Abstract Background we investigate the levels of adiponectin, resistin, tumor necrosis factor-alpha (TNF-α) and signs of puberty progression after 12-weeks of combined exercise and 4-weeks of detraining. Methods Thirty overweight and obese girls (age7-9) with precocious puberty who injected Differlin were randomly divided into two groups (exercise and control). At the beginning, blood samples were obtained from all subjects and serum levels of adiponectin, resistin and TNF-α were measured. Exercise group performed 60minutes of combined (aerobic and resistance) exercises 3 times a week for 12-weeks.Control group did not receive any exercise. 48 hours after the last training session and after four weeks of detraining, blood samples were collected in the second and third stage, respectively. Blood samples were collected from the control group in both steps. BMI, cholesterol, triglyceride, signs of puberty progression (bone age, uterine lengths, ovarian volumes), luteinising hormone (LH) and follicle-stimulating hormone (FSH) were measured on all three occasions. Results In the exercise group, adiponectin significantly increased and resistin significantly decreased after 12-weeks. After 4-weeks of detraining, adiponectin significantly decreased and resistin significantly increased. TNF-α levels did not change significantly during the study. There was no significant difference in all of the factors in the control group. Throughout the 16-week study period, the rate of puberty and LH significantly decreased in both exercise and control groups but FSH and LH/FSH significantly decreased in exercise group, alone. Conclusion Combined exercise increased adiponectin and decreased resistin and rate of puberty. After 4-weeks of detraining, these effects diminished but did not disappear. Abstract Background The prevalence of precocious puberty is increasing. Obesity has been demonstrated to be associated with changes in the adipokine profile and incidence of early puberty in girls. This study assessed the pubertal signs, the levels of adiponectin, resistin, and tumor necrosis factor-alpha (TNF-α) after 12 weeks of combined exercise and 4 weeks of detraining in overweight and obese girls with precocious puberty. Methods Thirty overweight and obese girls (aged 7–9) with precocious puberty, who had received Triptorelin, were randomly divided into two groups (15 exercise and 15 control). Initially, serum levels of adiponectin, resistin, TNF-α, luteinising hormone (LH), and follicle-stimulating hormone (FSH) and the signs of puberty progression (bone age, uterine length, and ovarian volume) were measured. The exercise group performed 60 min of combined (aerobic and resistance) exercise three times/week for 12 weeks. The control group did not receive any exercise. 48 h after the last training session and after 4 weeks of detraining, all research variables were measured (also in the control group). The statistical method used for data analysis was repeated measures ANOVA. Results In the exercise group, adiponectin significantly increased and resistin significantly decreased after 12 weeks. After 4 weeks of detraining, adiponectin significantly decreased, but resistin significantly increased. TNF-α levels did not change significantly during the study. There was no significant difference in all of the factors in the control group. Abstract Background In this study, we investigated the importance of netrin-1 levels in ulcerative colitis (UC) in clinical activity of the disease, and its association with other proinflammatory cytokines IL-6 and TNF-α. Methods This study is a type of case–control study. Sixty-seven patients with UC (36 of them activation, 31 of remission) and 50 healthy controls were included in the study. UC patients; ‘Truelove Witts clinical activity index by remission (n = 31), mild activation (n = 21), moderate activation (n = 6) and severe activation (n = 9) were divided into groups. Netrin, IL-6 and TNF-α measurements in plasma samples were performed using enzyme-linked immunosorbent assay kit. Results Between the patient group and the control group; there was a statistically significant difference between netrin-1, IL-6, TNF-α, neutrophil, platelet (p &lt; 0.05 for all). The plasma netrin-1 mean of UC with severe activation group (139.21 ± 48.09 pg/ml) was statistically significantly higher than that of the mild activation (p = 0,037), remission group (p = 0,001) and control group(p = 0,011). The plasma netrin-1 mean of UC with moderate activation group was statistically significantly higher than that of the mild activation(p = 0,045) and remission group(p = 0,004). Conclusion Our results reveal that plasma netrin-1 levels have been shown to be associated with UC activation, similar to proinflammatory cytokines such as TNF-α and IL-6, in UC.

Abstract Background The aim of this study was to investigate the effect of 12 weeks of aerobic training, 4 weeks of detraining and use gonadotropin-releasing hormone agonist (GnRHa) on serum C-reactive protein (CRP) and cortisol levels in girls with central precocious puberty (CPP). Methods Forty-five girls (aged 6–8 years) with precocious puberty were randomly divided into three groups (medication, training and medicine + training groups). Fifteen healthy girls (without precocious puberty) were also included as the control group. Serum CRP and cortisol levels were measured at baseline by the enzyme-linked immunosorbent assay (ELISA) technique. Then, the experimental groups performed an aerobic training program for 3 days/week 20–75 min per day at 45–75% maximum heart rate for 12 weeks. The medication groups also received GnRHa during the study, once a month (1 mL every 4 weeks) by intramuscular injection. Serum CRP and cortisol levels were measured again 48 h after the last training session and also after 4 weeks of detraining. Results Analysis of variance (ANOVA) with repeated measures showed a significant decrease in CRP (p = 0.02) and cortisol levels (p = 0.01) in the training group and the medicine + training group. Detraining led to return of CRP and cortisol levels to the pre-training levels (p = 0.001). No significant difference in serum CRP (p = 0.43) and cortisol levels (p = 0.06) was observed in the medication group. Further, no significant difference was observed between groups in CRP and cortisol. Conclusions Long-term regular moderate training decreases inflammation indices, and detraining eliminates the benefits of training in girls with precocious puberty.

Abstract A recent study showed that expression of Toll-like receptor and interferon-gamma associated genes is significantly increased in patients with chronic ITP. Interferon-gamma is an important protein which takes place in immunoregulation. +874A/T polymorphism in the first introne of interferon gamma gene is found to be associated with the development and clinical phenotype of some autoimmune diseases such as diabetes mellitus, thyroiditis, multiple sclerosis, and SLE. The aim of our study was to investigate whether interferon gamma +874A/T polymorphism is a risk factor for the development of ITP and whether it affects the clinical course and response to the treatment. Thirty five children with acute ITP and 40 children with chronic ITP who were followed for at least 6 months were included. Control group consisted 90 healthy children. Two millilitres of blood sample was taken into sterile tubes containing 0.1% EDTA from each child and all blood samples were stored at −20 until analysis. DNA was isolated from blood samples and interferon gamma +874A/T polymorphism was studied with real-time PCR and LightCycler TM. Twenty one patients had AA, 35 patients had AT, and 19 patients had TT genotype. In the control group, 47 children had AA, 36 children had AT, and 7 children had TT genotype. There was a statistical difference between ITP and control group regarding the genotype (p=0.001). The frequency of A and T alleles in ITP group was 52% and 48%, respectively. The frequency of A and T alleles in control group was 72.7% and 27.8%, respectively. The frequency of allele distribution was statistically different between the ITP and control groups (p&lt;0.0001). There was a statistical significant difference between acute ITP and control group regarding the frequency of AA, AT, and TT gene polymorphisms and allele frequency (p=0.002, p=0.002). Similarly, there was a statistical significant difference between chronic ITP and control group regarding the frequency of AA, AT, and TT gene polymorphisms and allele frequency (p=0.008, p=0.002). The frequency of AA, AT, and TT gene polymorphisms and allele frequency showed no statistical difference between acute and chronic ITP groups (p=0.285, p=0.896). There was no correlation between interferon gamma +874A/T polymorphism and severity of bleeding (mild, moderate and severe) (p=0.09). There was no correlation between interferon gamma +874A/T polymorphism and response to long term treatment in patients with chronic ITP (p=0.568). In conclusion, there was a significant difference between patients with ITP and children in control group regarding interferon gamma +874A/T polymorphism and in the light of recent data involving other autoimmune disorders, we think that interferon gamma +874A/T polymorphism may be a risk factor for ITP.

Background: Regular physical activity is recognized as a non-pharmacological treatment of inflammation and glycemic profile. Objectives: This exercise aimed to determine the effect of aerobic training on serum interleukin-6 (IL-6), glucose and insulin resistance in obese females. Patients and Methods: Thirty-two middle-aged obese women (body mass index [BMI], 32 ± 3 kg/m2) aged 35 to 45 years were randomly assigned to exercise (aerobic training, n=16) and control (n=16) groups. Exercise subjects were completed a 12 weeks aerobic training program as 3 sessions per week at 60%-80% of HRmax and control subjects received no training. Pre- and post-training of fasting serum IL-6, insulin, insulin resistance, and glucose concentration were measured and compared between 2 groups. Results were compared using t test at a significance level of P<0.05. Results: Exercise group obtained significant decreases in BMI, body fat %, and abdominal obesity as well as the concentrations of serum IL-6 and fasting glucose (P<0.05). No significant difference was observed between pre- and post-training of insulin and insulin resistance in the exercise group (P >0.05). There were no alterations in these variables in the control group (P>0.05). Conclusions: These data suggest that aerobic training may improve glycemic profile in absence of change in insulin resistance of women with abdominal obesity

Objectives: The current study aims to examine the effect of combined exercises and ginkgo biloba supplementation for 8 weeks on Brain-Derived Neurotrophic Factor (BDNF) level in depressed older men. Methods & Materials: Forty depressed older men were randomly divided into four groups of exercise (n=10), supplementation (n=10), exercise + supplementation (n=10), and control (n=10). In the pre-test phase, blood samples were taken from the subjects. Then the participants in the exercise group performed combined exercises for eight weeks, 3 sessions per week. Those in the supplementation group, the subjects were given 80 mg of ginkgo biloba extract daily for eight weeks. In the exercise + supplementation group, the subjects received both interventions simultaneously. Blood samples were taken again 24 hours after the end of intervention. Collected data were analyzed using paired t-test, one-way ANOVA, and Bonferroni Post Hoc Test. Results: Eight weeks of combined exercise alone, ginkgo biloba supplementation alone, and combined exercise and ginkgo biloba supplementation simultaneously significantly increased BDNF level in depressed older men (P<0.05). The increase in the BDNF level of subjects in the exercise + supplementation group was higher (84.78) compared to the exercise (74.37) and supplementation (72.20) groups. Conclusion: Simultaneous use of combined exercises and ginkgo biloba supplementation can be used by geriatricians to improve brain function of depressed older men.

Abstract: Aerobic exercise with mild to moderate intensity will improve health and fitness. Aerobic exercise stimulates an increase in the concentration of IL-8 and will attract neutrophils out of circulation and infiltrate into the damaged tissue for physical activity. To determine the effect of mild and moderate aerobic exercise on levels of interleukin 8 and the number of neutrophils in adolescents, semi-experimental research laboratory with the entire population of male students of SMAN I Banjarbaru. Determination of the sample with inclusion and exclusion criteria obtained 31 samples (9 mild aerobic exercise, moderate aerobic exercise 12 and 10 as a control). The implementation of data collection (blood plasma) immediately after the sample is finished doing aerobic exercise. Blood plasma samples were analyzed by flowcytometri to see the number of neutrophils and elisa to see the levels of Interleukin 8 (IL-8). The results showed that mean plasma levels of IL-8 in mild aerobic group (605.69 ± 123.28) and moderate (718.75 ± 132.55) is lower than the control group (720.80 ± 213.11). Kruskal Wallis statistic test in the three groups no significant difference with p = 0.320. The average number of neutrophils mild aerobic exercise group (52.42 ± 8.29) and moderate (63.60 ± 8.73) was higher than the control group (50.11 ± 5.55), which means that there is an increase in the number of neutrophils after aerobic exercise , with one-way ANOVA statistical test showed different significant with p = 0.001. LSD test found significant differences between mild and moderate aerobic group with p = 0.003, 95% CI. There is no difference between the control group mild aerobic with p = 0.519. There are differences between the groups of moderate aerobic and control with p = 0.000. It can be concluded that mild and moderate aerobic exercise did not affect the increased plasma levels of interleukin-8 in adolescents. Mild aerobic exercise had no effect on the increase in the number of neutrophils in adolescents. Moderate aerobic exercise influence on the increase in the number of neutrophils in adolescents.Keywords :  Aerobic exercise, Interleukin 8, Neutrophyl Abstrak: Latihan aerobik dengan intensitas ringan sampai sedang akan meningkatkan kesehatan dan kebugaran tubuh. Latihan aerobik merangsang peningkatan konsentrasi IL-8 dan akan menarik netrofil dari sirkulasi dan menyusup kejaringan yang rusak karena aktivitas fisik.  Untuk mengetahui pengaruh latihan aerobik ringan dan sedang terhadap kadar interleukin 8 dan jumlah netrofil pada remaja, dilakukan penelitian semi eksperimental laboratorik dengan populasi seluruh pelajar laki-laki SMAN I Banjarbaru. Penentuan besarnya sampel dengan kriteria inklusi dan ekslusi didapatkan 31 sampel (9 latihan aerobik ringan, 12 latihan aerobik sedang, dan 10 sebagai kontrol). Pelaksanaan pengambilan data (plasma darah) segera setelah sampel selesai melakukan latihan aerobik. Plasma darah sampel dianalisa dengan flowcytometri untuk melihat jumlah netrofil dan pemeriksaan elisa untuk melihat kadar Interleukin 8 (IL-8). Hasil penelitian menunjukkan bahwa rerata kadar IL-8 plasma pada kelompok aerobik ringan (605,69 ± 123,28) dan sedang (718,75 ± 132,55) lebih rendah dari kelompok kontrol (720,80 ± 213,11). Secara uji statistik Kruskal Wallis ketiga kelompok tidak ada perbedaan bermakna dengan nilai p = 0,320. Jumlah rerata netrofil kelompok latihan aerobik ringan (52,42 ± 8,29)  dan sedang (63,60 ±  8,73) lebih tinggi daripada kelompok kontrol (50,11 ± 5,55), artinya ada peningkatan jumlah netrofil setelah latihan aerobik. Dengan uji statistik ANOVA satu arah didapatkan hasil yang bermakna berbeda dengan nilai p = 0.001. Uji LSD didapatkan  perbedaan bermakna antara kelompok aerobik ringan dan sedang dengan nilai p = 0,003, IK 95 % . Tidak terdapat perbedaan antara kelompok aerobik ringan dengan kontrol dengan nilai p = 0,519. Terdapat perbedaan antara kelompok aerobik sedang dan kontrol dengan nilai p = 0,000. Dapat disimpulkan bahwa latihan aerobik ringan dan sedang tidak berpengaruh terhadap peningkatan kadar Interleukin-8 plasma pada remaja. Latihan aerobik ringan tidak berpengaruh terhadap peningkatan jumlah netrofil pada remaja. ABSTRACT Introduction: Fine motor skills are important for children not only in the activities of daily living, but also for learning activities. In the present study, the effects of supervised physical training were investigated in normal children. Objective: To evaluate the effects of supervised training by combining full-body exercise and the eye-hand coordination activities to improve fine motor skills in a group of five-year-old normal children. Methods: Fifty-two children were selected and randomized in exercise and control groups. The exercise group participated in three 30-minute training sessions per week for 24 weeks. Results: The fine motor skills and hand grip strength of the exercise group were significantly increased, while there was no significant change in the control group during the experimental period. Conclusion: The results indicate that the current exercise training program is effective and can be applied to 5-year-old normal children to improve their fine motor skills. In addition, this program has simple physical activities that are appropriate to the physical and mental level of child development. The 30-minute training session would be easily implemented in the kindergarten program. Level of Evidence I; High quality randomized trial with statistically significant difference or no statistically significant difference but narrow confidence intervals.

Objective In recent years, aerobic exercise has been considered as a method of anti-aging. The aging mechanism in nervous system is closely related to increased activation of neuroglia cells and progressing neuroinflammation with aging.This study tried to shed some light on relationship between chronic inflammation accumulation and aerobic exercise so as to understand how exercise intervened nervous aging through inflammation. In this study, we used D- galactose aging models, applying two intervention ways: aerobic exercise during aging process, after that, to explore the changes of astrocytes, microglia and TNF-α expression. This study aims to provide certain evidence that sport plays a role in anti-aging. Methods Fifty male Sprague-Dawley rats at age of eight weeks, are randomly divided into five groups: control group(A), Aging group(S), Aging while exercising group(YS), After aging Quiet group(SA), After aging exercise group(SY). The aging groups were subcutaneously injected with D- galactose 150mg / kg / d for six weeks. Group YS do swimming exercise three times a week,once 60 minutes in the first six weeks. SY had same exercise intervention in the 7-12 week after injection. We took the hippocampus of rats in A, S, YS at the end of the sixth week, and of those in SA, SY at the end of twelfth week. tested the expression of GFAP, CD11b and TNF-α by immunohistochemical staining. Results 1.GFAP immunohistochemistry: Compared with group A,The expression of GFAP in group S, YS, SA and SY was significantly increased (P<0.01). Compared with group S, The expression of GFAP in group YS significantly decreased by 20.5% (P<0.01). There is no significant difference between group SA and SY(P > 0.05). 2.CD11b immunohistochemistry: Compared with group A, The expression of CD11b in group S, YS, SA and SY was significantly increased (P<0.01). Compared with group S, The expression of CD11b in group YS decreased by 18.4% (P < 0.05).There is no significant difference between group SA and SY(P > 0.05). 3.TNF-α Immunohistochemistry: Compared with group A,The expression of TNF-α in group S, YS, SA and SY was significantly increased (P<0.01). Compared with group S, The expression of TNF-α in group YS decreased by 30.1% (P < 0.01).There is no significant difference between group SA and SY(P > 0.05). Conclusions Exercise could effectively delay aging progression through improving neuroinflammation, but hard to reverse it, so the earlier in age to exercise, the better to delay aging.

Kunci regulator terhadap respons inflamasi diketahui melalui aktivasi interleukin-1β (IL-1β). Makrofag merupakan sel fagosit mononuklear berperan dalam sistem imun innate dan adaptif. Sitokin yang disekresikan makrofag sebagai respons terhadap patogen antara lain IL-1, IL-6, IL-12, TNF-α, dan chemokine. Minyak atsiri temu putih (Curcuma zedoaria Rosc.) diduga memiliki efek anti inflamasi. Tujuan penelitian ini adalah untuk mengetahui kadar IL-1β pada makrofag terekspose aggregat bakteri actinomycetemcomitans setelah pemberian minyak atsiri temu putih. Subjek penelitian adalah 10 ekor tikus Wistar jantan yang dibagi menjadi 2 kelompok (perlakuan dan kontrol), masing-masing kelompok terdiri atas 5 ekor. Kelompok perlakuan diberi minum minyak atsiri temu putih dosis 30,6 μl/ml dan kelompok kontrol diberi aquabides selama 14 hari. Gingiva anterior rahang bawah tikus diolesi A.actinomycetemcomitans sebanyak 100 μl dalam CMC 2% pada hari ke-7 setelah pemberian minum bahan uji dan kontrol selama 7 hari. Pada hari ke-15, tikus pada masing-masing kelompok dianestesi lalu makrofag dikoleksi dari cairan peritoneal. Kadar IL-1β makrofag diukur menggunakan ELISA kit (R&D Systems, USA), selanjutnya data dianalisis menggunakan uji-t. Hasil penelitian menunjukkan terdapat penurunan kadar IL-1β setelah perlakuan. Perbandingan kelompok perlakuan dan kontrol menunjukkan perbedaan bermakna (p < 0,05). Disimpulkan bahwa minyak atsiri temu putih kemungkinan memiliki efek anti inflamasi melalui penurunan kadar IL-1β makrofag.IL-1β level of macrophage exposed to A. actinomycetemomitans decreases after administration Curcuma Zedoaria volatile oil. Activation of interleukin-1β (IL-1β) is a key regulator of the inflammatory response. Macrophage is a phagocytic mononuclear cell that plays an important role in innate and adaptive immune response. The cytokine secreted by macrophages in response to pathogen are IL-1, IL-6, IL-12, TNF-α and chemokine. Curcuma zedoaria volatile oil may have anti inflammation effect. The aim of this study was to investigate IL-1β level of macrophage exposed to A. actinomycetemcomitans after administration Curcuma zedoaria volatile oil. The subjects were 10 male Wistar rats, which divided into two groups (treatment and control), each group 5 rats. In the treatment group, 30,6 μl/ml Curcuma zedoaria volatile oil was administered per oral for 14 days and the control group used aquabidest. In the 7th days, 100 μl A. actinomycetemcomitans in CMC 2% were applied on the anterior gingival mandible for 7 days. Rats were anesthetized in the 15th days then macrophage was collected from peritoneal. Interleukin-1β level of macrophage was measured using ELISA kit (R&D Systems, USA). Data were analyzed using t-test. The result showed IL-1β level decreased after treatment. The comparison between treatment and control was significant difference (p<0.05). It can be concluded that Curcuma zedoaria volatile oil may have anti inflammation effect through reducing the IL-1β level of macrophage.
