The objective of our experiment was to evaluate preservation and revitalization strategies for rumen inoculum anticipating research and veterinary applications. Rumen fluid samples were collected from 12 harvested cattle. Liquid samples were divided into five 500-mL aliquots which were randomly allocated to one of five treatments in a 2 × 2 + 1 augmented factorial design. Factors included preservation method [freezing (FZN) or lyophilization (LYO)] and preservative (glycerol; + or –). A fresh control (CON) was maintained from each sample. Feedstuffs used in this experiment were alfalfa hay, Coastal bermudagrass hay, cracked maize, rice bran, and soybean meal. Reference feedstuffs were subjected to batch culture in vitro true digestibility (IVTD) and in vitro NDF digestibility (IVNDFD) assays using inoculum from each of the five treatments. There was an effect (P < 0.05) of preservation method, preservative, and their interaction for both IVTD and IVNDFD of each of the five references feedstuffs. Freezing or lyophilization of rumen inoculum reduced (P < 0.05) IVTD and IVNDFD of reference feeds relative to the CON. Despite lower degradation of feeds when frozen or lyophilized rumen fluid was used rather than fresh, differences between them in IVTD and IVNDFD suggest that, in the absence of fresh inoculum, preserved rumen fluid may be a viable option for veterinary applications, such as transfaunation, but likely will not be viable for research applications.

Abstract Microbes and chemical constituents in the rumen fluid play an important role when re-establishing the rumen microbial population. Identification of a viable preservation method could have lasting implication of veterinary and research applications. Our objective was to determine the efficacy of preservation methods on rumen liquor. Twelve paunch samples were collected from slaughtered cattle at the Tarleton State University Meat Laboratory, Stephenville, TX. Rumen content was collected from different locations within the rumen, strained through eight layers of cheesecloth, and divided into five 500-mL aliquots. Aliquots were randomly allocated to one of five preservation methods (freezing or lyophilisation) and preservative (glycerol; yes or no). A fresh control was maintained from each sample. Three reference feeds and two roughage sources were used in the batch culture procedure. Reference feeds were dried to a constant weight at 55°C and ground to pass through a 2-mm screen. Ground samples were subjected to batch culture in vitro true digestibility (IVTD) assays using each of the four ruminal fluids (plus the control) for digestibility estimates. Original samples and incubated residues were assayed for DM, CP, NDF, and ADF. Data were analyzed in a 2 × 2 + 1 augmented factorial design using GLIMMIX procedure of SAS. There were no differences (P > 0.05) in IVTD when using protein or energy feeds; however, there were differences in IVTD for all treatments for fiber concentrates and forage reference feeds. Despite lower degradation of feeds when frozen or lyophilized rumen fluid was used, preserved rumen fluid may be a viable option in the absence of fresh inoculum for veterinary and research applications

ABSTRACT. The study aimed to determine the protein-energy synchronization (PES) index of the various energy source of feed concentrate in vitro, as a database for the preparation of ruminant rations based on the PES index. The research was conducted from May to August 2020. The research was carried out experimentally through three stages: proximate analysis, in vitro digestibility test, and index calculation. The materials used were rumen fluid of three Jawa Randu Goats, taken shortly after being slaughtered at Sokaraja Goat Slaughterhouse, and seven types of energy sources of feed concentrate. Each feedstuff was duplicated for 3 replications, then the digestibility data for organic matter and protein of each feedstuff were collected at the 2nd, 4th, 6th, 8th, 12th, 24th, and 48th hour by in vitro fermentation. The data collection results were then regressed and the results were included in the PES index equation. The variable measured was the PES synchronization index. The results showed that the PES index of rice bran was 0.54, rice polish 0.50, pollard 0.57, corn 0.87, dried cassava 0.94, dried cassava dreg 0.90, and bread flour 0.94. The study concluded that the energy source of feed concentrate are potential as the medium to the high category of PES index values in the preparation of ruminant rations, where corn, dried cassava, dried cassava dreg, and bread flour have a PES index in the high category, while rice bran, rice polish, and pollard in the medium category.  (Indeks sinkronisasi protein-energi berbagai bahan pakan konsentrat sumber energi bagi ruminansia) ABSTRAK. Tujuan penelitian ini adalah menginventarisasi indeks sinkronisasi protein-energi (SPE) berbagai bahan pakan konsentrat sumber energy, sebagai basis data penyusunan ransum ruminansia berbasis indeks SPE secara in vitro. Penelitian dilaksanakan mulai bulan Mei hingga Agustus 2020. Penelitian dilaksanakan secara eksperimental melalui tiga tahap yaitu analisis proksimat, kecernaan in vitro, dan perhitungan indeks. Materi yang digunakan adalah cairan rumen 3 Kambing Jawa Randu yang diambil sesaat setelah disembelih di Rumah Potong Hewan Sokaraja, serta 7 jenis bahan pakan konsentrat sumber energi. Masing-masing bahan pakan diduplikasi sebanyak 3 ulangan, kemudian masing-masing diukur kecernaan bahan organik dan protein pada waktu fermentasi in vitro ke 2, 4, 6, 8, 12, 24 dan 48 jam. Data yang dikoleksi selanjutnya di uji regresi dan hasilnya dimasukkan dalam persamaan indeks SPE. Variabel yang diukur adalah indeks SPE. Hasil penelitian menunjukkan bahwa indeks SPE dedak sebesar 0,54, bekatul 0,50, pollard 0,57, jagung 0,87, gaplek 0,94, onggok kering 0,90 dan tepung roti 0,94. Penelitian menyimpulkan bahwa, konsentrat sumber energi memiliki potensi nilai indeks sinkronisasi protein-energi pada kategori sedang hingga tinggi dalam penyusunan ransum ruminansia. Jagung, gaplek, onggok kering dan tepung roti memiliki indeks sinkronisasi protein-energi pada kategori tinggi, sedangkan dedak, bekatul dan pollard pada kategori sedang.

Abstract A study was conducted to evaluate effects of increasing concentration of food grade glycerol on rumen environment and nutrient digestibility. Three ruminally cannulated Jersey steers were used in this study. The study was conducted from March to May 2019. Experimental design was a 3x3 Latin square with a 2wk adjustment period followed by a 1wk collection period. Diet was coastal bermudagrass hay based. Different forage types were introduced in the incubation process to evaluate digestibility. Glycerol was administered once a day at 0, 15, or 20% of DMI (dry matter intake). dNDF (digestible NDF) and dDM (digestible dry matter) was determined using an ANKOM Daisy II incubator inoculated with 200g fresh rumen fluid and incubated for 12, 24, 48 and 72 h at 39°C. Each vessel contained ground forage samples in filter bags in triplicate. After incubation, filter bags were rinsed with cold water and dried for 24h in a 55°C forced air oven. Data were analyzed using the Proc MIXED procedure of SAS version 9.4. There was no difference dNDF in effect of different levels of glycerol between forage types by diet. But a numerical tendency was observed that dNDF was decreased at 20% inclusion rates in comparison to 0 and 15% inclusion of glycerol in the diet. Neither steer nor run was significantly different in the study. However as expected digestibility over time was significantly different (P < 0.001). A significant increase was observed in DMI with the increased levels of glycerol in the diet (P = 0.003), both the 15% and 20% levels of glycerol increased in DMI in comparison to the control (0%). It appears based on these study results that digestibility may be inhibited, as levels of dietary glycerol increase in the diet and more work needs to be done to find the optimal level of glycerol supplementation.

Products of rumen fermentation and protein microbial of dairy cattle feed with rice bran ammonization and some feedstuffs as an energy sourcesABSTRACT. This study aims to examine the energy sources of feed ingredients that can increase the production of Volatile Fatty Acids (VFA), N-NH3, microbial protein synthesis, total gas production and metabolic energy. The material used is as a source of rumen fluid inoculum from Frisian Holstein cows (FH) females, amoniasi rice straw, salt, mineral mix brand "Ultra Minerals' production Eka Farma Semarang, onggok wet and dry, corn, and rice bran. Observed variable is the concentration of (VFA), N-NH3, rumen microbial protein synthesis, and total gas production. Based on the analysis of diversity seen any significant effect (P0.05) on total VFA concentration, N-NH3 and total gas but had no effect (P0.05) on microbial protein synthesis. Conclusion of research is the provision of energy sources with rice bran treatment, onggok wet and dry corn flour can be used as fermentable carbohydrates on feed hay amoniasi in vitro.

In a study that included C4 tropical grasses, C3 temperate grasses and C3 pasture legumes, in vitro dry matter digestibility of extrusa, measured as in vitro dry matter loss (IVDML) during incubation, compared with that of the forage consumed, was greater for grass extrusa but not for legume extrusa. The increase in digestibility was not caused by mastication or by the freezing of extrusa samples during storage but by the action of saliva. Comparable increases in IVDML were achieved merely by mixing bovine saliva with ground forage samples. Differences were greater than could be explained by increases due to completely digestible salivary DM. There was no significant difference between animals in relation to the saliva effect on IVDML and, except for some minor differences, similar saliva effects on IVDML were measured using either the pepsin–cellulase or rumen fluid–pepsin in vitro techniques. For both C4 and C3 grasses the magnitude of the differences were inversely related to IVDML of the feed and there was little or no difference between extrusa and feed at high digestibilities (>70%) whereas differences of more than 10 percentage units were measured on low quality grass forages. The data did not suggest that the extrusa or saliva effect on digestibility was different for C3 grasses than for C4 grasses but data on C3 grasses were limited to few species and to high digestibility samples. For legume forages there was no saliva effect when the pepsin–cellulase method was used but there was a small but significant positive effect using the rumen fluid–pepsin method. It was concluded that when samples of extrusa are analysed using in vitro techniques, predicted in vivo digestibility of the feed consumed will often be overestimated, especially for low quality grass diets. The implications of overestimating in vivo digestibility and suggestions for overcoming such errors are discussed.

This review summarises the use of the Ankom DaisyII incubator (ADII; Ankom Technology Corporation Fairport, NY, USA), as presented in studies on digestibility, and its extension to other species apart from ruminants, from its introduction until today. This technique has been modified and adapted to allow for different types of investigations to be conducted. Researchers have studied and tested different procedures, and the main sources of variation have been found to be: the inoculum source, sample size, sample preparation, and bag type. In vitro digestibility methods, applied to the ADII incubator, have been reviewed, the precision and accuracy of the method using the ADII incubator have been dealt with, and comparisons with other methods have been made. Moreover, some hypotheses on the possible evolutions of this technology in non-ruminants, including pets, have been described. To date, there are no standardised protocols for the collection, storage, and transportation of rumen fluid or faeces. There is also still a need to standardise the procedures for washing the bags after digestion. Moreover, some performance metrics of the instrument (such as the reliability of the rotation mechanism of the jars) still require improvement.

Previous studies (El Shaer, Omed and Axford, 1987; Akhter, Owen, Fall, O'Donovan and Theodorou, 1994) with the two-stage in vitro procedure of Tilley and Terry (1963) have shown a high correlation between digestibilities of forages as determined using either sheep rumen liquor, sheep faeces or cow faeces as the microbial inoculum. In the first study of the of the present investigation one objective was to examine the repeatability of these digestibility measurements when made on different occasions. A second objective was to assess whether the correlations between faecal and rumen fluid based inocula could be improved if microorganisms were obtained from pairs rather than individual animals. The objective in the second study using forages of known in vivo digestibility, was to investigate the effect of freezing or freeze-drying of faeces on the repeatability of digestibilities of forages determined in vitro using micro-organisms from cow faeces.

Three experiments were conducted to study the effects of substituting rumen fluid (RF) with faecal suspension (FS) as an inoculum for determination of in vitro dry matter digestibility (IVDMD, experiment 1) and gas production (exp. 2). Barley grain (BG), Persian clover (PC), alfalfa (ALF), bromegrass (BR) and barley straw (BS) were used to evaluate the efficacy of the two inocula. In exp. 1, IVDMD was determined using RF or FS containing 3, 6 and 9% fresh cattle faeces as inocula. Except for BS, IVDMD values obtained with either RF or FS containing 3% faeces were not different (P > 0.05). In contrast, FS containing 6 or 9% faeces resulted in lower (P < 0.05) IVDMD than those obtained with RF. Total VFA, butyric and valeric acids of in vitro supernatant were higher (P < 0.05) in RF than FS incubated buffers. Molar proportions of acetic and propionic acids were variable among feeds. Mean acetic:propionic ratio was similar (except for BG) for RF or FS. In exp. 2, substitution of RF with FS containing 6, 11 or 16% cattle faeces as inoculum generally resulted in an increased lag time for four feeds (PC was not included). The rate of gas production was lower when FS was used instead of RF for ALF and BR. For BG however, use of FS resulted in a higher (P < 0.05) rate of gas production than when RF was used. Total gas production values were similar within all the feeds regardless of the source of inoculum. In experiment 3, the relative size of the total and cellulolytic populations were higher for RF than for FS, with a greater diversity of genera of bacteria isolated from RF. Results from this study indicate that FS has the potential to be used instead of RF to obtain IVDMD and gas production for BG and forage hay but not for poor quality roughages such as BS. Rumen fluid from two sheep was used to determine in vitro dry matter digestibilities of 25 samples each from alfalfa hay (Medicago sativa L.) and barley grain (Hordeum vulgare L.). The sheep were fed either a good quality hay ad libitum and 1 kg rolled barley per day, or only the hay ad libitum, in a switchback design. The in vitro dry matter digestibilities of both alfalfa hay and barley were lower (P < 0.01) when the donor animal was fed hay and barley than when fed only hay.

Tiger nut (Cyperus esculentus L.), or chufa, is a plant that is found in nature and is cultivated for its edible tubers. The purpose of this study was to determine the chemical composition, nutritive value, and in vitro digestibility of three tiger nut varieties using the in vitro gas production technique. These varieties were Sarışeker (yellow), Introduction 1, and Balyumru (brown). Rumen fluid was obtained from two cannulated Holstein animals. Time-dependent in vitro gas production was monitored at 3, 6, 9, 12, 24, 48, 72, and 96 hours of incubation. The varieties differed in dry matter (DM), crude ash (CA), ether extract (EE), neutral detergent fibre (NDF), and non-fibre carbohydrate (NFC) content (P <0.05). They also differed in the instantaneous volume of gas produced and in time-dependent gas production. Balyumru produced more gas at the onset of incubation than Introduction 1 and Sarışeker. However, over time, the gas produced by digestion of Introduction 1 exceeded the other two varieties. The amounts of gas produced at each time-point were intercorrelated. It is recommended that these results should lead to further evaluation in in vivo studies. Keywords: chemical composition, energy content, in vitro gas production
