Effect of thermal treatment on antioxidant activity and colour of carrot purées

AbstractTotal antioxidant activity, levels of bio-active compound groups and instrumental colour of carrot purée subjected to thermal treatment (70°C/2 min) were measured. The method applied to the dosage of ascorbic acid was with 2,6-diclorophenolindophenol. Total phenols (TP) in purée were determined using the Folin- Ciocalteau method and antioxidant activity by the use of DPPH free radical method. The colour of the samples was measured using a Hunter-Lab colour meter. Heat treatment caused a rapid decrease in ascorbic acid. Phenolic contents were in general unaffected by thermal treatment. Colour parameters were significantly affected by thermal treatment. This provides a helpful tool for understanding the effect of processing on colour variation of carrot purée in a broader spectrum. Industrial relevance: This research paper provides scientific evidence of the influence of thermal treatments in retaining important bioactive compounds.

AbstractThe aim of the study was to assess the effect of three weeks of storage on the chemical and rheological properties of apple juices obtained from Idared and Jonatan apples variety. Total antioxidant activity, levels of bio-active compound groups and the viscosity were measured to characterize the investigated juices. The method applied for the determination of ascorbic acid concentration was with 2, 6-diclorophenolindophenol. Total phenols (TP) in apple juices were determined using the Folin-Ciocalteau method and antioxidant activity by the use of DPPH free radical method. The viscosity of apple juices was investigated by a rotational viscometer, Brookfield viscometer (Brookfield Engineering Inc., Model RV-DV I Prime) with RV spindles. During three weeks of storage, different rates of all measured properties have been observed decreasing for both studied apples varieties juices. The juices from Jonatan apples have higher antioxidant activities that are correlated with the higher content in polyphenols and lower values of viscosity.

This study was designed to evaluate the antioxidant potential ofLoranthus pulverulentus. Stem bark, leaves, and seeds ofLoranthus pulverulentuswere extracted in methanol:water (90 : 10) and partitioned with n-hexane, chloroform, ethyl acetate, and n-butanol successively using partition chromatography. Total phenolic contents and antioxidant potential were checked using standard protocols. Total phenolic contents of all extracts were determined, using Folin–Ciocalteu reagent, and ranged between 151 ± 2.1 and 396 ± 1.6 for stem bark, 137 ± 0.9 and 430 ± 2.2 for, and 39 ± 0.6 and 231 ± 1.7 for seeds. The antioxidant potential of extracts was evaluated; namely, DPPH, FRAP, and total antioxidant models. The ethyl acetate extract of stem-bark, leaves, and seeds showed the highest activity in DPPH (94.5 ± 2.1%, 96.30 ± 0.9%, and 92.30 ± 1.1%, IC5015.9 ± 0.5 μg, 14.5 ± 0.8, and 102.7 ± 1.3, resp.), FRAP (7.7 ± 0.6, 7.5 ± 0.7 and 6.6 ± 0.7, resp.), and total antioxidant (0.95 ± 0.09, 1.19 ± 0.09, and 0.686 ± 0.08, resp.). Strong correlations were observed between total phenols versus total antioxidant activity, DPPH, and FRAP withR2values ranging from 0.8185 to 0.9951 (stem-bark), 0.6728 to 0.8648 (leaves), and 0.8658 to 0.9910 (seed) which indicated that phenolic contents are the major constituents responsible for antioxidant activity.

Introduction: Cyclea peltata (Lam.) Hook.f. & Thomson is an important plant listed in Ayurveda. The plants are used in digestive, diuretic and inflammatory disorders. Aim: The present study was to study the immunomodulatory, anticancerous and antioxidative properties of the fraction isolated from Cyclea peltata. Methods: The immunomodulatory activity was evaluated in lymphocytes and THP-1 macrophage cell lines. The anticancer activity of the fraction was determined by MTT assay. The antioxidant activity was evaluated by DPPH and total antioxidant assays. Results: The fraction showed the presence of flavanoids which induced lymphocyte proliferation and also could induce nitrite production in THP-1 cell lines at 1mg/ml. It also exhibited good anticancerous activity at 100 μg/ml after 48h of incubation.  The antioxidant activity was less at this concentration and was found to be equivalent to 79± 0.03 μg/ml of ascorbic acid. Conclusion: The fraction containing flavonoid, isolated from Cyclea peltata have shown to have good immunomodulatory, antioxidant and anticancerous property.

The increased consumption of acerola in Brazil was triggered because it is considered as a functional food mainly for its high ascorbic acid content, but the fruit also has high nutritional value, high levels of phenolic compounds, total antioxidant activity, anthocyanins and carotenoids in its composition. The objective was to evaluate the chemical, physical-chemical and antioxidant activity of eight varieties of acerola tree. The acerolas used in the research were the harvest 2015, 8 varieties: BRS 235 - Apodi, Mirandópolis, Waldy - CATI 30, BRS 238 - Frutacor, Okinawa, BRS 236 - Cereja, Olivier and BRS 237 - Roxinha, from the Active Bank Germplasm APTA Regional Alta Paulista in Adamantina - SP. Avaluated the following attributes: pH, titratable acidity, soluble solids, reducing sugar, instrumental color, ascorbic acid, total phenolics, flavonoids and antioxidant activity. The design was completely randomized, 8 varieties and 3 replications of 20 fruits each. Acerola fruit of the analyzed varieties prove to be good sources of phenolic compounds and antioxidant activity, ensuring its excellent nutritional quality relative to combat free radicals. The variety BRS 236 - Cereja presents higher ascorbic acid content, antioxidant activity and phenolic compounds, and the lowest value for flavonoid, which was higher than the other cultivars, especially Olivier and Waldy CATI-30.

Cassia absus (Linn)Cassia absus(Linn) (family Fabaceae ) is generally known as “chaksu ” inan ayurvedic traditional system.The current study,aerial parts of different concentrates(Pet.ether, ethyl acetate and methanol) of Cassia absus, was evaluated for its in-vitro antioxidant potential byDiphenylpicrylhydrazyl radical,nitric oxide activity andtotal antioxidant activitytaking ascorbate as the standardfor all the three methods. The IC50 value was originated that methanolic concentrates of Cassia absusmore efficient inDiphenylpicrylhydrazyl radical,nitric oxide activity, total antioxidant activitycompared EA&PEconcentrates.The methanolic concentrates of Cassia absus& ascorbic acid exhibited antioxidant potential possessing IC50230µg/ml &130µg/ml (Nitric oxide). 205µg/ml &57µg/ml (total antioxidant),195µg/ml & 66µg/ml (Diphenylpicrylhydrazyl radical)respectively. The difference in the scavenging potential of the extracts can be due to variation in the percentage of bioactive compounds present in different solvents. Invitroantioxidant studiesobviouslyshow the methanolic concentrates of Cassia absushave better antioxidant activity. This result indicates that aerial parts of methanolic concentratesCassia absuscould serve as a natural antioxidant, which may be useful in preventfree radical-induced diseases.

ABSTRACT The family farming from the Borborema Territory-PB, Brazil, produces sweet oranges that supply the regional market. In this context, it is necessary to define standards of identity and quality, as well as to quantify the bioactive compounds in the fruit, in view of adding value and creating more competitive markets. This work sought to evaluate the quality, bioactive compounds and total antioxidant activity (TAA) of oranges from family farming. A 3x3 factorial completely randomized design was used, with three cultivars (Baía, Comum, Mimo-do-Céu) and three maturity stages (predominantly green, green/yellow, yellow), with 60 replications of 1 fruit for the physical evaluations, and 4 of 15 fruit for the others. The whole fruits were evaluated by color index, length, diameter, fresh weight and firmness. The juice was assessed for yield, pH, soluble solids (SS), titratable acidity (TA), SS/AT ratio, and ascorbic acid. Total extractable polyphenols (TEP) and ABTS•+ and DPPH• total antioxidant activity (TAA) were measured in the juice and albedo. The 'Baía' and 'Mimo-do-Céu' oranges presented quality parameters aligned with the CEAGESP standards. On average, the ascorbic acid content was higher than 45 mg 100 g-1, with ‘Mimo-do-Céu’ presenting the highest content (50.26 mg 100 g-1). During maturation, the firmness decreased, and the SS, TEP and TAA of the juice and the albedo increased. In general, the TEP content was about eight-fold higher in the albedo than juice, corresponding to the much higher TAA in this portion, thereby highlighting its higher functional potential, especially for ‘Baía’ orange.

Flemingia macrophylla locally known as ‘Charchara’ in Bangladesh is a medicinal herb. Being a member of the Fabaceae family it claims a number of medicinal uses including hypoglycemic activity, neuroprotective effect and gynecological remedies. Methanolic extract of F. macrophylla was investigated to evaluate its antioxidant and general toxic properties in this study. Antioxidant potential was evaluated using total antioxidant capacity, total phenol contents, total flavonoid contents and DPPH (1,1-diphenyl-2-picrylhydrazyl) assays. Both leaf and stem extracts of F. macrophylla were found to possess significant amount of phenolics and flavonoids, expressed as gallic acid equivalents (GAE) and quercetin equivalent (QE), respectively. Interestingly total phenol content was equal for both leaf and stem extract and it was 43.8 mg GAE/g. Total flavonoid content was found to be 64.4 and 39.1 mg QE/g for leaf and stem extract, respectively. The total antioxidant capacity was expressed as ascorbic acid equivalents (AAE) and for leaf it was 5.067 mg/gm AAE and for stem 0.8167 mg/gm AAE. DPPH scavenging activity was determined by comparing with ascorbic acid. The IC50 values were 19.95, 125.89 and 707.94 ?g/ml for ascorbic acid, leaf and stem extract, respectively. The results of the present study on methanolic extracts of F. macrophylla revealed the presence of moderate antioxidant activity and extract of leaves produced better results than stem of the plant. In brine shrimp lethality bioassay, methanolic extract of F. macrophylla exhibited insignificant cytotoxicity. Cashew nuts have high nutritional value. This nut providesmacronutrients, micronutrients and a large variety of antioxidantssuch as phenolic compounds. There are losses in some compoundsduring the processing of fruits and there are very few studies on thisaspect. Given the above, this research aimed to determine the eff ectof the processing steps on cashew nut characteristics with emphasison total antioxidant activity. Signifi cant diff erences were observedbetween the processing steps for all the chemical and physicalchemicalcharacteristics studied. Cashew nuts contained high levelsof lipids and energy, low content of moisture and water activity andmedium content of phenolic compounds (51.33 GAE/100 g) andantioxidant activity by the ABTS•+ radical (6.49 μM Trolox/g) at theend of the processing. With regard to the total antioxidant activity,it was observed that the ABTS•+ radical method can be consideredappropriate to measure the antioxidant activity of cashew nuts, sincethe results presented lower coeffi cients of variation and expressproportionate results to other methods.
