Pleurotus florida (Oyster mushroom) belongs to the family Pleurotaceae and widely consumed all over the world. The aqueous ethanolic extract of the fruiting body of P.florida was evaluated for hypolipidemic activity. Hyperlipidemia was induced with the help of Triton WR 1339 (100mg/Kg b.w.) administered via intraperitoneal injection. Atorvastatin (2.5 mg/Kg b.w) was used as the standard drug. Different concentrations of aqueous ethanolic extract of P. florida (500, 250, and 100 mg/Kg b.w) were given orally before triton administration. The serum lipid profile was assayed and showed significant hypolipidemic activity compared to the control group (Triton alone). The activity of HMG CoA reductase was assayed using hydroxylamine hydrochloride. Hepatic HMG CoA reductase activity was significantly decreased in the treated group as compared to the control. The inhibition of lipid peroxidation was also assayed. A significant reduction in lipid peroxidation was seen in groups treated with extract compared to control. Lovastatin was also isolated from fruiting bodies and culture filtrate and screened using Thin Layer Chromatography. Lovastatin (sigma) was used as the standard. The finding suggests the significant hypolipidemic activity of the aqueous ethanolic extract of P. florida.

The present study aimed to determine the effect of an ethyl acetate extract of Mikania micrantha stems (EAMMS) in hypercholesterolemia-induced rats. Rats were divided into a normal group (NC) and hypercholesterolemia induced groups: hypercholesterolemia control group (PC), simvastatin group (SV) (10 mg/kg) and EAMMS extract groups at different dosages of 50, 100 and 200 mg/kg, respectively. Blood serum and tissues were collected for haematological, biochemical, histopathological, and enzyme analysis. Total cholesterol (TC), triglycerides (TG), low-density lipoprotein cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL-C), aspartate aminotransferase (AST), alanine aminotransferase (ALT), urea, creatinine, malondialdehyde (MDA) level, as well as enzymes of HMG-CoA reductase (HMGCR) and acetyl-CoA acetyltransferase 2 (ACAT2), were measured. Feeding rats with high cholesterol diet for eight weeks resulted in a significantly (p < 0.05) increased of TC, TG, LDL-C, AST, ALT and MDA levels. Meanwhile, the administration of EAMMS extract (50, 100 and 200 mg/kg) and simvastatin (10 mg/kg) significantly reduced (p < 0.05) the levels of TC, TG, LDL-C and MDA compared to rats in the PC group. Furthermore, all EAMMS and SV-treated groups showed a higher HDL-C level compared to both NC and PC groups. No significant difference was found in the level of ALT, AST, urea and creatinine between the different dosages in EAMMS extracts. Treatment with EAMMS also exhibited the highest inhibition activity of enzyme HMGCR and ACAT2 as compared to the control group. From the histopathological examination, liver tissues in the PC group showed severe steatosis than those fed with EAMMS and normal diet. Treatment with EAMMS extract ameliorated and reduced the pathological changes in the liver. No morphological changes showed in the kidney structure of both control and treated groups. In conclusion, these findings demonstrated that EAMMS extract has anti-hypercholesterolemia properties and could be used as an alternative treatment for this disorder.

Introduction: Euphorbia antiquorum is a fleshy shrub belonging to the family Euphorbiaceae. It is widely distributed in the hotter parts of India. It is used as a substitute for the Ayurvedic drug ‘Snuhee’ whose accepted botanical source is Euphorbia neriifolia Linn. The leaf, stem, latex and root of Snuhee are used in treatment of diseases and conditions like, diabetes, coryza, and oedema. Objective: The present study is aimed to investigate the renal protective effect of  E. antiquorum alcoholic extracts of the stem   on alloxan (150mg/kg) induced diabetic Wistar rats. Methods: All animals were treated for 21 days and sacrificed on the last day. Unwanted placebo effect in case of the control group was blocked by executing appropriate vehicle treatment and same animal handling pattern. Extract and standard drug doses given orally were suspended in 0.9% normal saline. Blood samples were drawn from retro orbital sinus using Diethyl-ether anesthesia at weekly intervals for 3 weeks. Blood samples were labeled in such a way that analysis team was kept blind about groups and nature of the study design. Body weight measurement and blood sampling were done on 1st, 7th, 14th and 21st day of the study. Conclusion: 21 days daily treatment with a test drug of ethanolic extract of stems of Euphorbia antiquorum significantly reduced the elevated Blood glucose level, Urinary Albumin, urine creatinine, serum creatinine in alloxan induced diabetic rats. Background: Inhibitors of HMG-CoA-reductase reduce cardiovascular mortality through the mechanisms yet elucidated. Most ischemic events are secondary to disruption of atherosclerotic plaques highly infiltrated by macrophages.Objectives: To study the effect of the 3-hydroxy-3-methyl-glutaryl coenzyme A (HMG-CoA)-reductase inhibitor simvastatin on the potential mechanisms involved in the recruitment of monocytic cells into the vessel wall.Methodology: This experimental study was carried out in the Laboratory of the Department of Pharmacology & Therapeutics, BSMMU, Shahbag, Dhaka, during the period from 1st July 2008 to 30th June 2009. Fifty healthy Long-Evans Norwegian male rats aged between 3-4 months and weighing between 180-200gm were randomly selected and divided into 3 groups (A, B & C). Control group A (n=10) were fed on standard rat diet for 8 weeks, Vehicle fed group B (n=10) were fed soybean oil at a dose of 1ml once daily for 8 weeks and 2% cholesterol fed group C (n=30) were fed a 2% cholesterol enriched diet (suspension of cholesterol powder in soybean oil) at a dose of 100mg/ml of 2ml once daily for 8 weeks. After 8 weeks 10 rats of each group were sacrificed and remaining 20 rats of group C were continued to the part II of the experiment and divided into two groups. Group-I (cholesterol fed control group, n=10) and Group-II (simvastatin treated group). Group-I fed 0.5ml of o.1% cholesterol enriched diet once daily to maintain atherosclerotic state & Group-II treated with 1ml of 60mg/ml simvastatin at a dose of 300mg/Kg/day along with 0.5ml of 0.1% cholesterol enriched diet. After 8 weeks all the rats of two group were sacrified. Blood collected from each rats for estimation of serum lipid profile by enzymatic methods and aorta was preserved for histopathological examination and Intima-media ratio was measured using Image-proplus software (Silva et al, 2000).Results: Simvastatin induced a significant reduction in serum lipids (p<0.001) and in atherosclerotic lesion size (p<0.001). Aortic macrophage infiltration was abolished by the treatment.Conclusions: In a rat atherosclerosis model, simvastatin diminished the neointimal inflammation and this could contribute to the stabilization of the atherosclerotic plaque. This may be an additional explanation for the reduction of acute ischemic events in patients treated with statins. The purpose of this study was to investigate antihyperlipidemic and antioxidative potentials of onion (Allium cepaL.) extract fermented with a novelLactobacillus caseiHD-010. In general, fermented onion extract is used for its antioxidative activity (ORAC), inhibitory effect on adipocytes differentiation, quercetin contents, and antihyperlipidemic activities. However, the effect of fermented onion extract on hyperlipidemia after oral administration using ApoE-deficient mice has not been reported yet. To understand the effect of fermented onion extract on hyperlipidemia, we used benzafibrate (10 mg/kg, bw/day) as a positive control in the present study. Serum was collected every week to analyze levels of low density lipoprotein (LDL), high density lipoprotein (HDL), triglyceride (TG), and cholesterol, 3-hydroxy-3-methylgutaryi-CoA (HMG-CoA) reductase activity, and cholesterol ester transport protein (CETP) activity. In the fermented onion-treated group, HDL level was significantly increased while levels of TG and LDL were significantly decreased compared to those in the control group. In addition, the inhibition activity of HMG-CoA reductase was increased 20% in the fermented onion-treated group at 100 mg/kg. CETP activity has been observed to be significantly inhibited in the fermented onion-treated groups compared to that in the control group. These results suggest that fermented onion has a preventive/therapeutic effect on hyperlipidemic disease. It might have potential to be developed as a functional food.

  Objective: The purpose of this study was to evaluate the effect of Terminalia chebula fruit extract on liver antioxidant enzymes in ethanol-induced hepatotoxicity in rats.Method: Rats were divided into six different groups each having six. Group 1 served as a control, Group 2 received 40% ethanol (2 ml/100 g, oral), in sterile water, Groups 4, 5, and 6 served as extract treatment groups and received 50, 100, and 200 mg/kg, orally, ethanolic fruit extract of T. chebula (TCE) and Group 3 served as standard group and received silymarin 25 mg/kg orally. All the treatment protocols followed 21 days, and after which rats were sacrificed, the liver was taken for antioxidant and histological studies, respectively.Results: The ethanol-treated group rats (G2) showed variable decrease in antioxidant parameter (catalase, glutathione, and glutathione reductase) levels. Administration of ethanolic TCE significantly prevented ethanol-induced elevation in the levels of malondialdehyde lipid peroxidation and decreased antioxidant parameters in experimental groups of rats. The effect of extract was compared with a standard drug, silymarin. The changes in antioxidant parameters were supported by histological profile.Conclusion: It is concluded that the ethanolic fruit TCE protects against ethanol-induced oxidative liver injury in rats.

Aims: This study aimed at investigating the antioxidative, anticonvulsive and histological effects of ethanolic fruit extract of Dennettia tripetala on the pre-frontal lobe of the brain in isoniazid-induced (300 mg/kg, i.p) seizure in adult wistar rat. Introduction: Neuronal hyper-excitability and excessive production of free radicals have been implicated in the pathogenesis of a considerable range of neurological disorders, including epilepsy. The high rate of oxidative metabolism, coupled with the low antioxidant defenses and the richness in polyunsaturated fatty acids, makes the brain highly vulnerable to free radical damage. Study Design: This is an original research conducted in Department of Anatomy, Faculty of Basic Medical Sciences, Enugu State University of Science and Technology (ESUT), Parklane, Enugu State, Nigeria, between June and August, 2019. Methodology: A total number of twenty four wistar rats were used for this experiment, the animals were grouped into six groups with four animals per group, Group I served as the negative control, Group II served as the positive control, Group III received the standard drug as well as the Isoniazid, while group IV,  V and VI  were treated with  ethanolic extract of Dennettia tripetala at different dosages; 250 mg/kg, 500 mg/kg and 750 mg/kg, intraperitoneally (i.p) respectively and its effects compared with a standard drug (Pyridoxine) treated group. Results: The extract significantly prolonged the onset of seizure at high dose administration (750 mg/kg) but completely prevented seizure occurrence at low and medium dose administration (250 mg/kg and 500 mg/kg, i.p) when induced with isoniazid (300 mg/kg, i.p.). Conclusion: The results obtained from this work suggest that ethanolic extract of Dennettia tripetala has anticonvulsant activity, and this supports the use of the formulation traditionally in the treatment of convulsions, thus should be considered for clinical trials.

Objective: Few studies have explored the diuretic property of fennel (Foeniculum vulgare). Hence, the aim of this study was to evaluate the antiurolithiatic property of ethanolic extract of fennel seeds in male Wistar albino rats.Methods: Prophylactic and curative urolithiasis models were used with 5 groups of 6 rats in each model. Ethanolic extract of fennel seeds in three doses 100, 200, and 300 mg/kg was used. Cystone 750 mg/kg was used as a standard drug. All drugs were administered orally. Zinc discs were surgically implanted in the bladder in all rats. After recovery, rats in the prophylactic model received three different doses of ethanolic extract of fennel seeds along with 1% ethylene glycol for 2 weeks whereas the rats in the other model received 1% ethylene glycol for 2 weeks followed by an ethanolic extract of fennel seeds in three doses for the next 2 weeks. Both models had a control group receiving 1% ethylene glycol. At the end of study period, rats were sacrificed and vesical calculi collected, weighed, and statistically evaluated using one-way ANOVA.Results: In both the models, all three doses of an extract of fennel seeds were effective in reducing stone formation as compared to control group with p<0.05. In both the models, all three test doses were comparable with cystone, but 300 mg/kg extract in prophylactic showed significance (p <0.05) when compared to standard.Conclusion: Fennel seeds can be used prophylactically as well as curatively in the treatment of urolithiasis. However, further studies and clinical trials are warranted to explore this property.
