  Twenty chicks at (45) days old were used and divided into two equal groups , one act as treatment (Baytril) dosed orally (10 mg/kg ) for 5 days, while the control group dosed with distilled water.  Duodenum was isolated in vi tro examined with autonomic agents.  The result showed significant decrease in intestinal motility between treatment and control group after addition of atropine, acetycholine, atropine flowed acetycholine and after addition propranolol and propranolol flowed adrenaline.  Concluded from this study that using of Enrofloxacin case a negative alteration tonic and rhythmic contraction of intestinal (smooth muscles).

The increased shortening velocity and capacity of airway smooth muscle (ASM) from ragweed pollen-sensitized dogs, which may be responsible for its in vivo airway hyperresponsiveness, have been shown to be associated with higher actomyosin adenosinetriphosphatase activity and greater level of phosphorylation of the 20-kDa myosin light-chain (MLC20) at rest and during contraction. Current studies show that the elevated level of phosphorylation may be the result of an increased myosin light-chain kinase (MLCK) activity due to excessive quantity of MLCK. There were no significant changes in total activity of calmodulin, a protein that binds and activates MLCK, in sensitized dog ASM (SASM) compared with control ASM (CASM). When normalized to the relative calmodulin content in the tissues, the specific calmodulin activities (means +/- SE) in sensitized tracheal smooth muscle (STSM) and sensitized bronchial smooth muscle (SBSM) and in their controls were not different (STSM 0.359 +/- 0.117, CTSM 0.339 +/- 0.136. SBSM 0.201 +/- 0.098, and control bronchial smooth muscle 0.213 +/- 0.056 nmol Pi.calmodulin content-1.min-1, respectively). Intracellular Ca2+ levels indicated by fura 2 fluorescent dye remained unaltered in SASM. We conclude that airway hyperresponsiveness may result from higher MLCK content in SASM rather than from changes in Ca(2+)-calmodulin activities, which is an example of alteration in Ca2- sensitivity of ASM.

Background: Recently, the role of inflammation triggered by handling of the intestine various gastrointestinal (GI) surgeries is generally accepted as the key event in postoperative ileus (POI). Because, prokinetics have been increased the smooth muscle contractions and may act by attenuating the inflammatory process in the GI tract, they have been used the treatment of POI in human and animals. There are many in vivo analysis techniques of GI motility. However, there have not yet been studied associated with the evaluation of the inflammatory response. Therefore, it was aimed to evaluate the efficiencies of 3 different prokinetics from inflammatory response during experimentally-induced POI.Materials, Methods & Results: Twenty healthy lambs (30-45 days old) were randomly assigned to four groups. In all groups, enterotomy was performed on the ileum. Erythromycin and metoclopramide were administered to the ERT and MET groups before the surgery, respectively, while lidocaine was administered to the LID group as bolus before and continuous rate infusion during the surgery. Physiological saline was administered to the lambs in control group as placebo before the surgery. Blood samples were collected before surgery (~30-45 min), at the end of surgery (0 h), and at the postoperative 1, 3, 5, 10, 48, 72 and 96 h. The concentrations of serum amyloid A (SAA), haptoglobin (HPT), fibrinogen (FIB) as acute phase proteins (APPs), thiobarbituric acide reactant substrate (TBARs), myeloperoxidase (MPO) as reactive oxygen species, and transforming growth factor-beta (TGF- β) as a cytokine were measured with ELISA reader. In terms of time points, it was found that FIB was statistically higher in ERT group at the 1st h, in MET and LID groups at the 10th h, and in LID group at the 48th and in MET group at the 72 h (P < 0.05). It was found that SAA was higher in MET group at the 1st, 3rd, 5th, 10th, 24th, 48th and 72nd h. HPT was higher in CNTR group until 72th h and MET group at 48th, 72nd and 96th h. TBARs concentrations were statistically higher in MET and LID groups at 0 hour, in ERT and MET groups at the 1st h, in MET group at the 3rd h, in MET and LID groups at the 5th and 10th h, in MET group at the 48th, 72nd and 96th h (P < 0.05). MPO concentrations was higher in LID group at the 3rd, 5th, 10th and 96th h, and in ERT group at the 72nd h (P < 0.05). TGF-β concentrations were particularly high in MET group at the 3rd, 5th, 48th and 72nd h, and in LID group at the 10th, 24th, and 96th h (P < 0.05).Discussion: APPs (HPT, SAA, FIB), which are important regulators of inflammation in cows and sheep, were higher generally in MET and LID groups and inflammation persists in these two groups and, therefore, metoclopramide and lidocaine are less effective in early postoperative POI treatment. Because, significant increase in serum TBARs and MPO concentrations was considered as an important indicator of oxidative stress and inflammatory response MPO concentrations was particularly high until 10th h in LID group, and TBARs concentrations was high both MET and LID groups throughout the study, this was correlated with higher neutrophil infiltration in the postoperative early period than the other groups. It is known that TGF-β, an inflammatory cytokine, is correlated with various smooth muscle disorders in humans. In this study, TGF-β concentration were higher in the MET and LID groups. High concentration of this cytokine might have led to decrease contractions in smooth muscles, thereby slowing down the intestinal transition. In conclusion, based on the presence of pro-inflammatory markers in this study, erythromycin seems to be the most suitable prokinetic drug in lambs. Moreover, lidocaine and metoclopramide are not as successful in small ruminants as reported in other species.

Introduction: Acidulated phosphate fluoride (APF) gels are commonly used as preventive caries materials in paediatric dentistry while glass ionomer cements (GICs) are widely used as a restorative material. The purpose of this study was to determine the effect of acidulated phosphate fluoride application towards the microhardness of GIC. Methods: This study was an experimental laboratory study where ten specimens were prepared from high viscosity GIC, which was Fuji IX. Specimens were stored in 50 ml distilled water at first 24 hours at 37°C, and then specimens were divided into fluoride group which was immersed in 25 ml of 1.23 % APF gel and control group which was stored 25 ml distilled water for 24 hours at 37°C. The Micromet II Microhardness Tester, Buehler, IL, USA that was standardised for Vickers hardness test was used to test the specimens at 100-gram load. Data were analysed using the t-test comparison test. Results: The level of microhardness of the fluoride group (14.34) was much lower compared to the control group (43.21) with a highly significant difference (p<0.01). Conclusion: The application of 1.23% APF gel on high viscosity GIC reduces the microhardness level compared to the control group.

In smooth muscle tissues, calcium-activated chloride channels (CaCC) provide the major anionic channel. Opening of these channels leads to chloride efflux and depolarization of the myocyte membrane. In this way, activation of the channels by a rise of intracellular [Ca2+], from a variety of sources, produces increased excitability and can initiate action potentials and contraction or increased tone. We now have a good mechanistic understanding of how the channels are activated and regulated, due to identification of TMEM16A (ANO1) as the molecular entity of the channel, but key questions remain. In reviewing these channels and comparing two distinct smooth muscles, myometrial and vascular, we expose the differences that occur in their activation mechanisms, properties, and control. We find that the myometrium only expresses “classical,” Ca2+-activated, and voltage sensitive channels, whereas both tonic and phasic blood vessels express classical, and non-classical, cGMP-regulated CaCC, which are voltage insensitive. This translates to more complex activation and regulation in vascular smooth muscles, irrespective of whether they are tonic or phasic. We therefore tentatively conclude that although these channels are expressed and functionally important in all smooth muscles, they are probably not part of the mechanisms governing phasic activity. Recent knockdown studies have produced unexpected functional results, e.g. no effects on labour and delivery, and tone increasing in some but decreasing in other vascular beds, strongly suggesting that there is still much to be explored concerning CaCC in smooth muscle.

To investigate effectsof Yangyinyiqi Mixture on pulmonary fibrosis caused by bleomycin. SD ratswere divided randomly into: model group(distilled water,1 mL·0.1 kg-1), dexamethasone acetate group(dexamethasone acetate, the dosage was reduced gradually), low-dose group(Yangyinyiqi Mixture, 11 g·kg-1), moderate-dose group(Yangyinyiqi Mixture, 22 g·kg-1), high-dose group (Yangyinyiqi Mixture, 44 g·kg-1) and control group(distilled water, 1 mL·0.1 kg-1). Yangyinyiqi Mixture and dexamethasone acetate were intragastrically administrated. Lung tissue was collected for histopathological examination. Compared with control group, collagen markedly increased andHYP content significantly increased on 7th day in model group (p<0.01). On 28th day, collagen was diffusely deposited, alveolar was destroyed, and HYP content significantly increased (p<0.01). Compared with model group, bleomycin-induced suffering injury caused MMP-9 expression levels to rapidly increase (7and 14 days, p<0.01). TIMP-1 markedly increased (7and 14 days, p<0.01) and stayed at a high levelto28th day. Yangyinyiqi Mixture exerted an effect against pulmonary fibrosis, which could involved prevention of collagen deposition through inhibitingMMP-9 and TIMP-1 expression.

Vasa deferentia from age-matched spontaneously hypertensive rats (SHR) compared with three strains of control rats were found to be hyperresponsive to low concentrations, 10 and 20 mM, of KCl (K+) but not to noradrenaline (NA); the SHR tissue also demonstrated a lower ED50 to K+. Significant differences in maximum tension between SHR and the control animals in response to K+ were not seen although maximum tension developed to K+ was always greater than that developed to NA in either SHR or a control group. The loss of contractile response to K+ in the absence of extracellular calcium was more rapid in the SHR than in the control animals but no such difference was noted for the loss of response to NA. Differences in the Ca2+ dependence of KCl responses were found for 15- to 17-week-old SHR but not for 5- to 7-week-old SHR while differences in the Ca2+ dependence of the NA response were noted in the younger age group of SHR but were less evident compared with the K+ response in the older age group of rat. The 15- to 17-week-old SHR, but not 5- to 7- or 9- to 11-week groups, also demonstrated a hypersensitivity to Ba2+ whereas both SHR and normotensive control tissues demonstrated an extracellular Ca2+-independent and D 600 insensitive contractile response to lanthanum (La3+). The contractile response of the vas deferens to La3+ could, in part, be related to an effect of raised H+ but a pH-independent action of La3+ was also evident from studies with Tris-buffered solutions. Although the action of La3+ was similar in both SHR and control rats the SHR tissue was shown to be more sensitive to H+. The results demonstrate an altered threshold sensitivity and altered responsiveness of nonvascular smooth muscle in the SHR which parallels, in some respects, previous studies indicating altered vascular smooth muscle responsiveness to a variety of physiological and nonphysiological stimuli in the SHR. Furthermore, these studies may indicate that a generalized genetic alteration in the sensitivity of the smooth muscle cell to chemical stimulation and Ca2+ utilization may persist in the SHR. Whether such differences relate to the elevated blood pressure and etiology of hypertension is not yet clear.

Objectives:  This study was performed to investigate the efficacy of using glycine powder air-polishing (GPAP) or  antiseptics (CHX) adjunctively to scaling and  root planing (SRP) in the treatment of periodontitis. Methods: In this trial, ninety patients (between 28-80 years old) who had at least three teeth with 3 to 7- mm periodontal pockets, were ıncluded from the Department of Periodontology. The patients were divided into three groups randomly. In the control group ultrasonic instrumentation was performed with distilled water and hand instrumentation, in the CHX group ultrasonic instrumentation was performed with CHX and hand instrumentation, in the GPAP group in addition to SRP with ultrasonic performed with distilled water and hand instrumentation, GPAP was performed for 10 seconds per periodontal pocket using a Perio-Flow device.  All treatments were performed in one session.  Resuls: The scores of  plaque index(PI),gingival index (GI), probing depth (PD), bleeding on probing (BOP) and clinical attachement level (CAL) were decreased in all groups after 1 month and the results were statistically significant in intergoup comparisions. PI and CAL scores were statistically significant in intragroup comparisions in GPAP group. Altough GI, PD and BOP scores were lower in the GPAP group than CHX and control group, the differences after 1 month were not statistically significant.  Conclusion: Within the limits of this study, it was seen that SRP was effective alone in the treatment of periodontitis. However, using a CHX with ultrasonic devices has little beneficial effects on periodontal parameters when compared with control and GPAP groups. GPAP groups was found more effectively in PI and CAL scores.

A newly developed immunosuppressant, FTY720, has a unique mechanism that is quite different from those of conventional immunosuppressants, and is presumed to be mediated through decreases in the number of peripheral lymphocytes, especially helper T cells. This study was performed to ascertain whether this innovative drug could prolong islet allograft survival. The donors were inbred Lewis rats and the recipients were ACI rats rendered hyperglycemic with intravenous streptozotocin. In the study group, FTY720 dissolved in distilled water was orally administered at a dose of 5 mg/kg to the recipient ACI rats 1 day before and on the day of grafting. In the control group, only distilled water was orally administered to the recipient ACI rats on the day before and the day of grafting. Two thousand islets were transplanted into the portal vein of the recipient rats in the study and control groups immediately after isolation. The graft survival time in the study group was significantly longer than that in the control group, indicating that FTY720 retains a potent effect on the prolongation of islet allograft survival. FTY720 could become a useful immunosuppressant for future clinical islet allotransplantation.
