This work was conducted to investigate the efficacy of chlorophyll fluorescence (CF) sorting to improve seed germination, seedling emergence, and vigor of seeds produced from different maturity fruits of four different cultivars. Four harvest dates from each cultivar were evaluated by harvesting orange (immature), bright red (half-mature), dark red (mature), and dark red and soft (overmature) fruits. Seeds were either sorted or nonsorted after harvesting and standard laboratory germination, seedling emergence, and controlled deterioration tests were conducted. CF sorting significantly increased laboratory germination, seedling emergence, and seed vigor. Maximum improvements were obtained from seeds harvested from half-mature and mature stages. Mean germination improvement among cultivars between CF-sorted and nonsorted seeds were 14% in the immature seeds, 11% in half-mature seeds, 6% in mature seeds, and 9% in overmature seeds. Improvements in seedling emergence were 21%, 17%, 9%, and 10% and 4%, 11%, 10%, 14% for seed vigor (CD germination) in the all maturity stages of seed lots, respectively. CF has the potential to upgrade seed quality in pepper lots as a non-destructive sorting technology.

Abstract: The aim of this work was to evaluate the effect of different spray volumes, via industrial treatment, on the physiological quality of soybean seeds with different levels of vigor. The experimental design was completely randomized in a 2x5 factor scheme, with four replications. Factors consisted in two levels of seed vigor (high and low) and five spray volumes (0, 600, 1200, 1800 and 2400 mL. 100 kg-1). Products used to obtain the volumes were: fungicide (carbendazin + thiram); insecticide (imidacloprid + thiodicarb); nematicide (abamectin); micronutrients (cobalt, molybdenum and zinc); polymer (peridiam); biostimulants (kinetin + gibberellic acid) and inoculum (Bradyrhizobium japonicum). The used cultivars were BRS 360 RR and BRS 1010 IPRO. The physiological seed quality was determined by the following evaluations: germination, first count of the germination test, seedling emergence in sand, emergency speed index, total length of shoot and root of the seedlings. In the different spray volumes, high vigor soybean seeds have higher physiological quality than low vigor seeds. The increased spray volumes in seed treatments reduce the physiological quality of low vigor soybean seeds.

ABSTRACT The physiological performance of seeds is related to their physiological quality and seed vigor, while their health quality may interfere with germination and early seedling establishment in the field due to the interaction of microorganisms associated with seeds. The study aimed to evaluate the physiological performance and health quality of cucumber seeds and to verify the relationship between these attributes. The physiological quality of cucumber seeds was evaluated by standard germination tests, first count of germination, controlled deterioration test, electrical conductivity test, seedling emergence, emergence speed index, and traditional accelerated aging and aging modified with saline for 48, 72, and 96 h. The health quality of cucumber seed lots was evaluated by blotter test. Stratification of cucumber seed lots by seedling emergence was similar to seedling emergence by controlled deterioration test, first count of germination, and electrical conductivity results. The cucumber seed lots evaluated showed high germination rates; however, lots 1 and 3 had a better performance in vigor tests than lots 2 and 4. Fungi detected in the blotter test were Alternaria sp., Aspergillus sp., Cladosporium sp., and Penicillium sp. Seed lots 2 and 4 had low vigor evaluated by seedling emergence and controlled deterioration, and showed a higher incidence of Penicillium sp. in the evaluation of health quality of seeds. The incidence of Penicillium sp. may negatively affect the vigor of cucumber seeds evaluated by seedling emergence and by controlled deterioration test.

Germination and vigor tests are essential components of seed quality control programs adopted by seed industries. We studied different procedures to perform the controlled deterioration test to identify differences in vigor among okra seed lots. Four seed lots of Colhe Bem cultivar and five seed lots of Santa Cruz 47 cultivar were submitted to the following tests: germination, seedling emergence, controlled deterioration (seeds with moisture contents of 18, 21 and 24% at 45°C for 24 and 48 hours) and moisture content. The controlled deterioration test is efficient to evaluate the physiological potential of okra seeds, and the combination of 24% water, 45°C during 24 hours is recommended.

Fruit maturity for seed production can occur at various times because of the continual flowering of pepper plants. Accordingly, seeds with different maturity are acquired as the fruits are collected in a single harvest. Immature seeds obtained in this harvest may lead to a decrease in the quality of seed lots. Therefore, this research aimed to evaluate the influence of four different endophytic bacteria strains (Pseudomonas fluorescens strain L5b, Pseudomonas gessardii strain L13, Bacillus subtilis strain Bs1 and Bacillus mojavensis strain ApBm) on germination and seedling vigor of immature and mature bell pepper seeds. To obtain seeds with different maturity levels, fruits were collected 45–49 days after flowering for immature seeds and 65–69 days for mature seeds. The effectiveness of these bacteria strains was examined by coating seeds with four different endophytic bacteria strains separately. Additionally, to see the activity of endophytic bacteria more clearly, a mock treatment with sterile water was added to the experiment as a control (+) group. Bio-priming (especially strain Bs1 and L13) improved germination and seedling emergence characteristics of both immature and mature seed lots compared to control groups (p < 0.05). The results demonstrate that bio-priming with beneficial endophytic bacteria can be used to stimulate the quality of both immature and mature seeds from the pepper.

ABSTRACT The use of growth-promoting bacteria can increase the yield and quality of seeds of several annual crops, such as soybean. Based on that, the present study aimed to evaluate the effects of the inoculation of Bacillus subtilis strains on seed treatment, evaluating the yield and quality of seeds of two soybean cultivars. The experiment was conducted in a conventional system, in an Oxisol of clayey texture, in a crop under sprinkler irrigation. The experimental design was randomized blocks, arranged in split plots with four repetitions. Treatments were composed, in the plots, of two soybean cultivars: M7110 IPRO and Brasmax Desafio RR. Within the levels of subplots, the treatments consisted of inoculation and doses of inoculant: Bacillus subtilis Pant001 strain at doses of 0, 2, 3, 4 and 5 mL kg-1, and QST713 strain at the recommended dose of 2 mL kg-1. Inoculation with the strains Pant001 and QST713 increased the yield of both soybean cultivars tested, besides improving seed quality due to the increase in the concentration of total storage proteins, seedling emergence percentage and seed vigor by the accelerated aging test.

Maturity at harvest determines seed viability and vigor. However, separating seeds from different stages of development can be difficult using existing seed sorting technologies. New technology non-destructively sorts seeds based on their chlorophyll fluorescence (CF), so seeds with the same dry weight but with different physiological maturates can be separated. We determined whether chlorophyll content of muskmelon (Cucumis melo L. cv. Top Mark) seeds changes during development and whether those changes were related to viability and vigor. Seed viability and vigor were determined using an Association of Official Seed Analysts wet paper towel germination test. `Top Mark' seeds from nine stages of development were run through the SeedMaster Analyzer (Satake USA Inc., Houston, Texas), which calculated the chlorophyll content of each seed. The CF signal was fed into a computer to obtain a frequency histogram. Forty, 45, and 55 days after anthesis (DAA) seeds had germination percentages of 96%, 98%, and 100%, respectively, the highest in the study. Fifty-five DAA had greater seed vigor and viability and contained the lowest CF values; 207 on the 1000-value scale. The less-mature seeds contained higher chlorophyll content and had the lowest seed vigor and viability. Seed vigor and chlorophyll content were negatively correlated in this study. All seeds with high CF values had low vigor, but not all seeds with low CF values have high vigor. Seed aging during storage can reduce viability and vigor independent of chlorophyll content. Based on chlorophyll content, the SeedMaster Analyzer can non-destructively remove immature, low-vigor seeds that have the same physical characteristics and weight as more mature seeds. Chlorophyll fluorescence technology may allow the seed industry to further improve seed quality and maximize vigor.

AbstractA method is proposed for assessing the maturity and quality of seeds, based on measuring the amount of chlorophyll fluorescence (CF) signals of intact seeds. In general, the amount of chlorophyll is directly related to the degreening process and thus the maturity. Cabbage seeds (Brassica oleracea var. capitata) were separated into three subsamples based on the CF signals of the individual intact seeds. Seeds with the lowest amount of CF had the highest percentage of germination and normal seedlings. In a controlled deterioration test, the subsample with the lowest CF signal had slightly lower germination and normal seedling percentages than the non-treated seeds, whereas the seeds with the highest CF signals had much lower germination and normal seedling percentages. Advantages of the CF method for determining seed maturity and seed quality are its high sensitivity and fully non-destructive nature and the high speed at which the fluorescence is generated and measured.

Objective: To investigate the impact of different slope directions on the quantity and quality of the soil seed bank and seedling germination process of Castanopsis hystrix plantations. Method: Fixed sample plots in forest stands of Castanopsis hystrix were established on different slope directions (sunny slope, semi-sunny slope, semi-shady slope, and shady slope). The characteristics of the forest stand were investigated, and per-wood scaling was carried out. The temporal dynamics of the seed rain and seed bank were quantified using seed rain collectors and by collecting soil samples from different depths. The quantity and quality of the seeds were determined, and the vigor of mature seeds was measured throughout the study. Results: (1) The diffusion of Castanopsis hystrix seed rain started in mid-September, reached its peak from late October to early November, and ended in mid-December. (2) The dissemination process, occurrence time, and composition of the seed rain varied between the different slope directions. The seed rain intensity on the semi-sunny slope was the highest (572.75 ± 9.50 grains∙m−2), followed by the sunny slope (515.60 ± 10.28 grains∙m−2), the semi-shady slope (382.13 ± 12.11 grains∙m−2), and finally the shady slope (208.00 ± 11.35 grains∙m−2). The seed rain on the sunny slope diffused earliest and lasted the longest, while the seed rain on the shady slope diffused latest and lasted the shortest time. Seed vigor and the proportion of mature seeds within the seed rain were greatest on the semi-sunny slope, followed by the sunny slope, semi-shady slope, and the shady slope. (3) From the end of the seed rain to August of the following year, the amount of total reserves of the soil seed banks was highest on the semi-sunny slope, followed by the sunny slope then the semi-shady slope, and it was the lowest on the shady slope. The amount of mature, immature, gnawed seeds and seed vigor of the soil seed bank in various slope directions showed a decreasing trend with time. The seeds of the seed bank in all slope directions were mainly distributed in the litter layer, followed by the 0–2 cm humus layer, and only a few seeds were present in the 2–5 cm soil layer. (4) The seedling density of Castanopsis hystrix differed significantly on the different slope directions. The semi-sunny slope had the most seedlings, followed by the sunny slope, semi-shady slope, and the shady slope. Conclusions: The environmental conditions of the semi-sunny slope were found to be most suitable for the seed germination and seedling growth of Castanopsis hystrix, and more conducive to the regeneration and restoration of its population.

Abstract: Initial seed vigor and sowing density interact in establishment of plants in the field, and can thus affect expression of the plasticity of the wheat crop and the yield and physiological quality of the seeds produced. The aim of this study was to evaluate the effect of seed vigor levels in combination with sowing densities on the yield and physiological potential of wheat seeds. The cultivars BRS Gralha-Azul and BRS Sabiá were used in a randomized block experimental design in a 2 × 4 factorial arrangement, with four replications. The treatments consisted of seeds of high and low vigor and four sowing densities (150, 250, 350, and 450 seeds.m-2). The following evaluations were made: seed yield, germination, first germination count, seedling length, seedling dry matter, accelerated aging, emergence speed index, and seedling emergence in sand. An increase in sowing density favors the seed yield of both cultivars; however, it reduces the vigor of the seeds produced by the cultivar BRS Gralha-Azul, especially of the seeds produced by plants originating from high vigor seeds. For the cultivar BRS Sabiá, an increase in sowing density decreases the germination performance of seeds produced by plants originating from low vigor seeds.

Storage is an important step in the production of soybean seeds, mainly in tropical regions, where high temperatures can drastically reduce seed quality. The storage at cooler temperatures may be a feasible alternative to preserve seed quality during storage. The objective of this study was to evaluate the physiological quality of soybean seeds artificially cooled by a dynamic process and stored in an environment, with non controlled temperature and relativity humidity. Two seed lots of cultivars Monsoy 8757, TMG 115 RR and BRS Valiosa RR were used. After processing and during packaging the seeds were cooled to 18 ºC by a dynamic process. Seed samples were collected after 0, 60 and 120 days of storage, using the following parameters germination, accelerated aging, seedling emergence in sand, speed of emergence index, electrical conductivity, tetrazolium and seed health. Although soybean seeds artificially cooled by the dynamic method may have slightly superior performance in maintaining physiological quality during storage in non-refrigerated warehouse, for lots of high physiological quality, these advantages are not observed.
