Klebsiella pneumoniae as an opportunistic pathogen can cause nosocomial infection.  The main concern on this bacterium is directed on the extended-spectrum β-Lactamase (ESBL)-producing Klebsiella pneumoniae. The therapy of ESBL-producing Klebsiella pneumoniae infections is very limited because of its multidrug resistance. It had been found new local isolates Streptomyces sp-MWS1, Streptomyces sp-MWS3, and Streptomyces sp-MWS6 in mangrove East Coast of Surabaya. These isolates have potential to produce antibiotics (Retnowati, 2008). This study was aimed to prove these isolates may inhibit the growth of ESBL-producing Klebsiella pneumonia. The test of antibacterial activity of Streptomyces sp-MWS1, Streptomyces sp-MWS3, and Streptomyces sp-MWS6 using the modification Agar print method against clinical isolates of ESBL-producing Klebsiella pneumoniae. The diameter of inhibition zone (mm) formed shows activity of these isolates. The profiles of antibacterial activity of Streptomyces sp-MWS1, Streptomyces sp-MWS3, and Streptomyces sp-MWS6 were different in terms of periode required to reach peak activity, duration of activity and inhibition zone diameter produced during 10 days of observation. Streptomyces sp-MWS3 reached the peak activity most rapidly on day 3 with the largest inhibition zone diameter of 9 mm in ESBL-producing Klebsiella pneumoniae. There were significant differences in inhibition zone diameter between Streptomyces sp-MWS1, Streptomyces sp-MWS3, and Streptomyces sp-MWS6 against ESBL-producing Klebsiella pneumoniae.

ABSTRACT Red galangal rhizome is a plant that has various properties including antifungal and antibacterial. Red galangal rhizomes contain a class of flavonoids, phenols and terpenoids, which can be, used as basic ingredients of modern medicines. Flavonoids and phenols are also known to inhibit microbes. Flavonoids can inhibit microbes that have been resistant to antibiotics. This study aims to determine the antibacterial activity of red galangal rhizome extract (Alpinia purpurata K.Schum) and test its inhibitory effect on the growth of Klebsiella pneumoniae bacteria in sputum isolates in patients with pneumonia resistant ceftriaxone antibiotics. Red galangal rhizome plant was extracted by maceration method using 96% ethanol. Antibacterial inhibitory tests were carried out using the disc and well method. The results showed that the red galangal extract of rhizome had a inhibitory effect on the bacterium Klebsiella pneumoniae. On the disc method for concentrations of 100%, 75%, 50%, 25% and 12.5% the diameter of the inhibition zone is 9.6 mm, 8.68 mm, 8.5 mm, 8.5 mm and 7.6 mm. The inhibition ability of bacterial growth on the disc method is categorized as moderate because it has an average inhibition zone diameter of 5-10 mm. Whereas in the well method for concentrations of 100%, 75%, 50%, 25% and 12.5% the diameter of the inhibition zone was 25.8 mm, 20 mm, 15.6 mm, 12.7 mm and 9.7 mm, respectively. The inhibition ability of bacterial growth in the well method for 100% concentration is categorized as very strong because it has an average inhibition zone diameter of >20 mm, for concentrations of 75%, 50% and 25% it is categorized as strong because it has an average inhibition zone of 10- 20 mm and for a concentration of 12.5% it is categorized as moderate because it has an average inhibition zone diameter of 5-10 mm. Keywords: Red galangal rhizome, Alpinia purpurata K.Schum, Klebsiella pneumonia  ABSTRAK Rimpang Lengkuas merah merupakan tanaman yang memiliki berbagai khasiat di antaranya sebagai antijamur dan antibakteri. Rimpang Lengkuas merah mengandung golongan senyawa flavonoid, fenol dan terpenoid yang dapat digunakan sebagai bahan dasar obat-obatan moderen. Senyawa flavonoid dan fenol juga diketahui dapat menghambat mikroba. Flavonoid dapat menghambat mikroba yang telah resisten terhadap antibiotik. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri ekstrak rimpang Lengkuas merah (Alpinia purpurata K.Schum) dan menguji daya hambatnya terhadap pertumbuhan bakteri Klebsiella pneumoniae isolat sputum pada penderita pneumonia resisten antibiotik seftriakson. Tanaman rimpang Lengkuas merah di ekstraksi dengan metode maserasi menggunakan pelarut etanol 96%. Uji daya hambat antibakteri dilakukan dengan metode cakram dan sumuran. Hasil penelitian menunjukkan bahwa ekstrak rimpang Lengkuas merah memiliki daya hambat terhadap bakteri Klebsiella pneumoniae. Pada metode cakram untuk konsentrasi 100%, 75%, 50%, 25% dan 12,5% diameter zona hambatnya ialah 9,6 mm, 8,68 mm, 8,5 mm, 8,5 mm dan 7,6 mm. Abstract Background Detection of ceftazidime/avibactam (CAZ/AVI) antibacterial activity is absolutely vital with the rapid growth of carbapenem resistant Enterobacteriaceae (CRE). But now, there is no available automated antimicrobial susceptibility testing card for CAZ/AVI, so Kirby-Bauer has become an economical and practical method for detecting CAZ/AVI antibacterial activity against Enterobacteriaceae. Result In this study, antimicrobial susceptibility testing of CAZ/AVI against 386 Enterobacteriaceae (188 Klebsiella pneumoniae, 122 Escherichia coli, 76 Enterobacter cloacae) isolated from clinical patients was performed by broth microdilution. Of the 386 strains, 54 extended spectrum β lactamases negative (ESBL(−)), 104 extended spectrum β lactamases positive (ESBL(+)), 228 CRE. 287 isolates were susceptible to CAZ/AVI and 99 isolates were resistant to CAZ/AVI. At the same time, to obtain optimal content avibactam (AVI) disk containing ceftazidime (30 μg), inhibition zone diameter of four kinds of ceftazidime (30 μg) disk containing different AVI content (0 μg, 10 μg, 25 μg, 50 μg) were tested by Kirby-Bauer method. The microdilution broth method interpretation was used as the standard to estimate susceptible or resistance and then coherence analysis was carried out between Kirby-Bauer and broth microdilution. The result shows the inhibition zone diameter of 30 μg/50 μg disk, susceptible isolates: 20.5 mm–31.5 mm, resistance isolates: 8.25 mm–21.5 mm. The inhibition zone diameter of 30 μg/25 μg disk, susceptible isolates: 19.7 mm–31.3 mm, resistance isolates: 6.5 mm–19.2 mm. The inhibition zone diameter of 30 μg/10 μg disk, susceptible isolates: 19.5 mm–31 mm, resistance isolates: 6.5 mm–11 mm. The inhibition zone diameter of ceftazidime (30 μg), susceptible isolates: 6.5 mm–27.5 mm, resistance isolates 6.5 mm. Conclusion Our results show that 30 μg/50 μg, 30 μg/25 μg, 30 μg/10 μg CAZ/AVI disk have significant statistical differences to determinate CAZ/AVI antibacterial activity, but for 30 μg/50 μg disk, there has a cross section between susceptible isolates (minimum 20.5 mm) and resistance isolates (maximum 21.5 mm). For 30 μg/25 μg disk, it is hard to distinguish the difference between susceptible isolates (minimum 19.7 mm) and resistance isolates (maximum 19.2 mm), so 30 μg/10 μg CAZ/AVI disk is more conducive to determinate antibacterial activity.

ABSTRACTWhite galangal rhizome is a plant that has properties such as antifungal and antibacterial. White galangal rhizome contains of active compounds, namely flavonoid, phenol and terpenoid compounds which can inhibit microbes. This study aimed to determine the antibacterial activity of ethanol extracts of white galangal rhizome against Klebsiella pneumoniae urine isolate in patients with urinary tract infections. White galangal rhizome plants were extracted using 96% ethanol solvent. Antibacterial activity test was performed using the disc and wells method. The result showed that ethanol extracts of white galangal rhizome has antibacterial activity against the bacteria Klebsiella pneumoniae. In the disc method with concentrations of 80%, 60%, 40%, 20% and 10% the diameter of the inhibition zone formed are 9,8 mm; 9 mm; 7,8 mm; 7,9 mm and 7,7 mm. Inhibition of the disc method is categorized as medium because the diameter of the zone formed is only around an average of 5-10 mm. Whereas in the wells method for concentrations of 80%, 60%, 40%, 20% and 10% the diameter of the inhibition zone are 11,3 mm; 10,3 mm; 9,3 mm; 6,3 mm and 2,6 mm. The inhibitory ability at concentrations of 80% and 60% is categorized as strong because it has an average of 10-20 mm, for concentrations of 40% and 20% are categorized as medium, because it has an average inhibition zone diameter of 5-10 mm, and for concentration of 10% is categorized as weak because it has a inhibition diameter <5 mm.Keywords: White Galangal Rhizome (Alpinia galanga L. Pada metode cakram dengan konsentrasi 80%, 60%, 40%, 20% dan 10% diameter zona hambat yang terbentuk ialah 9,8 mm; 9 mm; 7,8 mm; 7,9 mm dan 7,7 mm. Penghambatan pada metode cakram dikategorikan sedang karena diameter zona yang terbentuk hanya berkisar pada rata-rata 5-10 mm. Sedangkan pada metode sumuran untuk konsentrasi 80%, 60%, 40%, 20% dan 10% diameter zona hambatnya ialah 11,3 mm; 10,3 mm; 9,3 mm; 6,3 mm dan 2,6 mm. Kemampuan penghambatan pada konsentrasi 80% dan 60% di kategorikan kuat karena memiliki diameter zona hambat rata-rata 10-20 mm, untuk konsentrasi 40% dan 20% di kategorikan sedang, karena memiliki diameter zona hambat rata-rata 5-10 mm, dan untuk konsentrasi 10% di kategorikan lemah karena memiliki diameter zona hambat <5 mm. Kata Kunci: Rimpang lengkuas putih (Alpinia galanga L. ABSTRACTObjective: The purpose of this study was to evaluate the antibacterial activity of chitosan, hydrolyzed coconut oil and their combination againstBacillus cereus and Escherichia coli.Methods: The materials used in this study were powder of chitosan (obtained from prawn shell produced by Laboratory of Research Centre FMIPAUniversity of Sumatera Utara) and virgin coconut oil (VCO) product of Siti Nurbaya-Indonesia. VCO was partially hydrolyzed by Lipozyme TL IM(active at sn-1,3 position) and the result called hydrolyzed virgin coconut oil (HVCO). The bacteria used in this study were B. cereus and E. coli. Theantibacterial activity of chitosan in 1% acetic acid and HVCO in dimethylsulfoxide was tested by Kirby–Bauer agar diffusion method using paper discwith diameter of 6 mm.Results: The results showed that the minimum inhibitory concentration of chitosan against B. cereus and E. coli is at concentration of 0.05% withinhibition zone diameter of 6.86 mm and 7.56 mm, respectively. MIC of HVCO against B. cereus is at concentration of 0.25% with inhibition zonediameter of 6.40 mm, and against E. coli is at a concentration of 0.50% with inhibition zone diameter of 6.20 mm. The inhibition zone diameter ofchitosan 0.05% and HVCO 0.25% in combination against B. cereus is 8.33 mm which is higher than half the sum of chitosan 0.05% and HVCO 0.25%(6.63 mm). The inhibition zone diameter of chitosan 0.05% and 0.5% HVCO in combination against E. coli is 8.53 mm which is higher than half thesum of chitosan 0.05% and HVCO 0.5% (6.53 mm).Conclusion: The findings of this study indicate that chitosan is more antibacterial than HVCO, and the interaction between chitosan and HVCO incombination demonstrated to be synergistic against B. cereus and E. coli.Keywords: Antibacterial, Chitosan, Coconut oil, Combination, Bacillus cereus, Escherichia coli.®

Patchouli oil obtained from patchouli plants (Pogostemon cablin Benth.) has been widely used as an antibacterial agent in liquid and solid soap preparation. In this research, patchouli oil was added to liquid soap formula which use sodium lauryl ether sulphate (SLES) surfactant. The purpose of this study was to determine the physical characteristics and antibacterial activity of patchouli oil liquid soaps based on SLES surfactant. The Liquid soaps were prepared into four formulas with variation of patchouli oil concentration that were F0 (0.00% w/w), F1 (0.05% w/w), F2 (0.50% w/w), and F3 (1.00% w/w). The liquid soaps were evaluated including organoleptic, microscopic property, physical stability by using centrifuge test, pH, foam height and stability, viscosity and rheology, and antibacterial activity against Staphylococcus aureus ATCC 25923 by using disc diffusion method and chloramphenicol as a positive control. The results showed that all liquid soap formulas had following characteristics: translucent except F3 slightly murky, homogen, stable after centrifuge test, pH between 6.55 – 6.66 that were no significantly different and met SNI standards of liquid soap, foam height between 2.67 – 3.23 cm that were no significantly different and stable after 5 minutes of testing, viscosity between 2237 – 2397 cPs relatively equal in all formulas with pseudoplastic rheological properties. The patchouli oil liquid soaps had antibacterial activity against Staphylococcus aureus ATCC 25923, where F0, F1, F2, F3 and positive control result in inhibition zone diameter of 11.26±2.60 mm, 14.60 ± 2.45mm, 15.51 ± 0.44 mm, 17.97 ± 0.71 mm and 19.00 ± 3.92 mm respectively. Based on the ANOVA one way test, the addition of 1% patchouli oil gave a significant effect on the inhibition zone diameter compared to F0 (without patchouli oil)

Racun ayam is one of the plants which has several activities in its fruits, one of which is an antibacterial. This study aims to determine the antibacterial activity of ethanol extract of racun ayam leaves. The extraction was done by maceration method using ethanol as a solvent. Antibacterial activity assay was carried out on S. aureus, E. coli, and P. aeruginosa bacteria by disk diffusion method at the concentrations of 80, 100, and 120 ppm. The results showed that the ethanol extract of B. javanica leaves had the greatest inhibition zone on E. coli with inhibition zone diameter values ​​at 80, 100, and 120 ppm were 11.7 mm, 14.5 mm and 19.67 mm respectively. Inhibition zones in S. aureus could only be seen at 100 and 120 ppm which had inhibition zone diameters at 1.17 mm and 2.17 mm respectively, whereas in P. aeruginosa they did not have any inhibitory zone values. The results of statistical analysis showed a significance of the inhibition zone diameter values ​​of E. coli bacteria starting at 80 ppm with a sig value of 0.034. Due to a sig value was <0.05, it was categorized as significantly different from the negative control. The bacteria of S. aureus which had ​​<0.05 for sig values only found on 100 ppm and 120 ppm. Bacteria P. aeruginosa which had sig value >0.05 was not significantly different at all concentrations

Food borne disease is a disease with symptoms such as nausea, diarrhea and vomiting. The main cause of FBD was food contamination of Staphylococcus aureus. Jarak pagar (Jatropha curcas Linn) leave is a plant that have antibacterial activity against S. aureus because it contains saponin, flavonoid, and tannin. This research aimed to determine the antibacterial activity of the ethanolic extract of Jatropha curcas Linn. leaves, formulated them as gel hand sanitizer, and evaluated the antibacterial activity of the formula. The sample was collected, determined, and extracted with ethanol 70% using maceration, and the phytochemical screening conducted to the concentrate extract. The antibacterial activity test conducted by well diffusion method with concetration 50, 100, dan 150 mg/mL. Then the effective extract group has formulated to gel hand sanitizer. The gel hand sanitizer formula tested the antibacterial activity and physical properties by organoleptic, pH, stability, and spreadablility. The result showed that the extract with concentration 50, 100, dan 150 mg/mL has inhibition zone diameter was 9.9, 15.7, and 16.4 mm, respectively. The gel hand sanitizer of Jatropha curcas Linn extract was formulated by using 100 mg/mL concentration of extract. The gel was physically stable and had inhibition zone diameter was 15.6 mm

Daun Kelor (Moringa oleifera Lam.) bermanfaat untuk menghambat aktivitas pertumbuhan bakteri karena adanya kandungan senyawa flavonoid , alkaloid, dan fenol. Penelitian ini bertujuan untuk menguji aktivitas antibakteri dari ekstrak etanol daun Kelor dan sediaan masker gel peel-off ekstrak etanol daun Kelor terhadap bakteri Staphylococcus aureus. Penelitian ini menggunakan metode eksperimental laboratorium, ekstrak etanol daun kelor dan sediaan masker gel peel-off ekstrak etanol daun Kelor dibuat dengan konsentrasi 1, 3, 5, 7, dan 9%. Ekstrak daun Kelor diperoleh dengan cara maserasi dengan menggunakan pelarut etanol 96%. Pada pengujian antibakteri menggunakan metode sumuran. Diameter zona hambat paling besar yang ditimbulkan oleh ekstrak etanol daun Kelor yaitu pada konsentrasi 7% dengan diameter zona hambat sebesar 5,75 mm, sedangkan pada sediaan masker gel peel-off tidak menunjukkan adanya daya hambat. Dapat disimpulkan bahwa ekstrak etanol daun Kelor memiliki aktivitas antibakteri yang termasuk dalam kategori sedang.Moringa oleifera Lam. This study uses laboratory experimental methods, Moringa leaf ethanol extract and gel mask peel-off preparation for Moringa leaf ethanol extract made with concentrations of 1, 3, 5, 7, and 9%. Moringa leaf extract was obtained by maceration using 96% ethanol solvent. In antibacterial testing using the well method. The biggest inhibition zone diameter caused by ethanol extract of Moringa leaves is at a concentration of 7% with a inhibition zone diameter of 5.75 mm, whereas the peel-off gel mask preparation did not show any inhibitory power. It can be concluded that the ethanol extract of Moringa leaves has antibacterial activity which is included in the medium category.

AbstractChinese petai plants contain alkaloids that have antibacterial abilities including Staphylococcus aureusATTC 25923 PK/5 bacteria. Plaster is a dosage form whose active ingredients need to be developed from natural ingredients, especially Chinese petai leaves. The purpose of this study was to optimize the plaster of Chinese petai leaf extract and antibacterial Staphylococcus aureusATTC 25923 PK/5and to determine the antibacterial activity of the extract plaster with various doses. The test method for antibacterial is the well method. The data obtained is the diameter of the inhibitory power of the extract in the plaster. Data analysis to determine antibacterial activity using spss. The results of the phytochemical test showed that the Chinese petai leaf extract contained active compounds, namely tannins, saponins, alkaloids, flavonoids and steroids. Chinese petai leaf extract was also able to inhibit Staphylococcus aureusATTC 25923 PK/5bacteria in the extract obtained an inhibition zone of 5.1 mm at a concentration of 25% and plaster obtained an inhibition zone of 3.46 mm, a concentration of 50% obtained an inhibitory zone diameter of 5.63 mm and at plaster obtained an inhibition zone diameter of 4.5 mm, a concentration of 75% obtained an extract inhibition zone diameter of 6.56 mm on plaster obtained an inhibition zone diameter of 5.53 mm, a concentration of 100% obtained an extract inhibition zone diameter of 7 mm and on plaster The diameter of the inhibition zone was 6.46 mm. Tujuan penelitian ini melakukan optimasi plester ekstrak daun petai cina dan antibakteri Staphylococcus aureusATTC 25923 PK/5dan mengetahui aktivitas antibakteri plester ekstrak dengan berbagai dosis. Metode uji terhadap antibakteri adalah metode sumuran. Data yang didapatkan adalah diameter daya hambat ekstrak dalam plester. Analisis data untuk mengetahui aktivitas antibakteri menggunakan ANOVA. Hasil uji fitokimia menunjukkan bahwa ekstrak daun petai cina memiliki kandungan senyawa aktif, yakni tanin, saponin, alkaloid, flavonoid dan steroid. Ekstrak daun petai cina juga mampu menghambat bakteri Staphylococcus aureusATTC 25923 PK/5pada ekstrak didapatkan zona hambat sebesar 5,1 mm pada konsentrasi 25% dan plester didapatkan zona hambat sebesar 3,46 mm, konsentrasi 50%didapatkan diameter zona hambat ekstrak sebesar 5,63 mm dan pada plester didapatkan diameter zona hambat sebesar 4,5 mm, konsentrasi 75% didapatkan diameter zona hambat ekstrak sebesar 6,56 mm pada plester didapatkan diameter zona hambat sebesar 5,53 mm, konsentrasi 100% didapatkan diameter zona hambat ekstrak sebesar 7 mm dan pada plester didapatkan diameter zona hambat sebesar 6,46 mm. Hal ini menunjukkan bahwa ekstrak daun petai cina memiliki potensi sebagai antibakteri terhadap pertumbuhan bakteri Staphylococcus aureusATTC 25923 PK/5.Kata kunci:Petai Cina, Ekstrak, Plester, Antibakteri

Introduction: Many medicinal plants produce antioxidants and antimicrobial properties which protect the host from cellular oxidation reactions and other pathogens highlighting the importance of search for natural antimicrobial drugs. Method : The antibacterial activity of 25, 50 and 100 mg/ml aqueous extract of garlic and onion on S.aureus, S. Typhi,P. aeruginosa and E. coli isolates were carried out using agar well diffusion method. The antibacterial activity of the aqueous plant extract were measured by zones of inhibition and the antibiotic sensitivity test of the bacteria isolates was done using the disc diffusion method. Results: Inhibition zone diameter of 18 mm, 19 mm, 21 mm and 27 mm were observed for E.coli, P.aeruginosa, S.aureus, and S.Typhi respectively using 100 mg/ml of garlic while for onion, 11.5 mm, 12 mm, 12 mm and 13 mm inhibition zone diameter were observed for the same bacteria isolates respectively. It was observed that the increase in concentration of garlic is needed for better antibacterial activity while increasing concentration does not have a significant effect on the antibacterial activity of onion. The results of the antibiotic tests showed that most of the tested organisms exhibited multiple drug resistance to the conventional antibiotics including cotrimoxazole, chloramphenicol, amoxillin, streptomycin and septrin. Conclusion: Garlic extract showed greater antimicrobial potential compared with onion extract and the widely used antibiotics. Thus, aqueous garlic extract may be more effective in the treatment of bacterial infections in order to overcomethe problem of multidrug resistance of bacterial pathogens.
