Oxalic and phytic acid are phytochemicals considered to be anti-nutritional factors as they are predominantly found as oxalates and phytates bound to minerals like calcium and potassium. Studies have associated excessive oxalate consumption with increased urinary excretion of oxalate (hyperoxaluria) and calcium oxalate kidney stone formation, and excessive phytate consumption with decreased bioaccessibility and bioavailability of certain minerals and reduced utilization of dietary protein. However, other studies suggest that dietary consumption of phytate may be beneficial and inhibit formation of calcium oxalate kidney stones. In light of these conflicting reports, dietary intake of oxalate and phytate enriched plants should be considered in relation to potential health outcomes following consumption. Terminalia ferdinandiana is one such plant and is investigated here with respect to oxalate, phytate, and mineral contents. Assessment of oxalate and phytate contents in T. ferdinandiana fruit, leaf, and seedcoat tissues through hydrolysis into acid forms revealed oxalic acid contents ranging from 327 to 1,420 mg/100 g on a dry weight (DW) basis whilst phytic acid contents ranged from 8.44 to 121.72 mg/100 g DW. Calcium content in the different tissues ranged from 131 to 1,343 mg/100 g. There was no correlation between oxalic acid and calcium, however a significant, positive correlation was observed between phytic acid and calcium (r = 0.9917; p < 0.001), indicating that tissues rich in phytic acid also contain higher levels of calcium. The high content of phytic acid in comparison to oxalic acid in T. ferdinandiana fruit found in this study and the dietary significance of this in terms of calcium bioavailability, needs to be investigated further.

Green vegetables are very beneficial for health, but sometimes it is not understood that some vegetables also contain anti-nutrients. For example, oxalic acid can be found in some green vegetables, such as spinach. Oxalic acid can be absorbed from food and as it is a toxin it has to be removed from the body via the kidneys. Under certain conditions, oxalate can combine with calcium in the kidneys forming stones which can lead to impaired kidney function. This study aims to compare two techniques for making vegetable juice, using a high-speed blender or a masticating juicer. The content of oxalic acid, calcium, pH and titratable acidity (TA) was measured in both juices. Total oxalic acid was measured using HPLC, calcium content was analyzed using a Coupled Plasma Optical Emission Spectrophotometer. Dry matter, pH and TA were measured based on AOAC methods. The highest total oxalate (OA) was found in juice made using a masticating juicer of 7638.27 mg OA/100 g dry weight. The highest calcium content was found in the fiber fraction or in the remaining juice using the masticating juicer technique, which was 10.04 mg/100 g juice. Meanwhile, the TA value of the thick juice produced by the high speed blender was not significantly different from the runny juice produced by the masticating juicer. The smoothie-like green juice prepared using a high-speed blender was healthier as the soluble and insoluble oxalate extracted in the juice was lower than in the clear juice prepared using a masticating juicer.

Abstract Background Patients with idiopathic calcium oxalate stones are advised to consume a low-oxalate diet to prevent recurrence. In this study, on patients with calcium oxalate stones we have attempted to determine the effect of calcium content of diet on the formation of calcium oxalate crystals in urine by in vitro supersaturation study of fresh postprandial urine samples and observing the morphology of the crystals formed using polarized optical microscopy. Methods The trial was conducted as a prospective interventional randomised crossover clinical trial in a repeated measures design. Sixty patients with calcium oxalate stones and no metabolic abnormalities in urine treated by lithotripsy at a tertiary care centre during the period May 2016 to May 2019 were recruited. Following a 14 h overnight fasting, urine samples were collected after providing the patient with either a low- or high-calcium meal for breakfast followed four hours later, by high-oxalate meal for lunch. Urine was tested for multiple parameters including urine pH, specific gravity, calcium/creatinine ratio and supersaturation of urine with sodium oxalate followed by optical density measurement by spectrophotometry and microscopic analysis of crystals formed. Results Optical density values and calcium/creatinine ratio of urine samples obtained after high-calcium meal are significantly higher than in corresponding sample obtained after low-calcium meal (p < 0.001). These findings were reflected in the morphology of formed crystals in their size, shape and number. When urinary calcium levels were low, no crystals were formed during supersaturation study of postprandial urine samples following a high-oxalate diet. Conclusions High calcium content in diet significantly contributes to kidney stone formation. There is a lower risk of kidney stone formation with a low-calcium meal even on consumption of a high-oxalate diet.

Calcium oxalate monohydrate (COM) is the primary constituent of the majority of renal stones. Osteopontin (OPN), an aspartic acid-rich urinary protein, and citrate, a much smaller molecule, are potent inhibitors of COM crystallization at levels present in normal urine. Current concepts of the role of site-specific interactions in crystallization derived from studies of biomineralization are reviewed to provide a context for understanding modulation of COM growth at a molecular level. Results from in situ atomic force microscopy (AFM) analyses of the effects of citrate and OPN on growth verified the critical role of site-specific interactions between these growth modulators and individual steps on COM crystal surfaces. Molecular modeling investigations of interactions of citrate with steps and faces on COM crystal surfaces provided links between the stereochemistry of interaction and the binding energy levels that underlie mechanisms of growth modification and changes in overall crystal morphology. The combination of in situ AFM and molecular modeling provides new knowledge that will aid rationale design of therapeutic agents for inhibition of stone formation.

Oxalic acid and crystals of calcium oxalate were produced during growth of Leucostoma cincta and L. persoonii on potato dextrose agar and in peach bark tissues. The identification of calcium oxalate was based on solubility characteristics, the results of KMnO4 titration, positive staining with silver nitrate – dithiooxamide, and crystal morphology as observed with light and scanning electron microscopes. Oxalic acid was detected by gas chromatography. This is the first report of oxalic acid production by both Leucostoma species causing peach canker. Calcium oxalate crystals observed on or near hyphae in culture were similar to crystals in artificially inoculated peach bark tissues. Addition of oxalic acid solutions alone to inner bark tissues caused maceration and necrosis. These results indicate a role for oxalic acid in the early stages of pathogenesis by Leucostoma spp. Tetragonal (bipyramidal) and prismatic calcium oxalate crystals formed on bark wounds treated with oxalic acid solutions were similar to those observed in infected tissues and in culture media amended with oxalic acid.

Nephrolithiasis is a highly prevalent condition, but its incidence varies depending on race, gender, and geographic location. Approximately half of patients form at least one recurrent stone within 10 years of the first episode. Renal stones are usually composed of calcium salts (calcium oxalate monohydrate or dihydrate, calcium phosphate), uric acid, or, less frequently, cystine and struvite (magnesium, ammonium, and phosphate). Calcium oxalate stones, the most commonly encountered ones, may result from urinary calcium oxalate precipitation on the Randall plaque, which is a hydroxyapatite deposit in the interstitium of the kidney medulla. Uric acid nephrolithiasis, which is common among patients with metabolic syndrome or diabetes mellitus, is caused by an excessively acidic urinary pH as a renal manifestation of insulin resistance. The medical evaluation of the kidney stone patient must be focused on identifying anatomic abnormalities of the urinary tract, associated systemic diseases, use of lithogenic drugs or supplements, and, mostly, urinary risk factors such as low urine volume, hypercalciuria, hyperuricosuria, hypocitraturia, hyperoxaluria, and abnormalities in urine pH that can be affected by dietary habits, environmental factors, and genetic traits. Metabolic evaluation requires a urinalysis, stone analysis (if available), serum chemistry, and urinary parameters, preferably obtained by two nonconsecutive 24-hour urine collections under a random diet. Targeted medication and dietary advice are effective to reduce the risk of recurrence. Clinical, radiologic, and laboratory follow-ups are needed to prevent stone growth and new stone formation, to assess treatment adherence or effectiveness to dietary recommendations, and to allow adjustment of pharmacologic treatment. Accumulation of heavy metals in fish is considered a critical problem for human health. Therefore, the study aimed to quantify the concentrations of iron (Fe), zinc (Zn), manganese (Mn), and lead (Pb) in Oreochromis niloticus and Clarias gariepinus from two areas in Al Sharqia governorate, Egypt, from September 2017 to August 2018. A human health risk assessment was conducted to evaluate the potential hazards associated with fish consumption. Metals concentrations (mg/kg dry weight) in muscles of catfish ranged 1.88-221.26 for Fe; 1.78-19.77 for Zn; BDL-238.51 for Mn; BDL-22.75 for Pb. In muscles of tilapia fish metals concentrations ranged 7.96-149.10 for Fe; 1.20-19.77 for Zn; BDL-230.82 for Mn; BDL-25.93 for Pb. Pb had Hazard quotients (HQs) which indicated potential health risks to tilapia consumers at both study areas and catfish consumers at the Faqous area. Fishermen were at higher risk compared to the other consumers.

Plants of five cultivars of wheat were grown under controlled-environmental conditions in order to analyse the effect of cultivar and of temperature and illuminance after anthesis on the accumulation of nitrogen and phosphorus by grains in relation to dry matter. The water relations of the grain during maturation were also examined, using calcium content as an index of water entry. The nitrogen and phosphorus contents of grains increased linearly throughout the grain growth period. The percentage of nitrogen and phosphorus in grains fell sharply during the first few days after anthesis but rose progressively thereafter. The higher the temperature, and the lower the illuminance, the higher was the percentage of nitrogen in the grain of all cultivars. Such conditions also reduce final grain size, but their effects on nitrogen concentration in the grain were apparent early in grain development. No evidence was found of a flush of nitrogen or phosphorus into the grain late in its development. Water entry into the grain continued at a steady rate until maximum grain dry weight was reached, then ceased suddenly. No evidence was found of an increased rate of water loss by the grain at that stage, and the rapid fall in water content at the cessation of grain growth may have been due to blockage of the chalazal zone of entry into the grain by the deposition of lipids. Accumulation of dry matter, nitrogen and phosphorus and entry of water into the grain all ceased at the time of lipid deposition in the chalazal zone.

1. Anomalous transmembrane anion transport has been observed in erythrocytes of patients with idiopathic calcium nephrolithiasis. 2. To verify whether cation transport is also abnormal, we investigated the frusemide-sensitive Na+ efflux from Na+-loaded erythrocytes and the natriuretic response to acute intravenous frusemide administration in calcium oxalate renal stone formers. 3. Frusemide administration induced a statistically significant smaller increase in the fractional excretion of Na+ in patients than in control subjects. Abnormal kinetic properties of erythrocyte Na+-K+-2Cl− co-transport were observed in approximately 60% of stone formers. The Km for Na+ of Na+-K+-2Cl− co-transport correlated with urinary Ca2+ excretion. 4. The abnormal kinetic properties of Na+-K+-2Cl− co-transport may be relevant for stone formation, hampering renal Ca2+ reabsorption in the distal nephron and determining critical physicochemical conditions for calcium/oxalate crystallization.
