Efficacy of Treatments for Delaying Senescence of Wheat Leaves: II. Senescence and Grain Yield under Field Conditions

Abstract Background The heterologous expression of isopentenyl transferase (IPT) under the transcriptional control of the senescence-associated receptor-like kinase (SARK) promoter delayed cellular senescence and, through it, increased drought tolerance in plants. To evaluate the effect of pSARK::IPT expression in bread wheat, six independent transgenic events were obtained through the biolistic method and evaluated transgene expression, phenology, grain yield and physiological biomass components in plants grown under both drought and well-irrigating conditions. Experiments were performed at different levels: (i) pots and (ii) microplots inside a biosafety greenhouse, as well as under (iii) field conditions. Results Two transgenic events, called TR1 and TR4, outperformed the wild-type control under drought conditions. Transgenic plants showed higher yield under both greenhouse and field conditions, which was positively correlated to grain number (given by more spikes and grains per spike) than wild type. Interestingly, this yield advantage of the transgenic events was observed under both drought and well-watered conditions. Conclusions The results obtained allow us to conclude that the SARK promoter-regulated expression of the IPT gene in bread wheat not only reduced the yield penalty produced by water stress but also led to improved productivity under well-watered conditions.

ALS-inhibiting herbicides usually provide adequate weed control in irrigated rice fields. After consecutive years of use, the Cyperaceae species, globe fringerush (Fimbristylis miliacea) began to show resistance to ALS (acetolactate synthase) inhibitors. Globe fringerush is one of the most problematic herbicide-resistant weeds in irrigated rice in the state of Santa Catarina in the South of Brazil. The objective of this research was to examine cross resistance of globe fringerush to ALS inhibitors, under field conditions. Two experiments were conducted in a rice field naturally infested with ALS-resistant globe fringerush in Santa Catarina, in the 2008/09 and 2009/10 cropping seasons. The experimental units were arranged in randomized complete block design, with five replicates, consisting of two factors (herbicide and dose) in a 4 x 5 factorial arrangement. ALS herbicides included bispyribac-sodium, ethoxysulfuron, pyrazosulfuron-ethyl and penoxsulam. Six-leaf globe fringerush was sprayed with herbicide doses of 0, 0.5, 1, 2 and 4X the recommended doses in a spray volume of 200 L ha-1. The number of rice culm, filled and sterile grains, plant height, dry shoot biomass and grain yield were recorded. Globe fringerush control was evaluated 28 and 70 days after herbicide application (DAA); shoots were harvested at 13 weeks after herbicide application and dry weight recorded. Competition with globe fringerush reduced the number of culm and rice grain yield. The globe fringerush biotype in this field was resistant to all ALS herbicides tested. Penoxsulam had the highest level of activity among treatments at 28 and 70 DAA, but the control level was only 50% and 42%, respectively, in the second year of assessment. This was not enough to prevent rice yield loss. Alternative herbicides and weed control strategies are necessary to avoid yield losses in rice fields infested with ALS-resistant biotypes of globe fringerush.

An experiment was conducted under laboratory and field conditions in order to evaluate the effects of different priming treatments, specifically KNO3 (1%), KCl (2%), KH2PO4 (1%), ZnSO4 (0.05%), PEG-6000 (20%), IBA (100 ppm), Mannitol (4%), GA3 (100 ppm) and distilled water, on seed germination properties and several agro-morphological plant characteristics of red lentil. Seeds not primed were used as a control. GA3 treatment increased shoot length. The control (non-primed seeds) treatment resulted in increased seedling root number and length. Distilled water, ZnSO4 and control treatments increased germination rate and percentage. In the pot experiments, GA3 treatment increased plant height and seedling emergence rate, whereas KCl treatment improved the number of nodules, as well as root and shoot dry weight when compared to the control. ZnSO4 treatment increased yield components and grain yield in field conditions. The results of this study showed that ZnSO4, GA3 and PEG-6000 seed priming treatments may be useful tools due to their positive effects on germination rate, germination percentage, yield component and grain yield of lentil.

Abstract Background Soil salinity is one of the main environmental conditions that affects rice production. Identifying the genetic loci that affect rice salt tolerance (ST)-related traits at the seedling stage, especially under saline field conditions, is crucial for ST rice breeding by pyramiding ST genes that act at different developmental stages. Results Large phenotypic variations were observed in 708 rice accessions, and yield and its related traits were considerably limited when exposed to salt stress. In a genome-wide association study (GWAS), 2255 marker-trait association signals were detected for all measured traits, and the significant SNPs were distributed in 903 genes. Of these, 43 genes processed same functional annotation, and the gene ontology terms “biological processes” and “molecular function” with the known genes responsive to salt stress in rice. Further haplotype analysis detected 15 promising candidates significantly associated with the target traits, including five known genes and 10 novel genes. We identified seven accessions carrying favorable haplotypes of four genes significantly associated with grain yield that performed well under saline stress conditions. Conclusions Using high density SNPs within genes to conduct GWAS is an effective way to identify candidate genes for salt tolerance in rice. Five known genes (OsMYB6, OsGAMYB, OsHKT1;4, OsCTR3, and OsSUT1) and two newly identified genes (LOC_Os02g49700, LOC_Os03g28300) significantly associated with grain yield and its related traits under saline stress conditions were identified. These promising candidates provide valuable resources for validating potential ST-related genes and will facilitate rice breeding for salt tolerance through marker-assisted selection.

Source limitation during grain filling is important for both management and breeding strategies of grain crops. There is little information on the sensitivity of grain weight of temperate cereals to variations in source–sink ratios, and no studies are available on the comparative behaviour of temperate cereals growing together in the same experiment. The objective of the current study was to evaluate, under field conditions, the response of grain weight to different source–sink ratios during grain filling in high-yielding cultivars of bread wheat, durum wheat, and triticale at 2 contrasting locations. Two experiments were carried out at C. Obregon and El Batan in Mexico. In each location, 6 genotypes (2 bread wheat, 2 durum wheat, 2 triticale) were evaluated. A week after anthesis, 2 source–sink (control and halved spikes) treatments were imposed. Location and genotype significantly (P < 0.01) affected grain yield and components. Significant grain weight increases (P < 0.05) were found only in 2 cases in El Batan. The highest response of 17% was found in triticale, with less than 10% in most of the other genotypes. The effect of genotype and location is discussed.
