Relevance: the article substantiates the need to develop and experimentally test the effectiveness of the author’s methodological system. Theoretical analysis of the problem under study is carried out on the basis of scientific works on the formation of life safety cultures in young people and the development of pedagogical and methodological systems. Aim: of the article was to experimentally test the developed methodological system of forming a safety culture of professional activity for future builders and to substantiate its future prospects. Methods: theoretical; empirical (self-evaluation methods; peer evaluation); mathematical and statistical (frequency analysis; criterion test of statistical hypotheses; Spearman’s rank correlation coefficient; Statistical Package for the Social Science for the social sciences, Microsoft Excel). Results: a purposeful, personally-oriented, culturally appropriate and self-organizing methodological sys- tem has been developed that provides a continuous process of improving the complex of values and motives, knowledge and skills, as well as the development of professionally important qualities for the prevention and overcoming of dangerous situations and ecological threats literate behaviour, building effective communication in the professional environment during their training in vocational (vocational) educational institutions. The author defines the criteria and the corresponding indicators for assessing the levels of formation of a safety culture of professional activity in future builders. Conclusions: the results of the experiment confirmed the positive influence of the developed methodological system on the growth of the level of safety culture formation of professional activity in them. In the future, it is envisaged to study and apply the best foreign practices on its formation and development in future specialists, construction workers and teachers, to create an informational educational environment for its formation among vocational education institutions.

Abstract Nuclear factor kappa B (NFkB) regulates transcription of many genes involved in the immune response, including cytokines (e.g. IL-6) and growth factors. NFkB suppresses apoptosis as well, through various mechanisms including activation of cellular inhibitor of apoptosis-1 (cIAP-1). NFkB can be activated by the activation of Ras/phosphatidylinositol 3′-kinase (PI3K)/protein kinase B pathway in acute myeloblastic leukemia (AML). It was reported that in large proportion of primary AML cells, NFkB is constitutively activated, and the target genes are upregulated. Flt3 is a member of receptor tyrosine kinases. Flt3 receptor phosphorylation/activation results in the activation of downstream kinase pathways, like Ras, PI3K, leading to abnormal cell growth and aberrant gene regulation. Increased levels of Flt3 transcript are observed in a large number of AML specimens, and the over-expression of Flt3 contributes to the phosphorylation of Flt3 and activation of this pathway. In this study, we addressed the role of Flt3 over-expression in the activation of NFkB, which is a target molecule of various kinase pathways. We first tested the effect of Flt3 expression to NFkB responsive reporter by transient transfection and revealed the significant induction of the reporter. Next, we generated Flt3 transgenic BaF3 (BaF3-Flt3) cells that stably over-expressing Flt3 to confirm this effect. The introduction of NFkB-responsive reporter, as well as serum response element (SRE)-luciferase fused reporter into these cells resulted in the activation of the reporters. These results suggest that Flt3 over-expression mediated signal transduction activates NFkB pathways and affects growth factor pathways as well. Real time quantitative PCR demonstrated that in BaF3-Flt3 cells, mRNA expression of IL-6 and cIAP-1, both known target genes of NFkB, were increased. AG1296 or PDTC, potent inhibitors of Flt3 and NFkB respectively, completely abrogated the induction of these genes, suggesting the importance of Flt3-NFkB pathway to these effects. Moreover, we measured the expression level of Flt3 and NFkB target genes in 24 primary AML samples. As a result, there was a tendency of moderate to weak positive correlation between Flt3 and NFkB target genes. (Flt3 vs IL-6 [Spearman’s rank correlation coefficient: r= 0.35], Flt3 vs cIAP-1 [r=0.16]). These facts may also indicate the positive influence of Flt3 over-expression to NFkB target genes. Overall, these suggest a role of Flt3 over-expression in the activation of NFkB pathway in AML.

Social media platforms have had a significant impact on the public image of sports in recent years. Through the relational dynamics of the communication on these networks, many users have emerged whose opinions can exert a great deal of influence on public conversation online. This research aims to identify the influential Twitter users during the 2016 UCI Track Cycling World Championships using different variables which, in turn, represent different dimensions of influence (popularity, activity and authority). Mathematical variables of the social network analysis and variables provided by Twitter and Google are compared. First, we calculated the Spearman’s rank correlation coefficient among all users (n = 20,175) in pairwise comparisons. Next, we performed a qualitative analysis of the top 25 influential users ranked by each variable. As a result, no single variable assessed is sufficient to identify the different kinds of influential Twitter users. The reason that some variables vary so greatly is that the components of influence are very different. Influence is a contextualised phenomenon. Having a certain type of account is not enough to make a user an influencer if they do not engage (actively or passively) in the conversation. Choosing the influencers will depend on the objectives pursued.

Abstract. The research problem lies in the need to study the contradiction between the high values of life-meaning orientations and the reflexivity of employees of the internal affairs bodies. Purpose of the article: to investigate the indicated contradiction and reveal the peculiarities of the subjectivity of the personality of employees. The analysis and interpretation of the characteristics of the personal development of employees in lifelong education was carried out by the method of clustering the sample using the values of the tests "Life-meaning orientations" D. Krambo, L. Makholik, adaptation - D.А. Leontiev, reflexivity (A.V. Karpov), as well as the subjectivity of a professional's personality and focus on the personal development of an employee, including in lifelong education based on the author's methodology. The work used the correlation analysis of the studied features by means of nonparametric statistical criteria (Spearman's rank correlation coefficient), cluster analysis. As a result of the study, it was revealed that reflexivity, orientation towards development in professional activity and focus on continuing education in clusters are expressed unevenly. The hypothesis of a strict inverse relationship between manifestations of reflexivity and life-meaning orientations was not confirmed. At the same time, a feature of the subject-personal orientation of employees is an insufficient connection between reflexivity and high values of meaningful orientations, subordination of the personal principle to the "professional role and position" (according to the terms of V.A. Lefebvre) due to the subordination and discipline in the profession, as well as the substitution of the moral imperative with legal norms. Strategies with extremely high values of life-meaning orientations of adult employees of internal affairs bodies, which have a negative correlation with reflexivity, regardless of subjectivity or the severity of orientation towards development in the profession, are assessed by us as unfavorable for personal development.

The coronavirus crisis hit the world and affected all aspects of our lives, including consumers’ habits, preferences, and shopping behaviors. The survey, which involved 937 respondents from two countries, examined how the pandemic affected shopping behavior and consumer preferences in Italy and Slovakia. This paper aims to explore the impact of social media on consumer behavior, more specifically, it examines the influence of social media on the preference of specific e-shops during the first wave of the COVID-19 pandemic. Spearman’s rank correlation coefficient was used to determine a statistically significant relationship between the variables and the Mann–Whitney U test and the Kruskal–Wallis H test to assess the significance of differences between respondents in terms of demographic characteristics (residence, age, and gender). The results revealed the existence of statistically significant differences in the use of social media during the first wave of the COVID-19 pandemic in terms of various demographic factors as well as a relatively weak relationship between the social media used and the purchase in the e-shop promoted on the social media.

There is considerable debate about the social license to compete with horses and controversy about training methods for dressage horses. The objectives were to: 1. apply the Ridden Horse Pain Ethogram (RHpE) to dressage horses competing at elite Grand Prix level; 2. compare RHpE and judges’ scores; and 3. document deviations in gaits from Fédération Equestre Internationale (FEI) guidelines. Video recordings of 147 competitors from nine World Cup competitions were assessed. Spearman’s rank correlation coefficient tested the correlation between RHpE and judges’ scores. The median RHpE score was 3 (IQR 1, 4; range 0, 7). There was a moderate negative correlation (Spearman rho −0.40, p < 0.001) between the RHpE scores and the judges’ scores. Mouth open with separation of the teeth for ≥10 s (68%), head behind vertical ≥10° ≥ 10 s (67%), an intense stare for ≥5 s (30%) and repeated tail swishing (29%) were the most frequent RHpE behaviours. Deviations from FEI guidelines were most frequent in passage, piaffe, canter flying-changes, canter pirouettes and “halt-immobility-rein back five steps-collected trot”. In conclusion, most horses appeared to work comfortably for the majority of the test. Further investigation of the influence of a double bridle compared with a snaffle bridle on head position and mouth opening is merited.

The main idea of the presented work is the assumption that innovative behavior is largely predetermined by the synthesis of values and motives, the relationship of which has not been sufficiently studied. The article presents a theoretical and empirical analysis of the value-motivational characteristics of managers with different levels of expression of innovativeness. On the basis of the theoretical study, the authors of the article formulated a hypothesis about the presence of significant differences in value-motivational characteristics among managers with different levels of innovativeness. The hypothesis determined the goal of the empirical study, which was to establish links between value-motivational characteristics and the specificity of the severity of innovativeness among middle managers working in manufacturing companies. The study involved 83 respondents from 25 to 50 years old, working in manufacturing companies in the Moscow region. The survey was carried out using the following methods: (1) methodology "Diagnostics of the real structure of personal value orientations" by S.S. Bubnova; (2) questionnaire "Diagnostics of the motivational structure of personality" by V.E. Milman; (3) "Scale of self-assessment of innovative personality traits" (N.M. Lebedeva, A.N. Tatarko). For statistical verification of the results, the Kruskal-Wallis H-test and Spearman's rank correlation coefficient were used. The empirical results obtained made it possible to identify managers with different levels of self-assessment of innovativeness among the respondents and to obtain in each of the groups specific data on the manifestation of value-motivational characteristics of behavior and activity. At the end of the article, the authors formulated recommendations to enhance the professional activity of managers with different levels of self-assessment of innovativeness. In particular, it is important to develop training programs that correct the value-motivational sphere of the individual, taking into account the synthesis of those values and motives of activity that are characteristic of groups with high, medium and low self-esteem of innovativeness.

The article considers the role and influence of professional creativity on the professional activities as an expression of the teacher’s creative abilities. The purpose of the article is a theoretical and empirical study of the influence of professional creativity on the effectiveness of a teacher’s professional activity. Attention is paid to the place and importance of creativity in the activities of a teacher. Based on an analysis of current literary sources, the phenomenon of professional creativity of a teacher is highlighted, a definition of this concept is given, and its structure and components are considered. The research methodology and its algorithm include a complex psychological and pedagogical experiment, diagnosing the stated parameters and statistical analysis of the results based on Spearman's rank correlation coefficient. The study of teachers’ creativity was carried out on the basis of the Institute of Foreign Languages of the Federal State Autonomous Educational Institution of Higher Education “The Peoples’ Friendship University of Russia” (RUDN) at the Center for Continuing Professional Education. For research purposes, an experimental and a control group were formed. Each group consisted of 35 teachers. The sample was homogeneous by age and gender. The results of the study allow us to conclude that there is a strong relationship between the professional creativity of a teacher and the effectiveness of the professional performance, which is confirmed by the results of statistical processing of the data obtained.

Abstract Background Type 2 diabetes mellitus (T2DM) is mainly affected by genetic and environmental factors; however, the correlation of long noncoding RNAs (lncRNAs) with T2DM remains largely unknown. Methods Microarray analysis was performed to identify the differentially expressed lncRNAs and messenger RNAs (mRNAs) in patients with T2DM and healthy controls, and the expression of two candidate lncRNAs (lnc-HIST1H2AG-6 and lnc-AIM1-3) were further validated using quantitative real-time polymerase chain reaction (qRT-PCR). Spearman’s rank correlation coefficient was used to measure the degree of association between the two candidate lncRNAs and differentially expressed mRNAs. Furthermore, the KEGG (Kyoto Encyclopedia of Genes and Genomes) pathway and GO (Gene Ontology) enrichment analysis were used to reveal the biological functions of the two candidate lncRNAs. Additionally, multivariate logistic regression analysis and receiver operating characteristic (ROC) curve analysis were performed. Results The microarray analysis revealed that there were 55 lncRNAs and 36 mRNAs differentially expressed in patients with T2DM compared with healthy controls. Notably, lnc-HIST1H2AG-6 was significantly upregulated and lnc-AIM1-3 was significantly downregulated in patients with T2DM, which was validated in a large-scale qRT-PCR examination (90 controls and 100 patients with T2DM). Spearman’s rank correlation coefficient revealed that both lncRNAs were correlated with 36 differentially expressed mRNAs. Furthermore, functional enrichment (KEGG and GO) analysis demonstrated that the two lncRNA-related mRNAs might be involved in multiple biological functions, including cell programmed death, negative regulation of insulin receptor signal, and starch and sucrose metabolism. Multivariate logistic regression analysis revealed that lnc-HIST1H2AG-6 and lnc-AIM1-3 were significantly correlated with T2DM (OR = 5.791 and 0.071, respectively, both P = 0.000). Furthermore, the ROC curve showed that the expression of lnc-HIST1H2AG-6 and lnc-AIM1-3 might be used to differentiate patients with T2DM from healthy controls (area under the ROC curve = 0.664 and 0.769, respectively). Conclusion The profiles of lncRNA and mRNA were significantly changed in patients with T2DM. The expression levels of lnc-HIST1H2AG-6 and lnc-AIM1-3 genes were significantly correlated with some features of T2DM, which may be used to distinguish patients with T2DM from healthy controls and may serve as potential novel biomarkers for diagnosis in the future.

We previously reported that superficial non-ampullary duodenal tumors (SNADETs) commonly had a whitish mucosal surface, named milk-white mucosa (MWM). The aim of this study was to evaluate the association of MWM with epithelial intracellular lipid droplets (immunohistochemically stained by adipose differentiation-related protein (ADRP)) and histological tumor grades. We reviewed endoscopic images and the histopathology of SNADETs resected en bloc endoscopically. We analyzed the correlation between the positive rates of endoscopic MWM in preoperative endoscopy and resected specimens, and ADRP-positive rates in the resected specimens. Associations between the MWM-positive rates and tumor grades, high-grade intraepithelial neoplasia (HGIN)/intramucosal carcinoma (IC), and low-grade intraepithelial neoplasia (LGIN) were analyzed. All the 92 SNADETs analyzed were <20 mm and histologically classified into 39 HGIN/IC and 53 LGIN. Spearman’s rank correlation coefficient showed a significant correlation between MWM-positive and ADRP-positive rates (p < 0.001). MWM-positive rates were significantly lower in the HGIN/IC than in the LGIN in preoperative endoscopy (p < 0.001) and resected specimens (p = 0.02). Our results suggest that endoscopic MWM is closely associated with epithelial intracellular lipid droplets and that the MWM-positive rate may be a predictor of histological grade in small SNADETs.

Summary Introduction: Hyperplasia of the pharyngeal tonsil is one of the main causes of mouth breathing, and accurate diagnosis of this alteration is important for proper therapeutic planning. Therefore, studies have been conducted in order to provide information regarding the procedures that can be used for the diagnosis of pharyngeal obstruction. Objective: To verify the correlation between nasopharyngoscopy and cephalometric examinations in the diagnosis of pharyngeal tonsil hyperplasia. Method: This was a cross-sectional, clinical, experimental, and quantitative study. Fifty-five children took part in this study, 30 girls and 25 boys, aged between 7 and 11 years. The children underwent nasofibropharyngoscopic and cephalometric evaluation to determine the grade of nasopharyngeal obstruction. The Spearman's rank correlation coefficient at the 5% significance level was used to verify the correlation between these exams. Results: In the nasopharyngoscopy evaluation, most children showed grade 2 and 3 hyperplasia of the pharyngeal tonsil, which was followed by grade 1. In the cephalometry assessment, most children showed grade 1 hyperplasia of the pharyngeal tonsil, which was followed by grade 2. A statistically significant regular positive correlation was observed between the exams. Conclusion: It was concluded that the evaluation of the pharyngeal tonsil hyperplasia could be carried out by fiber optic nasopharyngoscopy and cephalometry, as these examinations were regularly correlated. However, it was found that cephalometry tended to underestimate the size of the pharyngeal tonsil relative to nasopharyngoscopy.
