In this study, we investigated the mineral composition, in vitro antidiabetic activity, and antibacterial potential of the aerial part of Ajuga iva. The mineral content of the plant powder was quantified using Inductively Coupled Plasma Atomic Emission Spectrometry. Then, three aqueous extracts were prepared: decocted, infused, and macerated and five organic: methanolic, ethyl acetate, chloroformic, and petroleum ether obtained by Soxhlet, and methanolic macerated extract. The in vitro hypoglycemic effect of the extracts was evaluated on α-amylase, α-glucosidase, and β-galactosidase, then the antibacterial activity was studied by the disk diffusion method, the minimum inhibitory concentrations (MICs) and bactericidal concentrations (MBCs) against six pathogenic bacteria, and finally, the correlation between the chemical composition and the inhibitory activities of different extracts on three enzymes was evaluated using Principal Component Analysis (PCA). The results obtained revealed that iron (112.00 mg/l) is the main mineral element, followed by potassium (44.071 mg/l) and sodium (16.572 mg/l). The results of in vitro antidiabetic activity showed that ethyl acetate extract presented the highest inhibitory activities against α-amylase and α-glucosidase with IC50 equal to 1475.67±94.21µg/ml and 140.50±32.25µg/ml respectively, while methanolic macerated extract showed high inhibitory activity against β-galactosidase. The results of the PCA analysis showed that the in vitro antidiabetic activity shows a linear relationship with the content of polyphenols, flavonoids, and tannins in Ajuga iva extracts. The study of the antibacterial effect showed that the five organic extracts have an inhibitory effect towards the microorganisms tested with zones of inhibition vary between 7 and 14 mm, except for Staphylococcus aureus that presented resistance to the five extracts. Furthermore, the MICs obtained range from 1.56 to 50 mg/ml, and the MBCs vary from 50 to 200 mg/ml. These results indicate that polar organic extracts of Ajuga iva have significant antidiabetic and antibacterial activities.

Objectives: In order to investigate alternate therapeutic option, this study was carried out to assess the in vitro antibacterial activity of gel extract of Aloe barbadensis against multiple antibiotic resistant Pseudomonas aeruginosa isolated from wound specimens. Methods: A total of 180 different wound specimens collected in a hospital, were subjected to isolate and identify P. aeruginosa by cultural methods. Antibiotic susceptibility testing was done by Kirby- Bauer disc diffusion method to screen multidrug resistant isolates. A. barbadensis extracts were prepared using aqueous and organic solvents and their in vitro inhibitory action was evaluated by agar well diffusion methods. Results: Out of total, 38 (21.1%) of the wound specimens showed the occurrence of P. aeruginosa, among which 15 (39%) isolates were multi-drug resistant. Organic extracts of various concentrations (0.2 - 0.8 v/v %) inhibited 66.7% of MDR and all non-MDR (n = 23) P. aeruginosa with zone of inhibition ranging from 9.5 ±1.0 to 21.3 ± 2.2 mm but not by aqueous extract. A positive Pearson’s correlation (r=0.983) was found between antibacterial effect and concentrations of the extracts. The antibacterial activity of organic extracts was statistically associated with antibiotic resistance profile of the organism (p<0.05). Conclusion: Organic extracts of A. barbadensis revealed variable in vitro inhibitory action against both MDR and non-MDR P. aeruginosa isolated from wound specimens. Although further confirmation is needed, aloe gel extract may be applied as an alternate treatment option.

In this study, the antibacterial activity of various organic solvent extracts of Ocimum sanctum L leaves, flowers and shoots was determined in vitro using agar diffusion method and MIC tested against the isolated soil bacteria. The identified bacteria were Paenibacillussp. L32, Paenibacillussp. BF38, Bacillus megaterium, Terribacillussp. 3LF, Bacillus simplex and Bacillus cereus. Various organic extracts of Ocimum sanctum L revealed a good antibacterial activity against about all bacteria. Results showed that the best extract was methanol because highest inhibition zone were obtained by this extract and all studied bacteria were inhibited. At a concentration of 500 µg/disc, the highest microbial inhibition was found 19.2±.76 mm against Bacillus simplex for methanol extract, 15.7±0.58 mm on Terribacillus sp. 3LF for ethanol extract of flower, 14.0±2.0 mm on Terribacillus sp. 3LF for ethanol extract of leaves, 13.34±0.58 mm on Bacillus megaterium for n-hexane extract and 11.7±0.6 mm on Terribacillus sp. 3LF for chloroform extract, respectively and MIC (64, 128, 256 and 512 µg/ml, respectively). Most of the cases, antibacterial activity with commercial antibiotics such as amoxicillin and erythromycin, organic extracts exhibited similar or higher antibacterial activity than standard drug. The results of this study suggest that the organic extracts of Ocimum sanctum L leaves can be a source of natural antimicrobial agents with potential applications.

The threat of antibiotic-resistance calls for novel antibacterial agents. This study was aimed at screening medicinal plants for their antibacterial properties, phytochemical content and safety. Leaves of Allophylus abyssinicus (Hochst.) Radlk., Dicliptera laxata C.B.Clarke, Ligustrum vulgare L., Solanecio gigas (Vatke) c. Jeffrey and Gymnanthemum myrianthum (Hook.f.) H.Rob.; leaf and stem-bark of Olinia rochetiana A. Juss. and the seed of Cucurbita pepo L. were used. Chloroform and ethanol were used to extract G. myrianthum, D. laxata and O. rochetiana; ethyl acetate and methanol for the rest, and water for all. The extracts were tested against clinical/standard strains of Escherichia coli, Pseudomonas aeruginosa, Salmonella typhi and Staphylococcus aureus by the agar-diffusion method. The minimum inhibitory concentrations (MIC) and minimum bactericidal concentrations (MBC) were determined. Acute toxicity to mice was checked and preliminary phytochemical screening was done. Thirteen extracts, out of 24, were active (inhibition zone >7 mm) at differing levels (9.67±0.33-25.66±0.57 mm) against at least one bacterial strain. The MICs and MBCs were 1.95-15.6 mg/mL and 7.8-125 mg/mL respectively. The aqueous extract of S. gigas, methanol extracts of L. vulgare and A. abyssinicus, and ethanol extract of O. rochetiana leaf were the most active (MIC 1.95mg/ml) against S. aureus. Ethyl acetate extracts of A. abyssinicus, L. vulgare and S. gigas; aqueous of C. pepo, O. rochetiana and G. myrianthum; and all D. laxata had no antibacterial activity. P. aeruginosa was the least susceptible to any extract, although the methanol and aqueous extracts of S. gigas performed better against it. Preliminary phytochemical screening of selected extracts for phenols, flavonoids, tannins, steroids, terpenoids, steroidal glycosides, alkaloids, saponins, resins and glycosides showed positivity at least for four of these phytochemicals with glycoside and terpenoids in nearly all extracts and resin in none. The plants were not toxic to mice at 2000 mg/kg. Further consideration of S. gigas, L. vulgare, A. abyssinicus and O. rochetiana is recommended in light of their promising potential and safety.

Objective: In folk medicines, different herbs and plants have been used for many thousands of years .Now it is important to investigate these plants and herbs scientifically which have been used in traditional medicines. The aim of this study was to investigate the antibacterial activity and preliminary phytochemical screening of root of Ipomea turpethum extracted in methanol and its fractions.Methods: The methanol extract was further extracted with three solvents ethyl acetate, chloroform and hexane and analyzed for their antibacterial activity using by agar well diffusion method. They were tested against six bacteria; Echrichia coli, Staphylococcus aureus, Staphylococcus epidermidus, Proteus vulgaris, Pseudomonas auroginosa and Salmonella typhi. The susceptibility of microorganisms to all three fractions was compared with each other and with standard antibotic (Ampicillin). The fractions of Ipomea turpethum was also qualitatively analyzed for the presence of chemical components, i.e. saponins, alkaloids, flavonoids, tannins and glycosides.Result: Among all fractions methanol exhibited highest antibacterial activity (average zone of inhibition 23.53mm ± 1.3) while ethyl acetate exhibited least antibacterial activity (average zone of inhibition 18.50mm ± 3.5). Minimum inhibitory concentration of methanol, ethyl acetate, chloroform and hexane fractions was found in the range of 650ug/ml to 2500ugl/ml against microorganisms.Conclusion: Results obtained from this preliminary in-vitro experiment indicate that, all three fractions of Ipomea turpethum has good antibacterial activity against all microorganisms used. By phytochemical analysis of extract, it has been found to contain some nutrient and chemical components which support its ethenomedicinal use but further work is required for development of new antibiotic compounds.Bangladesh Journal of Medical Science Vol.17(1) 2018 p.93-97

Albizia coriaria Welw ex. Oliver is a customary African medicinal plant, which has a long history of utilization in the management of oxidative stress-induced and bacterial diseases. However, there is no report on the phytochemicals, antioxidant, and antibacterial activities of its leaves. The aim of this study was therefore to compare the phytochemicals, antioxidant, and antibacterial potential of A. coriaria leaves from Jinja, Kole, and Mbarara districts of Uganda. Shade-dried leaf samples were ground into powder and successively extracted with ethyl acetate, ethanol, and distilled water. Phytochemical screening indicated the presence of alkaloids, phenols, saponins, flavonoids, cardiac glycosides, tannins, and terpenes as the major secondary metabolites in the extracts. Total phenolic and flavonoid contents and total in vitro antioxidant activity were found to be the highest for ethanolic extracts, with the highest contents (101.72 ± 0.22 mg GAE/g DW; 13.23 ± 0.03 mg QE/g DW) and antioxidant potential (IC50 = 18.65 ± 0.06 mg/mL) being for leaves from Mbarara district. Antibacterial activity of the extracts determined by agar disc diffusion method revealed that ethanolic extracts had higher antibacterial activities with mean zones of inhibition of 6.00 ± 1.73 to 10.00 ± 1.73 mm, 5.00 ± 1.00 to 12.30 ± 1.53 mm, 17.00 ± 0.00 to 25.00 ± 2.65 mm, and 9.00 ± 1.73 to 16.00 ± 1.73 mm for Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, and Salmonella typhi, respectively. Ethyl acetate extracts of A. coriaria leaves from Kole and Mbarara had lower antibacterial activities, while aqueous extracts and ethyl acetate extract of leaves from Jinja showed no antibacterial activity. The current study for the first time established that A. coriaria leaves possess therapeutic phytochemicals with significant in vitro antioxidant and antibacterial activities, which lend credence to their use in traditional management of oxidative stress-induced conditions and bacterial diseases in Uganda. Structural elucidation of the responsible pure compounds for the observed bioactivities as well as toxicity studies of the extracts is recommended.

Aim: This study aimed to evaluate the antibacterial and antitubercular activities of ethylacetate and ethanol leaf extracts of Senna occidentalis. Study Design: Fresh leaves of Senna occidentalis collected from Suleja, Niger state were used for this study against some medically important micro-organisms viz; Staphylococcus aureus, Escherichia coli, Bacillus subtilis, Pseudomonas aeruginosa, Salmonella paratyphi, Klebsiella pneumoniae, Mycobacterium bovis and Mycobacterium smegmatis. Place and Duration of Study: The study was conducted in Abuja, Nigeria at the Department of Microbiology and Biotechnology, National Institute for Pharmaceutical Research and Development [NIPRD], from February 2019 to April 2019. Methodology: Senna occidentalis leaves were extracted successively with ethyl-acetate and ethanol. The obtained extracts were tested in vitro for antibacterial activity by agar well diffusion method, while anti-tubercular screening was carried out by broth micro-dilution method. A fixed-dose concentration of chloramphenicol was used as a control drug against the bacterial isolates while isoniazid was used as control drug against the mycobacterium isolates. Results: The in vitro antibacterial screening showed that the crude extracts exhibited varying activity against the different microbes with highest zone of inhibition at 12 mm, and anti-tubercular activity with MICs ranging from 97.6-390.6 μg/mL.  Among these extracts, ethyl-acetate extract showed significant antibacterial activity against most of the test micro-organisms. The most susceptible micro-organism was P. aeruginosa (12mm zone in ethyl-acetate at 80 mg/mL) followed by B. subtilis (10 mm zone in ethyl-acetate extract at 80 mg/mL) and E. coli (9 mm zone in ethyl-acetate extract at 80 mg/mL). The ethanol extract was the most effective in inhibiting the growth of M. smegmatis and M. bovis with MICs of 97.6 μg/mL and 195.3 μg/mL. Conclusion: The activities observed could be attributed to the presence of some active metabolites contained in the extracts which could be useful in drug development for therapeutic purposes.

Objective: The objective of this study was to evaluate the antibacterial activity of phytochemical extracts and endophytic fungi of Carapa guianensis against Enterococcus faecalis. Carapa guianensis leaves and stems were collected to obtain phytochemical extracts and fungal metabolites and evaluated for in vitro antibacterial activity against E. faecalis using the disc diffusion method and dentin blocks with bacterial biofilm. Methods: Thirty dentin blocks were prepared and contaminated for 60 days with E. faecalis. The specimens were randomly divided into 6 experimental groups according to the test solution used: G1 – hexane stem extract of Carapa guianensis; G2 – methanol stem extract of Carapa guianensis; G3 – methanol leaf extract of Carapa guianensis; G4 – ethyl acetate extract of the endophytic fungus Penicillium isolated from Carapa guianensis; G5 – negative control, with no addition of bacterial inoculum; G6 – positive control. Results: Bacterial growth was analyzed by spectrophotometry after 14 days of direct contact between the extracts and dentin blocks. The hexane-stem, methanol-stem, methanol-leaf, and ethyl-acetate endophytic fungus Penicillium extracts inhibited bacterial growth in 100% of the samples. Conclusion: The present study demonstrated the antibacterial potential of phytochemical extracts and endophytic fungi of Carapa guianensis against E. faecalis.

AbstractStaphylococcus aureus and coagulase-negative staphylococci are among the major causes of mastitis in sheep. The main goal of this research was to determine the in vitro antibacterial activity of several essential oils (EOs, n 30), then five of them were chosen and tested alone and in blends against staphylococci isolates. Five bacteria were isolated from episodes of ovine mastitis (two S. aureus and three S. xylosus). Biochemical and molecular methods were employed to identify the isolates and disk diffusion method was performed to determine their antimicrobial-resistance profile. The relative percentage of the main constituents in the tested essential oils and their blends was detected by GC-EIMS analysis. Antibacterial and bactericidal effectiveness of essential oils and blends were evaluated through minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC). All of them showed sensitivity to the used antimicrobials. The EOs with the highest antibacterial activity were those belonging to the Lamiaceae family characterized by high concentrations of thymol, carvacrol and its precursor p-cymene, together with cinnamon EO, rich in cinnamaldehyde. In terms of both MIC and MBC values, the blend composed by Thymus capitatus EO 40%, Cinnamomum zeylanicum EO 20%, Thymus serpyllum EO 20% and Satureja montana EO 20% was found to be the most effective against all the isolates. Some essential oils appear to represent, at least in vitro, a valid tool against ovine mastitis pathogens. Some blends showed a remarkable effectiveness than the single oils, highlighting a synergistic effect in relation to the phytocomplex.

Modifying the composition of dental restorative materials with antimicrobial agents might induce their antibacterial potential against cariogenic bacteria, e.g., S.mutans and L.acidophilus, as well as antifungal effect on C.albicans that are major oral pathogens. Essential oils (EOs) are widely known for antimicrobial activity and are successfully used in dental industry. The study aimed at evaluating antibacterial and antifungal activity of EOs and composite resin material (CR) modified with EO against oral pathogens. Ten EOs (i.e., anise, cinnamon, citronella, clove, geranium, lavender, limette, mint, rosemary thyme) were tested using agar diffusion method. Cinnamon and thyme EOs showed significantly highest antibacterial activity against S.mutans and L.acidophilus among all tested EOs. Anise and limette EOs showed no antibacterial activity against S.mutans. All tested EOs exhibited antifungal activity against C.albicans, whereas cinnamon EO showed significantly highest and limette EO significantly lowest activity. Next, 1, 2 or 5 µL of cinnamon EO was introduced into 2 g of CR and microbiologically tested. The modified CR showed higher antimicrobial activity in comparison to unmodified one. CR containing 2 µL of EO showed the best antimicrobial properties against S.mutans and C.albicans, while CR modified with 1 µL of EO showed the best antimicrobial properties against L.acidophilus.

Studies were conducted to identify Pseudomonas fluorescens isolates from a collection of bacteria isolated from bacterial haemorrhagic septicaemia infected carp and catfish, evaluate their antibiotic sensitivity pattern and screen the antibacterial activity of some medicinal plant extracts against the isolates.. A total of 10 isolates were identified as P. fluorescens by morphological, physiological and biochemical tests. In vitro antibiotic sensitivity test of the P. fluorescens isolates were conducted by disc diffusion method for seven antibiotics where, all of the isolates were found to be sensitive only against streptomycin and gentamycin but, most of the isolates (80%) were found resistant to chloramphenicol (C). Moreover, eighty percent of the isolates showed resistance to multiple antibiotics. A total of 118 plant extracts were screened for their antibacterial activity against the P. fluorescens isolates where the isolates exhibited sensitivity to 30 samples. Leaf extracts of Tamarindus indicus, Terminalia chebula, Citrus aurantifolia, Eugenia caryophyllata and Spondias pinnata were found to inhibit the growth of all of the P. fluorescens isolates.
