Bull fertility is an important economic trait in sustainable cattle production, as infertile or subfertile bulls give rise to large economic losses. Current methods to assess bull fertility are tedious and not totally accurate. The massive collection of functional data analyses, including genomics, proteomics, metabolomics, transcriptomics, and epigenomics, helps researchers generate extensive knowledge to better understand the unraveling physiological mechanisms underlying subpar male fertility. This review focuses on the sperm phenomes of the functional genome and epigenome that are associated with bull fertility. Findings from multiple sources were integrated to generate new knowledge that is transferable to applied andrology. Diverse methods encompassing analyses of molecular and cellular dynamics in the fertility-associated molecules and conventional sperm parameters can be considered an effective approach to determine bull fertility for efficient and sustainable cattle production. In addition to gene expression information, we also provide methodological information, which is important for the rigor and reliability of the studies. Fertility is a complex trait influenced by several factors and has low heritability, although heritability of scrotal circumference is high and that it is a known fertility maker. There is a need for new knowledge on the expression levels and functions of sperm RNA, proteins, and metabolites. The new knowledge can shed light on additional fertility markers that can be used in combination with scrotal circumference to predict the fertility of breeding bulls. This review provides a comprehensive review of sperm functional characteristics or phenotypes associated with bull fertility.

Studies of nonhuman primate communication are often motivated by the desire to shed light on the evolution of speech. In contrast to human speech, the vocal repertoires of nonhuman primates are evolutionarily highly conserved. Within species-specific constraints, calls may vary in relation to the internal state of the caller or social experience. Receivers can use signalers’ calls to predict upcoming events or behavioral dispositions. Yet nonhuman primates do not appear to express or comprehend communicative or informative intent. Signalers are sensitive to the relation between their own actions and receivers’ responses, and thus, signaling behavior can be conceived as goal directed. Receivers’ ability to integrate information from multiple sources renders the system flexible and powerful. Researchers who take a linguistic or biological perspective on nonhuman primate communication should be aware of the strengths and limitations of their approaches. Both benefit from a focus on the mechanisms that underpin signaling and responses to signals.

Diabetes mellitus is a group of heterogeneous metabolic disorders characterized by chronic hyperglycaemia mainly due to pancreatic β cell death and/or dysfunction, caused by several types of stress such as glucotoxicity, lipotoxicity and inflammation. Different patho-physiological mechanisms driving β cell response to these stresses are tightly regulated by microRNAs (miRNAs), a class of negative regulators of gene expression, involved in pathogenic mechanisms occurring in diabetes and in its complications. In this review, we aim to shed light on the most important miRNAs regulating the maintenance and the robustness of β cell identity, as well as on those miRNAs involved in the pathogenesis of the two main forms of diabetes mellitus, i.e., type 1 and type 2 diabetes. Additionally, we acknowledge that the understanding of miRNAs-regulated molecular mechanisms is fundamental in order to develop specific and effective strategies based on miRNAs as therapeutic targets, employing innovative molecules.

AbstractTrichoderma reesei is an ascomycete fungus known for its capability to secrete high amounts of extracellular cellulose- and hemicellulose-degrading enzymes. These enzymes are utilized in the production of second-generation biofuels and T. reesei is a well-established host for their production. Although this species has gained considerable interest in the scientific literature, the sugar transportome of T. reesei remains poorly characterized. Better understanding of the proteins involved in the transport of different sugars could be utilized for engineering better enzyme production strains. In this study we aimed to shed light on this matter by characterizing multiple T. reesei transporters capable of transporting various types of sugars. We used phylogenetics to select transporters for expression in Xenopus laevis oocytes to screen for transport activities. Of the 18 tested transporters, 8 were found to be functional in oocytes. 10 transporters in total were investigated in oocytes and in yeast, and for 3 of them no transport function had been described in literature. This comprehensive analysis provides a large body of new knowledge about T. reesei sugar transporters, and further establishes X. laevis oocytes as a valuable tool for studying fungal sugar transporters.

The evolution of salt tolerance is interesting for several reasons. First, since salt-tolerant plants (halophytes) employ several different mechanisms to deal with salt, the evolution of salt tolerance represents a fascinating case study in the evolution of a complex trait. Second, the diversity of mechanisms employed by halophytes, based on processes common to all plants, sheds light on the way that a plant’s physiology can become adapted to deal with extreme conditions. Third, as the amount of salt-affected land increases around the globe, understanding the origins of the diversity of halophytes should provide a basis for the use of novel species in bioremediation and conservation. In this review we pose the question, how many times has salt tolerance evolved since the emergence of the land plants some 450–470 million years ago? We summarise the physiological mechanisms underlying salt-tolerance and provide an overview of the number and diversity of salt-tolerant terrestrial angiosperms (defined as plants that survive to complete their life cycle in at least 200 mM salt). We consider the evolution of halophytes using information from fossils and phylogenies. Finally, we discuss the potential for halophytes to contribute to agriculture and land management and ask why, when there are naturally occurring halophytes, it is proving to be difficult to breed salt-tolerant crops.

This article deals with the textual legacy of Dutch Brazil, in particular the ethnographic descriptions in one of the most popular works about the colony: Barlaeus’ Rerum per Octennium in Brasilia et alibi nuper gestarum. Barlaeus never set foot in Brazil, but was an important Dutch intellectual authority in the seventeenth century. To compose the Rerum per Octennium, he relied on a wide variety of available sources, not only firsthand observations, but also classical, biblical and other contemporary sources. From these, he made a careful selection to produce his descriptions. Recent research shows that the Dutch participated in networks of knowledge and imagination as well as in a more familiar early modern trading network. This article reveals that Barlaeus’ descriptions not only circulated as knowledge, but also produced new knowledge. The Rerum soon became one of the standard works about the colony due to the importance of its author and its composition. Furthermore, the article discusses the rhetorical techniques used in some selected descriptions in order to shed light upon the strategies Barlaeus used in his discourse on the strange reality of the New World. For example, his ethnographic descriptions employed parallel customs or events from the classical Antiquity or the Bible. In these comparisons he displays both his intellectual capacities and shows his desire to comprehend this exotic reality.

Macadamia is the only Australian native crop tree. Unlike some model crop trees like apple or peach trees, the understanding of the physiological mechanisms regulating bud break and flowering in macadamia has never been investigated. We have recently attempted to fill this gap by improving the genetic resources available for macadamia. We identified several components involved in flowering and bud break, including the FLOWERING LOCUS T (FT). FT is a mobile protein, synthesized in the leaves which triggers flowering in terminal and axillary buds. In macadamia, we found two FT-related genes, FTa and FTb namely. FTa and FTb are differentially regulated; FTa being more abundant in axillary buds than leaves, which is unexpected based on the studies performed in model plants. Analysis of the promoter sequences of FTa and FTb, revealed that the FTa regulation may be closer to FT regulation in the model plant Arabidopsis thaliana. Gene expression in buds revealed that the expression of TERMINAL FLOWER1 (TFL1), a gene previously reported to inhibit flowering, was induced prior to bud break and flowering. The patterns of marker gene expression in buds highlighted that the bud dormancy decreased before May, after which flowering induction occurs. Altogether these results shed light on the regulation of bud break and flowering in macadamia and demonstrate that some regulatory mechanisms may be different from other models.

Uruguay is one of the main exporters of beef and dairy products, and cattle production is one of the main economic sectors in this country. Rotavirus A (RVA) is the main pathogen associated with neonatal calf diarrhea (NCD), a syndrome that leads to significant economic losses to the livestock industry. The aims of this study are to determine the frequency of RVA infections, and to analyze the genetic diversity of RVA strains in calves in Uruguay. A total of 833 samples from dairy and beef calves were analyzed through RT-qPCR and sequencing. RVA was detected in 57.0% of the samples. The frequency of detection was significantly higher in dairy (59.5%) than beef (28.4%) calves (p < 0.001), while it did not differ significantly among calves born in herds that were vaccinated (64.0%) or not vaccinated (66.7%) against NCD. The frequency of RVA detection and the viral load were significantly higher in samples from diarrheic (72.1%, 7.99 log10 genome copies/mL of feces) than non-diarrheic (59.9%, 7.35 log10 genome copies/mL of feces) calves (p < 0.005 and p = 0.007, respectively). The observed G-types (VP7) were G6 (77.6%), G10 (20.7%), and G24 (1.7%), while the P-types were P[5] (28.4%), P[11] (70.7%), and P[33] (0.9%). The G-type and P-type combinations were G6P[11] (40.4%), G6P[5] (38.6%), G10P[11] (19.3%), and the uncommon genotype G24P[33] (1.8%). VP6 and NSP1-5 genotyping were performed to better characterize some strains. The phylogenetic analyses suggested interspecies transmission, including transmission between animals and humans.

What are the effects of party defections on the attitudes of politicians who remain loyal to the party? We answer by combining multiple sources of data into a comprehensive novel data set on parliamentary party switching, to estimate how this affects the perceived distance between a politician and his party. Focusing on the theory of cognitive dissonance and the black sheep effect, we hypothesize that politicians perceive themselves closer to their parties when those parties recently suffered defections. The effect should be greater among incumbent politicians as they directly experience divisions, but also among officials dissatisfied with the leadership as their dissonance should be stronger. Statistical analyses of data from two elite surveys, on a sample of 13,256 politicians belonging to 92 parties that ran in 28 elections held between 2005 and 2015 in 14 countries, provide support for our hypotheses and shed light on the consequences of intra-party defections.

In Japan, national outbreak of novel influenza A (H1N1) triggered serious social disruption. The public perceived overwhelming fear and their behaviors were severely affected. Countless events were put off, with massive economic losses due to activity cancellations. The heightened fear may have been a mixture of risk communication consequences, geographic characteristics (island nation), and culture-bound fear related to shimaguni konjo, or “island mentality”; according to a Japanese cultural norm, the “outside” is considered “impure” and is often covered-up, criticized, and avoided. These consequences shed light on cultural effects on collective behaviors, along with the importance of risk communication strategies.

AbstractWe test the interpretation of pronominal forms in L2 speakers of English whose L1 is Spanish. Previous research on learners of nonnull subject languages has shown conflicting results. The aim of the present study is to reconcile previous evidence and shed light on the factors that determine learners’ difficulty to interpret pronominal forms in the L2. In six comprehension experiments, we found that intermediate L2 speakers did not show increased difficulty compared to native speakers in integrating multiple sources of information (syntactic, discourse, pragmatic) to resolve ambiguous pronouns in intrasentential anaphora and cataphora conditions. However, we also found that when two referents with equal prominence are introduced using a conjoined noun phrase in the preceding context, the learner’s performance is significantly different than the performance of the native speakers, both in intrasentential and intersentential anaphora. We suggest that L2 speakers may encounter difficulties evaluating the salience of the antecedents during pronoun resolution.
