package type used, showed the best results. Thus, macaw palm seeds can be classified as intermediates seeds.

Fungal incidence in stored common bean (Phaseolus vulgaris L.) is the main concern in order to preserve seed health and viability. The main aim of this study was to analyse these quality parameters in hermetically stored seeds at 10.2, 13.1, 16.2, 18.5% moisture content (MC) and 25, 30, 35, 40°C, through seed germination and health tests. Water activity recorded at 10.2 and 18.5% MC were 0.448 and 0.700, respectively. Low seed moisture content reduced Alternaria spp. incidence at 25-30°C. Highest incidence of Fusarium spp. (7.5%) occurred at 16.2% MC and 35-40°C. Highest incidences of Rhizoctonia spp. (8-10%) were recorded at 16.2-18.5% MC and 30-40°C. Penicillium spp. and Aspergillus spp. were predominant throughout the experiment and the highest incidences (80-100%; 20-30%, respectively) were scored at 18.5% MC and 30-35°C and 13.1-18.5% MC at 35°C, respectively. The higher the seed MC the higher the fungi incidence while lower seed MC decreased the incidences by 25%. Storage conditions below 30°C and 13.0% MC appear suitable to preserve common bean seed in relation to viability and health, up to a 8-month period.

Combinational traits of seed size and seed-coat hardness in Citrus garrawayi (F.M.Bailey) (syn. of Microcitrus garrowayi) were investigated as markers for estimation of seed morphological and physiological maturity. Seed size (length) and coat hardness correlated well with changes in seed coat and embryo morphological development, dry-weight accumulation, decreases in moisture content and a significant increase in germinability. Seed moisture content decreased from 82 ± 1% in immature seeds to 40 ± 1% at seed maturation. The outer integument of immature seeds consisted of thin-walled epidermal fibres from which outgrowths of emerging protrusions were observed. In comparison, mature seed coats were characterised by the thickening of the cell walls of the epidermal fibres from which arose numerous protrusions covered by an extensive mucilage layer. Immature seeds, with incomplete embryo and seed-coat histodiffereniation, had a low mean germination percentage of 4 ± 4%. Premature seeds, with a differentiated embryonic axis, were capable of much higher levels of germination (51 ± 10%) before the attainment of mass maturity. Mature seeds, with the most well differentiated embryonic axis and maximum mean dry weight, had the significantly highest level of germination (88 ± 3%).

Seed development was examined in Malva parviflora. The first flower opened 51 days after germination; flowers were tagged on the day that they opened and monitored for 33 days. Seeds were collected at 12 stages during this period and used to determine moisture content, germination of fresh seeds and desiccation tolerance (seeds dried to 10% moisture content followed by germination testing). Seed moisture content decreased as seeds developed, whereas fresh (max. 296 mg) and dry weight (max. 212 mg) increased to peak at 12–15 and ~21 days after flowering (DAF), respectively. Therefore, physiological maturity occurred at 21 DAF, when seed moisture content was 16–21%. Seeds were capable of germinating early in development, reaching a maximum of 63% at 9 DAF, but germination declined as development continued, presumably due to the imposition of physiological dormancy. Physical dormancy developed at or after physiological maturity, once seed moisture content declined below 20%. Seeds were able to tolerate desiccation from 18 DAF; desiccation hastened development of physical dormancy and improved germination. These results provide important information regarding M. parviflora seed development, which will ultimately improve weed control techniques aimed at preventing seed set and further additions to the seed bank.

International research on vegetable seed vigor is not at the same level attained for grain crops species. This study was conducted to identify reliable procedures for the accelerated aging and controlled deterioration tests to rank onion (Allium cepa L.) seed lots according to their physiological potential. Six seed lots of the cultivars Aurora and Petroline were evaluated in the laboratory for germination, first count, seedling vigor classification, traditional and saturated salt accelerated aging (41ºC / 48 and 72 h), controlled deterioration (24% of water / 45ºC / 24 h) and seedling emergence tests. Seed moisture content after the saturated salt accelerated aging test was lower and uniform, which is considered an important advantage in comparison to the traditional procedure. The saturated salt accelerated aging (41ºC / 48 and 72 h) and controlled deterioration (moisture content adjusted to 24% / 45ºC / 24 h) tests were the best procedures to assess the physiological potential of onion seeds, and are indicated for use in quality control programs.

AbstractThe effects of development of leek seeds at 20/10°, 25/15° and 30/20°C (day/night) and drying of seed harvested at different developmental stages on subsequent performance were examined in each of 3 years. An increase in temperature from 20/10° to 30/20°C reduced mean seed weight from 2.90 to 2.55 mg as a result of a reduction in the duration of seed growth from 80 to 55 days; seed growth rate was unaffected. Seed moisture content reached a minimum, up to 35 days after the attainment of maximum seed dry weight and 115, 90 and 70 days after anthesis at 20/10°, 25/15° and 30/20°C, respectively. The curves relating seed moisture to time for each temperature regime were mapped onto a single line accounting for >90% of the variation in moisture content, using accumulated day-degrees >6°C instead of chronological time. Seeds were capable of germinating when seed moisture contents were >60% (fresh weight basis), but maximum viability and minimum mean time to germination were not attained until seed moisture contents at harvest had fallen to 20–30%. Germination was little affected by temperature of seed development. Drying immature seeds increased percentage germination. Growing seeds at 30/20°C and drying at 35°C and 30% RH raised the upper temperature limit of germination compared with growing at 20/10°C and drying at 15°C and 30% RH.

The Ellis-Roberts seed viability equation is used to predict seed survival after storage at specified temperatures and moisture contents. Seed priming, which can break dormancy and accelerate germination, can also reduce seed storage life. Because primed seeds were not used in developing the Ellis-Roberts equation, the reciprocal nature of specific seed moisture content (MC, fresh weight basis) and temperatures that applies to nonprimed lettuce (Lactuca sativa L.) seeds may not apply to primed seeds. To determine how priming affects lettuce seeds in relation to the viability equation, an experiment was conducted using two cultivars, ‘Big Ben’ and ‘Parris Island Cos’. Seeds primed in polyethylene glycol 8000 (–1.45 MPa, 24 h at 15 °C) and nonprimed seeds were first adjusted to 6% and 9% moisture contents and then stored at 48 and 38 °C for up to 30 days, respectively. These storage conditions (6% MC and 48 °C; 9% MC and 38 °C) were predicted by the viability equation to result in equal longevities. Subsequent viability assays at 20 °C revealed that nonprimed seeds in both storage environments exhibited similar losses in viability over time, thus validating the Ellis-Roberts equation and the use of these conditions to apply different but equal aging stress. Primed seeds of both cultivars deteriorated faster than nonprimed seeds as expected. However, primed seeds did exhibit different rates of deterioration between the storage environments. Primed seeds stored at 9% MC and 38 °C deteriorated faster than primed seeds stored at 6% MC and 48 °C. The rate of decline in probit viability percentage was three times greater in primed ‘Big Ben’ seeds stored at 9% MC and 38 °C than for those stored at 6% MC and 48 °C (–1.34 versus –0.26 probits per day, respectively). ‘Parris Island Cos’ seeds stored at 9% MC and 38 °C had twice the rate of deterioration that those stored at 6% MC and 48 °C (–1.19 and –0.49 probits per day, respectively). The results indicate that primed lettuce seeds were more sensitive to the adverse effects of higher seed MC than were nonprimed seeds during storage at elevated temperatures.

Pterocarya fraxinifolia (Poir.) Spach (Juglandaceae) is a model relict tree species native to South Caucasus and is a typical element of threatened riparian forests. Intensive land transformations, which are common in Transcaucasia, have resulted in loss of natural habitat and population decline of the species. One of the methods of ex situ conservation is seed banking. Cryopreservation in liquid nitrogen (−196 °C) is of particular interest, as it allows safe preservation of valuable plant genetic resources. However, the feasibility of seed cryopreservation is related to the desiccation tolerance and intrinsic composition of the seeds. In this study, we examined the physiological traits of Pterocarya fraxinifolia seeds, for which desiccation tolerance is unknown or controversial, and their feasibility for cryopreservation. Additionally, we tested stratification methods for dormancy assessment. Results showed that seeds survived desiccation to a moisture content of 2.8% with a germination rate of 64%. Stratification at a temperature of 3 °C for 8 weeks proved to be both fast and effective. Seed moisture content ranging from 2.8% to 18.1% was determined to be safe for cryopreservation. There was no difference in seedling emergence in seeds stored for 1 year regardless of the storage temperature (−3, −18, or −196 °C). Based on our results, Pterocarya fraxinifolia seeds can be classified as orthodox. This study demonstrates for the first time the feasibility of cryopreserving Pterocarya fraxinifolia seeds.

Weed management is a major production issue facing otebo bean growers in Ontario. Field trials were conducted at six Ontario locations during a 2-yr period (2003 and 2004) to evaluate the tolerance of otebo bean to the preplant incorporated (PPI) application of EPTC at 4,400 and 8,800 g ai/ha, trifluralin at 1,155 and 2,310 g ai/ha, dimethenamid at 1,250 and 2,500 g ai/ha,S-metolachlor at 1,600 and 3,200 g ai/ha, and imazethapyr at 75 and 150 g ai/ha. EPTC, trifluralin, dimethenamid, andS-metolachlor applied PPI resulted in minimal (less than 5%) visual injury and with exception of the low rate of dimethenamid causing a 16% reduction in shoot dry weight and the high rate causing an 8% plant height reduction had no adverse effect on plant height, shoot dry weight, seed moisture content, and yield. Imazethapyr applied PPI caused up to 7% visual injury and reduced plant height, shoot dry weight, and yield 8, 18, and 12% at 75 g/ha and 19, 38, and 27% at 150 g/ ha, respectively. Seed moisture content was also reduced by 0.4% with both rates. Based on these results, otebo bean is not tolerant of imazethapyr applied PPI at rates as low as 75 g/ha, the proposed use rate. EPTC, trifluralin, dimethenamid, andS-metolachlor applied PPI have a 2× rate crop safety margin for use in otebo bean weed management.

The after-ripening requirement of three fall rye (Secale cereale L.) cultivars, harvested at three seed moisture contents (50, 42, and 32%), was assessed by monitoring the changes in seedling emergence from sowings made over a period of 60 days. All cultivars exhibited a short after-ripening requirement of 1–7 days. The absolute duration of the after-ripening requirement was not affected by the range of seed moisture contents studied. Seed treatment with captan [N[(trichloromethyl) thiol]-4-cyclohexene-1,2-dicarboximide] significantly increased percent seedling emergence. The captan treatment had the greatest effect on seeds harvested at the lowest moisture content.
