Background: Acorus calamus (sweet flag) is a well-known traditional herb used in India. There were only limited studies done on both hydrophilic and hydrophobic extracts of A. calamus and the biological activities of phenolic and flavonoid compounds as well as alpha-asarone in the sweet flag.Objective: The present study was designed to explore the antioxidant and antibacterial activities of both hydrophilic and hydrophobic of A. calamus leaf and rhizome extracts. Lastly, the correlation between the biological activities and total phenolic content (TPC), total flavonoid content (TFC) as well as total alpha-asarone content (TAC) of three different extracts (water, hexane and methanol extracts) was revealed.Method: TAC was determined through using HPLC method, while TPC and TFC were determined through spectrophotometric method. DPPH-free radical scavenging assay (DPPH-FRS) and ferric reducing power assay (FRAP) were used to analyze antioxidant activity, whereby disc diffusion method was used to analyze the antibacterial activity.Results: Methanol extracts had significantly (p<0.05) highest levels of antioxidant activities, antibacterial activity TPC (4.466%1; 3.603%2) and TFC (12.819%1; 7.411%2) content. The significant (p<0.05) highest TAC was observed in hexane extracts (0.762%1; 2.975%2). TPC (p<0.05; r=0.855) and TFC (p<0.05; r=0.855) correlated to FRAP, while DPPH-FRS solely correlated to TFC (p<0.05; r=0.873).Conclusion: Both of the hydrophilic and hydrophobic extracts of A. calamus could be potent antioxidant agents for nutraceutical application and antibacterial agent for food application. Lastly, flavonoids and phenols extracted from sweet flag could be a good potent source of antioxidants for nutraceuticals or food additives as those compounds are correlated to the antioxidant activity.1 Leaf extract; 2Rhizome extract

In vitro antibacterial activity of 21 plant methanolic extracts were investigated by disc diffusion method against gram-negative bacteria Escherichia coli. Amongst the extracts tested, the Datura stramonium L. and Ocimum basilicum L. showed significant antibacterial activity against the bacterial pathogen. D. stramonium showed the highest antibacterial activity followed by O. basilicum extract. Phytochemical analysis of extracts showed the presence of alkaloids, tannins, saponins, flavonoids, and phenols. The 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging and ABTS analysis of potent extract D. stramonium showed appreciable antioxidant activity. For DPPH and ABTS assay the highest antioxidant activity was observed at 60 mg ml-1 concentration with a percent inhibition of 71.66 and 77.19 with IC50 value 9.71 and 22.16 μg ml-1 respectively. Moreover, the radical scavenging activity of the extract was lower than that observed for the synthetic antioxidants butylated hydroxyanisole (BHA )and butylated hydroxytoluene (BHT). The total phenolic content of D. stramonium extract was found to be 0.592 mg GAE/gm which was assayed using Folin ciocalteau reagent. The results provide evidence that the extract of D. stramonium and O. basilicum can be further recommended in the treatment of the infections caused by E. coli; further D. stramonium is also a potential source of natural antioxidants.

The Panchthar, a hilly district of Eastern Nepal, has several medicinal plants which are used by indegenous people for the treatments of different diseases. In the present work, phytochemical and biological activities of hexane, ethyl acetate, acetone, and methanol extracts of Litsea monopetala leaves from the Pachthar district were analyzed. The phytochemical screening demonstrated the presence of major secondary metabolites like flavonoids, alkaloids, terpenoids, polyphenols, glycosides, and proteins. The antibacterial activity was estimated by the disc diffusion method against Bacillus substilus and Escherichia coli. The result showed that ethyl acetate extract exhibit the highest zone of inhibition (ZOI) value of 18 mm/disc. Acetone extract also showed significant antibacterial activity at the concentration of 100 mg/mL. Antioxidant activity from DPPH radical scavenging assay displayed the inhibitory concentration (IC50) values of dried methanol and acetone extracts were 30.74 μg/mL and 202.86 μg/mL, respectively. The methanol extract has the highest total phenolic content of 360.14 mg GAE/g extract and the highest total flavonoid content of 27.13 mg QE/g extract. The present study revealed that Litsea monopetala plant from the Panchthar district is a source of several bioactive substances and can be used in several drugs formulation.

Objective: This study has investigated cosmeceutical properties namely, antibacterial, sun protection factor and total phenolics contents of some selected macrofungi. The studies were conducted on five reference cultures collected from Indian type culture collection center (IMTEC, Chandigarh, India) and five isolates collected from TERI-Deakin Nano Biotechnology Centre facility at TERI Gram, Gurugram, India. Methods: The cosmeceutical properties of the crude extracts from selected macrofungi were analyzed using standard bioassay techniques. Antibacterial activity was analyzed against Staphylococcus epidermidis, Escherichia coli, Micrococcus luteus, Bacillus megaterium, Pseudomonas aeruginosa, Staphylococcus aureus, and Acinetobacter baumannii using Agar well diffusion method. The sun protection factor was estimated and calculated using the Mansur equation. Free radical scavenging activity using DPPH was performed to assess the antioxidant activity of the extracts. Results: Ethyl acetate extracts of the broth from P. florida and TERI-G1 cultures showed a broad-spectrum antibacterial activity against S. epidermidis, E. coli, M. luteus and B. megaterium. Ethyl acetate extracts of the broth from TERI-G3 showed the highest SPF activity of 34.02 at 200 µg. ml-1 concentration. Ethyl acetate extract of the broth from F. velutipes, P. florida, P. ostreatus, and TERI-G1 showed comparable antioxidant activity of 66.86%, 79.51%, 82.02%, and 69.58% respectively when compared to ascorbic acid (85.83%) and quercetin (83.09%) taken as positive control in the study and their total phenolic contents were found to be 6.93, 43.68, 20.88 and 13.77 Gallic acid equivalent (GAE) per gram. The minimal inhibitory concentration of F. velutipes, P. florida, P. ostreatus, and TERI-G1 was found to be 3552.89 µg. ml-1, 1250 µg. ml-1, 2418.9 µg. ml-1 and 3219 µg. ml-1 respectively. Conclusion: The work is in progress to identify and characterize TERI-G1and TERI G3 cultures. Further studies on the anti-inflammatory, anti-tyrosinase, elastase inhibition properties of the cultures will be assessed to identify potential cosmeceutical active ingredients with promising applications in cosmeceutical products.

Plant extracts are an important alternative to antibiotics, which are ever more restricted because of their developing microbial resistance and some adverse effects that have been observed following frequent application. The aim of the present study was to determine the antifungal and antioxidant activity of the methanolic extracts of Acorus calamus, Chlorella vulgaris, Lemna minuta and Scenedesmus dimorphus. The antifungal activity of the extracts against strains of Aspergillus flavus, Aspergillus parasiticus, Aspergillus ochraceus, Aspergillus niger, Aspergillus carbonarius, Fusarium graminearum, Fusarium oxysporum, Penicillium chrysogenum and Alternaria alternata was evaluated via the agar well diffusion method. The antioxidant activity of the extracts was measured through the determination of three parameters—total phenolic content, total flavonoid content and radical scavenging potential (determined through UV/Vis analysis). A. calamus extracts had the highest antimicrobial activity against eight fungal strains, followed by the C. vulgaris, L. minuta and S. dimorphus extracts, which were inhibitory against two to three strains. Among the extracts from the species studied, the extract from S. dimorphus showed the highest antioxidant potential, as determined via the DPPH (1,1’-diphenyl-2-picrylhydrazil-radical) method. This correlated to its high total phenolic and flavonoid content. From A. calamus and L. minuta, methanolic extracts were obtained that exhibited similar values of the aforementioned parameters, followed by C. vulgaris extracts, which showed the lowest antioxidant activity. Based on the Pearson correlation coefficients, the impacts of the total phenolic content and the total flavonoid content on radical scavenging capacity are similar, and flavonoids were a significant part of the total phenolic compounds extracted from the plant materials studied.

In this study the effect of pollen source (mesquite and catclaw) on the sensory characteristics (appearance, color, aroma, taste, consistency and visible impurities), and physicochemical properties of raw propolis, and the phenolic content and biological activities of propolis extracts (PEs) was determined. The phenolic composition of PEs was determined by the total phenolic (TPC), flavone and flavonol (FFC), and flavanone and dihydroflavonol content (FDC). The individual phenolic components were analyzed by HPLC-DAD. The antioxidant activity was determined by the ferric-reducing power (FRAP) and free-radical scavenging activity (FRS). The antibacterial activity was evaluated against Gram-positive (Staphylococcus aureus and Listeria innocua) and Gram-negative (Echerichia coli and Salmonella thyphimurium) bacteria. The results showed that sensory characteristic and physicochemical properties of mesquite and catclaw propolis complied with international quality regulations. Fifteen phenolic compounds were identified, of which pinocembrin, naringenin, galangin, chrysin and quercetin were found a high concentration (> 3 mg/g). Mesquite propolis had the highest phenolic content (TFC and FDC), as well as antioxidant activity (> 2.5 mg Fe (II) equivalent/g; > 40% of DPPH radical inhibition) and antibacterial activity against Gram-positive bacterias in the order S. aureus > L. innocua (> 50% of inhibition for both bacterias at 500 µg/mL). These results indicating that pollen source affect the sensory characteristics and physicochemical properties of propolis, as well as the biological activity of their extracts.

ABSTRACT: In the present study, antibacterial and antioxidant [1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity] activities of a weed, slender amaranth (Amaranthus viridis L.) were investigated. Extracts of different plant parts were prepared in n-hexane, chloroform and ethyl acetate. Antibacterial activity was measured by using 100 mg mL-1 concentration extracts against 4 deadly phytopathogenic bacterial species viz. Pseudomonas syringae Van Hall, Ralstonia solanacearum Smith, Erwinia carotovora (Jones), Holland and Xanthomonas axonopodis Hasse. In antioxidants assays, 10, 20 and 30 mg mL-1 extracts were used keeping DPPH as control. In these bioassays, ethyl acetate fraction of A. viridis leaf exhibited the best antibacterial and antioxidants activity. Ethyl acetate leaf fraction showed the highest inhibition zone diameter (IZD) where it caused 21 mm IZD against P. syringae and 19 mm IZD against E. carotovora. This extract also showed 22, 52 and 84% antioxidant activity at 10, 20 and 30 mg mL-1 concentrations, respectively. Previously there is no report available that describes antibacterial activity of root extract of A. viridis against P. syringae. Moreover, antioxidant activity of stem and root extracts in n-hexane, chloroform and ethyl acetate was investigated first time in the world. It was concluded that the biological activities observed during the present investigation may be due to the presence of bioactive constituents that can be harnessed as natural antibacterials and antioxidants.

The methanol and hexane extract of leaf of Annona reticulata, vernacularly known as ‘Sareefa’ were subjected to biological assay and column chromatography respectively. Column chromatography of the hexane extract of the leaf of A. reticulata resulted in the isolation of one pure compound PF1. The compound PF1 was suggested as β-sitosterol by comparing TLC, IR and melting point with authentic sample. The antibacterial activity against two bacteria, Staphylococcus aureus (gram positive) and Escherichia coli (gram negative) was done by agar well diffusion method and Zone of Inhibition (ZOI) was observed. The ZOI obtained ranged from 8 to 12 mm. The methanol extract of leaf showed moderate antibacterial activity on comparison with standard antibiotic streptomycin. The Antioxidant activity of the extract was tested using scavenging activity of DPPH (1, 1-Diphenyl-2- Picrylhydrazyl) radical method. The IC50 value of A. reticulata was found (41 µg/ml). The standard compound has shown IC50 value as 120 µg/ml. The antimicrobial and antioxidant activity of the extract was positively associated with the total phenolic and flavonoid contents of the extract. This study revealed that methanolic leaf extract of A. reticulata comprise effective potential source of natural antioxidant, which might be helpful in preventing the progress of various oxidative stresses.Journal of Institute of Science and TechnologyVolume 21, Issue 1, August 2016, page: 157-163

Ruta angustifolia was used in this study in order to evaluate the antimicrobial activity and antioxidant properties and its correlation with the polyphenolic content. Two Gram-positive bacteria (Staphylococcus aureus ATCC 25923, Bacillus cereus ATCC 11778) and two Gram–negative bacteria (Pseudomonas aeruginosa ATCC 27853, Escherichia coli ATCC 8739) were used to determine the antibacterial activity. Aqueous maceration extract was used for antioxidant activities and methanolic maceration extract was used for antibacterial activity. The antioxidant properties and activities were evaluated by using total phenolic content (TPC), total flavonoid content (TFC), DPPH free radical scavenging activity and beta-carotene bleaching method. Whereas, the antibacterial activity was examined using disc diffusion method against selected microorganism at concentration 1.0 mg/disc. The results showed the phenolic content of R. angustifolia extract was 18.89 g GAE/100 g extract while the flavonoid content was 14.170 g QE/100 g extract. R. angustifolia exhibited good radical scavenging with IC50 value of 2.04 mg/ml. The result for disc diffusion method showed no inhibition zone against all the strains of bacteria at 1.0 mg/disc concentration of the extract. Based on the results, it can be concluded that the R. angustifolia aqueous extract has the antioxidant properties and there is correlation between polyphenolic content of the extract with its antioxidant activity. However, R. angustifolia methanolic extract did not show any antibacterial activity.

Abstract Background Essential oils are chemical products produced by odoriferous glands from a variety of plants. These essential oil have many health benefits i.e. antiseptic, anti-inflammatory and antimicrobial activities. So due to these medicinal properties present study was designed to analyze essential oil of Thymus vulgaris and Mentha arvensis for their chemical composition and biological activities. Materials and methods Essential oil from these plants were extracted by hydrodistillation method, and analyzed by GC-MS. To test the microbial activity of these oil disk diffusion method and micro wells method were used. For free radical scavenging DPPH assay was used. However total phenolic content was measured by colorimetric method. Results The GC-MS analysis of T. vulgaris oil showed the presence of 47 chemical compounds among which thymol, terpinene, p-cymene and carvacrol were major. However essential oil of M. arvensis showed the presence of 28 constituents, among which the Menthone, Menthol, Isomenthone, Eucalyptol, neo-Menthol, cis-Piperitone oxide, Linalool, Thymol, Limonene, and α-Phellandrene were major. Essential oil from both these plant tested for antimicrobial activity showed that the T. vulgaris oil was effective against seven bacterial strains and the essential oil of M. arvensis was effective against six bacterial strain. The antioxidant activity of both samples by DPPH assay which showed positive result. Conclusion As both species showed the presence of active components, positive microbial activities, and antioxidant activity so, research should be carried on for further biological activities of these oil for betterment of living beings.

Objective: In the present study, the essential oil, methanol extract, and methanol fractions (n-hexane, chloroform, ethyl acetate, and n-butanol) obtained from Commelina nudiflora were investigated for the free radical scavenging effects and phytochemical analysis.Methods: The antioxidative effect of the essential oil, methanol extracts and methanol fractions were evaluated using 2, 2 diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay. Total phenolic and flavonoid contents were determined using Folin-Ciocalteau and aluminium chloride reagents respectively. The phytochemical analyses of the essential oil, methanol extracts and methanol fractions were performed by gas chromatography and mass spectrometry (GCMS). Results: The antioxidant, total phenolic and total flavonoid contents of butanol, ethyl acetate and chloroform fractions were higher followed by methanol extract, hexane fraction and essential oil. Phytochemical analysis indicated the presence of alkaloid, saponin, steroid, phytosterols, triterpenoids and tannins etc. The identified bioactive constituents of essential oil, methanol extract and methanol fractions of C. nudiflora were indole, 2-methoxy-4-vinylphenol, 2-pentadecanone, 6,10,14-trimethyl, phenol, benzyl alcohol, eugenol, phenol, 2, 4-bis (1,1-dimethylethyl), hexadecanoic acid, ethyl ester (palmitic acid ester), n-hexadecanoic acid (palmitic acid), 9, 12-octadecadienoic acid, (linoleic acid) and phytol. All identified bioactive compounds and their derivatives were generally reported with antimicrobial, antioxidant, anti-inflammatory and antitumor properties.Conclusion: The obtained data suggest that the essential oil, methanol extract and methanol fractions of C. nudiflora possess remarkable antioxidant activities and vital phytochemicals. Thus the plant can be a utilized as a potential source of nutraceutical with antioxidant activity.
