Studies have suggested that modulation of gut microbiota is a viable therapeutic possibility for diabetes. This study evaluated the ability of an edible plant, Gongronema (G.) latifolium Benth (Asclepiadaceae), to modulate the gut microbiome and reduce blood glucose of alloxan-induced diabetic rats. Thirty (30) young, male, albino rats were divided into 6 groups of 5 rats each: Group 1 comprised normal rats; Groups 2 to 4, diabetic rats treated with 200, 400 and 800 mg/Kg body weight of hydro-alcoholic leaf extract, respectively; Group 5, diabetic rats treated with 0.2 mg/Kg glibenclamide (an anti-diabetic drug); and Group 6 comprised untreated diabetic rats. Following induction of diabetes with alloxan injections, the treatments were administered twice daily on a 12-hourly basis by orogastric intubation for 21 days. Thereafter, faecal samples were collected from the animals and subjected to metagenomic analysis, to ascertain the composition and relative abundance of the gut microbiota. There were five dominant bacterial phyla in the rat gut: Firmicutes, Bacteroidetes, Actinobacteria, Spirochaetea and Proteobacteria. Induction of diabetes resulted in observable dysbiosis in the rats. However, treatment of the diabetic rats with G. latifolium extract, ameliorated the state of dysbiosis and resulted in significant increase in species like Lactobacillus (L.) johnsonii, L. reuteri and Prevotella corpri, which are associated with improved glucose metabolism. The plant extract produced the best result at the dose of 400 mg/Kg. The results from this study show that G. latifolium may be used as a therapeutic option for restoration of the microbiome in diabetic patients.

In recent years,green tea and ginger have become a subject of interest because of their beneficial effects on human health. The aim of the present study was to compare the effect of long term administration of green tea and ginger each alone to the effect of their combination on blood glucose level in streptozotocin-induced diabetic rats. Method: A group of 50 male albino rats was divided into five groups (10 rats each group). The normal control group( NC) administered tap water, other animals were injected by streptozotocin 45mg/Kg body weight intraperitonially to induce diabetes mellitus and then divided into four groups ,diabetic control (DC) without treatment, diabetic group administered green tea extract for four weeks (DGT), diabetic group administered ginger extract for four weeks(DGI), and diabetic group administered mixture of green tea and ginger for four weeks (DGG), then we compare the blood glucose level at 1st,2nd,3rd, and 4th week of experiment. Results:We observed that in groups whose water drink was substituted by green tea and ginger extract, the blood glucose level was significantly( p less than 0.05) reduced as compared to diabetic animals. Importantly, we observed that blood glucose level was near control level when green tea was administered simultaneously with ginger extract.Conclusion: Combination of green tea and ginger may be of great value as hypoglycemic agent in diabetic patients, the synergism of their effect on glucose regulation process is underlying this results.

People living with diabetes mellitus across the world are expected to rise to 592 million by 2035 and the pandemic disease is worst in the developing countries like Nigeria, where the cost of treating/managing diabetes mellitus is relatively out of the reach of many diabetic patients. The study investigated the effect of Anogeissus leiocarpus Guill and Perr ethanolic leaf extract on blood glucose and other biochemical indices in alloxan-induced diabetic adult male Wistar albino rats. Diabetes mellitus was induced in overnight-fasted rats by a single intraperitoneal injection of alloxan monohydrate (150 mg/kg body weight). Diabetic rats were then treated with the extract for 14 days. Blood glucose, lipid profile and liver glycogen levels were determined. After the treatment with the extract, the fasting blood glucose levels significantly reduced (p < 0.05) in treated diabetic rats (62.2 ± 18.4 mg/dl) compared to untreated diabetic rats (73.8± 8.2 mg/dl). There was also significant (p < 0.05) reduction in the hyperlipidemic parameters. The results revealed that A. leiocarpus ethanolic extract has antidiabetic and hypolipidemic effects in alloxan-induced diabetic rats.Dhaka Univ. J. Pharm. Sci. 17(1): 65-72, 2018 (June)

The aim of the present study was to investigate the protective effects of Sesbania grandiflora flower (SGF) extract on erythrocyte membrane in Streptozotocin (STZ)-induced diabetic rats. Adult male albino rats of Wistar strain, weighing 190–220 g, were made diabetic by an intraperitonial administration of STZ (45 mg/kg). Normal and diabetic rats were treated with SGF, and diabetic rats were also treated with glibenclamide as drug control, for 45 days. In this study plasma insulin and haemoglobin levels were decreased and blood glucose, glycosylated haemoglobin, protein oxidation, lipid peroxidation markers, and osmotic fragility levels were increased in diabetic rats. Moreover, erythrocytes antioxidant enzymes such as superoxide dismutase, catalase, glutathione peroxide, glutathione reductase, glutathione-S-transferase, and glucose-6-phosphate dehydrogenase activities and non-enzymatic antioxidants such as vitamin C, vitamin E, reduced glutathione (GSH), and oxidized glutathione (GSSG) levels were altered. Similarly, the activities of total ATPases, Na+/K+-ATPase, Ca2+-ATPase, and Mg2+-ATPase were also decreased in the erythrocytes of diabetic rats. Administration of SGF to STZ-induced diabetic rats reduced blood glucose and glycosylated haemoglobin levels with increased levels of insulin and haemoglobin. Moreover, SGF reversed the protein and lipid peroxidation markers, osmotic fragility, membrane-bound ATPases activities, and antioxidant status in STZ-induced diabetic rats. These results suggest that SGF could provide a protective effect on diabetes by decreasing oxidative stress-associated diabetic complications.

Postprandial hyperglycemia is a predisposing factor for vascular dysfunction and organ damage.α-glucosidase is a hydrolytic enzyme that increases the glucose absorption rate and subsequently elevates blood glucose levels. Garlic (Allium sativumL.) is a rich source of several phytonutrients, including thiosulfinate (THIO). The aim of this study was to evaluate the ability of THIO, a potent inhibitor of intestinalα-glucosidase, to reduce postprandial blood glucose. Male albino rats were randomly assigned to five different groups (n=10/group). Group 1 served as the control group. Groups 2–5 were injected intraperitoneally with a single dose of streptozotocin (STZ) to induce diabetes. Group 2 comprised untreated diabetic rats. Groups 3 and 4 contained diabetic rats that were given THIO orally (20 mg/kg body weight/day and 40 mg/kg body weight/day, resp.). Group 5 was the positive control having diabetic rats treated orally with acarbose (10 mg/kg body weight/day; positive control). Diabetic rats treated with THIO displayed a significant blood glucose reduction (p<0.001and < 0.01 by analysis of variance, resp.) and a significant elevation in insulin compared with that of untreated rats. THIO is an effective noncompetitive intestinalα-glucosidase inhibitor that promotes hypoglycemic action (p<0.001) in STZ-injected rats. THIO is a promising agent for the management of postprandial hyperglycemia.

Decreased cardiac function in streptozotocin-diabetic rats has been used as a model of diabetes-induced cardiomyopathy, which is a secondary complication in diabetic patients. The present study was designed to evaluate the therapeutic effect of a new organic vanadium complex, bis(maltolato)oxovanadium(IV), (BMOV), in improving heart function in streptozotocin-diabetic rats. There were four groups of male, Wistar rats: control (C), control treated (CT), diabetic (D), and diabetic treated (DT). Treatment consisted of BMOV, 0.5 mg/mL (1.8 mM) for the first 3 weeks and 0.75 mg/mL (2.4 mM) for the next 22 weeks, in the drinking water of rats allowed ad libitum access to food and water. BMOV lowered blood glucose to < 9 mM in 70% of DT animals without any increase in plasma insulin levels, and mean blood glucose and plasma lipid levels were significantly lower in DT vs. D rats. Tissue vanadium levels were measured in plasma, bone, kidney, liver, muscle, and fat of BMOV-treated rats. Plasma vanadium levels averaged 0.84 ± 0.07 μg/mL (16.8 μM) in CT rats and 0.76 ± 0.05 μg/mL (15.2 μM) in DT animals. The highest vanadium levels at termination of this chronic feeding study were in bone, 18.3 ± 3.0 μg/g (0.37 μmol/g) in CT and 26.4 ± 2.6 μg/g (0.53 μmol/g) in DT rats, with intermediate levels in kidney and liver, and low, but detectable levels in muscle and fat. There were no deaths in either the CT or DT group, and no overt signs of vanadium toxicity were present. Tissue vanadium levels were not correlated with the glucose-lowering effect. Isolated working heart parameters of left ventricular developed pressure (LVDP) and rate of pressure development (+dP/dT, and −dP/dT) indicated that BMOV treatment resulted in significant correction of the heart dysfunction associated with streptozotocin-induced diabetes in rat.Key words: bis(maltolato)oxovanadium(IV), vanadium, diabetes, streptozotocin, myocardial dysfunction.

Aims: To investigate the effect of food blends (plantain, soybean and ginger) on the blood glucose, lipid profile and haematological indices on streptozotocin induced diabetic rats. Methodology: A total of 35 rats of mean body weight 219.07 g separated into7 groups (5 per group) where induced by a single intraperitoneal (I.P) injection of streptozotocin (0.1 g dissolved in 5 ml of freshly prepared sodium citrate buffer 0.1 M, pH 4.5) at a dose of 40 mg/kg body weight after fasting for 12 hours and fed with flours/blends. The flours were produced from plant materials for different treatments/blends (blend A=100% unripe plantain, B=80% unripe plantain, 14% soybean, 6% ginger, C=70% unripe plantain, 26% soybean, 4% ginger, D= 60% unripe plantain, 38% soybean, 2% ginger, E= 50% unripe plantain, 50% soybean) and the phytochemicals and minerals content were determined. Blood glucose was determined at 5 days interval for 25 days. Diabetes was confirmed in rats with blood glucose concentrations >200 mg/dl. After 25 days rats were anaesthetized with chloroform vapour and blood samples collected by cardiac puncture for haematology and lipid profile determination. Results: The results showed that unripe plantain, soya beans and ginger in adequate proportion(C=70% unripe plantain, 26% soybean, 4% ginger or D= 60% unripe plantain, 38% soybean, 2% ginger) could help to reduce blood glucose, improve haematological parameters and lipid profile. Significant reduction was observed in the blood glucose level of rats fed blends C and D from 286 to 85 mg/dl and 307 to 90 mg/dl respectively at the end of experiment. These results also demonstrated that the inclusion of ginger at 6% causes rise in blood glucose level. Total cholesterol (TC) increased in all the blends. However, the lowest concentration of TC was observed in blends C and D. The highest packed cell volume (60%) and Haemoglobin (20 g/dl) level observed in rats fed blend C was significantly higher than the normal control fed conventional feeds. The increase in packed cell volume (PCV) (50%) and Hb (17 g/dl) in diabetic rats demonstrated that the formulated blend C was able to raise PCV and Hb above 50% and 17 g/dl (Normal control NC) respectively. Significant increase (P<0.05) in low density lipoprotein cholesterol (LDLc) was also observed in all the blends with blend C having the least (4.0 mg/dl) close to NC (2.0 mg/dl). Conclusion: From the results it is evident that blend C will manage and improve the health status of diabetic patients.

The effect of Protein isolate from fermented melon seeds (Ogiri Protei Isolates; OPI) of Cucumeropsis manii on blood glucose, lipid profile, and antioxidant enzyme activities in streptozotocin (STZ)-induced diabetic rats was investigated. Thirty Male Wistar rats were divided into five equal groups. GThe first control group with no exposure. The second group of rats with Streptozotocin-induced non-treated diabetes. The 3rd and 4th groups of rats with Streptozotocin-induced diabetes supplemented with Ogiri protein isolates (200, 600 mg/kg in diet). And the 5th group of rats with Streptozotocin-induced diabetes administered glibenclamide in a dose 500 ug/kg in diet [17]. The OPI was administered for 6 weeks. The administration of OPI reduced the blood glucose concentration of the STZ-induced diabetic rats. Sera and hepatic superoxide dismutase, activities of the STZ-induced diabetic rats were significantly (P< 0.05) increased in comparison with the diabetic control rats. Lipid peroxidation of the supplemented OPI diabetic rats was significantly (P< 0.05) decreased in comparison with the diabetic control rats as the administration of OPI to the STZ-induced diabetic rats significantly increased the enzymes’ activities. The concentration of low-density lipoproteins in the OPI supplemented rats was significantly elevated. These data demonstrate that OPI supplements might be beneficial for correcting hyperglycemia but the consumption of OPI can modulate some tissue lipids in a direction not beneficial for CVD risk in patients with diabetes.

The antihyperglycemic agent linagliptin, a dipeptidyl peptidase-4 (DPP-IV) inhibitor, has been shown to reduce inflammation and improve endothelial cell function. In this study, we hypothesized that DPP-IV inhibition with linagliptin would improve impaired cerebral perfusion in diabetic rats, as well as improve insulin-induced cerebrovascular relaxation and reverse pathological cerebrovascular remodeling. We further postulated that these changes would lead to a subsequent improvement of cognitive function. Male Type-2 diabetic and nondiabetic Goto-Kakizaki rats were treated with linagliptin for 4 wk, and blood glucose and DPP-IV plasma levels were assessed. Cerebral perfusion was assessed after treatment using laser-Doppler imaging, and dose response to insulin (10−13 M–10−6 M) in middle cerebral arteries was tested on a pressurized arteriograph. The impact of DPP-IV inhibition on diabetic cerebrovascular remodeling was assessed over a physiologically relevant pressure range, and changes in short-term hippocampus-dependent learning were observed using a novel object recognition test. Linagliptin lowered DPP-IV activity but did not change blood glucose or insulin levels in diabetes. Insulin-mediated vascular relaxation and cerebral perfusion were improved in the diabetic rats with linagliptin treatment. Indices of diabetic vascular remodeling, such as increased cross-sectional area, media thickness, and wall-to-lumen ratio, were also ameliorated; however, improvements in short-term hippocampal-dependent learning were not observed. The present study provides evidence that linagliptin treatment improves cerebrovascular dysfunction and remodeling in a Type 2 model of diabetes independent of glycemic control. This has important implications in diabetic patients who are predisposed to the development of cerebrovascular complications, such as stroke and cognitive impairment.

  Objective: This study was conducted to scientifically evaluate the antidiabetic and antioxidant effects of ethanolic leaf extract of Erythrina abbysinica (EEA).Methods: Acute and sub-chronic effects of EEA at 100, 200, and 400 mg/kg/bwt and glibenclamide (GL) at 5 mg/kg/bwt. were evaluated in both normal and streptozotocin (STZ)-induced diabetic male Wistar rats (250–300 g). The acute studies were performed using oral glucose tolerance test (OGTT). In sub-chronic studies, animals were orally administered with EEA and GL daily for 6 w. Brine shrimp assay was used to determine the toxicity of EEA. 1, 1-diphenyl-2-picrylhydrazyl, ferric reducing capacity of plasma, and thiobarbituric acid reactive substances assays were used to determine antioxidant properties of EEA.Results: Following OGTT, EEA significantly (p<0.05) and dose-dependently (100, 200, and 400 mg/kg/bwt) decreased blood glucose levels in both normal and STZ-induced diabetic rats when compared with positive and negative control counterparts at all-time points, whereas GL significantly (p<0.05) decreased blood glucose only in normal rats but not in diabetic rats. Daily, oral administration of EEA for 6 w significantly (p<0.05) and dose-dependently (100, 200, and 400 mg/kg/bwt) decreased blood glucose levels in STZ-induced diabetic rats when compared with the diabetic control group. EEA revealed weak toxicity with a lethal concentration50 value of 997 μg/ml). Furthermore, EEA showed significant free radical scavenging, total antioxidant, and anti-lipid peroxidative capacities.Conclusion: The study has shed more light on the scientific basis for the use of E. abbysinica in management of diabetes in some communities of Eastern Cape of South Africa.

Objective: The main objective of the study was to investigate the effect of ethanol seed extracts of Nelumbo nucifera Gaertn (Nymphaeaceae) as antihyperglycemic, anti-hyperlipidemic and antioxidant activity in streptozotocin-induced diabetic rats.Methods: Diabetes was induced in wistar albino rats by administration of streptozotocin (single intraperitoneal dose of 60 mg/kg B. W). The ethanol extract of N. nucifera seed at a dose of 200 and 400 mg/kg body weight was administrated at a single dose per day to STZ induced diabetic rats for a period of 42 d. The different pharmacological parameters were evaluated. The effect of ethanol seed extract of N. nucifera on insulin, blood glucose, urea, creatinine, HbA1C, serum protein, albumin, globulin, serum enzymes, serum lipid profiles, lipid peroxides (LPO) and other antioxidant enzymes like catalase (CAT), glutathione peroxidase (GPx), superoxide dismutase (SOD) and reduced glutathione (GSH) were measured in the diabetic rats.Results: In the acute toxicity study ethanol seed extract of Nelumbo nucifera were non-toxic at 2000 mg/kg in rats. The ethanol extract of Nelumbo nucifera seed showed significant reduction in blood glucose (p<0.05), serum enzymes (SGPT, SGOT, ALP) (p<0.05), lipid parameters (TC, TG, VLDL, LDL) (p<0.05) except HDL and significantly increased insulin (p<0.05), HDL (p<0.05), GPx, GSH, SOD and CAT (p<0.05) at the dose of 400 mg/kg when compared with the diabetic-induced control.Conclusion: The present study suggested that the NNSE has significant (p<0.05) antihyperglycemic, antihyperlipidemic and antioxidant activity in STZ induced diabetic rats. These results clearly indicate that Nelumbo nucifera is effective against free radical-mediated diseases, thus replacing the synthetic ones.
