Volatility of the petroleum market has renewed research into butanol as an alternate fuel. In order to increase the selectivity for butanol during glycerol fermentation with Clostridium pasteurianum, butyric acid can be added to the medium. In this manuscript, different methods of extracellular butyric acid addition are explored, as well as self-generation of butyric acid fermented from sugars in a co-substrate strategy. Molasses was used as an inexpensive sugar substrate, and the optimal molasses to glycerol ratio was found to allow the butyric acid to be taken back up into the cells and increase the productivity of butanol from all carbon sources. When butyric acid is added directly into the media, there was no significant difference between chemically pure butyric acid, or butyric acid rich supernatant from a separate fermentation. When low concentrations of butyric acid (1 or 2 g/L) are added to the initial media, an inhibitory effect is observed, with no influence on the butanol selectivity. However, when added later to the fermentation, over 1 g/L butyric acid is taken into the cells and increased the relative carbon yield from 0.449 to 0.519 mols carbon in product/mols carbon in substrate. An optimized dual substrate fermentation strategy in a pH-controlled reactor resulted in the relative carbon yield rising from 0.439 when grown on solely glycerol, to 0.480 mols C product/mols C substrate with the dual substrate strategy. An additional benefit is the utilization of a novel source of sugars to produce butanol from C. pasteurianum. The addition of butyric acid, regardless of how it is generated, under the proper conditions can allow for increased selectivity for butanol from all substrates.

The lignocellulosic biomass comprises three main components: cellulose, hemicellulose, and lignin. Degradation and conversion of these three components are attractive to biotechnology. This study aimed to prospect fungal lignocellulolytic enzymes with potential industrial applications, produced through a temporal analysis using Hymenaea courbaril and Tamarindus indica seeds as carbon sources. α-L-arabinofuranosidase, acetyl xylan esterase, endo-1,5-α-L-arabinanase, β-D-galactosidase, β-D-glucosidase, β-glucanase, β-D-xylosidase, cellobiohydrolase, endoglucanase, lichenase, mannanase, polygalacturonase, endo-1,4-β-xylanase, and xyloglucanase activities were determined. The enzymes were produced for eight filamentous fungi: Aspergillus fumigatus, Trametes hirsuta, Lasiodiplodia sp., two strains of Trichoderma longibrachiatum, Neocosmospora perseae, Fusarium sp. and Thermothelomyces thermophilus. The best producers concerning enzymatic activity were T. thermophilus and T. longibrachiatum. The optimal conditions for enzyme production were the media supplemented with tamarind seeds, under agitation, for 72 h. This analysis was essential to demonstrate that cultivation conditions, static and under agitation, exert strong influences on the production of several enzymes produced by different fungi. The kind of sugarcane, pretreatment used, microorganisms, and carbon sources proved limiting sugar profile factors.

We constructed a novel design integrating the administration of a clinical self-assessment scale with simultaneous acquisition of functional Magnetic Resonance Imaging (fMRI), aiming at cross-validation between psychopathology evaluation and neuroimaging techniques. We hypothesized that areas demonstrating differential activation in two groups of patients (the first group exhibiting paranoid delusions in the context of paranoid schizophrenia—SCH—and second group with a depressive episode in the context of major depressive disorder or bipolar disorder—DEP) will have distinct connectivity patterns and structural differences. Fifty-one patients with SCH (n = 25) or DEP (n = 26) were scanned with three different MRI sequences: a structural and two functional sequences—resting-state and task-related fMRI (the stimuli represent items from a paranoid-depressive self-evaluation scale). While no significant differences were found in gray matter volumes, we were able to discriminate between the two clinical entities by identifying two significant clusters of activations in the SCH group—the left Precuneus (PreCu) extending to the left Posterior Cingulate Cortex (PCC) and the right Angular Gyrus (AG). Additionally, the effective connectivity of the middle frontal gyrus (MFG), a part of the Dorsolateral Prefrontal Cortex (DLPFC) to the Anterior Insula (AI), demonstrated a significant difference between the two groups with inhibitory connection demonstrated only in SCH. The observed activations of PreCu, PCC, and AG (involved in the Default Mode Network DMN) might be indirect evidence of the inhibitory connection from the DLPFC to AI, interfering with the balancing function of the insula as the dynamic switch in the DMN. The findings of our current study might suggest that the connectivity from DLPFC to the anterior insula can be interpreted as evidence for the presence of an aberrant network that leads to behavioral abnormalities, the manifestation of which depends on the direction of influence. The reduced effective connectivity from the AI to the DLPFC is manifested as depressive symptoms, and the inhibitory effect from the DLPFC to the AI is reflected in the paranoid symptoms of schizophrenia.

SummaryStudies on the immunomodulatory activities of ruminant milk and colostral whey fractions were undertaken. By comparing with boiled colostral whey in a preliminary experiment, a putative heat-labile immunostimulatory factor for antibody responses was found to be present in ovine colostral whey. Studies were then undertaken in sheep in which the efferent prefemoral lymphatic ducts were cannulated bilaterally, and immune responses in the node were measured following subcutaneous injection in the flank fold of whey protein preparations of various purities. A significant sustained decline of efferent lymphocyte output was observed following injection with autologous crude milk whey or colostral whey preparations, but no changes were observed in interferon-gamma levels in lymph plasma. Two bovine milk whey fractions (lactoperoxidase and lactoferrin) of high purity were compared in bilaterally cannulated sheep. A transient decline over the first 6 h was seen in the efferent lymphocyte output and lymph flow rate after injection of both fractions. A significant difference was seen between the two fractions in interferongamma levels in lymph at 6 h after injection. However, no significant changes in the proportion of the various efferent lymphocyte phenotypes were seen following either treatment. Whereas both fractions showed a significant inhibitory effect in a dose-dependent manner on the proliferative response of T lymphocytes, but not B lymphocytes, to mitogenic stimulation in vitro, no similar changes were seen following in vivo stimulation with these two fractions.

Rosa damascena is very important for essential oil production, medicinal properties and it is also widely cultivated as a garden rose. The Rose species is mainly propagated by stem cutting. In the present study, the effect of different levels of 0, 500 and 1,000 mg l-1 (quick dip method for 20 s) of indole butyric acid (IBA) on the rooting of 12 wild genotypes (including: Kurdistan 1 to Kurdistan 12) of R. damascena was investigated. The results show that the rooting ability of R. damascena differs significantly between the twelve genotypes. The highest rooting (79.56%) and callus production (69.08%), number of roots (8.33), root fresh and dry weights (361.80 and 244.74 mg, respectively) were recorded in Kurdistan 5 genotype with 1,000 mg l-1 IBA. The maximum root length (5.84 cm) was observed in Kurdistan 5 genotype with 500 mg l-1 IBA that showed a significant difference compared to the control treatment (0.96 cm). The highest number of leaves per bud (7.33 at 500 mg l-1 IBA) and number of buds (5.00 at 1,000 mg l-1 IBA) were recorded in Kurdistan 1 genotype. The current study demonstrated that the different genotypes of R. damascena were in a difficult-toroot state, which suggests that cutting treatment with 1,000 mg l-1 IBA overcame the problem of the difficult-to-root state, and it can also enhance the rooting percentage in the studied genotypes.

An inhibitory effect of various concentrations (0.0, 0.5, 1.0, 1.5 and 2,0%) of mint (Mentha piperita L.) and caraway (Carvum carvi L.) on the growth of A. fumigatus, A. flavus and A. ochraceus was examined during 10 days of cultivation in YES medium at temperature of 25?C. Mint showed stronger inhibitory effect than caraway. Total dry weight (g/l) after 10 days of the growth of A. fumigatus in YES medium with 0.5% of mint decreased by about 95%, A. flavus by 97% and A. ochraceus by about 82%. Addition of higher concentrations of mint (1.0, 1.5 and 2.0%) reduced the growth of all tested species. It was poor and hardly visible. pH values of the media increased with the increase of mint concentrations. A. fumigatus showed the highest sensitivity towards caraway and A. flavus the lowest. Total dry weight (g/l) after 10 days of growth of A. fumigatus in medium with 0.5% of caraway decreased by about 72% in comparison to the control. In media with higher concentrations of caraway, its growth was found to be very poor. Concentration of 1.0% of caraway reduced A. flavus growth by 15% and of 1.5% by 92%, in regard to the control. In medium with 2.0% of caraway the growth of A. flavus was observed as poor and hardly visible. The growth of A. ochraceus in medium with 0.5% of caraway decreased by about 85% comparing with control and further decrease was noticed by the increase of concentrations. In medium with 1.5% of caraway a reduction of about 95% of growth was found and under 2.0% of caraway it was poor. pH of the media also increased with the increase of caraway concentrations. Applied concentrations of mint and caraway inhibited completely the production of AB1 by A. flavus.

This preliminary study again provides evidence that it is the method and not the media that matters the most in learning effectiveness. The present work examines classroom community in order to determine how sense of community differs between students enrolled in traditional face-to-face and those enrolled in asynchronous learning network (ALN) courses. Subjects consist of 326 adult learners who were enrolled in a mix of 14 undergraduate and graduate courses at two urban universities. As operationalized by the Sense of Classroom Community Index (SCCI), there appears no significant difference in classroom community between the two groups of subjects. However, a discriminant analysis shows a significant overall difference in community structure between the two groups. Variations between groups on feelings of similarity of needs, recognition, importance of learning, connectedness, friendship, thinking critically, safety, acceptance, group identity, and absence of confusion are the characteristics contributing mostly to this difference in learning effectiveness.

Background: Butyric acid, a major short-chain fatty acid (SCFA), has an important role in the microbiota–gut–brain axis and brain function. This study investigated the role of butyric acid in treatment response in drug-naïve first episode schizophrenia.Methods: The study recruited 56 Chinese Han schizophrenia inpatients with normal body weight and 35 healthy controls. Serum levels of butyric acid were measured using Gas Chromatography-Mass Spectrometer (GC-MS) analysis at baseline (for all participants) and 24 weeks after risperidone treatment (for patients). Clinical symptoms were measured using the Positive and Negative Syndrome Scale (PANSS) for patients at both time points.Results: At baseline, there was no significant difference in serum levels of butyric acid between patients and healthy controls (p = 0.206). However, there was a significant increase in serum levels of butyric acid in schizophrenia patients after 24-week risperidone treatment (p = 0.030). The PANSS total and subscale scores were decreased significantly after 24-week risperidone treatment (p's < 0.001). There were positive associations between baseline serum levels of butyric acid and the reduction ratio of the PANSS total and subscale scores after controlling for age, sex, education, and duration of illness (p's < 0.05). Further, there was a positive association between the increase in serum levels of butyric acid and the reduction of the PANSS positive symptoms subscale scores (r = 0.38, p = 0.019) after controlling for potential confounding factors.Conclusions: Increased serum levels of butyric acid might be associated with a favorable treatment response in drug-naïve, first episode schizophrenia. The clinical implications of our findings were discussed.

In Indonesia, 93.5% of the population aged ≥10 years still consume less fruits and vegetables 5 servings per day for 7 days a week (RISKESDAS, 2013).  The preliminary study conducted at SDN Mekarjaya I shows that the level of knowledge of school children about vegetables and fruits by 60% is still low and 93.3% of vegetable and fruit consumption is still lacking. In addition, SDN Mekarjaya I has never been used as a place of prior research. Therefore, it is necessary to conduct research on the knowledge of fruit vegetables in the fourth grade students of SDN Mekarjaya I. One of them is by doing counseling with the media of puzzle game. This research was conducted to know the increase of knowledge about vegetables and fruits before and after given counseling with puzzle game media in fourth grade students of SDN Mekarjaya I. This research was conducted by Pre-exsperiment method using "One group pretest and posttest" research design. Sampling by purposive sampling is 66 people. The statistical test used is paired sample t-test. Based on the results of the analysis shows that there is a significant difference of knowledge where p-value 0.000 or p <0,05 means there is a meaningful difference between before and after given counseling with puzzle game media.This puzzle game media can be developed by the teachers as a medium of learning in the classroom so that students are more motivated in following the learning process and make students do not feel bored

The study was set back by the low the creativity of children age 5-6 years in the Adzkia Air Bangis. one of the key factors this is suspected because it is not suitable or the media used by the teacher is not interestig. This study aims to determine the effectiveness of the application of rock painting activities in developing the creativity of children aged 5-6 years in kindergarten Adzkia Air Bangis. the type of research is quantitative usng pseudo-experiment methods (quasi experiment) with pre-test design and post-test. The research population is 55 people. While the sampel 10 children. Sampling retrieval techniques are used sampling samples. The data uses t-test samples samples. As a result of the coclusion to the data collaction and management, 1) before being given an treatment (pre-test) the development of children’s creativity can be categorized as starting to develop but has not developed as expected. This is based on the result of research by researchers at Adzkia Air Bangis Kindergarten 2) after being given treatment (post-test) the development of chidren’s creativity can be categorized as developing according to expectations. 3) there is significant difference between the result of the pre-test and post-test after doing the paired sampe t-test sig 0,000 < 0,05.  Advice for teachers helping teachers by using experimental learning methods to develop the creativity of children aged 5-6 years trough rock painting activities.

The effect of different carbon sources on morphology and cellulase and xylanase production ofPenicillium echinulatumwas evaluated in this work. Among the six carbon sources studied, cellulose and sugar cane bagasse were the most suitable for the production of filter paper activity, endoglucanases, xylanases, andβ-glucosidases. However, sucrose and glucose showedβ-glucosidase activities similar to those obtained with the insoluble sources. The polyacrylamide gels proved the enzymatic activity, since different standards bands were detected in the media mentioned above. Regarding morphology, it was observed that the mycelium in a dispersed form provided the greatest enzymatic activity, possibly due to greater interaction between the substrate and hyphae. These data are important in understanding the physiology of fungi and could contribute to obtaining enzyme with potential application in the technology of second generation ethanol.
