Objective: To evaluate the anti diabetic effect of cinnamon extract in alloxaninduced diabetic animal model (albino rats) in comparison with oral hypoglycemic drugs. StudyDesign: An Experimental study. Place of Study: Al Tibri Medical College, Isra University, KarachiCampus. Duration of Study: December 2012 to December 2013. Materials and Methods: Total60 Albino rats of both genders were divided into 6 groups consisting of 10 rats in each group.Each group of animals was further divided into two sub groups containing 5 rats in each groupResults: The results obtained from the data indicated that there is significant reduction in bloodglucose level rats treated with low dose of cinnamon extract. The animals of low dose cinnamonextract (200mg/kg. bw) when compared with other groups; there is a reduction in the bloodglucose level in alloxan induced diabetic rats. Also tolbutamide and acarbose treated groupsshowed better antidiabetic effects as compared with cinnamon extract treated groups (pvalue<0.007 and p value<0.012 respectively), but cinnamon extract treated group showedsynergetic effects when it was given in combination with tolbutamide or acarbose havingsignificant p value<0.001 and p value<0.011 respectively. Conclusions: Tolbutamide andAcarbose showed better anti diabetic effect in comparison with cinnamon extract treated groupswhen used individually. This effect was enhanced when cinnamon was used in combination witheither tolbutamide or acarbose.

Atorvastatin, a lipid lowering agent, possesses various pleiotropic vasculoprotective effects, but its role in coronary angiogenesis is still controversial. Our objective was to study the effects of atorvastatin on the angiogenic responsiveness of coronary endothelial cells (cEC) from normal and diabetic rats. Male Wistar rats were distributed among 9 groups; (i) normal rats, (ii) 30 day diabetic rats, (iii) 60 day diabetic rats, (iv) normal rats administered a low dose of atorvastatin (1 mg/kg body mass, per oral (p.o.), for 15 days); (v) 30 day diabetic rats administered a low dose of atorvastatin; (vi) 60 day diabetic rats administered a low dose of atorvastatin; (vii) normal rats administered a high dose of atorvastatin (5 mg/kg, p.o., for 15 days); (viii) 30 day diabetic rats administered a high dose of atorvastatin; (ix) 60 day diabetic rats administered a high dose of atorvastatin. Each group was further divided into 2 subgroups, (i) sham ischemia–reperfusion and (ii) rats hearts that underwent ischemia–reperfusion. Angiogenic responsiveness the and nitric oxide (NO) releasing properties of the subgroups of cECs were studied using a chorioallantoic membrane assay and the Griess method, respectively. Atorvastatin treatment significantly increased VEGF-induced angiogenic responsiveness and the NO-releasing properties of cECs from all of the subgroups, compared with their respective non-treated subgroups except for the late-phase diabetic rat hearts that underwent ischemia–reperfusion, and the high dose of atorvastatin treatment groups. These effects of atorvastatin were significantly inhibited by pretreatment of cECs with l-NAME, wortmannin, and chelerythrine. Thus, treatment with a low dose of atorvastatin improves the angiogenic responsiveness of the cECs from normal and diabetic rats, in the presence of VEGF, via activation of eNOS–NO release.

Intraperitoneal transplantation of collagenase-digested, isogeneic, neonatal rat pancreatic tissue successfully reversed streptozotocin-induced diabetes in 77% of recipients. The low serum immunoreactive insulin, hyperglycaemia, glycosuria and weight loss, characteristic of the diabetic animal, were corrected and the reduced activities of hepatic glucokinase and pyruvate kinase, and the low glycogen concentration of the liver of diabetic rats were restored to normal. Forty-three per cent of the successfully transplanted rats became normoglycaemic within 1 month of transplantation whereas 57% took from 1 to 6 months to achieve normoglycaemia and displayed a mild glucose intolerance when subjected to a glucose load. The rats which had not become normoglycaemic 6 months after transplantation showed some amelioration of the diabetic state, as shown by increased serum immunoreactive insulin and hepatic glycogen concentration and a slow weight gain compared with diabetic controls.

Currently, cardiovascular diseases are the leading cause of global mortality, while diabetes mellitus remains an important cause of cardiovascular morbidity. A recent study showed that patients with diabetes mellitus treated with mineralocorticoid receptor antagonists have improved coronary microvascular function, leading to improved diastolic dysfunction. In this study, we evaluated the influence of acute administration of spironolactone on myocardial function in rats with streptozotocin-induced diabetes mellitus, with special emphasis on cardiodynamic parameters in diabetic rat hearts. The present study was carried out on 40 adult male Wistar albino rats (8 weeks old). Rats were randomly divided into 4 groups (10 animals per group): healthy rats treated with 0.1 μmol/L of spironolactone, diabetic rats treated with 0.1 μmol/L of spironolactone, healthy rats treated with 3 μmol/L of spironolactone, and diabetic rats treated with 3 μmol/L of spironolactone. Different, dose-dependent, acute responses of spironolactone treatment on isolated, working diabetic and healthy rat heart were observed in our study. In healthy rats, better systolic function was achieved with higher spironolactone dose, while in diabetic rats, similar effects of low and high spironolactone dose were observed.

Background: Vernal keratoconjunctivitis (VKC) is a recurrent, bilateral, external, ocular inflammation primarily affecting young adults living in warm dry climates. The objectives of the research was to compare the two ophthalmic solutions of olopatadine hydrochloride (0.1%) and epinastine hydrochloride (0.05%) on clinical signs of vernal keratoconjuntivitis and to determine side effects of both the drugs.Methods: The study was carried out in 40 patients who attended the out-patient department (OPD) Ophthalmology, Darbhanga Medical College and Hospital, Laheriasarai from July 2007 to December 2008.  Forty patients with symptoms of VKC (ocular itching, ropy discharge, papillary hypertrophy, gelatinous thickening and horner-trantas spots were selected and included in our study.Results: Mean score of palpebral hyperemia at 0, 14, 28 and 42 days in olopatadine treated eye were 2.1, 1.4, 0.8 and 0.4 respectively having p value <0.01, and <0.01 and <0.05 respectively, while mean score at same stages in placebo eye were 2.1, 2, 1.9, and 1.5 having value >0.05. Epinastine treated group mean score of palpebral hyperemia were <0.01 and <0.01 respectively in epinastine treated eye whereas in placebo treated eye, mean score were 2.1, 2.0, 1.8 and 1.6 respectively having p value >0.05 in all stages. Statistically insignificant reduction at day 14 while very significant reduction at day 28 and 42 was observed in epinastine treated eye as compared to placebo.Conclusions: The present study had shown that both olopatadine and epinastine were effective in treating clinical signs of VKC as compared to placebo.

Background: Diabetes mellitus deeply changes the genes expression of integrin (Itg) subunits in several cells and tissues such as monocytes, arterial endothelium, kidney glomerular cells, retina. Furthermore, hyperglycemia could impress and reduce the rate of successful assisted as well as non-assisted pregnancy. Endometrium undergoes thorough changes in normal menstrual cycle and the question is: What happens in the endometrium under diabetic condition? Objective: The aim of the current study was to investigate the endometrial gene expression of α3, α4, αv, Itg β1 and β3 subunits in diabetic rat models at the time of embryo implantation. Materials and Methods: Twenty-eight rats were randomly divided into 4 groups: control group, diabetic group, pioglitazone-treated group, and metformin-treated group. Real-time PCR was performed to determine changes in the expression of Itg α3, α4, αv, β1, and β3 genes in rat’s endometrium. Results: The expression of all Itg subunits increased significantly in diabetic rats’ endometrium compared with control group. Treatment with pioglitazone significantly reduced the level of Itg subunits gene expression compared with diabetic rats. While metformin had a different effect on α3 and α4 and elevated these two subunits gene expression. Conclusion: Diabetes mellitus significantly increased the expression of studied Itg subunits, therefore untreated diabetes could be potentially assumed as one of the preliminary elements in embryo implantation failure.

Background: Herbal medicine is used in all parts of the world mainly for prevention and treatment of various disorders due to better cultural suitability, lower cost and less side effects. Objectives: The aim of this study was to determine the hypoglycemic and kidney-protective effects of the aqueous extract of Trigonella foenum and Cinnamon on diabetic rats. Methods: In this experimental study, rats were randomly divided into 6 groups as follows: Group 1: control group in which animals received chow diet, group 2: diabetic rats, group 3: diabetic rat + 2% T. foenum extract (w/w), group 4: diabetic rat + 8% of Trigonella foenum extract (w/w), group 5: diabetic rat + 2% Cinnamon extract (w/w) and group 6: diabetic rat + 8% of Cinnamon extract (w/w). Aqueous extract of T. foenum leaves and Cinnamon were administered to diabetic rats for 4 weeks. The malondialdehyde (MDA) level and total antioxidant capacity were also measured in kidney of the animals. In addition, morphological changes of the kidney were also analyzed by the light microscope. Results: Trigonella foenum and Cinnamon extract in diabetic animals significantly reduced MDA levels and restored antioxidant capacity (P<.05). T. foenum and Cinnamon also normalized plasma urea and creatinine concentration in diabetic rats (P<.05). Administration of T. foenum and Cinnamon extract especially at the dose of 8 mg/kg normalized histopatholgical changes of kidney in diabetic animal. Conclusions: The findings of this experiment showed that T. foenum extract and Cinnamon restored antioxidant capacity and structural changes of kidney.

Objective: To determine the anti-hyperglycemic effects of interleukin-1 inhibitor (diacerein) in alloxan induced diabetic albino wistar rats. This experimental study was performed at the Department of Animal Husbandry and Veterinary Sciences, Sindh Agriculture University, Tando Jam within 6 months from April 2016 to September 2016. Total of 160 adult Albino Wistar Rats having an average of 200 to 300 grams body weights were selected. Animals were categorized into 4 groups as; Group A (n=15): Control rats – receive 0.9% normal saline as placebo Experimental Groups Group B (n=15): Experimental Control (Diabetic rats) - Alloxan50 mg/kg body weight intraperitoneal. Group C (n=15): Diabetic rats + Diacerein (30 mg/kg/day) orally daily. Group D (n=15): Diabetic rats + Diacerein (50 mg/kg/day) orally daily. Animals were kept and treated as per the NIH Guideline for Use and Care of Laboratory Animals. Diabetes mellitus was induced via a single intraperitoneal injection of 50 milligram/kg alloxan monohydrated dissolved in aseptic 0.9% saline. After 72 hours, blood specimens were taken from the caudal vein of the rats and glucose level>200 mg/dL was taken as diabetes. Experimental rats were given diacerein approximately 30 and 50 mg orally for 6 weeks. At the completion of experiment the body weight was measured of each animal by electronic measuring balance and blood sample was taken from each animal of all groups to assess the blood glucose level and HbA1c level. Data were recorded via self-made proforma and analysis was done by using SPSS version 20. Results: Average body weight of Diabetic control (Group B) was 193.33±22.50 grams, which was lower in contrast to Diacerein treated group C 202.47±25.70 grams and significantly lower as compared to Diacerein treated group D as  212.6±23.43 grams. A significant increase in blood glucose levels 182.07±10.63 mg/dl was noted in the Diabetic control (Group B) compared to Diacerein treated group C (110.13± 8.54 mg/dl) and group D (85.87±8.41 mg/dl) (P=0.001). HbA1c was markedly raised in the Group B- diabetic controls, while diacerein treated diabetic rats (groups C and D) showed a significant decrease in HbA1c (P=0.001). Conclusion: It was concluded that Diacerein achieves the Euglycemic state by reducing the levels of blood glucose and glycated hemoglobin (HbA1c) in Alloxan-Induced diabetes mellitus in Wistar Albino Rats.

Abstract Background: The majority of people in Africa receive their basic healthcare through herbal treatments. Herbal medicine may negatively impact fetal development irreparably. Methods: This study examined teratogenic potential of Solenstemma argel extract in pregnant rats. pregnant rats were treated with Solenstemma argel from 7th to 16th day of gestation. Dosage used was 500g/kg, intraperitoneal.Results: Solenstemma argel extract treated group showed fetal abnormalities appeared as body clots, limbs abnormalities and Resorption of fetuses that appears in 25% of the fetuses (P-value = 0.01) which is significantly differ from control group. furthermore, Histopathological findings of liver sections from fetuses of Solenstemma argel - treated mothers showed loose liver texture and hepatocyte hemorrhage.Conclusions: In this study we explore the teratogenic potential of S. argel extract during the organogenesis period in pregnant rats.

The present work investigated the protective effects of Costus afer Ker Gawl. aqueous leaf extract (CALE) on lipid profile and hematological changes induced by exposure to low-dose heavy metal mixture in male albino rats. The experimental animals were divided into six weight matched groups. The normal (group 1) and toxic (group 2) controls received deionized water and metal mixture (20 mg/kg PbCl2, 1.61 mg/kg CdCl2, and 0.40 mg/kg HgCl2), respectively. Test rats in groups 3, 4, and 5 were treated with metal mixture and CALE (750, 1500, and 2250 mg/kg, respectively), and group 6 received metal mixture and ZnCl2. All treatments were administered through oral gavage for 12 weeks. LDHMM caused a marked increase ( p < 0.05 ) in cholesterol, triglyceride, low-density lipoprotein (LDL), and very low-density lipoprotein (VLDL) levels and a decrease in high-density lipoprotein (HDL), percentage body weight gain, and feed and fluid intake. Also, a significant decrease in RBC, Hb, and PCV, a significant increase in WBC, and no significant increase in platelet PLT were observed in the metal mixture-treated group. But in CALE treated groups, their levels were found to attain almost normal values as found in normal control which is also similar to the zinc-treated group. Costus afer may hold a promise in improving lipid profile and hemodynamic picture in cardiovascular diseases.

Connexin 43 (Cx43) is a potential target for accelerating wound healing. This study aimed at evaluating the therapeutic efficiency of topical application of Gap27, a Cx43 mimetic peptide, on corneal tissue reorganization during wound healing in streptozocin-induced Diabetes in albino rats and its effect on the infiltration of inflammatory cells. Fifty adult male albino Wistar rats were divided equally into two groups: non-diabetic and diabetic. Twenty rats from each group were subjected to corneal injury: 10 untreated and 10 treated with Gap27. The remaining five rats from each group served as negative controls (intact corneas). All rats were sacrificed 3 days after injury. Histological studies were performed to assess signs of cell degeneration, the infiltration of inflammatory cells. Histomorphometric studies were performed to quantify the expression of Cx43. Gap27 promoted corneal wound healing in non-diabetic and diabetic rats. It reduced mononuclear cell infiltration and improved corneal tissue remodeling. However, minor structural changes were still seen in diabetic corneas after treatment with Gap27. Blocking Cx43 was a valuable tool to restore corneal tissue structure, reduce the infiltration of inflammatory cells in non-diabetic and diabetic rats.
