BACKGROUND In 2020, the discoverers of CRISPR/Cas9 technology, Jennifer A. Doudna and Emmanuelle Charpentier, were awarded the Nobel Prize in Chemistry. This technology has revolutionized the field of biomedical science since its birth and contributed to the treatment of a variety of complex diseases. Scholars have carried out many studies on the development trend of CRISPR technology, bibliometrics, visualization and other aspects. However, there are few studies on the hot spot analysis and topic changes in the biomedical field, which is not conducive to scholars' thinking. OBJECTIVE This study aimed to:(1) mining and analyzing the research hotspots of CRISPR/Cas9 system in each stage; (2) sorting out and visualizing the evolution direction of the hot research topics of CRISPR/Cas9 gene editing technology in the past decade; (3) exploring the hidden research hotspots of CRISPR/Cas9 gene editing technology, so as to provide reference for further research METHODS In this study, research papers related to CRISPR/Cas9 gene editing technology in biomedical field published on the Web of Science core datasets from 2010 to 2020 were used; literature research method, analysis method and expert consultation method and other methods were comprehensively utilized; CiteSpace software and VOS viewer software were selected as visualization tools to analyze hot topics and topic evolution in research. RESULTS The generated co-occurrence map and cluster map were analyzed by keyword analysis strategy to determine the hot topics in each development stage. The topic evolution types and evolution paths are studied based on the categories and keywords of hot topics in each stage and the characteristics of their advancement along the time axis, and the topic evolution diagram and evolution path diagram were presented respectively. CONCLUSIONS Based on the above research results, firstly, in recent years, scholars have attached great importance to the research on this technology in the field of biomedical science, and the research topics have been deepened and enriched. Secondly, the development of CRISPR/Cas9 system shows different hot research directions in different periods, which is in line with the development of The Times. Finally, the theme evolution of CRISPR/Cas9 in the field of biomedicine generally shows a trend from single to multiple, and from theory to practice. Hidden research hotspots focus on the detection and treatment of infectious diseases, aging mitigation, drug resistance and other common diseases.

BACKGROUND Medical informatics has become a discipline that attracted researchers worldwide. It’s necessary to understand the development of its research hotspots and the future research trend. OBJECTIVE This research aimed to explore the evolution of research topics in medical informatics by analyzing relevant research articles published from 1964 to 2020. METHODS We collected research articles from 27 representative medical informatics journals indexed by the Web of Science Core Collection. The research topics of medical informatics were extracted based on LDA model and the topic evolution patterns were analyzed based on similarities between research topics. RESULTS A total of 56466 publications were identified. We found that medical informatics was in a period of rapid development. Health data analysis and health behavior intervention were the research hotspots all the time. While in recent years, the application of emerging computer technologies and mobile health tools attracted more research interests. CONCLUSIONS Our study provided a comprehensive understanding of the research hotspots and the evolution pattern among them in medical informatics, which was helpful for researchers to grasp research trends and design their studies.

CRISPR/Cas9 has become one of the most promising techniques for genome editing in plants and works very well in poplars with an Agrobacterium-mediated transformation system. We selected twelve genes, including SOC1, FUL, and their paralogous genes, four NFP-like genes and TOZ19 for three different research topics. The gRNAs were designed for editing, and, together with a constitutively expressed Cas9 nuclease, transferred either into the poplar hybrid Populus × canescens or into P. tremula. The regenerated lines showed different types of editing and revealed several homozygous editing events which are of special interest in perennial species because of limited back-cross ability. Through a time series, we could show that despite the constitutive expression of the Cas9 nuclease, no secondary editing of the target region occurred. Thus, constitutive Cas9 expression does not seem to pose any risk to additional editing events. Based on various criteria, we obtained evidence for a relationship between the structure of gRNA and the efficiency of gene editing. In particular, the GC content, purine residues in the gRNA end, and the free accessibility of the seed region seemed to be highly important for genome editing in poplars. Based on our findings on nine different poplar genes, efficient gRNAs can be designed for future efficient editing applications in poplars.

Tracking scientific and technological (S&T) research hotspots can help scholars to grasp the status of current research and develop regular patterns in the field over time. It contributes to the generation of new ideas and plays an important role in promoting the writing of scientific research projects and scientific papers. Patents are important S&T resources, which can reflect the development status of the field. In this paper, we use topic modeling, topic intensity, and evolutionary computing models to discover research hotspots and development trends in the field of blockchain patents. First, we propose a time-based dynamic latent Dirichlet allocation (TDLDA) modeling method based on a probabilistic graph model and knowledge representation learning for patent text mining. Second, we present a computational model, topic intensity (TI), that expresses the topic strength and evolution. Finally, the point-wise mutual information (PMI) value is used to evaluate topic quality. We obtain 20 hot topics through TDLDA experiments and rank them according to the strength calculation model. The topic evolution model is used to analyze the topic evolution trend from the perspectives of rising, falling, and stable. From the experiments we found that 8 topics showed an upward trend, 6 topics showed a downward trend, and 6 topics became stable or fluctuated. Compared with the baseline method, TDLDA can have the best effect when K is 40 or less. TDLDA is an effective topic model that can extract hot topics and evolution trends of blockchain patent texts, which helps researchers to more accurately grasp the research direction and improves the quality of project application and paper writing in the blockchain technology domain.

Because the enterprise management incentive mechanism operates in the complexity and nonlinearity of the enterprise system, its life evolution process is full of uncertainty and chaos. The theory of management entropy and management dissipative structure reveals two trends in the development of corporate organizational structure and incentive mechanism. Through the analysis of information asymmetry in corporate activities, we can know that information asymmetry absolutely exists. In order to weaken the information asymmetry, the enterprise must design an effective management system to make the company management more systematic. Based on the analysis of the basic theories of management entropy and management dissipative structure, this paper applies the two to the study of corporate incentives. Then, based on the self-organization theory of dissipative structure, we discussed the orderly evolution mechanism of the enterprise management system, and judged the development trend of corporate management performance from the perspective of system changes. The order degree calculation model can then evaluate the efficiency of corporate management and provide a new perspective for corporate management. Judging the role and development stage of management entropy and management dissipative structure in the enterprise’s incentive mechanism, so that the incentive can play an effective role, and provide theoretical support and practical methods for the enterprise to establish a good incentive mechanism.

This paper discusses a possibility to evolve the current Air Traffic Flow and Capacity Management towards a more proactive approach. This new method focuses on reducing the expected probability of air traffic control intervention based on “hot spot” identification and mitigation at strategic level by applying subliminal changes on the times of arrival at the crossing or merging points (junctions). The concept is fully aligned with the trajectory-based operation principles. The approach assumes that the changes on the times of arrival only demand small speed changes from the involved aircraft. In this study, the hot spots are defined as clusters of aircraft expected to arrive to the junctions. Two aircraft are said to be in the same cluster if their proximity and closure rate are below a given threshold. Some exercises are proposed and solved by applying this method. The obtained results show its ability to remove the potential conflicts by applying simple linear programming. This approach seeks to change the current capacity limiting factor, established by the number of aircraft occupying simultaneously each sector, to another parameter where the level of traffic complexity, flowing towards junctions, is identified and mitigated at strategic level. The speed changes, used as the control variable and computed before or during the flight, are designed to provide an adjustment on aircraft’s required time of arrival at the junctions in order to have a de-randomised and well-behaved (conflict free) traffic. This will enable improvements in airspace capacity/ safety binomial. It is recognised that this measure alone is unable to produce a conflict free airspace, and then other collaborative and coordinated actions, such as adjusting and swapping departing times at the departing airports (before the aircraft are taking off), offsetting some flights from nominal route, and allowing multi-agent separation management (while they are in flight) should be applied together with this method.

PurposeThe purpose of this paper is to make an overview of 474 publications and 512 patents of FTL from 1987 to 2020 in order to provide a conclusive and comprehensive analysis for researchers in this field, as well as a preliminary knowledge of FTL for interested researchers.Design/methodology/approachFirstly, the FTL algorithms are classified and its functions are introduced in detail. Secondly, the structures of the publications are analyzed in terms of the fundamental information and the publication of the most productive countries/regions, institutions and authors. After that, co-citation networks of institutions, authors and papers illustrated by VOS Viewer are given to show the relationship among those and the most influential of them is further analyzed. Then, the characteristics of the patent are analyzed based on the basic information and classification of the patent and the most productive inventors. In order to obtain research hotspots and trends in this field, the time-line review and citation burst detection of keywords carried out by Cite Space are made to be visual. Finally, based on the above analysis, it draws some other important conclusions and the development trend of this field.FindingsThe research on FTL algorithm is still the top priority in the future, and how to improve the performance of SSD in the era of big data is one of the research hotspots.Research limitations/implicationsThis paper makes a comprehensive analysis of FTL with the method of bibliometrics, and it is valuable for researchers can quickly grasp the hotspots in this area.Originality/valueThis article draws the structural characteristics of the publications in this field and summarizes the research hotspots and trends in this field in recent years, aiming to inspire new ideas for researchers.
