Consumption of lipid-rich foods can increase the blood cholesterol content. β-glucans have hypocholesterolemic effect. However, subtle changes in their molecular branching can influence bioactivity. Therefore, a comparative investigation of the cholesterol-lowering potential of two β-glucans with different branching patterns and a cholesterol-lowering drug, namely simvastatin was undertaken employing the zebrafish (Danio rerio) model of diet-induced hypercholesterolemia. Fish were allocated to 5 dietary treatments; a control group, a high cholesterol group, two β-glucan groups, and a simvastatin group. We investigated plasma total cholesterol, LDL and HDL cholesterol levels, histological changes in the tissues, and explored intestinal transcriptomic changes induced by the experimental diets. Dietary cholesterol likely caused the suppression of endogenous cholesterol biosynthesis, induced dysfunction of endoplasmic reticulum and mitochondria, and altered the histomorphology of the intestine. The two β-glucans and simvastatin significantly abated the rise in plasma cholesterol levels and restored the expression of specific genes to alleviate the endoplasmic reticulum-related effects induced by the dietary cholesterol. Furthermore, the distinct patterns of transcriptomic changes in the intestine elicited by the oat and microalga β-glucans impacted processes such as fatty acid metabolism, protein catabolic processes, and nuclear division. Oat and microalgal β-glucans also altered the pattern of lipid deposition in the liver. Our study provides insights into the effectiveness of different β-glucans to alleviate dysfunctions in lipid metabolism caused by dietary cholesterol.

The effect of feeding liveLactobacillus reutericells containing active bile salt hydrolase (BSH) on plasma cholesterol levels was studied in pigs. During an experiment lasting 13 weeks, twenty pigs were fed on a high-fat, high-cholesterol, low-fibre diet for the first 10 weeks, and a regular pig diet for the last 3 weeks. One group of animals received, twice daily, 11·25 (SD 0·16) log10colony forming units of the potential probiotic bacteria for 4 weeks (from week 3 until week 7). From week 8 onwards, the treated group was again fed on the same diet as the control group without additions. The total faecalLactobacilluscounts were only significantly higher in the treated pigs during the first 2 weeks ofL. reuterifeeding. Based on limited data, it was suggested that the administeredLactobacillusspecies had caused a temporary shift within the indigenousLactobacilluspopulation rather than permanently colonizing the intestinal tract. The probiotic feeding brought about significant lowering (P≤ 0·05) of total and LDL-cholesterol concentrations in the treated pigs compared with the control pigs, while no change in HDL-cholesterol concentration was observed. The data for faecal output of neutral sterols and bile salts were highly variable between the animals of each group, yet they indicated an increased output in the treated pigs. Although the blood cholesterol levels went up in both groups during the 3 weeks following theLactobacillusadministration period, significantly lower serum total and LDL-cholesterol levels were observed in the treated pigs. During the final 3 weeks of normalization to the regular diet, cholesterol concentrations significantly decreased in both animal groups and the differences in total and LDL-cholesterol concentrations between the groups largely disappeared.

Hypercholesterolemia is a well-established risk factor for atherosclerotic cardiovascular disease (ASCVD). Low-density lipoprotein cholesterol (LDL-C) has been labeled as “bad” cholesterol and high-density lipoprotein cholesterol (HDL-C) as “good” cholesterol. The prevailing hypothesis is that lowering blood cholesterol levels, especially LDL-C, reduces vascular deposition and retention of cholesterol or apolipoprotein B (apoB)-containing lipoproteins which are atherogenic. We review herein the clinical trial data on different pharmacological approaches to lowering blood cholesterol and propose that the mechanism of action of cholesterol lowering, as well as the amplitude of cholesterol reduction, are critically important in leading to improved clinical outcomes in ASCVD. The effects of bile acid sequestrants, fibrates, niacin, cholesteryl ester transfer protein (CETP) inhibitors, apolipoprotein A-I and HDL mimetics, apoB regulators, acyl coenzyme A: cholesterol acyltransferase (ACAT) inhibitors, cholesterol absorption inhibitors, statins, and proprotein convertase subtilisin kexin 9 (PCSK9) inhibitors, among other strategies are reviewed. Clinical evidence supports that different classes of cholesterol lowering or lipoprotein regulating approaches yielded variable effects on ASCVD outcomes, especially in cardiovascular and all-cause mortality. Statins are the most widely used cholesterol lowering agents and have the best proven cardiovascular event and survival benefits. Manipulating cholesterol levels by specific targeting of apoproteins or lipoproteins has not yielded clinical benefit. Understanding why lowering LDL-C by different approaches varies in clinical outcomes of ASCVD, especially in survival benefit, may shed further light on our evolving understanding of how cholesterol and its carrier lipoproteins are involved in ASCVD and aid in developing effective pharmacological strategies to improve the clinical outcomes of ASCVD.

ABSTRACT This study aims to determine the effect of infusion of conjoined pumpkin leaves (Sechium edule) on reducing total blood cholesterol of male white rats (Rattus norvegicus). The subjects of this study were 15 male white rats with an average body weight of 200 grams which were divided into 5 groups, each group consisted of 3 mice. The method used is a laboratory experiment with a completely randomized design. The results were obtained from 2 measurements of blood cholesterol levels, namely measurements before and after treatment. The treatment begins with the provision of high-fat foods for 48 days. On the 49th day a blood cholesterol level was measured before treatment. Furthermore, treatment was given to each group, namely aquades in the negative control group, simvastatin in the positive control group, and squash leaves infusion with their respective doses in the dose group I (40%), the dose group II (20%), and the dose group III (10%). Measurement of cholesterol levels after treatment was carried out on day 54. Data were analyzed by Paired t-test and One Way ANOVA. The analysis showed that there were no significant differences between treatment groups. Judging from the change in average and percentage, 40% infusion dose of siamese pumpkin leaves gives the best reduction in cholesterol levels. Keywords: Cholesterol, pumpkin leaves, male white mouse infusion. ABSTRAK Penelitian ini bertujuan untuk mengetahui pengaruh pemberian infusa daun labu siam (Sechium edule ) terhadap penuruan kolesterol darah total tikus putih jantan (Rattus norvegicus). Subjek penelitian ini berupa tikus putih jantan berjumlah 15 ekor dengan berat badan rata-rata 200 gram yang dibagi dalam 5 kelompok, masing-masing kelompok sebanyak 3 ekor. Metode yang digunakan yaitu eksperimen laboratorium dengan rancangan acak lengkap. Hasil penelitian diperoleh dari 2 kali pengukuran kadar koleterol darah yaitu pengukuran sebelum dan sesudah perlakuan. Perlakuan dimulai dengan pemberian makanan tinggi lemak selama 48 hari. Pada hari ke49 dilakukan pengukuran kadar kolesterol darah sebelum perlakuan. Selanjutnya diberikan perlakuan pada tiap kelompok yaitu aquades pada kelompok kontrol negatif, simvastatin pada kelompok kontrol positif, dan infusa daun labu siam dengan dosis masing-masing pada kelompok dosis I (40 %), kelompok dosis II (20 %), dan kelompok dosis III (10%). Pengukuran kadar kolesterol sesudah perlakuan dilakukan pada hari 54. Data diananlisis dengan Paired t-test dan One Way ANOVA. Hasil analisa menunjukan tidak terdapat perbedaan yang bermakna antar kelompok perlakuan. Dilihat dari perubahan rerataan dan presentase, dosis infusa  40% daun labu siam memberikan penurunan kadar koleterol terbaik. Kata kunci : Infusa daun Labu siam, kolesterol, tikus putih jantan.

Background: Much alternative therapy such as cupping therapy in the community as one of the cheap treatment efforts without side effects makes the challenge and the goal for nursing staff improve nursing services. The unfinished scientific verification of cupping therapy's alternative therapy makes nurses need to prove decreased blood pressure and cholesterol levels in the elderly. Objective: Scientific proof of cholesterol and blood pressure decrease after Cupping Therapy in an elderly group in Wedomartani Village, Sleman Yogyakarta Indonesia. Method: quantitative type with pseudo experimental research design. Pre-test and Post-test nonequivalent control group. The treatment group was given wet cupping therapy, while the control group was given dry cupping. Respondents were selected using a purposive sampling technique. Blood pressure measurement uses a digital tensimeter and cholesterol measurement using a digital blood cholesterol level measuring device. Results: The elderly blood cholesterol levels after the average therapy decreased 42.89 mg/dl, whereas blood cholesterol levels in the control group decreased an average of 20.95 mg/dl. The blood pressure of the elderly systole in the intervention group after the average therapy decreased 10.74 mmHg, whereas the control group's systole blood pressure decreased an average of 1.58 mmHg. Diastole blood pressure in the treatment group decreased 3,48 mmHg, in the diastole blood pressure control group increased 5,26 mmHg. Conclusions: Wet and dry cupping are both significantly associated with decreased blood cholesterol and blood pressure levels, but wet cupping lowers blood cholesterol levels by a mean difference of 21.95 mg/dl and mean systole blood pressure of 3.74 mmHg and pressure diastole blood difference of mean 9.1 mmHg. Accompanied with nutrition transition, non-HDL-C levels of individuals in Asian countries has increased rapidly, which has caused the global epicenter of nonoptimal cholesterol to shift from Western countries to Asian countries. Thus, it is critical to underline major genetic and dietary determinants. In the current study of 2,330 Chinese individuals, genetic risk scores (GRSs) were calculated for total cholesterol (TC; GRSTC, 57 SNPs), LDL-C (GRSLDL-C, 45 SNPs), and HDL-C (GRSHDL-C, 65 SNPs) based on SNPs from the Global Lipid Genetics Consortium study. Cholesterol intake was estimated by a 74-item food-frequency questionnaire. Associations of dietary cholesterol intake with plasma TC and LDL-C strengthened across quartiles of the GRSTC (effect sizes: −0.29, 0.34, 2.45, and 6.47; Pinteraction = 0.002) and GRSLDL-C (effect sizes: −1.35, 0.17, 5.45, and 6.07; Pinteraction = 0.001), respectively. Similar interactions with non-HDL-C were observed between dietary cholesterol and GRSTC (Pinteraction = 0.001) and GRSLDL-C (Pinteraction = 0.004). The adverse effects of GRSTC on TC (effect sizes across dietary cholesterol quartiles: 0.51, 0.82, 1.21, and 1.31; Pinteraction = 0.023) and GRSLDL-C on LDL-C (effect sizes across dietary cholesterol quartiles: 0.66, 0.52, 1.12, and 1.56; Pinteraction = 0.020) were more profound in those having higher cholesterol intake compared with those with lower intake. Our findings suggest significant interactions between genetic susceptibility and dietary cholesterol intake on plasma cholesterol profiles in a Chinese population.

Aqueous extracts of liver mitochondria were administered to rats by intraperitoneal, subcutaneous, and oral routes, and C14-acetate or mevalonate was injected intraperitoneally 3 hours later. Cholesterol synthesis in vivo from C14-acetate was depressed by the inhibitory substance in the mitochondrial extracts. Synthesis from C14-mevalonate was not inhibited.Active extracts were prepared from livers of rat, rabbit, pig, and sheep. An inhibitory substance is present in blood serum and administration of active mitochondrial extracts depressed blood cholesterol levels. Pending its identification, the active principle has been provisionally termed I.C.S. (inhibitor of cholesterol synthesis).

Whole egg is a food source of dietary cholesterol and inconsistent research findings exist about the effect of dietary cholesterol from whole egg on blood cholesterol concentration. We assessed the effect of co-consuming cooked whole egg (CWE) on dietary cholesterol absorption from two randomized-crossover studies. For study 1, 16 men consumed raw vegetables with no egg, 75 g CWE, or 150 g CWE. For study 2, 17 women consumed cooked vegetables with no egg or 100 g CWE. Triacylglycerol-rich lipoprotein fractions (TRL) were isolated from collected blood. In study 1, total-cholesterol areas under the curve (AUC)0–10h in TRL were not different but triacylglycerol AUC0–10h in TRL was greater for 150 g CWE vs. 75 g CWE and no egg. Similarly, in study 2, total-cholesterol AUC0–10h in TRL was not different but triacylglycerol AUC0–10h in TRL was greater for 100 g CWE vs. no egg. In both studies, whole egg consumption did not affect plasma total-cholesterol AUC0–10h, while triacylglycerol AUC0–10h was increased. These results suggest that the dietary cholesterol in whole egg was not well absorbed, which may provide mechanistic insight for why it does not acutely influence plasma total-cholesterol concentration and is not associated with longer-term plasma cholesterol control.

Some seaweeds exert antioxidant and hypocholesterolaemic properties. The effects of diets including restructured meats (RM) containing Wakame (W) or Nori (N) algae on arylesterase (AE) activity and lipoprotein concentration and composition were tested. In the present study, six groups of ten male growing Wistar rats each were fed a mix of 85 % AIN-93M diet and 15 % freeze-dried RM for 35 d. The control group (C) consumed control RM, the W and N groups consumed RM with 5 % W and 5 % N, respectively. The cholesterol-enriched C (CC), W (CW) and N (CN) groups consumed their corresponding basal diets with supplementary cholesterol (2·43 %) and cholic acid (0·49 %). Cholesterol in the diet induced lower (P < 0·001) growth ratios. Both W and N diets significantly increased AE activity. VLDL-cholesterol values were lower in N rats than in W rats. AE activity increased (P < 0·001) in CC and CW rats but not in CN rats compared with their corresponding counterparts. AE was lower (P < 0·05) in the CN group than in the CC and CW groups. The CN diet partially blocked (P < 0·001) the hypercholesterolaemic induction observed in CC and CW diets and reduced TAG levels (at least P < 0·05) with respect to those of CC rats. Although dietary cholesterol supplementation increased total cholesterol, VLDL-cholesterol and (intermediate-density lipoprotein+LDL)-cholesterol (all P < 0·001) in all rats, the CN diet moderately improved the lipoprotein profile of hypercholesterolaemic rats. Changes in AE activity and plasma cholesterol in CN rats but not in CW rats suggest a possible relationship between the two parameters. It is concluded that inclusion of RM enriched with N may be used in hypercholesterolaemic diets to improve lipoprotein metabolism.

Cholesterol homeostasis is maintained through concerted action of the SREBPs and LXRs. Here, we report that RNF145, a previously uncharacterized ER membrane ubiquitin ligase, participates in crosstalk between these critical signaling pathways. RNF145 expression is induced in response to LXR activation and high-cholesterol diet feeding. Transduction of RNF145 into mouse liver inhibits the expression of genes involved in cholesterol biosynthesis and reduces plasma cholesterol levels. Conversely, acute suppression of RNF145 via shRNA-mediated knockdown, or chronic inactivation of RNF145 by genetic deletion, potentiates the expression of cholesterol biosynthetic genes and increases cholesterol levels both in liver and plasma. Mechanistic studies show that RNF145 triggers ubiquitination of SCAP on lysine residues within a cytoplasmic loop essential for COPII binding, potentially inhibiting its transport to Golgi and subsequent processing of SREBP-2. These findings define an additional mechanism linking hepatic sterol levels to the reciprocal actions of the SREBP-2 and LXR pathways.
