The impact of different temperatures (0, 40, 60, 80 and 100° C for an hour) and radiation (0, 2.5, 5.0, 7.5 and 10.0 kGy) and combined treatment (temperature and radiation) for decontamination of bacteria from dried prawn, Macrobrachium lamarrei was also investigated under laboratory condition.The total viable bacterial (TVB), total staphylococcal (TS), total coliform (TC), total faecal coliform (TFC), total Aeromonas ( TA) and total fungal (TF) count were ranged from 2.7×108 to 5.3×108 cfu/g, 2.2×106 to 6.0×106 cfu/g, 2.9×104 to 9.5×104 cfu/g, 2.8×103 to 8.1×103 cfu/g, 5.7×103 to 8.5×103 cfu/g and 1.6×104 to 3.8×104 cfu/g respectively. Seventy eight bacterial strains were isolated and identified out of which 18 (23.07 %) were Staphylococcus aureus, 11 (14.10% ) were Micrococcus varians, 8 (10.25%) were Aeromonas hydrophila, 5 (6.41%) were Klebsiella ozaenae, 7 (8.97%) were Bacillus subtilis, 7 (8.97%) were Escherichia coli, 5 (6.41%) were Bacillus megaterium, 7 (8.97%) were Klebsiella edwardsii, 6 (7.69%) were Pseudomonas aeruginosa and 4 (5.12%) were Micrococcus radiodurans. Inactivated was more favorable at 100° C for TC, TFC, TA and TF and at 5.0 kGy for TA and TF and combined treatments (40° C+2.5 kGy, 60° C+2.5 kGy and 80° C+2.5 kGy) for TFC, TA and TF respectively. TS, TC and TVB were completely eliminated in treatment 40° C+5.0 kGy, 60° C+2.5 kGy, 80° C+2.5 kGy, 40° C+7.5 kGy, 60° C+7.5 kGy and 80° C+7.5 kGy respectively. Results demonstrated that the combination treatments were more effective than single treatment for eliminating the associated microorganisms/bacteria from dried prawn.Jahangirnagar University J. Background: This study aims to investigate the combination effect of a novel sirtuin inhibitor (BZD9L1) with 5-fluorouracil (5-FU) and to determine its molecular mechanism of action in colorectal cancer (CRC). Methods: BZD9L1 and 5-FU either as single treatment or in combination were tested against CRC cells to evaluate synergism in cytotoxicity, senescence and formation of micronucleus, cell cycle and apoptosis, as well as the regulation of related molecular players. The effects of combined treatments at different doses on stress and apoptosis, migration, invasion and cell death mechanism were evaluated through two-dimensional and three-dimensional cultures. In vivo studies include investigation on the combination effects of BZD9L1 and 5-FU on colorectal tumour xenograft growth and an evaluation of tumour proliferation and apoptosis using immunohistochemistry. Results: Combination treatments exerted synergistic reduction on cell viability on HCT 116 cells but not on HT-29 cells. Combined treatments reduced survival, induced cell cycle arrest, apoptosis, senescence and micronucleation in HCT 116 cells through modulation of multiple responsible molecular players and apoptosis pathways, with no effect in epithelial mesenchymal transition (EMT). Combination treatments regulated SIRT1 and SIRT2 protein expression levels differently and changed SIRT2 protein localization. Combined treatment reduced growth, migration, invasion and viability of HCT 116 spheroids through apoptosis, when compared with the single treatment. In addition, combined treatment was found to reduce tumour growth in vivo through reduction of tumour proliferation and necrosis compared with the vehicle control group. This highlights the potential therapeutic effects of BZD9L1 and 5-FU towards CRC. Conclusion: This study may pave the way for use of BZD9L1 as an adjuvant to 5-FU in improving the therapeutic efficacy for the treatment of colorectal cancer.

The study revealed that the total viable bacterial count, staphylococcal count, coliform count, faecal coliform count, Aeromonas count and total fungal counts varied from 8.8×107 to 1.27×108, 2.7×106 to 4.2×106, 9.0×104 to 1.03×105, 4.8×105 to 7.3×105, 2.7×105 to 5.0×105 and 1.6×103 to 3.5×103 cfu/gm, respectively. Seventy six bacterial strains were isolated and identified, where Staphylococcus aureus, Micrococcus varians, Aeromonas hydrophila, Klebsiella ozaenae, Bacillus subtilis, Escherichia coli, Bacillus megaterium, Klebsiella edwardsii, Pseudomonas aerugenosa and Micrococcus radiodurans were 16 (21.06%), 11 (14.47%), 9 (11.82%), 6 (7.89%), 6 (7.89%), 8 (10.53%), 6 (7.89%), 4 (5.26%), 7 (9.21%) and 3 ( 3.95% ), respectively. Total coliform, faecal coliform, Aeromonas bacteria and total fungi were found to be totally inactivated by the irradiation dose of 2.5 kGy onwards and total viable bacteria and staphylococcal bacteria were eliminated completely at the irradiation dose of 7.5 kGy. The bacterial counts were shown to be gradually decreased in all the samples from initial to final storage. The combined treatments were found more effective than that of the single treatment for reducing or eliminating the associated bacteria. The moisture contents of the studied fish were found nearly similar during initial and final storage periods and also at different radiation doses but the total protein contents decreased during final storage period and also at different radiation doses. Introduction: The present study is a case report of a 50-year-old man (SM) with aphasia whose word retrieval was severely impaired. The aim of the study was to compare the effect of a combined treatment program (attention training and language treatment) and a single program (language treatment alone) on the naming picture materials. Materials and Methods: This case was affected by a hemorrhagic cerebrovascular accident in the territory of Middle Cerebral Artery branches of the left hemisphere. Two naming treatments, i.e. the single and combined treatments sequentially, were provided for SM; during each of them a 50-item wordlist was practiced (Lists A and B). 12 treatment sessions were implemented for him during each treatment program. These two lists along with another list (List C), left for assessment of generalization of treatments to untrained items, were probed 6 times during and after each program. The whole picture set was normalized in another study described in the text. Results: Our patient’s naming ability progressed during the treatment programs. However, this progression was more salient in combined treatment program. More specifically, List B had the mean score of 34.25 in combined treatment program that was higher than mean of List A in single treatment program (14.5). The slope of List B scores was also higher than that of List A (3.7 vs 1.2). List C showed more generalization of combined treatment to untrained items than single treatment based on its mean scores (27 vs 18.5). Conclusion: the observed improvements of naming ability were felt to be the result of combining attention training into the language treatment which is lead to sustained attention, less distraction and more concentration.

As a result of the model experiment, an increase in the enzymatic activity of meadow chernozem of the impact zone of Ataman Lake with the introduction of a strains mixture of metal-resistant microorganisms into the soil was established. The experiment has shown that the application of bacterial strains increases the dehydrogenase activity of contaminated soil by 51.8% compared to the variant without remediation

Gasoline come primarily from petroleum cuts, it is the preferred liquid fuel in our lives. Two gasoline samples of octane numbers 91 and 95 from Saudi Arabia petrol stations were studied. This study was achieved at three different temperatures 20oC, 30oC and 50oC representing the change in temperatures of the different seasons of the year. Both the evaporated gases of light aromatic hydrocarbons (BTEX) of gasoline samples inside the tank were subjected to analyze qualitatively and quantitatively via capillary gas chromatography. The detailed hydrocarbon composition and the octane number of the studied gasoline samples were determined using detailed hydrocarbon analyzer. The idea of research is indicating the impact of light aromatic compounds in gasoline on the toxic effect of human and environment on the one hand, and on octane number of gasoline on the other hand. Although the value of octane number will be reduced but this will have a positive impact on the environment as a way to produce clean fuel.

AbstractAnisakidae, marine nematodes, are underrecognized fish-borne zoonotic parasites. Studies on factors that could trigger parasites to actively migrate out of the fish are very limited. The objective of this study was to assess the impact of different environmental conditions (temperature, CO2 and O2) on larval motility (in situ movement) and mobility (migration) in vitro. Larvae were collected by candling or enzymatic digestion from infected fish, identified morphologically and confirmed molecularly. Individual larvae were transferred to a semi-solid Phosphate Buffered Saline agar, and subjected to different temperatures (6 ℃, 12 ℃, 22 ℃, 37 ℃) at air conditions. Moreover, different combinations of CO2 and O2 with N2 as filler were tested, at both 6 °C and 12 °C. Video recordings of larvae were translated into scores for larval motility and mobility. Results showed that temperature had significant influence on larval movements, with the highest motility and mobility observed at 22 ℃ for Anisakis spp. larvae and 37 ℃ for Pseudoterranova spp. larvae. During the first 10 min, the median migration of Anisakis spp. larvae was 10 cm at 22 ℃, and the median migration of Pseudoterranova spp. larvae was 3 cm at 37 ℃. Larval mobility was not significantly different under the different CO2 or O2 conditions at 6 °C and 12 ℃. It was concluded that temperature significantly facilitated larval movement with the optimum temperature being different for Anisakis spp. and Pseudoterranova spp., while CO2 and O2 did not on the short term. This should be further validated in parasite-infected/spiked fish fillets.

AbstractIn this study, dual-phase (DP, ferrite + martensite) microstructures were obtained by performing intercritical heat treatments (IHT) at 750 and 800 °C followed by quenching. Decreasing the IHT temperature from 800 to 750 °C leads to: (i) a decrease in the volume fraction of austenite (martensite after quenching) from 0.68 to 0.36; (ii) ~ 100 °C decrease in martensite start temperature (Ms), mainly due to the higher carbon content of austenite and its smaller grains at 750 °C; (iii) a reduction in the block size of martensite from 1.9 to 1.2 μm as measured by EBSD. Having a higher carbon content and a finer block size, the localized microhardness of martensite islands increases from 380 HV (800 °C) to 504 HV (750 °C). Moreover, despite the different volume fractions of martensite obtained in DP microstructures, the hardness of the steels remained unchanged by changing the IHT temperature (~ 234 to 238 HV). Applying lower IHT temperature (lower fraction of martensite), the impact energy even decreased from 12 to 9 J due to the brittleness of the martensite phase. The results of the tensile tests indicate that by increasing the IHT temperature, the yield and ultimate tensile strengths of the DP steel increase from 493 to 770 MPa, and from 908 to 1080 MPa, respectively, while the total elongation decreases from 9.8 to 4.5%. In contrast to the normalized sample, formation of martensite in the DP steels could eliminate the yield point phenomenon in the tensile curves, as it generates free dislocations in adjacent ferrite.

This study investigated the impact of immune-related pathway inhibition, among them indolamine 2,3-dioxygenase (IDO), alone and together with immune cells on growth and viability of colorectal cancer (CRC) cells. A panel of patient-derived CRC cell lines with different molecular characteristics (CpG island methylator phenotype, chromosomal, and microsatellite instability) was included. Initial phenotyping of CRC cell lines (n=17) revealed high abundance of immunosuppressive checkpoint-molecules in general, but an individual profile for IDO. Presence of immune-related molecules was independent of the molecular subtype. Selective treatment of CRC cell lines showing high or low IDO expression (n=2 cell lines each) was performed with single agents and combinations of Indoximod, Curcumin, and Gemcitabine with and without the addition of peripheral blood lymphocytes (PBL) in an allogeneic setting. All substances affected CRC cell growth in a cell line specific manner. The combination of Curcumin and Gemcitabine proved to be most effective in tumor cell elimination. Functional read-out analyses identified cellular senescence, after both single and combined treatment. Curcumin alone exerted strong cytotoxic effects by inducing early and late apoptosis. Necrosis was not detectable at all. Addition of lymphocytes generally boosted antitumoral effects of all IDO-inhibitors, with up to 80 % cytotoxicity for the Curcumin treatment. Here, no obvious differences became apparent between individual cell lines. Combined application of Curcumin and low-dose chemotherapy is a promising strategy to kill tumor target cells and to stimulate antitumoral immune responses.

A positive correlation has been established between increased oxidative stress and cardiovascular diseases in diabetes mellitus. We evaluated the effects of single or combined treatments with vitamin A (retinol acetate, 30 mg/kg/day, for 12-weeks) and insulin (8-10 IU/rat/day for the final 6-week) on vasomotor activity, oxidative stress and retinol metabolism in 12-week streptozotocin diabetic rats. The vasomotor activity was determined by measuring invitroresponsiveness of aorta rings to phenylephrine (PE) and acetylcholine (ACh) in the absence or in the presence of hydrogen peroxide (H2O2). Preincubation withH2O2(10 μM) produced a significant decrease in PE (1 mM)-induced contraction in untreated-diabetic but not in control rats. Single treatment with insulin counteracted this effect ofH2O2and also reversed the increased contractile response of diabetic aorta to PE, while vitamin A was found to be ineffective.H2O2(10 μM) also inhibited ACh (1 mM)-stimulated endothelium- dependent relaxation two fold more in diabetic than in control aorta. In the prevention ofH2O2-induced inhibition of vascular relaxation to ACh, vitamin A alone was markedly effective while insulin alone was not. The combination of vitamin A plus insulin removed the inhibitory action ofH2O2in diabetic aorta. Diabetic animals displayed an increased level of aorta thiobarbituric acid reactive substance (TBARS) in association with decreased levels of plasma retinol and retinol-binding protein (RBP). Single treatment with insulin, in spite of allowing recovery of normal growth rate and improved glucose and retinol metabolism in diabetic rats, was unable to control TBARS production to the same extent as vitamin A alone. Our findings suggest that the maintenance of ACh-stimulated endothelium-dependent vasorelaxant tone in normal physiological levels depends largely on the prevention and/or inhibition of peroxidative stress, which is achieved by combined treatment with vitamin A plus insulin. The use of vitamin A together with insulin provides a better metabolic control and more benefits than use of insulin alone in the reduction of diabetes-induced vascular complications.
