The present study was conducted to record incidence and severity of citrus canker and to screen citrus species against this disease. A detailed survey was conducted in 19 nurseries and 13 commercial orchards of Peshawar division during 2011. It was observed that almost all the nurseries were severely affected by the disease. The highest disease incidence (100%) and severity (31.54%) were observed in New Tarnab nursery in (Mention name of district here). Among the orchards, Shado II showed comparatively more disease severity (2.92%) as compared to other orchards. Bacteria, isolated from symptomatic plant samples on nutrient agar medium produced small, shiny yellow colonies with smooth margins. Biochemical tests further confirmed the bacteria as Xanthomonas axonopodis pv. citri. Detached and attached leaf assays were made to check the resistance of four citrus species i.e. Citrus sinensis (sweet orange), C. aurantifola (kaghazi lemon), C. paradise (Grape fruit) and C. medica (Lime). C. medica was moderately resistant whereas C. sinensis was found to be the most susceptible against the disease. Present study will be useful for developing best possible solution for the management of citrus canker in Peshawar division.

Amylase is abundantly present in nature. The main source of this enzyme is the microbial origin. It is known that about two-third of the industrial enzymes (amylase, protease, cellulose, penicillinase, chitinase, etc,) are produced by the Bacillus spp. The present work comprised in the identification of amylase producing Bacillus spp and exposure of the producers to various parameters for the maximum yield of the enzyme. To isolate and identify the amylase and protease producing strain, soil samples were collected from different vegetation from the altitude at 4367.35 feet above sea level. The isolates were screened and various biochemical tests and morphological observations were done to identify the isolates. The enzymes were produced by the submerged state fermentation (SmF) from the isolates and purified by dialysis. Effects of temperature, pH, and different carbon and nitrogen sources of the medium using SmF were optimized. Among 95 isolates, 36 were identified. Among the identified isolates, Bacillus subtilis produced the maximum yield and thus, it was optimized for the amylase production. The maximum amylase production was found at 42 C temperature, in fructose as a carbon sugar, peptone as a nitrogen source and at pH 7. Almost all the enzyme producers inhabited the roots of leguminous plants. In the present study, starch is used with the nutrient agar medium to help in cell immobilization for maximum production of amylase by strains of Bacillus. More sophisticated process of purification such as chromatography and electrophoresis will yield more enzyme as compared to the dialysis.

Abstract Purpose Collective evidence from single-centre studies suggests an increasing incidence of pilonidal sinus disease in the last decades, but population-based data is scarce. Methods We analysed administrative case–based principal diagnoses of pilonidal sinus disease and its surgical therapy between 2005 and 2017 in inpatients. Changes were addressed via linear regression. Results The mean rate of inpatient episodes of pilonidal sinus disease per 100,000 men increased from 43 in 2005 to 56 in 2017. In females, the mean rate of inpatient episodes per 100,000 women rose from 14 in 2005 to 18 in 2017. In the whole population, for every case per 100,000 females, there were 3.1 cases per 100,000 males, but the numbers were highly variable between the age groups. There was considerable regional variation within Germany. Rates of inpatient episodes of pilonidal sinus disease were increasing in almost all age groups and both sexes by almost a third. Surgery was dominated by excision of pilonidal sinus without reconstructive procedures, such as flaps, whose share was around 13% of all procedures, despite recommendations of the national guidelines to prefer flap procedures. Conclusion Rates of inpatient episodes of pilonidal sinus disease in Germany rose across almost all age groups and both sexes with relevant regional variation. The underlying causative factors are unknown. Thus, patient-centred research is necessary to explore them. This should also take cases into account that are solely treated office-based in order to obtain a full-spectrum view of pilonidal sinus disease incidence rates.

Vitis labruscana ‘Concord’ is a grape cultivar widely grown in the United States for processing into juice and other grape products. Concord grapes are sporadically but sometimes severely damaged by the grape powdery mildew pathogen, Uncinula necator. Although the foliage is often reported to be moderately resistant to powdery mildew, severe fruit infection occurs in some years. We observed the seasonal development of powdery mildew on leaves, rachises, and berries of unsprayed Concord grapevines. Inoculations of flower and fruit clusters revealed a brief period of berry susceptibility and a protracted period of rachis susceptibility. The rachis remained highly susceptible to infection, and the severity of rachis infection increased throughout the growing season until the rachis formed a periderm shortly before harvest. In contrast, berries were nearly immune to infection within 2 weeks after fruit set. Rachis and berry infections were detected before the disease was observed on foliage, and the incidence of rachis and berry infection often exceeded disease incidence observed on foliage until after fruit acquired substantial ontogenic resistance. Excellent control of fruit infection, and adequate control of leaf infection, was achieved by two fungicide applications targeted at the peak period of fruit susceptibility. Although Concord is thought to be moderately resistant to powdery mildew, the rachis is highly susceptible, and may be the avenue by which prebloom infections make their way onto the developing fruit. Late-season infection of the rachis neither spread to the fruit, nor did it cause fruit to drop prematurely, and may be of little economic consequence on fruit destined for processing. Although fruit of V. vinifera cultivars have been reported to remain susceptible to infection until berry sugar levels reach 8 to 15%, Concord fruit become nearly immune to infection nearly 6 weeks before this stage of development. Because powdery mildew does not become conspicuous on foliage until late summer, it is generally regarded as a late-season problem on Concord grapes, and previous management programs have reflected this belief. However, the greatest contribution to control of fruit infection is due to fungicides applied during the peak period of fruit susceptibility, from bloom until shortly after fruit set, long before the disease is observed on foliage.

The purpose and objectives of the study is to identify the hormonal, biochemical and ultrasound features of the gestation course in women with beneficial tumours of the uterus. Results. We conducted a complete clinical examination and prospective observation of 182 pregnant women. Of which, 98 puerperas with a verified diagnosis of uterine fibroids and/or endometriosis, which developed before gestation, but did not prevent the onset of pregnancy, were included into the main group. The comparison group included 84 women with physiological pregnancy. The studies showed that 14 pregnant women in the main group had C677T (Ala222Val) T/T mutation, while only 2 women in the comparison group had a decrease in enzyme activity due to genetic mutation. Accordingly, the homocysteine level was almost 3 times lower in the comparison group than in the main group. The women with hyperhomocisteinemia (HHC) and uterine fibroid in the main group showed the lowest estriol level and hCG level, while women without HHC had higher estriol level and hCG levels. It was found that almost all patients with HHC had subclinical hypothyroidism. Ultrasound imaging and biochemical tests at the beginning of the 2nd trimester: no ultrasound markers of fetal anomalies were identified in women from both groups; however, signs of retrochorial hematoma were detected in 57 women from the main group, which was confirmed by clinical manifestations and previous ultrasound imaging in earlier gestation periods. Conclusion. Thus, placenta formation in women with HHC and uterine fibroids is accompanied by relative hormonal insufficiency, which is clinically manifested as a threat of miscarriage in the early stages, but carrying a child is possible due to appropriate management of a patient as part of the preserving therapy. However, the issue of preventing the development of fetoplacental insufficiency is a valid one for further investigation.


Citrus canker is the most devastating bacterial disease. In Pakistan, where canker is endemic, cultural practices and chemical control is vital module of integrated management system. But due to non-prudent use of chemicals wicked impacts start to appear on human health and environment there was need of some alternative management which should be eco-friendly and has no adverse effect for human. Therefore, our present study was based on screening of different cultivars of citrus and allelopathic management of citrus canker. The results revealed that Cara cara navel and kinnow both performed as moderately susceptible response in field condition than all other cultivars of citrus in screening experiment. Consequently, disease incidence was observed increasing by increasing the lesion area and these cultivars can be suggested as a source of resistance against canker pathogen. In allelopathic management we observed that ethanolic extracts were more efficient than aqueous extracts and their efficacy was also increasing by increasing the concentration. Ethanolic extracts of jatropha (13.33cm) followed by amaltas (12.5cm), Arjun (11.13cm), Bougenvilla (7.21cm) have great potency against the pathogen. So these ethanolic extracts can be used as good and alternative management of citrus canker disease.

A study was undertaken to evaluate effectiveness of foliar spray with 33 plant extracts against leaf spot (Tikka) of groundnut caused by Cercospora arachidicola and Cercosporidium personatum. Bavistin and BAU-Biofungicide were included in the experiment as checks and spray of plain water represented control. Almost all treatments gave considerable reduction in disease incidence and increase in growth parameters, pod and haulm yield compared to control. The most effective materials were Bavistin 50 WP, BAU-Biofungicide, leaf extract of neem, tomato, datura black, and datura white. The materials decreased spot number per leaf, defoliation per plant, incidence of leaf spot, and number of infected leaf per plant by 35.45 -60.07, 42.06-72.20, 51.97–63.58, and 38.33 to 46.89 % and increased pod yield and haulm yield by 64.37-111.41 and 32.35- 74.71 %, respectively. The reasons for an increase in barley loose smut in high rainfall areas of Western Australia were investigated in field trials from 1986 to 1988 by examining the effects of environment, cultivar and adequacy of chemical control. Disease was 4-18 times greater in 2 seed lines produced in very high rainfall areas (>750 mm/year) compared with that produced in high (450-750 mm/year) or low (<325 mm/year) rainfall areas. The effectiveness of 5 fungicide seed treatments was assessed. No fungicide seed treatment controlled disease completely. Triadimenol at 225 mg a.i./kg and carboxin at 940 mg a.i./kg were most effective, providing 93-96% disease control. Treatments were significantly (P<0.01) less effective in high rainfall areas of Western Australia. Barley cultivars released recently in Western Australia were found to be susceptible to loose smut; we suggest that the replacement of the moderately resistant Dampier with these cultivars has contributed to an increased incidence of disease.

Fusarium head blight (FHB) is one of the most destructive wheat fungal diseases, causing yield loss, quality reduction, and accumulation of mycotoxins. The aim of this research was to summarize the occurrence of major Fusarium mycotoxins: deoxynivalenol (DON), 3-acetyldeoxynivalenol (3-AcDON), nivalenol (NIV), and zearalenone (ZEN) in two consecutive years to search the relationship between disease incidence and severity with mycotoxins found in control and inoculated grains and corresponding malt. In addition, deoxynivalenol-3-glucoside (D3G) in one-year research was measured. Tested wheat varieties showed infection scores of 3% (‘U1’ and ‘Sirban Prolifik’) to 79% (‘Golubica’) for Type I resistance evaluation. There were few moderately resistant varieties in view of their areas under the disease progress curve, which can be considered Type III resistance (‘Sirban Prolifik’ and ‘U1’). According to the data quantified by LC–MS/MS, DON decreased in infected malt in comparison to corresponding grain, while ZEN occurred only in infected malt samples. Both 3-AcDON and NIV increased in inoculated malt in comparison to corresponding grain, due to a combination of plant metabolism and de novo synthesis by molds during malting. Based on the results, we can draw a few conclusions: the resistance to Fusarium decreased quantified concentrations of DON; ZEN gets synthetized during malting; unregulated 3-AcDON and NIV increase during malting; more resistant varieties have converted DON to D3G more successfully. Modified mycotoxins should be also included to legislation, since they could be transformed back to the corresponding mycotoxins under food processing conditions or during digestion.

Resistance mechanisms of Pseudomonas aeruginosa to ceftolozane/tazobactam (C/T) were assessed on a collection of 420 nonredundant strains non-susceptible to ceftazidime (MIC > 8 μg/ml) and/or imipenem (> 4 μg/ml), collected by 36 French hospital laboratories over a one month period (GERPA study). Rates of C/T resistance (MIC > 4/4 μg/ml) were equal to 10% in this population (42/420 strains), and 23.2% among the isolates resistant to both ceftazidime and imipenem (26/112). A first group of 21 strains (50%) was found to harbor various extended-spectrum β-lactamases (1 OXA-14; 2 OXA-19; 1 OXA-35; 1 GES-9; 3 PER-1), carbapenemases (2 GES-5; 1 IMP-8; 8 VIM-2), or both (1 VIM-2/OXA-35; 1 VIM-4/SHV-2a). All the strains of this group belonged to widely distributed epidemic clones (ST111, ST175, CC235, ST244, ST348, ST654), and were highly resistant to almost all the antibiotics tested except colistin. A second group was composed of 16 (38%) isolates moderately resistant to C/T (MIC from 8/4 to 16/4 μg/ml), of which 7 were related to international clones (ST111, ST253, CC274, ST352, ST386). As demonstrated by targeted mass spectrometry, cloxacillin-based inhibition tests and gene blaPDC deletion experiments, this resistance phenotype was correlated to an extremely high production of cephalosporinase PDC. In part accounting for this strong PDC upregulation, genomic analyses revealed the presence of mutations in regulator AmpR (D135N/G in 6 strains) and enzymes of the peptidoglycan recycling pathway such as AmpD, PBP4, and Mpl (9 strains). Finally, all the 5 (12%) remaining C/T resistant strains (group 3) appeared to encode PDC variants with mutations known to improve the hydrolytic activity of the β-lactamase to ceftazidime and C/T (F147L, ΔL223-Y226, E247K, N373I). Collectively, our results highlight the importance of both intrinsic and transferable mechanisms in C/T resistant P. aeruginosa. Which mutational events lead some clinical strains to massively produce the natural cephalosporinase PDC remains incompletely understood.

The management of citrus canker, caused by Xanthomonas citri subsp. citri, has been widely studied in endemic areas due to the importance of the disease in several citrus producing countries. A set of control measures is well-established, but no study has investigated the efficiency of each measure individually and their combination for disease suppression. This study comprised a 3-year field study to assess the relative contribution of three measures for the control of citrus canker and reduction of crop losses. Windbreak (Wb), copper sprays (Cu), and leafminer control (Lc) were assessed in eight different combinations in a split-split plot design. The orchard was composed of ‘Valencia’ sweet orange trees grafted onto ‘Rangpur’ lime. Casuarina cunninghamiana trees were used as Wb. Cu and Lc sprays were performed every 21 days throughout the year. Individually, Cu showed the highest contribution for canker control, followed by Wb. Lc had no effect on reducing citrus canker. Wb+Cu showed the highest efficiency for control of the disease. This combination reduced the incidence of diseased trees by ~60%, and the incidence of diseased leaves and fruit by ≥ 90% and increased the yield in 2.0 to 2.6-fold in comparison with the unmanaged plots. Cu sprays were important for reducing disease incidence and crop losses, whereas Wb had an additional contribution in minimizing the incidence of cankered, non-marketable fruit. The results indicated that the adoption of these measures of control may depend on the characteristics of the orchard and destination of the production.
