Effects of the volatile phase of thyme, cinnamon and clove essential oils on Colletotrichum acutatum were investigated. Mycelial disc was placed in the center of the Petri dish (V=66 ml) containing PDA. Different volumes of either non- or ethanol-diluted essential oils were placed on the inner side of the dish cover to obtain final concentrations of 153, 107, 76, 46, 15, 14, 12, 11, 7.6, 3.82, 1.53, 0.153 and 0.0153 ?l/L of air. The dishes were sealed with Parafilm and incubated in up-side-down position. After 7 days of incubation, mycelial growth was recorded by measuring the colony diameter. If no mycelial growth was recorded, the disc was transferred to a new PDA plate in order to evaluate whether the activity was either fungistatic or fungicidal. Mean growth values were obtained and then converted to inhibition percentage of mycelial growth compared with the control treatment. All the tested essential oils inhibited mycelial growth of C. acutatum in the dose dependent manner. Mycelial growth was totally inhibited by thyme oil in the concentration of 76 ?l/L of air. The same results were obtained by cinnamon and clove oil in the concentration of 107 ?l/L of air. Thyme and cinnamon oil had fungicidal effect in concentrations of 107 and 153 ?l/L respectively. The results obtained provide evidence on the antifungal in vitro effect of the tested essential oils as potential means for the control of C. acutatum.

The objectives of this work were to assess the in vitro effect of essential oils extracted from cinnamon, citronella, lemon grass, India clove, tea tree, thyme, neem and eucalyptus on the conidia germination and on mycelial growth of Cercospora coffeicola, and their efficacy to control the brown eye spot in coffee seedlings (cultivars Catucaí 2SL, Catuaí IAC 62 and Mundo Novo 379/19) in a greenhouse, as well as their effects on the initial germination and infection events by scanning electron microscopy. All essential oils promoted the inhibition of conidia germination with increasing concentrations. India clove, cinnamon, neem, thyme and lemon grass oils inhibited the mycelial growth of C. coffeicola. The cinnamon and citronella oils were the most promising for brown eye spot control in all cultivars. In scanning electron microscopy, the cinnamon and citronella oils reduced germination and mycelial development of C. coffeicola in vivo, eight and 16 hours after inoculation, promoting, in some cases, the leakage of the cellular content. Essential oils of cinnamon and citronella reduced the incidence and severity of brown eye spot, in addition to presenting direct toxicity to the pathogen.

Black root rot, caused by Macrophomina phaseolina (Tass.) Goid., is the most common root disease in soybean fields. This study aimed to determine the in vitro mycelial sensitivity, measured by the IC50 (concentration to inhibit 50% of the fungus mycelial growth) of a M. phaseolina isolate obtained from soybean, to different fungicides (thiram, iprodione, carbendazim, pyraclostrobin, fluquinconazol, tolyfluanid, metalaxyl and penflufen + trifloxystrobin), at six concentrations (0.01 mg L-1, 0.10 mg L-1, 1.00 mg L-1, 10.00 mg L-1, 20.00 mg L-1 and 40.00 mg L-1 of the active ingredient). The 0.00 mg L-1 concentration represented the control, without fungicide addition. The mycelial growth evaluation was performed with the aid of a digital pachymeter, by measuring the colonies diameter, when the fungus growth in the control treatment reached the Petri dish edge. The experimental design was completely randomized, with four replications. Concerning the fungitoxicity of active ingredients, a variation from non-toxic to highly fungitoxic was observed to the M. phaseolina isolate, with IC50 values ranging from 0.23 mg L-1 to > 40.00 mg L-1, being carbendazim the most efficient one (IC50 = 0.23 mg L-1). The fungus showed insensitivity to the active ingredients of fluquinconazole, metalaxyl, thiram and tolyfluanid.

Aims: The aim of this study was to evaluate the in vitro effect of different phosphite formulations and concentrations on the development of Colletotrichum musae. Sample: to evaluate the inhibition of germination, mycelial growth and sporulation of Colletotrichum musae. Study Design:  Treatments were conducted in a completely randomized design, with 4 replicates, each replicate consisting of 1 Petri dish. Place and Duration of Study:  Laboratory of Post-Harvest Pathology, State University of Montes Claros, between March and October 2017. Methodology: Three different phosphite formulations were used: FCu1 (4% Cu + 20% P2O5), FCu2 (4% Cu + 22% P2O5) at concentrations of 0.5;1.0; 1.5 and 2.0 mL L-1 and FK (42% P2O5 + 27.7% K2O) at concentrations of 0.5; 1.0; 1.5 and 2.0 mg.L-1. Products were incorporated into the respective culture media. Culture medium alone and culture medium + imazalil were used as controls. Petri dishes were housed in BOD chamber at 25°C under a 12 hours photoperiod. Results: Results were submitted to analysis of variance and regression, and means were compared by the Tukey test (P <0.05). Control was compared to the other treatments by the Dunnet's test (P <0.05). Among the tested phosphite formulations, copper and potassium phosphites were found to reduce the mycelial growth of Colletotrichum musae. FCu2 presents a fungicide-like effect from the concentration of 0.5 m.L-1 in the control of conidia production. As for the FCu1, a fungicide-like effect was observed in the control of germination from the concentration of 1.5 mL.L-1. Conclusion: A significant fungistatic effect was observed between the concentrations of the products in the mycelial growth, sporulation and germination obtaining control of up to 100% of the development of C. musae. Copper phosphites were as effective as fungicide in inhibiting fungal development.

Lavender, anise, chamomile, fennel, geranium, oregano, parsley, and sage essential oils were tested for their effectiveness against mushroom pathogens: Verticillium fungicola var. fungicola, Mycogone perniciosa, and Cladobotryum sp. Isolates were exposed to the volatile phase of the oils and then ventilated in order to determine if the effect of the oil was lethal to the pathogen. Oregano and geranium oils were the most toxic, having a fungicidal effect at 0.02-0.08 ?l/ml of air, depending on the pathogen. Oregano oil was characterized by high content of carvacrol and thymol, while citranelol and geraniol were the main components of geranium oil.

Several phytopathogenic fungi greatly affect the production of important crops across the globe, as they lead to huge losses. To control diseases caused by fungal phytopathogens, a wide range of synthetic fungicides are applied in the fields. However, these agrochemicals are harmful for ecosystems (aerial, aquatic and terrestrial), non-target organisms and humans. In addition, since these antifungals have one specific cellular target, fungi can acquire resistance to them via the accumulation of mutations. Plant extracts provide natural alternatives to the use of synthetic fungicides in agriculture. Several plants are rich in secondary metabolites, including alkaloids, coumarins, flavonoids, terpenoids and saponins, which confer antifungal activity. This sustainable option is biodegradable, environmentally friendly and proves to be safer, and it is less prone to development resistance since they often have several cellular targets. This study was conducted to investigate the antifungal activity of Urtica dioica extract against Colletotrichum acutatum, Colletotrichum gloeosporioides, Colletotrichum godetiae, Colletotrichum nymphaeae, Diplodia corticola and Phytophthora cinnamomi. Urtica dioica extract was prepared with 50% (v/v) ethanol, the solvent was evaporated at a low pressure, and the residue was dissolved in water. The extract was incorporated into PDA medium at different concentrations (100, 500, 1000 and 2000 µg/mL) and mycelial discs were placed in the center of each Petri dish. Growth was measured in terms of radial mycelial growth in the third, sixth and ninth days of incubation, at 25 °C and in the dark. Urtica dioica extract was able to inhibit the growth of all strains except C. nymphaeae. Growth inhibition was around 20% at 2000 µg/mL for the remaining Colletotrichum species. An inhibition of growth was also observed with D. corticola in a concentration-dependent manner, from 100 µg/mL to 2000 µg/mL and revealed statistically significant differences (p < 0.05) between these concentrations. Regarding the growth of P. cinnamomi, significant differences were observed between the 100 µg/mL and 2000 µg/mL extract (p < 0.0001 and p < 0.05 on day 3 and 6, respectively). The most pronounced mycelial growth reduction (39.9%) was observed on day 3, an effect that is significantly different from (24.9%; p < 0.05) the result observed on the sixth day of incubation. Overall, the results of this work suggest U. dioca as a potential, ecologically sustainable alternative to conventional fungicides to protect crops from damage caused by phytopathogenic fungi.

Essential oils promote the inhibitory control of several fungi, including those within the genus Colletotrichum, the causal agent of Anthracnose, a disease which may occur at any stage of development in various crops, reducing up to 70% of crop production in some cases. Thus, the use of alternative products constitutes an important strategy for the integrated management, promoting less persistent molecules in the environment and lower toxicity rates, providing health benefits to producers and consumers of agricultural products. In this context, the present study evaluated the in vitro fungitoxic effect of essential oils from Java citronella (Cymbopogon winterianus), clove (Syzygium aromaticum), eucalyptus (Eucalyptus globulus) and rose pepper (Schinus terebinthifolius) on the mycelial growth of Colletotrichum theobromicola. The experiment was performed under completely randomized design, in a 4x5 factorial scheme (4 essential oils x 5 concentrations), with five replications, and the experimental unit consisting of a Petri dish. The treatments were generated by combining the concentrations (0, 5, 10, 15, 25 and 50 μL mL-1) of essential oils (citronella, clove, eucalyptus and rose pepper). The plates were inoculated with the pathogen C. theobromicola and incubated for seven days at 25 ± 2 °C. To verify the difference between treatments, the percentage of mycelial growth inhibition (PGI) was estimated. The mycelial growth of C. theobromicola was significantly reduced with increasing concentrations of essential oils. At a concentration of 50 μL mL-1, the essential oil from S. terebinthifolius showed the best result inhibiting 54.57% of mycelial growth, followed by the oil from S. aromaticum (49.26%), C. winterianus (23.70%) and E. globulus (17.90%). All the studied oils showed antifungal activity.

The Colletotrichum spp. is a significant strawberry pathogen causing yield losses of up to 50%. The most common method to control plant diseases is through the use of chemical fungicides. The findings of plants antimicrobial activities, low toxicity, and biodegradability of essential oils (EO), make them suitable for biological protection against fungal pathogens. The aim is to evaluate the inhibition of Colletotrichum acutatum by thyme, sage, and peppermint EO in vitro on detached strawberry leaves and determine EO chemical composition. Our results revealed that the dominant compound of thyme was thymol 41.35%, peppermint: menthone 44.56%, sage: α,β-thujone 34.45%, and camphor: 20.46%. Thyme EO inhibited C. acutatum completely above 200 μL L−1 concentration in vitro. Peppermint and sage EO reduced mycelial growth of C. acutatum. In addition, in vitro, results are promising for biological control. The detached strawberry leaves experiments showed that disease reduction 4 days after inoculation was 15.8% at 1000 μL L−1 of peppermint EO and 5.3% at 800 μL L−1 of thyme compared with control. Our findings could potentially help to manage C. acutatum; however, the detached strawberry leaves assay showed that EO efficacy was relatively low on tested concentrations and should be increased.

Background and Aim: Respiratory bacterial agents represent one of the most harmful factors that ordinarily threaten the poultry industry and usually lead to great economic losses. Meanwhile, there is a global demand to avoid the highly emerging antibiotic resistance and antibiotic residues in edible meat. Whereas, the use of alternatives became of great priority, especially for those substances extracted from natural plant origin. The study aimed to evaluate the antibacterial effect of cinnamon oil as a herbal extract on different respiratory bacterial agents. Materials and Methods: One hundred and fifty biological samples were collected through targeted surveillance for respiratory diseased poultry farms representing three governorates, from which bacterial isolation and identification, DNA sequencing of representative strains were performed. Furtherly, phenotypic and genotypic evaluation of the antibacterial effect of cinnamon oil was performed by minimum inhibitory concentration, agar disk diffusion, and virulence genes expression real-time polymerase chain reaction. Results: Cinnamon oil gave rise to acceptable degrees of virulence genes downregulation of 0.15, 0.19, 0.37, 0.41, 0.77, and 0.85 for Staphylococcus aureus sed gene, Escherichia coli stx1 gene, Avibacterium paragallinarum HPG-2 gene, Pasteurella multocida ptfA gene, Mycoplasma gallisepticum Mgc2 gene, and Ornithobacterium rhinotracheale adk gene, respectively. Phenotypically, using agar disk diffusion assay and broth microdilution susceptibility, cinnamon oil showed also tolerable results as it stopped the growth of S. aureus, E. coli, P. multocida, and A. paragallinarum with varying zones of inhibition. Conclusion: The encountered results declared the successful in vitro effect of cinnamon oil that recommends its application for living birds for future use as a safe antibacterial in the poultry industry.

Cardiovascular diseases (CVD) leading to heart failure are associated with myocardial cell loss and cardiac fibrosis. Hydroxymethylglutaryl-Coenzyme-A Reductase (HMGR) inhibitors ("statins") are widely used to limit cardiovascular events in patients with hypercholesterolemia and CVD by altering their lipid profile. HMGR inhibition reduces cholesterol precursor L-mevalonate production, whose depletion induces autophagy, apoptosis, and endoplasmic reticulum stress in various cell types. However it is unclear if this is a class effect or a phenomenon specific to various compounds. We examined the in vitro effect of HMGR inhibition on human atrial fibroblast (hATF) viability with particular reference to hydrophilic vs lipophilic compounds. Hypothesis- Lipophilic statins induce cell death in primary hATF via mevalonate depletion; whereas hydrophilic statins do not have any effect on hATF viability. IRB approval was obtained for collection of hATF from consenting patients undergoing open heart surgery. Cells were treated with atorvastatin, simvastatin or pravastatin (0.1, 1.0 or 10 λM) for 24, 48, 72 or 96 hours. Expression of proteins involved in the regulation of apoptosis and autophagy was assessed using immunoblotting. Cell viability was assessed using MTT assay. Treatment of hATF with 0.1 - 10 λM atorvastatin or simvastatin (lipophilic statins) resulted in progressively reduced cell viability in time and dose-dependent manner. Viability could be rescued by coincubation with mevalonate. Expression of key apoptotic cascade proteins -Bcl2, Bax and cleaved Caspase3 showed a clear induction of apoptosis. Also, there was an increase in Atg5-12 expression at 24h indicating induction of early autophagic response. Pravastatin (hydrophilic statin) did not affect cell viability or autophagy and apoptosis. We conclude that statin-induced cell death is mediated by mevalonate depletion, which activates intrinsic apoptotic pathways in hATF. Lipophilic statins impair the viability of hATFs in vitro, whereas hydrophilic statins have no effect on cell growth and cell viability of hATFs. This may represent an additional pleiotropic effect of statins, and may represent a novel therapeutic option for the prevention and treatment of cardiac fibrosis.
