Abstract Background Significant progress has been made in advancing and standardizing tools for human genomic and biomedical research. Yet, the field of next-generation sequencing (NGS) analysis for microorganisms (including multiple pathogens) remains fragmented, lacks accessible and reusable tools, is hindered by local computational resource limitations, and does not offer widely accepted standards. One such “problem areas” is the analysis of Transposon Insertion Sequencing (TIS) data. TIS allows probing of almost the entire genome of a microorganism by introducing random insertions of transposon-derived constructs. The impact of the insertions on the survival and growth under specific conditions provides precise information about genes affecting specific phenotypic characteristics. A wide array of tools has been developed to analyze TIS data. Among the variety of options available, it is often difficult to identify which one can provide a reliable and reproducible analysis. Results Here we sought to understand the challenges and propose reliable practices for the analysis of TIS experiments. Using data from two recent TIS studies, we have developed a series of workflows that include multiple tools for data de-multiplexing, promoter sequence identification, transposon flank alignment, and read count repartition across the genome. Particular attention was paid to quality control procedures, such as determining the optimal tool parameters for the analysis and removal of contamination. Conclusions Our work provides an assessment of the currently available tools for TIS data analysis. It offers ready to use workflows that can be invoked by anyone in the world using our public Galaxy platform (https://usegalaxy.org). To lower the entry barriers, we have also developed interactive tutorials explaining details of TIS data analysis procedures at https://bit.ly/gxy-tis.

Abstract Many nurses, patients, and families continue to believe that physical activity increases the risk of falling. The purpose of this study was to test the hypothesis that residents who are exposed to Function Focused Care for Assisted Living (FFC-AL-EIT) and engage in moderate levels of physical activity would not be more likely to fall. This was a secondary data analysis using data from the first two cohorts of the FFC-AL-EIT study. The study included 508 residents the majority of whom were female (70%), white (97%), with a mean age of 87.72 (SD=7.47). Those who engaged in more moderate intensity physical activity were 1% less likely to fall (b = -.01, Wald =6.13, p =.01). There was no association between exposure to function focused care and falling (Beta =.41, Wald =2.35, p=.13). Further, engaging in moderate level physical activity was noted to be slightly protective of falling.

Abstract Regardless of presenting symptoms, there are concerns that BPSD is more often identified in males versus females and males are more likely to be treated with pharmacologic and non-pharmacologic interventions than females. In part this is due to the behaviors in men, specifically aggression, being more distressing for staff and more difficult to manage. The purpose of this study was to test for gender differences in identification and management of BPSD. This was a secondary data analysis using data from the EIT-4-BPSD study including 357 residents, 114 males and 243 females. Men had more aggressive behavior (p=.03) and women more refusal of care (p=.05) and repetitive verbal behavior (p=.03). Men received more mood stabilizers (p=.02) than women. Ongoing research is needed to evaluate if aggression in females may not be recognized or treated as aggressive women are less distressing for staff than these same behaviors in males.

<p><em>This study aims to determine the impact of the COVID-19 pandemic on the Indonesian economy. Until this research was written, 93 countries were found infected with COVID-19. The spread of COVID-19 which had spread eventually brought very bad risks to the world economy, including Indonesia, especially in terms of tourism, trade and investment. The method used in this research is descriptive quantitative method that uses an approach called secondary data analysis, which is a research methodology that uses secondary data as the main data source. Based on the results of the study, Indonesia is currently still in a stable economic situation. Strategic steps related to fiscal and monetary are also estimated to still have room to provide economic stimulus if needed. However, as the COVID-19 pandemic case developed, the market did fluctuate more in the negative direction. Not only that, the slow pace of Indonesia's export activities to China will also have a significant impact on the economy in Indonesia.</em></p><p><em><br /></em></p><p><em>Penelitian ini bertujuan untuk mengetahui dampak pandemi COVID-19 terhadap perekonomian Indonesia. The objective of this study was to examine the effect of the Corona Virus Disease (COVID-19) on household welfare in Cameroon. This was done using data from a survey of 264 households in Cameroon in 2020. A linear regression model and Heckman sample correction bias were used and the results showed that COVID-19 reduces household welfare in Cameroon. These findings have implications of reducing the impact of COVID-19 on household welfare if households should develop new sources of income and food strategies by engaging more on agricultural activities and government can take a strategy of allocating grants either by reducing the cost of necessities (water, electricity, foodstuff, and housing). The triple helix approach where government, institutions of technology, and businesses should develop a vaccine for the pandemic to bring back economic life to normal.
