Objective: This research aimed to evaluate the effect of fractionation of Merremia mammosa Lour. (Mm (Lour.)) extract on diabetic wound healing by observing collagen synthesis process and to search the most potent fraction.Methods: Wistar rats were divided into five groups (n=5), i.e., K-(negative control), K+ (positive control), K1 (ethyl acetate fraction), K2 (water fraction), and K3 (n-hexane fraction). The Mm (Lour.) was extracted with ethanol 70%, then fractionated by using three solvents which have different polarity. The rats were adapted in 7 days, then induced into diabetic by streptozotocin dose 40mg/kg body weight. The wound was made by Morton excision method. Treatment was given every two days and skin biopsy was done on day 11. Analysis of collagen density was done by photomicrograph of histopathology preparations in Masson’s trichome stained by using trinocular microscope with 400x magnification in 6 fields of view, then processed by imageJ software and analyzed by appropriate statistic tool.Result: The results of this research showed that fractionation of Mm (Lour.) extract significantly enhanced diabetic wound healing based on macroscopic (percentage of wound healing) and collagen density with p-value<0.05 when compared with negative control, especially the water fraction (p=0.000). The follow up post hoc analysis showed that there was no­ significant (p=0.989) or there was no meaningful difference in the group of water fraction when compared to positive control.Conclusion: Water fraction is the extract fraction of Mm (Lour.) which has the most significant influence on diabetic wound healing showed by enhancement of collagen synthesis.

Abstract In this paper, we extend the model of wound healing by Boon et al. (J Biomech 49(8):1388–1401, 2016). In addition to explaining the model explicitly regarding every component, namely cells, signalling molecules and tissue bundles, we categorized fibroblasts as regular fibroblasts and myofibroblasts. We do so since it is widely documented that myofibroblasts play a significant role during wound healing and skin contraction and that they are the main phenotype of cells that is responsible for the permanent deformations. Furthermore, we carried out some sensitivity tests of the model by modifying certain parameter values, and we observe that the model shows some consistency with several biological phenomena. Using Monte Carlo simulations, we found that there is a significant strong positive correlation between the final wound area and the minimal wound area. The high correlation between the wound area after 4 days and the final/minimal wound area makes it possible for physicians to predict the most probable time evolution of the wound of the patient. However, the collagen density ratio at the time when the wound area reaches its equilibrium and minimum, cannot indicate the degree of wound contractions, whereas at the 4th day post-wounding, when the collagen is accumulating from null, there is a strong negative correlation between the area and the collagen density ratio. Further, under the circumstances that we modelled, the probability that patients will end up with 5% contraction is about 0.627.

 AbstractClover flower honey is a herbal therapy used as an alternative wound treatment. The objective of this study was to determine the effect of Clover flower honey in accelerating incision wound healing, enhancing reepithelization degree, increasing fibroblast density, and collagen. This was a true experimental research, using 25 mice divided into 5 groups (n=5); positive control group (feracrylum 1%), negative control group (aquadest), 100% honey group, 50% honey group, and 25% honey group. Wound healing activity was measured macroscopically with calipers, dan microscopically through pathological anatomy preparations to observe reepithelization degree and collagen density. This study showed wound incision lengths were different significantly between Clover flower honey 100%, 50%, and 25% compared to aquadest (p ≤ 0.05), and feracrylum 1% (p ≤ 0.05). Reepithelization degree, fibroblast and collagen density measurements between Clover flower honey 100%, 50%, and 25% were different significantly compared with aquadest (p ≤ 0.05), and only the 100% honey group was different significantly with feracrylum 1% (p ≤ 0.05). We concluded topical Clover flower honey administration accelerated wound healing, increased reepithelization degree, fibroblast density, and collagen degree.Keywords : wound healing, reepithelization degree, fibroblast density, collagen density, Clover flower honey

ABSTRACT Remek daging (Hemigraphis colorata W.Bull) have been studied and used traditionally for wound healing. This study aimed to determine the effect of topical application of remek daging leaves ethanolic extract 70% on the burn wound. The animals used for this study were 30 rats, divided into five groups, namely 20, 10, 5%  remek daging extract ointment, negative control (vaseline flavum), and positive control (silver sulfadiazine 1%). Histology observations were held on days 3, 7, and 14 after burn wound induction. Histological observations showed an increase number of macrophages, fibroblasts, collagen density, and re-epithelialization in the extract ointment group significantly compare to the negative control (p <0.05). The application of ointment extract 20% to the rats showed comparable results to silver sulfadiazine 1% (p> 0.05). It can be concluded that remek daging ointment extract can accelerate the healing of burn wounds with the best results at a concentration of 20%. Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak etanol 70% daun remek daging secara topikal pada luka bakar tikus putih. Hewan yang digunakan adalah 30 ekor tikus yang dibagi menjadi 5 kelompok yaitu kelompok salep ekstrak daun remek daging 20, 10, 5% (%b/b), kontrol negatif (vaselin flavum) dan kontrol positif (silver sulfadiazine 1% (%b/b)). Pengamatan secara histologi dilakukan pada hari ke 3, 7 dan 14 setelah induksi luka bakar. Pengamatan histologi menunjukkan peningkatan jumlah makrofag, jumlah fibroblas, kepadatan kolagen dan ketebalan re-epitelisasi pada kelompok salep ekstrak daun remek daging secara signifikan dibandingkan kontrol negatif (p<0,05). Tikus yang diberikan perlakuan salep ekstrak 20% menunjukkan hasil sebanding dengan silver sulfadiazin 1% (p>0,05). Dapat disimpulkan bahwa salep ekstrak daun remek daging dapat mempercepat penyembuhan luka bakar dengan hasil terbaik pada konsentrasi 20%. Kata kunci: Hemigraphis colorata, luka bakar, makrofag, fibroblas, kolagen.

This study aims to determine the effect of Muntingia calabura‘s leaves extract ointment on the healing of cuts in the skin of hyperglycemic mice in terms of collagen density. This study used 24 male mice (Mus musculus) with alloxan induced hyperglycemia (150 mg/kg body weight) intraperitoneal. Skin incisions are made on the back using a scalpel with a length of ± 15 mm and a depth reaching the subcutaneous. The experimental animals were randomly divided into four treatments and each treatment consisted of six replications, namely the control was given a placebo, and the treatment of cherry leaf extract ointment with a concentration of 30%, 40%, and 50%. The process of wound healing was observed histopathological with collagen density parameters. Based on the results, there was an increase in collagen density in each treatment. The mean collagen density in treatment (T) 0% (T0) 0.60; at treatment 30% (T1) 0.73; at treatment 40% (T3) 1,46; at 50% treatment (T3) was 2.20. The results of the study through the Kruskal Wallis test stated that the cherry leaf extract ointment had a very significant effect on the healing of the incision wound of hyperglycemic mice as seen from the density of collagen. We conclude that the administration of 50% cherry leaf extract ointment is the most effective in accelerating the healing of the hyperlycemic mice incision wounds in terms of collagen density.

Untreated skin wound can inhibit the wound healing process. Chitosan and eel mucous have biodegradable, biocompatible, and antimicrobial properties. This study aimed to determined the potential of combination between chitosan membrane and eel mucous extract in skin wound re-epithelization and collagen fibers deposition. Thirty adult male mice were divided into 5 groups, 6 animals/group. Skin wo und were perform by punch biopsy in 0.5 cm diameter. Group I-IV respectively recieved: 10% povidone iodine; eel mucus extract and gauze; combination of 1 % chitosan membrane and eel mucus; 1% chitosan membrane. Group V without therapy. Therapy was given once a day for 10 days. At the 5th and 10th half of animals/group were euthanized. Wound skin area were collected for hematoxylin-eosin and Mallory-azan stainings. Epithelial thickness and collagen density were observed and analyzed statistically. The result s showed significant difference in epithelial thickness on the 5th and 10th days (P ≤ 0.05) in combination group. Percentage of collagen density at 10th day of combination group showed significantly difference (P ≤ 0.05) compared with control. The conclusion is combination of 1 % chitosan membrane and eel mucous extract has the most potential to re-epithelization and increase density of collagen fibers in skin wound healing.

Full-thickness wound are areas damage of skin associated with loss of epidermis and dermis. The wound healing mechanism consists proliferation, migration and remodeling. Hypoxic conditional medium of mesenchymal stem cells (HMSCs-CM) contains lots of soluble molecules, such as protein growth factor and cytokine anti-inflammation. The soluble molecule of HMSCs-CM plays a critical role in wound healing by upregulation of VEGF and collagen synthesis. The objective of this study was to evaluate the effect of HMSCs-CM on VEGF and collagen concentrations in rats with incised wounds. The methods of this study were an experimental animal study with post-test only control group design was performed involving 24 Wistar rats. The rats were randomized into four groups consisting of sham, control and two treatment groups (gel of HMSCs-CM at doses of 200 μL and 400 μL). The VEGF levels and collagen density were analyses using ELISA assay and Masson-trichome specific staining, respectively. One-way ANOVA and Post Hoc LSD were used to analyses the data. The results of this study showed that a VEGF levels was significant increased on day 6 with doses-dependent manner. Interestingly, the VEGF levels gradual decrease on day 9. In addition, the decreased of VEGF levels on day 9 in this study in line with our findings in which we found there was a trend in the decreased of collagen density, it indicated the completion of remodeling phase and there has been an acceleration in wound healing. This study demonstrated that HMSCs-CM were able to regulate VEGF levels and collagen synthesis in accelerate wound healing. Background: Disintegration of skin tissue can lead to disability and death. Recent studies on wound therapy applied stem cells and adipose derived stem cell conditioned medium (ADSC-CM) to improve wound healing. However, the role of ADSC-CM in wound healing mechanism in terms of angiogenesis, quantity of collagen, and epithelialization is not fully understood. Therefore, this study aimed to analyze the levels of growth factors (VEGF and EGF) in ADSC-CM and histological features of angiogenesis, epithelialization, and collagen density after skin incision in Sprague Dawley rats.Methods: Thirty rats were injured at the back (full thickness wound) and treated topically with ADSC-CM, culture medium, basal medium, and without treatment. Mice were sacrificed on days 3, 7, 14, 21 and 28. After sacrificed, tissue samples were examined microscopically to assess angiogenesis, epithelialization, and collagen density. Concentrations of VEGF and EGF in ADSC-CM were measured by ELISA.Results: Clinically, wound that was treated with ADSC-CM showed improvement in wound healing process. ADSC-CM treated wound showed the highest epithelialization ratio and the fastest wound closure.Conclusion: There were no statistical significant differences between groups that were treated with ADSC-CM and not. However, topical ADSC-CM treated wound revealed a better clinical improvement in epithelialization.

INTRODUCTION: Treatment for obesity essentially has to do with weight loss, which can be achieved through surgical procedures. Despite the considerable rise in the number of such procedures, the relationship between obesity and the healing process has not been totally clarified. PURPOSE: To investigate abdominal wound healing in obese Wistar rats on the seventh and fourteenth days following a laparotomy. METHODS: Thirty-six Wistar rats were randomly distributed into two groups, the control and experiment group. The control group were fed on either a standard diet for the species and the experiment group were put on a high calorie diet. After 116 days, all the animals were submitted to a laparotomy followed by laparorrhaphy. After euthanasia on the seventh or fourteenth day, fragments of the abdominal wound containing the scar were submitted to histopathological and tensiometric analysis. RESULTS: The average weight of the animals from the experiment group was higher than that of the control group (p<0.001). The difference in the resistance of the cutaneous scars was not significant. The aponeurotic scars were more resistant in the control group after seven days (p=0.011) and fourteen days (p=0.040). There was no difference in terms of intensity of the inflammatory reaction and the collagen density was similar in both groups. CONCLUSION: In rats, obesity lowered the resistance of the aponeurotic scars but not the skin scars. It did not interfere with the delayed inflammatory response and the collagen density.
