Comprehensive GC-MS and FT-IR Profiling of Coleus zeylanicus essential oil and investigation of its anti-fungal and anti - oxidant activities

To evaluate the leaf volatile constituents of essential oil of Coleus zeylanicus and evaluate their anti-oxidant and anti-fungal activity. The Chemical composition of Coleus zeylanicus essential oil was determined using GC-MS and FT-IR analytical techniques. The antioxidant activity was evaluated using DPPH scavenging assay. The anti-fungal effect was tested against two potential pathogenic fungal strains - Candida albicans and Malassezia furfur using agar well diffusion method. The essential oil was profiled by the presence of sesquiterpene hydrocarbons 90.67% of their total composition followed by oxygenated monoterpenes and monoterpene hydrocarbons as 5.3% and 2.1% respectively. The GC-MS results showed 14 compounds from Coleus zeylanicus leaf EO representing 98.07% of the total oil composition. The major component was identified as a-Gurjunene (35.94%), a-bisabolol (10.82%) and G-selinene (4.26%). EO showed remarkable antioxidant activity values of IC50 = 59.78± 3.21µg/ml by DPPH scavenging assay. The essential oil showed interesting anti-fungal effects against two pathogenic fungal strains. The most sensible strains to Coleus zeylanicus EO was Malassezia furfur (32.00±0.50mm) compared to that of Candida albicans (15.00±1.25mm). Hence, Coleus zeylanicus EO has potential application against fungal infection and oxidative stress-related diseases. However, further investigations are necessary to isolate and investigate the action mechanism of these bioactive compounds.

Medicinal activities of Thymus vulgaris L. were studied in the underlying research which was conducted in University of Central Punjab, Lahore. The powdered plant sample was collected and subjected to extraction and filtration by dissolving it in seven different solvents and thus the fractions obtained, were used for further analysis. The qualitative and quantitative analysis was carried out which revealed that this plant contained many biologically active compounds like saponin, alkaloids etc. These compounds have enormous bioactive roles. Antimicrobial activity was also determined in which four bacterial strains were used and the susceptibility of all the fractions of plant extract was determined by their inhibition zones which were calculated in mm. The results showed that water and chloroform extracts of plant has highest activity. The antioxidant activity was also determined by DPPH scavenging assay for which different concentrations of all plant extracts were made and their absorbance was noted to measure the percentage inhibition of each concentration, which revealed that this plant has significant antioxidant activity. The results have been represented using statistical approach that revealed that T. vulgaris plant has significant antibacterial and anti-oxidant activities and phytochemical analysis also proved that it has many useful compounds which have voluminous biological activity and these could be used in the formation of novel drugs.

Background and Aims: Acorus calamus L. is an indigenous herb in Nepal. It belongs to family Acoraceae and grows in wet land with scented rhizomes. It is also known as Sweet flag in English and commonly as Bojho in Nepal. The present investigation reveals the chemical compositions and antioxidant activity of rhizome essential oil of A. calamus. Methods: Essential oil of rhizomes of Acorus calamus L. from Kaski district, Nepal was extracted by hydrodistillation method and volatile constituents were analyzed Gas chromatography-Mass spectrometry. The antioxidant potential of essential oil was analyzed by 1,1-Diphenyl-2-Picrylhydrazyl (DPPH) scavenging assay. Results: A GC-MS analysis revealed the presence of β-asarone (22.38%), α-asarone (14.97%), 1-(4,6-dimethoxy-2,3-dimethylphenyl ethanone (14.24%), Isoelemicin (5.68%), cis-Methylisoeugenol (4.26%), α-calacorene (4.16%), and other 20 minor components. From DPPH assay, half maximal inhibitory concentration (IC50) value of essential oil was found to be 108.71 µg/mL. Conclusions: These findings have strengthened the A. calamus L. is good source of compounds like β-asarone, α-asarone and can be used as potential antioxidants. BIBECHANA 17 (2020) 89-95

This work involves the preparation of high yield iminochalcon compounds (B1-B15)  through two parts. The first part involves the preparation of 2,4-dihydroxy Chalcone (A1-A15) by the condensation of 2,4-dihydroxy acetophenone  with  aryl aldehyde in the presence of sodium hydroxide (40%) as a catalyst. The second part includes the preparation of  iminochalcon from the condensation of p-hydroxy aniline with 2,4-dihydroxy chalcone  derivatives  )A1-A15)  in the presence of some drops of conc. H2SO4. Thin-layer chromatography ((TLC)  was used to control the chemical reaction . These new derivatives were characterized by using FT-IR and  1H-NMR spectroscopy. These synthesized compounds were also assessed by the DPPH (2,2-diphenyl-1-picryl-hydrazyl-hydrate) free radical method, through which the compounds (B1-B15) were evaluated for their antioxidant activity. The compound B3 was found to have the strongest antioxidant activity  (IC50= 23.91 μg/mL) as compared to that of the common standard of ascorbic acid (IC50=31.95 μg/mL).

In the ancient time Eucalyptus globulus used for the various purpose. It is a tall and an evergreen tree that can grow up to the height of 70m and its diameter is about 4 to7ft. which was first found on the island of Tasmania in 1792 which associate to the Myrtaceae family which is commonly called as Tasmanian Blue Gum, southern blue gum or blue gum are the different names of eucalyptus globulus grow in the various parts of India. The particular species of Eucalyptus globulus have been used for various purposes. It had been found that eucalyptus is a rich source of phytochemical constituent and also possesses medicinal use. The different part of the eucalyptus is very nutritionally and highly valuable therapeutically property because of the presence of some specific chemical composition as its essential oil contain esters, carboxylic acids, aldehyde etc. phytochemical analysis of this tree has revealed that their leaf oil contain 1.8 cineole, cryptophone etc., essential oil had been extracted from its buds. The aim of this review paper is compiling all the information about eucalyptus such as anti-inflammatory, analgesic, anti-nociceptive antimicrobial, anti-fungal, anti-viral and anti-oxidant agent of the nature.

Tilianin (acacetine-7- O-β-D-glucopyranoside) was isolated from the ethyl acetate extract of Morinda tinctoria Roxb. and its structure established unambiguously by a series of 1D- and 2D-NMR analyses. This is the first record of this compound for this species. The antioxidant activity of tilianin, determined using the DPPH scavenging assay, was measured with an IC50 value of 57.5 μg/mL.

The fruits of some Cornus species (dogwoods) are used in traditional medicine and considered potential anti-diabetic and hypolipemic agents. The aim of the study was to determine the ability of extracts from Cornus alba (CA), Cornus florida (CF), and Cornus sanguinea (CS) to inhibit digestive enzymes namely α-amylase, pancreatic lipase, and α-glucosidase, as well as isolation of compounds from plant material with the strongest effect. In addition, the phytochemical profile and antioxidant activity of extracts from three dogwoods were compared with HPLC-DAD-MS/MS and DPPH scavenging assay, respectively. Among the aqueous-ethanolic extracts, the activity of α-amylase was the most strongly inhibited by the fruit extract of CA (IC50 = 115.20 ± 14.31 μg/mL) and the activity of α-glucosidase by the fruit of CF (IC50 = 38.87 ± 2.65 μg/mL). Some constituents of CA fruit extract, such as coumaroylquinic acid, kaempferol, and hydroxytyrosol derivatives, were isolated. Among the three species of dogwood studied, the greatest biological potential was demonstrated by CA extracts, which are sources of phenolic acids and flavonoid compounds. In contrast, iridoid compounds or flavonoid glycosides found in fruits of CF or CS extracts do not play a significant role in inhibiting digestive enzymes but exert antioxidant activity.

Abstract The synthesis of novel 1,4-naphthoquinone derivatives has attracted prominent interest in the field of medicinal chemistry since these compounds exhibit potent pharmacological activity as antibacterial, antioxidant, antifungal, and anticancer. Herein, a series of novel 1,4-naphthoquinone derivatives 4-7, 8a-c, and 9a-d containing heterocyclic moieties were synthesized in good yields and characterized by spectral and elemental analyses. All the new synthesized compounds were subjected to in-vitro antimicrobial testing against gram-positive, gram-negative, and fungal strains by calculating the average of the zone of inhibition. The antimicrobial results showed that compounds 8b, 9b, and 9c displayed the highest efficacy against both bacterial and fungal strains. Further studies have been conducted to estimate the antioxidant activity of the compounds using DPPH scavenging assay. The obtained results revealed that compounds 9d, 9a, 9b, 8c, and 6 exhibited the highest radical scavenging activity. Docking studies of the most active antimicrobial compounds within GLN- 6-P, recorded good scores with several binding interactions with the active site.

Abstract: Inhibition Activity Of Turmeric Essential Oil Against The Growth Of Candida Albicans. Turmeric has rhizomes which contain essential oils and curcuminoid that has anti-fungal activity. The purpose of this research was to know the influence of essential oil of turmeric Rhizome (Curcuma domestica Val.) against the growth of Candida albicans in vitro. This was an experimental research with diffusion method. Essential oil obtained through distillation methods and the concentration used was 20%, 40%, 60%, 80% and 100%. The treatment was repeated three times. Anova test was used. Inhibition zone is formed at a concentration of 20%, 40%, 60%, 80%, 100% were 6,16 mm, 6,50 mm, 6,66 mm, 7,00 mm and 7.83 mm, respectively Results in analysis by one way ANOVA showed there was a significant difference in the variance of essential oil concentration against the inhibition zone (p< 0.05).Abstrak: Daya Hambat Minyak Atsiri Rimpang Kunyit Terhadap Pertumbuhan Candida Albicans In Vitro. Kunyit merupakan  salah satu rimpang yang diketahui memiliki kandungan minyak atsiri serta curcuminoid yang bersifat anti jamur. Tujuan penelitian ini untuk mengetahui pengaruh minyak atsiri rimpang kunyit (Curcuma domestica Val.) terhadap pertumbuhan Candida albicans secara in vitro. Jenis penelitian ini adalah penelitian eksperimental dengan metode difusi cara sumuran. Minyak atsiri didapatkan melalui   metode destilasi dan konsentrasi  yang digunakan adalah 20%, 40%, 60%, 80% dan 100%. Perlakuan diulang sebanyak tiga kali. Hasil penelitian di analisis dengan Anova. Hasil zona hambat yang terbentuk pada konsentrasi minyak atsiri 20%, 40%, 60%, 80%, 100% berturut-turut adalah 6,16 mm, 6,50 mm, 6,66 mm, 7,00 mm,  dan 7,83 mm. Hasil analisis dengan one way anova menunjukkan terdapat perbedaan yang signfikan pada berbagai konsentrasi minyak atsiri terhadap zona hambat yang dihasilkan (p<0,05). Dari penelitian ini dapat disimpulkan bahwa berbagai konsentrasi minyak atsiri rimpang kunyit dapat menghambat pertumbuhan Candida albicans. Kemampuan ini berpotensi dikembangkan lebih lanjut sebagai alternatif dalam pengobatan terhadap penyakit karena infeksi Candida albicans.

Citronella fragrance is one of the many plants in Indonesia. Citronella fragrance is widely used by Indonesian people as a spice dish and has traditionally been used for various treatments. Citronella fragrance contains essential oils that have the potential to have anti-fungal effects and have traditionally been used for the treatment of skin diseases. Isolation of citronella essential oil with the method of vapor distillation. The tested fungi are opportunistic fungi that often infect human skin. In testing the antifungal activity of the citronella essential oil (Cymbopogonnardus (L.) Rendle) against the fungal causing skin infection. The research was done by using agar diffusion method and using the density disc as the diffusion medium against A research of antifungal�s activity of essential oil of citronella (Cymbopogon nardus (L.) Rendle ) to fungus that cause disease in humans has been done. This research used agar diffusion method to the fungus test Tricophyton rubrum, Tricophyton mentagrophytes and Candida albicans. The concentration that used 1%, 0,5%, 0,25% and 0,1%. The results showed that in concentration 1% essential oil of citronella (Cymbopogon nardus ( L.)Rendle) has the highest activity with Candida albicans with an average of inhibition diameter was 19,4mm � 0,15. In 0,1% concentration of essential oil of citronella (Cymbopogon nardus (L.) Rendle) does not have antifungal�s activity to Tricophyton mentagrophytes that without inhibition diameter around the paper discs. Meanwhile, in concentration 0,1%, the essential oil of citronella (Cymbopogon nardus (L.) Rendle) still have antifungal�s activity to Tricophyton rubrum and Candida albicans with an average of inhibition diameter 7,4 mm � 0,35 dan 8,5 mm � 0,15.
