Abstract Diabetes is a chronic disease characterized by a disorder in the metabolism of proteins, fats, and carbohydrates. The liver as a non-insulin dependent organ plays an important role in the regulation of blood fat and glucose. Most blood glucose lowering drugs that are introduced for treatment have side effects in long-term consumption. Therefore, to control diabetes and its complications, the use of herbal drugs is widely considered nowadays. The present study investigates the biochemical effects of oleuropein in diabetic male rats. In this study, 30 adult male Wistar rats with a weight range of 190±30 gr were equally divided into 3 groups randomly: 1) control group or intact animals, 2) diabetic animals, and 3) treatment group, which received 60 mg/kg oleuropein for 30 days by gastric gavage. Diabetes was induced in diabetic and treatment groups by injection of streptozotocin (60 mg/kg) intraperitoneally. At the end of the treatment, the levels of triglyceride, cholesterol, LDL, HDL, VLDL, blood glucose, HbA1C, and activity of AST and ALT were determined. The results showed that the serum lipid profile and blood glucose increased significantly in the diabetic group compared with the control group (p<0.05). Also, HbA1C and atherogenic index decreased significantly in the treatment group compared with the diabetic group (p<0.05). This study showed that oral administration of oleuropein has hypoglycemic effects, which can reduce the serum levels of the lipids profile and the atherogenic index in streptozotocin-induced diabetic male rats.

Introdution: Diabetes is the most common endocrine disorder characterized by hyperglycemia. Increasing the oxidative stress and changing the amount of antioxidants play important roles in pathogenesis of diabetes. Nowadays to control diabetes and its complications, the use of herbal drugs is considered widely. In this study, we investigated the effect of oleuropein on antioxidant enzymes activity of heart tissue in Streptozotocin induced diabetic male rats. Methods: In this study, 30 adult male Wistar rats with a weight range of 190±30 gr were randomly divided into 3 groups(n=10 in each group): 1) control group or intact rats, 2) diabetic rats, and 3) treatment group, which received 60 mg/kg oleuropein for 30 days by gastric gavage. Diabetes was induced by injection of streptozotocin (60 mg/kg) intraperitoneally. At the end of the treatment, serum concentrations of blood glucose and heart tissue antioxidant enzymes activity were determined. The obtained data were analyzed using  SPSS Inc., Chicago, IL; Version 18, statistical method of one way variance analysis and post hoc-Duncan test . Results: The results showed that serum concentration of glucose decrease significantly in treatment group compared with the diabetic group (p=0.000). Also, TAC, SOD and GPX activity increased significantly in the treatment group compared with the diabetic group (p=0.000). Conclusion: This study showed that oleuropein can prevent blood glucose increasing and reinforce antioxidant system of cardiac tissue in diabetic rats.

To evaluate the antifertility effect of crude mixture of A. precatorius seeds at the dose level of 50 mg/kg body weight in adult male rats, after oral administration to male rats for 40 days, the rats were sacrificed and hormonal profiles, serum biochemistry, sperm count and histological changes were recorded. A sharp decrease in the serum levels of testosterone (0.70 ± 0.17 ng/ml), FSH (0.70 ± 0.22 lU/L), and LH (0.87 ± 0.35 IU/L) was detected compared to control (FSH, LH and testosterone levels 0.93 ± 0.15 ng/ml, 0.76 ± 0.28 IU/L, 1.44 ± .011 IU/L, respectively). A significant reduction of epididymal sperm count (2.34 million/mL) was noted in treated rats as compared to control group (7.87 million/mL). Histology of testes showed marked atrophy of the testes, which was characterized by disruption of the seminiferous epithelium and atrophy of the Leydig cells. Crude mixture of A. precatorius seed has a negative impact on male reproductive functions. It might be suggested that crude mixture of A. precatorius seeds might have antifertility property for male rats.   Keywords: Abrus precatorius; antifertility; male rat; testosterone. AIM: This research aims to examine the effects of soy milk on mounting latency (ML), mounting frequency (MF), estrogen levels, androgen-binding protein (ABP) expression, and spermatogenesis in male rats (Rattus norvegicus). METHODS: Twenty-four male wistar rats (Rattus norvegicus) aged 4 weeks were divided into four groups. Control group (given a normal diet), P1; P2; P3 (given the normal diet and soy milk powder at doses of 7.1; 14.2; 21.3 g/KgBW/day, respectively) for 6 weeks. Observation of ML and MF were performed at 9 weeks 5 days of age, and rat surgery was performed at 10 weeks of age. Analysis of estrogen hormone levels was conducted by enzyme-linked immunosorbent assay (ELISA), ABP staining was using immunohistochemistry method, testicular spermatogenesis was observed using histopathological methods, and observation of spermatozoa was performed under the microscope.  RESULTS: The results showed no significant reduction of ML and MF, estrogen levels, and ABP expression (p ≤ 0.256; 0.865; 0.959, respectively) in male rat, but there was a significant decrease in the number, morphology, motility of spermatozoa, and testicular histophatology, (p ≤ 0.000, 0.003, 0.008, 0.000, respectively). CONCLUSION: The administrassion of soy milk in various doses (7.1;14.2;21.3 g/KgBW/day) in male Wistar rats (Rattus norvegicus) had showed significantly difference on histopathological evaluation using Johnson’s scoring system, sperm quantity and quality, while on mounting latency and frequency, estrogen levels, and ABP expressions did not show significantly difference between groups. That describe of isoflavone in soy milk can affect several aspects related to male endocrine and reproductive development.

The purpose of this study was to know the effect of Dayak onion tuber extract (Eleutherine palmifolia) given by per oral in lowering levels of histophatology damage kidney of albino male rat (Rattus norvegicus) strain Wistar which was induced by alloxan. Animals which were used in this research were 24 white male rats (Rattus norvegicus) strain Wistar, and then divided into 6 groups. The negative control group K (-) were given with aquabidest and CMC-Na 1% during the therapy period, the positive control group K (+) were given with alloxan 110 mg / kgbw, the group of drug control K (O) were given with alloxan and oral therapy  with metformin 9 mg / 200g bw / day, the treatment group 1 (P1) were given with alloxan and per oral therapy with extract of  Dayak onion tuber 100 mg / kgbw, the treatment group 2 (P2) were given with alloxan and per oral therapy with extract of Dayak onion tuber 200 mg / kgbw and the treatment group 3 (P3) were given with alloxan and per oral therapy with extract of Dayak onion tuber 400 mg / kgbw. The therapy was given for 14 days, then the animals were sacrificed with ketamine and then its kidney was taken for examination of hisphatology in kidney. Observations based on their depiction of renal histopathology tubular degeneration and necrosis, glomerular necrosis, intestitial infiltration and glomerular sclerosis. Data obtained from the scoring of histopathological appearence albino rat kidneys were analyzed by test Kruskal-Wallis and if there is a real difference followed by Mann-Whitney test using SPSS 20.0 for windows. The results showed that the extract of Dayak onion tuber 400 mg/ kgbw  can reduce the degree of kidney damage in albino male rat exposed to alloxan significantly.  Key words: Eleutherine palmifolia, alloxan, kidney, histhopathology

<p>Apple vinegar is estimated to be able to cure diabetes. Vinegar is able to control the increase of glicemic index. Apple has antioxidant substance such as quercetin and chlorogenic acid. Manalagi apple could be found easily in Indonesia.</p><p>            This research aims to reveal the effect of Manalagi apple vinegar (<em>Malus sylvestris </em>Mill) towards blood glucose level of alloxan induced Wistar white male rats (<em>Rattus norvegicus</em>).</p><p>            This research is experimental laboratory using <em>Experimental </em>methods. The samples are 30 white rats divided into three groups for 31 days, 1) group which only given standard diet; 2) group that induced by 120mg/dL alloxan; 3) group that induced by 120mg/dL alloxan and Manalagi apple vinegar 0.27ml/100gram weight for 14 days. The measurement of blood serum glucose level is done on day twenty four using homogenous colorimetric enzymatic test method for all groups.</p><p>            Blood glucose average in group with only given standard diet as negative control group (182.33 mg/dL), group which induced by 120mg/dL alloxan as positive control group (205.2 mg/dL), group which induced by 120mg/dL alloxan and Manalagi apple vinegar as treatment group (212.25 mg/dL). The result of <em>One-Way Anova </em>significance<em> </em>is 0.229.</p><p>            The conclusion of this research is that Manalagi apple vinegar treatment doesn’t able to decrease the blood glucose level of white Wistar male rats significantly.</p><p> </p><strong>Keywords </strong>: <em>Malus sylvestris </em>Mill, Blood glucose, Alloxan.

In diabetes mellitus there is an increase in oxidative stress in which oxidants exceed the antioxidant system resulting from the production of oxidant-antioxidant imbalance and/or limited antioxidant defenses. Abelmoschus esculentus (“okra”) is known as lowering blood glucose vegetable by Indonesian. A. esculentus has a high antioxidant activity that can: 1) lower H2O2 level; 2) decrease the amount of ROS; 3) lower •OH level. This study tried to prove that administration of A. esculentus may decrease fasting blood glucose, serum malondialdehyde, and increase superoxide dismutase activity of diabetic male white rats. This research is a pure experimental research, with randomized post test only control group designs, held in 28 days. Thirty one rats were divided into 4 groups randomly: KN = normal control group given 0.1% Na-CMC solution; KDM = DM control group injected with STZ, followed by 0.1% Na-CMC solution; P1 = treatment group 1 injected with STZ, followed by administration of A. esculentus at a dose of 775 mg/kgBW/day; P2 = treatment group 2 injected with STZ, followed by administration of A. esculentus at a dose of 1550 mg/kgBW/day. The result: administration of A. esculentus at doses of 775 mg/kgBW/day and 1550 mg/kgBW/day can not decrease fasting blood glucose and MDA serum level of diabetic male white rats. Giving A. esculentus at a dose of 1550 mg/kgBW/day may increase the activity of SOD erythrocytes of diabetic male white rats.. Conclusions: A. esculentus has the potential to lower blood glucose and MDA serum levels, also increase erythrocyte SOD activity.

ABSTRACT Non-alcoholic fatty liver can be triggered by used cooking oil consumption due to the formation of free radicals and the accumulation of fatty acids in the body. Astaxanthin is a powerful antioxidant that may be able to inhibit the pathogenesis of fatty liver. This study aims to determine the effect of astaxanthin in inhibiting fatty liver (steatosis) and levels of Gamma-Glutamyltransferase (GGT) in male Wistar rats given used cooking oil. An experimental study with Post-test Only Control Group Design was conducted on 36 male Wistar rats aged 3.5-4 months with an approximate bodyweight of 200-210 grams divided randomly into 2 groups. The control group was given 0.42 ml of used cooking oil + 0.5 ml of distilled water, and the treatment group was given 0.42 ml of used cooking oil + 0.2 mg of astaxanthin each day for 14 days. On day 15, blood tests and hepatic histopathology were performed to check  GGT serum levels and steatosis. The comparative test was conducted to compare the results of the control and treatment groups. The results showed that the mean steatosis and GGT levels in the treatment group were significantly lower than the control group. It can be concluded that giving astaxanthin can inhibit fatty liver (steatosis) and increase GGT serum levels in male Wistar rats given used cooking oil.   Keywords: Astaxanthin, Fatty liver, Gamma-glutamyltransferase, Used cooking oil

Abstract Background: Diabetes mellitus is one of the most important causes of Alzheimer’s disease and dementia. Portulaca oleracea (P.oleracea) is a rich source of antioxidants, which reduces inflammation and oxidative stress in diabetic rats. Exercise training has also been shown to improve mental function and enhance learning and memory efficacy. Therefore, this study was designed to explore the potential combined effect of P. oleracea and exercise training on neurobehavioral dysfunction in streptozotocin (STZ)-induced diabetic male rats. Methods: For this purpose, 50 male Wistar rats were divided into five groups: 1) healthy control group (Con), 2) sedentary diabetic group (D), 3) diabetic rats treated with P. oleracea(D+Po), 4) diabetic rats treated with exercise training (D+Ex), and 5) diabetic rats treated with P.oleracea and exercise training (D+Po+Ex) simultaneously. Animals in the exercise groups were subjected to progressive swimming training for 12weeks. P.oleracea was mixed with standard pellet food for 12weeks. Neurobehavioral dysfunction was investigated by elevated plus-maze, shuttle box, open field, and novel object recognition tests.Results: Compared with the normal control group, rats in the sedentary diabetic group showed a more passive avoidance memory deficit and more anxiety, and less exploration. Due to exercise training and treatment with P. oleracea, the neurobehavioral deficit in the trained diabetic rats receiving P. oleracea reached the normal levels of those in the healthy group.Conclusion: These data demonstrated that diabetes causes significant neurobehavioral deficit. Nevertheless, swimming training and P. oleracea synergistically ameliorate and reverse the neurobehavioral deficit in STZ-induced diabetic male rats.

Background: Diabetes mellitus can lead to histomorphometrical changes in the brain. Recent studies have shown that Aloe vera gel has antioxidant and neuroprotective effects, which is independent of glucose-lowering effects. Objectives: The present study aimed to investigate the effects of A. vera gel on histomorphometrical changes of cerebellum following streptozotocin (STZ)-induced diabetic male rats. Methods: A total of 25 male Wistar rats were randomly allocated into five groups as follows: (1) the control group received normal saline; (2) A. vera gel group; (3) diabetic group (normal saline); (4) treatment group diabetic rats, which received A. vera; and (5) diabetic rats which received insulin. A single dose of STZ [60 mg/kg; intraperitoneal (IP)] was used for the induction of diabetes in rats. All the treatments were administered daily for eight weeks. Subsequently, histomorphometrical changes were evaluated in the cerebellum of the rats. Results: The results showed that the number of granular and purkinje cells reduced in the cerebellum granulosa region, while the number of glial cells increased in the molecular region of the cerebellum in diabetic rats compared to the control group (P < 0.05). These changes were improved in treated rats by insulin or A. vera. Also, the thickness of molecular, purkinje, granular, and white matter layers at the apex of lobules and depth of sulcus in the diabetic group had a significant reduction compared to other groups (P < 0.001). Conclusions: Our results confirmed that improvement of the cerebellar tissue changes in diabetic rats following the use of A. vera gel is comparable to insulin. However, more investigations are required to determine the protective effects of A. vera gel against diabetes-induced cerebellum histomorphometrical changes.

Abstract: Grass jelly leaf is a traditional Indonesian beverage that is believed to be beneficial for our health. This study was carried out to observe the increased number of pancreatic  cells and the decreased fasting blood glucose in male Wistar rats with diabetes induced by streptozotocin (STZ). This was a pure experimental study with a post-test only control group design. Subjects were 36 male rats divided into 2 groups: the control group (P0), given glibenclamide dose 0.09 mg /200 g rat body weight and 2 ml aquadest as placebo; and the treatment group (P1), given glibenclamide 0.09 mg/200mg rat body weight and grass jelly leaf extract 54 mg/200 mg rat body weight. Pancreatic  cell count and blood glucose examination were performed after 28 days of treatment. The results showed that the number of pancreatic  cells in the treatment group (P1) was statistically higher than of the control group (P0) (56.72±5.644 vs 29.11±2.698 cells (P ≤0.001). In addition, the fasting blood glucose level of the treatment group (P1) was statistically lower than of the control group (P0) (79.50±37.75mg/dl vs 191.00±123.15mg/dl) (P <0.001). Conclusion: The administration of grass jelly extract increased pancreatic β cells and decreased fasting blood glucose level in diabetic male rats. Penelitian ini dilakukan untuk mengetahui adanya peningkatan jumlah sel β pankreas dan penurunan kadar gula darah puasa pada tikus Wistar diabetes yang diinduksi streptozotocin (STZ). Jenis penelitian ialah eksperimental murni dengan post-test only control group design. Subyek penelitian ialah 36 ekor tikus putih jantan, dibagi menjadi 2 kelompok, yaitu: kelompok kontrol (P0) yang diberikan glibenklamid oral dosis 0,09 mg/200gr BB tikus dan 2 ml aquadest sebagai plasebo; dan kelompok perlakuan (P1) yang diberikan glibenklamid oral dosis 0,09 mg/200 gr BB tikus dan ekstrak daun cincau dosis 54 mg/200gr BB tikus. Penghitungan jumlah sel β pankreas dan pengukuran gula darah dilakukan setelah 28 hari perlakuan. Hasil penelitian menunjukkan bahwa kelompok perlakuan (P1) memiliki jumlah sel β pankreas yang secara statistik lebih tinggi dibandingkan kelompok kontrol (P0) (56,72±5,644 sel/lapang pandang vs 29,11±2,698 sel/lapang pandang) (P ≤0,001). Selain itu, kelompok perlakuan (P1) memiliki kadar gula darah puasa yang secara statistik lebih rendah dibandingkan kelompok kontrol (P0) (79,50±37,75 mg/dl vs 191,00±123,15 mg/dl) (P <0,001). Simpulan: Pemberian ekstrak daun cincau (Mesona palustris BL) meningkatkan jumlah sel β pankreas dan menurunkan kadar gula darah puasa pada tikus (Rattus novergicus) jantan galur Wistar diabetes.Kata kunci: ekstrak daun cincau, gula darah puasa, sel beta pankreas, diabetes melitus
