Vitamin E as an inhibitor of oxidative damage to goose meat storage

The vitamin E effect on the lipid peroxidation product content and the endogenous antioxidant activity (at -18 C during the different types of storage) has been studied in goose meat. The goose meat of three samples has been used for storage. Meat of control sample has been obtained from geese fed by the standard diet. The 1st meat test sample of geese differs from the control group by two times higher content of vitamin E (40 mg / kg) in their diet from the 42nd to the 63rd day. Meat of the 2nd test sample obtained from the control group of geese is processed by a vitamin E solution (calculated at 100 mcg per g of meat) immediately before storage. The shelf life of meat is 210 days. It has been established that the intensive accumulation of the secondary lipid peroxidation products begins from the 90th day in the goose meat during its storage. In the goose diet a double increase of the vitamin E has contributed a significant (by 27.6%, p ≤ 0.05) TBA-AP decrease in the first meat test sample in comparison with the control group at the end of the experiment. The addition of vitamin E to the diet of geese has contributed the stabilization of the antioxidant pool in their meat. It has been confirmed by a 1.88-fold lower level of TBA-AP upon initiation of peroxide oxidation of Fe2+ and by the higher coefficient of antioxidant activity (36.0%, p ≤ 0.05) in this sample in comparison with the control on the 210th day. At the end of the experiment, the vitamin E content is higher 41.7 % (p ≤ 0.01) in the first sample than in the control, β-carotene - 15.0 % (p ≤ 0.05), and vitamin A is at the level of the control sample. Processed goose meat with a solution of vitamin E also provides reliable inhibition of peroxidation processes during the first half of the experiment. However, at the end of the experiment the content of TBA-AP reaches the level of the corresponding control indices in the 2nd test sample of meat. From the 120th day, there has been the more intensive use of endogenous antioxidants. The antioxidant activity coefficient decreasing to the control level on the 210th day in this meat sample is its conformation. The meat of this sample differs by a higher content of β-carotene from the control sample significantly (by 13.5%, p ≤ 0.05). Thus, to obtain a prolonged antioxidant effect during low-temperature storage of meat it is more advisable to add vitamin E to the diet of geese in the pre-slaughter period. Background: Preeclampsia is the most common and major medical complication of pregnancy with a high incidence of maternal and foetal morbidity and mortality. During pregnancy abnormally increased lipid peroxidation and free radical formation as well as significantly decreased antioxidants production in maternal blood may lead to pathogenesis of preeclampsia. So, we designed this study as little information is known about lipid peroxidation and antioxidant level in preeclampsia.Objectives: To assess the serum malondialdehyde (MDA) level as a lipid peroxidation product and vitamin E (antioxidant) level in women with preeclampsia as well as in normal pregnancy and to compare the values.Materials and Methods: The study was conducted on 60 women aged from 25 to 35 years in the department of Biochemistry, Budi Kemuliaan Maternity Hospital (BKMH) in Jakarta during the period April to July 2004. Twenty were normal pregnant women and 20 were preeclamptic patients. For comparison age matched 20 apparently healthy nonpregnant women were included in the study. The study subjects were selected from outpatient department (OPD) of Obstetrics and Gynaecology of BKMH in Jakarta. Serum MDA (lipid peroxidation product) level was measured by thiobarbituric acid reactive substances assay (TBRAS) method and vitamin E was estimated spectroflurometrically. Data were analyzed by unpaired Student’s t test between the groups by using SPSS version 12.Results: The mean serum MDA levels were significantly higher in normal pregnancy and also in preeclampsia than that of nonpregnant control group women (p<0.001). Again the serum MDA levels were significantly higher in preeclampsia than that of normal pregnant women (p<0.001). The serum vitamin E levels were significantly lower in preeclampsia and also in normal pregnancy than that of nonpregnant control women (p<0.001). Moreover, the serum vitamin E levels were significantly lower in preeclampsia compared to that of normal pregnancy which was also statistically significant (p<0.001)Conclusion: As in preeclamptic group level of oxidation product is high and antioxidant level is low, it can be assumed that in preeclampsia, serum antioxidants are excessively utilized to counteract the cellular damage mediated by free radicals production and deficient antioxidants as well as increased lipid peroxidations may be the important factors in the pathogenesis of preeclampsia. Introduction. Nowadays, it has been shown that one of the possible ways to increase the effectiveness of the treatment of malignant tumors is the use of combined treatment methods. Aim. To study the intensity of lipid peroxidation (LPO) and the activity of superoxide dismutase (SOD) in hyperthermia against the background of bee venom in the blood of tumor-bearing rats. Materials and methods. The experimental animals were divided into 5 groups: 1st — intact; 2nd — control (tumor-bearing animals (PS-1 tumor strain) with intraperitoneal (IP) administration of saline solution); 3rd, 4th and 5th groups — tumor-bearing animals with IP administration of 0.5 ml of bee venom against the background of hyperthermia 42, 43 and 44°C respectively. The content of diene and triene conjugates, Schiff bases (SB) and SOD activity in the blood of animals was determined. Results. From the 1st day after the end of the experiment, a statistically significant decrease in triene conjugates was recorded in all experimental groups compared to the control group. SB decreased on the 1st–7th day after the end of the experiment with the action of hyperthermia 42°C and bee venom, on the 7th–28th day — with the action of hyperthermia 43°C and bee venom, and on the 28th day — with the action of bee venom and hyperthermia 44°C, which was accompanied by an increase in SOD activity from the 7th day in all experimental groups compared to the control group. Conclusion. Hyperthermia in combination with the action of bee venom causes a decrease in lipid peroxidation products and an increase in antioxidant activity in the blood of tumor-bearing rats. The most effective action, in our opinion, is the use of bee venom against the background of hyperthermia of 43°C, at which a prolonged effect is recorded both with respect to a decrease in the concentration of SB and an increase in the activity of SOD.

The article consist data about effect of different levels of vitamin E and selenium in the diet of carps during their growing and also informationabout the influence on processes of lipid peroxidation and activity of antioxidant protection in their body.The experiment conducted in three experimental ponds. After wintering there were placed four ears mature females and males carp, six individuals in each group. Supplements of vitamin E administered at a rate of 25 mg/kg and drug «Sel–Plex» the rate of selenium – 0.3 mg/kg were added to further basic diet(BD) of female and male carp first experimental group (EG1). The second experimental group (EG2) received (BD) and vitamin E supplements in an amount of 75 mg / kg, and just as in EG1 – selenium – 0.3 mg/kg of feed.The control group received fish feed without additives vitamins and minerals.Feeding lasted for 30 days, due to the physiological needs of the fish feed. After spawning in fish of research and control group swere taken blood samples for biochemical research.The study led to a dose–dependent decrease (p < 0.01 – 0.001) content of TBA–active products and hydroperoxidase of lipids, but did not significantly effect onsuperoxide dismutase and glutathione peroxidase activity of blood So, the reduction of lipid peroxidation products in the carp’s blood of experimental groups probably was caused by the growth of non–enzymatic level, which is associated with fat–soluble vitamins.

Hepatotoxic chemicals damage liver cells primarily by producing reactive oxygen species. The decoction of the leaves ofTamarindus indicaL. is used for liver disorders. In this work we evaluated the hepatoprotective activity of a tablet formulation of this plant. Thirty-five Sprague Dawley rats were randomly divided into five groups (n=7). First group (I) is control group, fed with standard diet. Groups II to V (hepatotoxic groups) were subjected to a subcutaneous injection of CCl4(0.5 mL/kg). Group II was negative control, fed with standard diet; group III was subjected to administration of Silymarin 150 mg/kg and groups IV and V were treated with tablets in dose of 100 mg/kg and 200 mg/kg, respectively. Lipid peroxidation and the activity of superoxide dismutase, catalase, and reduced glutathione were evaluated. Serum levels of alanine aminotransferase, aspartate aminotransferase, gamma-glutamine transferase, alkaline phosphatase, and a lipid profile were evaluated too. The tablets inhibit lipid peroxidation. The redox balance (SOD-CAT-GSH) remains normal in the experimental groups treated with tablets. The liver function using dose of 200 mg/kg of tablets was better than the other experimental groups. These results justify, scientifically, the ethnobotanical use of the leaves ofTamarindus indicaL.

Teucrium poliumL. (Lamiaceae) (RDC 1117) is a medicinal plant whose species have been used for over 2000 years in traditional medicine due to its diuretic, diaphoretic, tonic, antipyretic, antispasmodic and cholagogic properties. The therapeutic benefit of medicinal plants is often attributed to their antioxidant properties. We previously reported that an aqueous extract of the leaves and stems of this plant could inhibit iron-induced lipid peroxidation in rat liver homogenate at concentrations that were not toxic to cultured hepatic cells. Others have reported that organic extracts of the aerial components of this plant could inhibit oxidative processes. Against this background, we felt further investigation on the antioxidant action of the extract ofT. poliumprepared according to traditional Arab medicine was warranted. Accordingly, we assessed (i) its ability to inhibit (a) oxidation of β-carotene, (b) 2,2′-azobis(2-amidinopropan) dihydrochloride (AAPH)-induced plasma oxidation and (c) iron-induced lipid peroxidation in rat liver homogenates; (ii) to scavenge the superoxide ($${\hbox{ O }}_{2}^{\bullet -}$$) radical and the hydroxyl radical (OH•); (iii) its effects on the enzyme xanthine oxidase activity; (iv) its capacity to bind iron; and (v) its effect on cell glutathione (GSH) homeostasis in cultured Hep G2 cells. We found that the extract (i) inhibited (a) oxidation of β-carotene, (b) AAPH-induced plasma oxidation (c) Fe2+-induced lipid peroxidation in rat liver homogenates (IC50 = 7 ± 2 μg ml−1); (ii) scavenged $${\hbox{ O }}_{2}^{\bullet -}$$(IC50 = 12 ± 3 μg ml−1) and OH• (IC50 = 66 ± 20 μg ml−1); (iii) binds iron (IC50 = 79 ± 17 μg ml−1); and (iv) tended to increase intracellular GSH levels resulting in a decrease in the GSSG/GSH ratio. These results demonstrate that the extract prepared from theT. poliumpossesses antioxidant activityin vitro. Further investigations are needed to verify whether this antioxidant effect occursin vivo.

Effects of GSE and vitamins C and E on aspirin- and ethanol-induced gastric ulcer and associated increases of lipid peroxidation in rats were compared. Two experiments were conducted. Rats were randomized into eight groups: a negative control and seven groups that received aspirin or ethanol for ulcer induction: one positive control (vehicle) and six with VC, VE, or GSE (25 and 250 mg/kg). Ulcer indexes and gastric levels of malondialdehyde (MDA) were quantified. VC, VE, and GSE (25 and 250 mg/kg) decreased aspirin, and ethanol-induced ulcers and MDA values compared with positive control group. The magnitude of aspirin ulcer reduction was comparable for all treatments, and MDA decrease with GSE was higher than with VC and tended to be greater, albeit none significantly, than with VE. GSE was more effective than VC and VE for lowering the ethanol ulcers, while the decrease of MDA levels with GSE was greater than with VC, but comparable to that achieved with VE. GSE protected against ethanol-induced gastric ulcers more effectively than VC or VE, while its protection against aspirin ulcers was comparable for all treatments. GSE produced the greatest reductions of gastric MDA in both models.

Evaluation of the phenolic compounds and antioxidant activity ofCedrela odorataL. andJuglans regiaL. bark extracts was performedin vitro.Juglans regiashowed greater extract concentration and higher antioxidant activity. Hypoglycemic activity in rats was assessed by generating a glucose tolerance curve and determining the area under the curve (AUC). Diabetes was later induced by an injection with streptozotocin (65 mg/kg of b.w.) and confirmed after 24 hours. The extract was administered (200 mg/kg b.w.) over 10 days, and blood glucose was monitored and compared with a control group. The glucose AUC showed a hypoglycemic effect ofJ. regiaandC. odoratain normal rats. Both extracts reduced hepatic lipid peroxidation in diabetic rats. Polyphenolic extracts reduced cholesterol levels in a hypercholesterolemic mouse model and decreased hepatic lipid peroxidation. Polyphenolic extract doses of 100 and 200 mg/kg b.w. were administered alone or with cyclophosphamide (CPA) 50 mg/kg ip, which was used as a positive control. Analyses were performed using leukocytes in a comet assay after 4 and 24 h of treatment. Genotoxic effects were evaluated by the comet assay, which showed that whileJ. regiaextract had no effect,C. odorataextract induced slight damage at 200 mg/kg, with the formation of type 0 and 1 comets.

This study investigated the in-vitro antioxidant activity of ethanol leaf extract of Justicia carnea and its effect on antioxidant status of alloxan-induced diabetic albino rats. The in-vitro antioxidant activity was assayed by determining the total phenol, flavonoids, ascorbic acid, β-carotene and lycopene contents and by using 2,2 diphenyl-1-picrylhydrazyl (DPPH) radical, reducing antioxidant power and inhibition of lipid peroxidation antioxidant systems. Oxidative stress was produced in rats by single intraperitoneal injection of 150 mg/kg alloxan and serum concentration of malonaldehyde (MDA), superoxide dismutase (SOD) and catalase (CAT) were determined. Five experimental groups of rats (n=6) were used for the study. Two groups of diabetic rats received oral daily doses of 100 and 200 mg/kg Justicia carnea leaf extract respectively while gilbenclamide (5 mg/ml); a standard diabetic drug was also given to a specific group for 14 days. From the result, the leaf extract contained a higher concentration of flavonoids followed byphenols, ascorbic acid, lycopene and β-carotene. The extract displayed more potent reducing power ability with EC50 of 40 µg/ml compared to BHA (EC50 of 400µg/ml). The percentage DPPH radical scavenging activity of the extract was also higher with EC50 of 200µg/ml and increased with increase in concentration while BHA had EC50of 320µg/ml. The inhibition of lipid peroxidation also increased with increase in concentration with EC50 of 58µg/ml and comparable with BHA (EC50=60µg/ml). The effect of the plant extract on antioxidant enzyme activities was concentration-dependent. Administration of 100mg/kg of the plant extract resulted in a significant decrease (p<0.05) in serum MDA concentration, while 200 mg/kg of the extract caused a significant (p˂0.05) increase in superoxide dismutase (SOD) and catalase activities with a non-significant increase (p>0.05) in the serum level of MDA when compared with the diabetic untreated group. These findings suggest that ethanol leaf extract of Justicia carnea have antioxidant properties and could handle diabetes-induced oxidative stress.
