PURPOSE: To evaluate the effects of coronal leakage on concentration of hydrogen ions (pH) and calcium release of several calcium hydroxide pastes, over different periods of time. MATERIAL AND METHODS:  Fifty extracted human mandibular central incisors (n=10) were instrumented up to the F2 instrument and assigned to the following intracanal dressing: G1- Calen, G2- Calen with 0.4% chlorhexidine (CHX), G3- Calcium hydroxide with camphorated paramonochlorophenol (CPMC) and glycerin, G4- Calen, but temporary filling material maintained during all test (positive control) and G5- Root canal without intracanal dressing (negative control). All groups were immersed in distilled water for 7 days. In sequence, the temporary filling materials were removed, except in controls groups. All specimens were individually mounted on a specific device and only its root again immersed in distilled water. Concentration of hydrogen ions and calcium release by calcium hydroxide pastes in distilled water were evaluated in 24h, 7, 14 and 28 days. The results were submitted to ANOVA test (p = 0.05). After 28 days, root canals from experimental groups were examined in SEM. RESULTS: G1, G2, G3 and G4 presented similar pH values and calcium release and did not differ from each other (p>0.05), up to 7 days. After this time G1, G2 and G3 presented values lower values than G4 (p<0.05). In SEM analysis, calcium hydroxide residues were observed in all experimental groups. CONCLUSIONS: After 7 days, coronal leakage decreased the concentration of hydrogen ions and calcium ion release provided by all calcium hydroxide pastes.

Abstract Introduction If the restoration, that seal the root canal from oral environment, failed or solubilize, the intracanal dressing could be partially removed, decreasing the efficiency of calcium hydroxide (CH). Objective To evaluate the alkalizing potential and calcium ions release of intracanal residues from different medications, in different periods. Materials and method Human incisors were endodontically instrumented and randomly divided (n=10). The root canals were filled with: G1⎯Calcium hydroxide (CH); G2⎯CH with 0.4% chlorhexidine digluconate (CHC); G3⎯CH with camphorated paramonochlorophenol and glycerin (HPG); G4⎯CH was maintained during all experiment, as positive control group (PC) and G5⎯ No medication was performed in the negative control group (NC). The specimens were immersed in distilled water for 7 days. The intracanal dressing was removed from the experimental groups using F1 instrument (Protaper). The teeth were stored in distilled water for 24 hours, 7, 14 and 28 days. pH value and calcium ions released were evaluated and the data were submitted to ANOVA one-way and Tukey tests. Result All experimental groups presented lower alkalizing potential and calcium ions release than PC (P<0.05). No significant difference was found among the experimental groups, regardless of the period (P>0.05). Conclusion The alkalizing potential and calcium ions release of residues from different intracanal dressing, in different periods, were similar and lower than the positive control group, that the medication was maintained inside the root canal, encouraging the replacement of intracanal dressing when the restoration fail.

Aim. The study aimed at evaluating the incidence of dentinal defects after root canal retreatment with ProTaper Universal retreatment (PTUR) and XP-endo Shaper and Finisher R (XP). Materials and Methods. Sixty extracted single-rooted human premolars were selected and divided into 4 groups of 15 teeth each. In the negative control group, the teeth were left unprepared. In the positive control group, the teeth were prepared with ProTaper Next and obturated with no further retreatment. In the PTUR and XP groups, the teeth were prepared and obturated followed by removal of the filling material at body temperature using PTUR and XP instruments, respectively. The roots were then sectioned at 3, 6, and 9 mm from the apex and observed under a digital microscope to detect defects. Results. PTUR group showed significantly higher ( p value <0.05) incidence of defects than the other groups. Comparison of no defects versus defects between groups in different areas of root canals demonstrated significant difference among the groups in the apical and cervical regions. Conclusion. Within the limitations of the present study, PTUR files created significantly more dentinal defects than XP files, with most of those defects at the cervical and apical areas of the root canals.

This study examined and compared wound healing between Thai propolis product and calcium hydroxide paste as pulp-capping agents after partial pulpotomy in New Zealand white rabbits. Forty incisor teeth from 10 rabbits were treated. Thirty-six teeth received class V cavity preparations with partial pulpotomy and application of either propolis or calcium hydroxide paste. Similar cavity preparations were performed in 2 teeth without any capping material as a positive control, whereas 2 teeth without the cavity preparation served as a negative control. Histological evaluation showed that both groups had dentin bridge formation. Dentinal tubules in the dentin bridge were more orderly arranged in the Thai propolis group than in the calcium hydroxide group. Wound healing and the median number of hyperemic blood vessels were not statistically significant different between the 2 groups. Thai propolis product may be used as a pulp-capping agent.

SUMMARY The aim of this study was to evaluate overall color change in bovine tooth fragments submitted to dental bleaching treatment performed simultaneously with the ingestion of beverages containing dyes. For this purpose, tooth fragments assembled into intraoral devices were submitted to at-home dental bleaching using 10% carbamide peroxide (CP) for 14 days and to immersion in staining beverages for 10 minutes daily. The specimens were divided into the following study groups according to bleaching treatment and staining substance (n=12): G I (negative control): no bleaching + distilled water; G II (positive control): bleaching + distilled water; G III: bleaching + coffee; and G IV: bleaching + grape juice. Twelve volunteers used the device continually, except during meals, oral hygiene, dental bleaching, and pigment challenge. Color readings were performed using a spectrophotometer both before the bleaching treatment and after each treatment week. The results were submitted to the normality test. The data obtained were submitted to analysis of variance and the Tukey or Kruskal-Wallis and Dunn tests (α=0.05). All bleached groups showed similar ΔE results at the end of treatment. Staining beverages generated negative ΔL mean values, and the lowest result was obtained in the treatment with coffee after 14 days. The Δa values in the groups that received treatment with staining beverages were higher when compared to the control groups. Dental bleaching associated with the consumption of staining substances may not affect overall tooth color change by the end of the treatment, although the consumption of staining substances did influence the different color dimensions.

This study aimed to evaluate the efficacy of different concentrations of essential oils combined with calcium hydroxide against Enterococcus faecalis. Thirteen experimental groups were formed: NC (negative control); PC (positive control); GC (growth control); SC (sterilization control); RO (Rosmarinus officinalis); ROH (calcium hydroxide + RO); ZO (Zingiber officinale); ZOH (calcium hydroxide + ZO); AB (Citrus aurantium bergamia); ABH (calcium hydroxide + AB); CO (Copaifera officinalis); COH (calcium hydroxide + CO); DWH (calcium hydroxide and distilled water). After reconstitution of the E. faecalis strain, microdilution testing was performed to define the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC). The data were tabulated in an Excel spreadsheet, and the MIC and MBC were calculated in accordance with the Bacteriological Analytical Handbook. MICs in the range of 0 to 100 mg/ml were only observed in the ROH group. The RO, ROH, AB, ZO, and ZOH presented absolute data for MBC. Bacterial growth was detected in the DWH group at all concentrations tested. The combination of the essential oils tested here with calcium hydroxide appears promising as an intracanal medication in endodontic treatment because of its effectiveness against Enterococcus faecalis. Essential oils are important in endodontic therapy since calcium hydroxide, the gold standard intracanal medication, is not effective against E. faecalis.

There has been little information and much confusion regarding the genotoxic effects of fluoride. The purpose of this study was to examine the spermatogenic influence of sodium fluoride (NaF) on the germ cells by means of the mouse sperm morphology test. Male mice of genotype B6C3F1 were obtained at about eight weeks of age and maintained on a low-fluoride diet (<0.2 ppm F) and distilled water ad libitum throughout the experiment. At approximately 13 weeks of age, the animals were randomly assigned to eight groups. Group I was intubated with the Maximum Tolerable Dosage (MTD) of NaF (70 mg/kg). Groups II through VI received NaF by stomach intubation at doses of 35, 20, 10, 1, and 0.1 mg/kg, respectively. Group VII served as a negative control and was intubated with distilled water. The positive control, Group VIII, was exposed to a known mutagen, cyclophosphamide (20 mglkg, i.p). The animals were treated daily for five days, and killed by cervical dislocation 35 days after the first exposure to chemicals. Slides of sperm from the cauda epididymides were prepared and blindly scored for morphological abnormalities. Weight of the testes was recorded, and the femurs were saved for fluoride (F) analysis. Analysis of bone F demonstrated the effective absorption of fluoride following intubation. The counts of abnormal sperm and the weights of the testes for mice exposed to NaF doses up to the MTD were not significantly different from those of the negative control. The results of this study showed that NaF did not have adverse effects on mouse sperm morphology.
