The sensitivity to thiacloprid and lambda-cyhalothrin ingested from prey organisms was studied in Coccinella septempunctata and Harmonia axyridis, since the effect of ingestion exposure to these insecticides is unknown in these species. All developmental stages of the ladybirds were fed on Acyrthosiphon pisum treated with half or full field rate of the insecticides. Almost all instars were killed within 3 h of the start of ingestion of lambda-cyhalotrin-treated prey. The action of thiacloprid was more extended in time in both coccinellids and more variable between their instars. Reducing the field rate of lambda-cyhalothrin seems to have no practical value for the survival of either of the coccinellid species. Contrastingly, using half instead of the full field rate of thiacloprid may enhance the chances of survival in L1, L2, and L4 larvae of both species. Of all developmental stages tested, the survival dynamics of the adults of either species are closest to one another, whereas the apparent difference in the species response to the dose rate of thiacloprid was found in the L4 stage.

The effect of continuous, intensive grazing by cattle on aphid populations was examined in the first growth cycle of ‘Alfagraze’ and ‘Apollo’ alfalfa which are tolerant and not tolerant to grazing, respectively. Populations were almost entirely pea aphid, Acyrthosiphon pisum (Harris). The effect of grazing on aphid populations was examined in small plot exclusion studies in 1991 and 1992, and the effects of grazing and use of an early insecticide application with ≤7 day grazing restriction were examined in large plot exclusion trials in 1993 and 1994. Grazing reduced aphid populations by 66% to 90% when numbers exceeded ~1 aphid per stem. Populations were not significantly reduced by grazing when numbers did not exceed 1 per stem. Permethrin reduced aphid numbers for up to 7 wks and was more effective than carbofuran. Effects of grazing were similar regardless of insecticide usage. Coccinellid beetle populations in 1993 and 1994 were composed of Coccinella septempunctata L., Harmonia axyridis (Pallas), and Coleomegilla maculata DeGeer. Coccinellid numbers were much lower in grazed than ungrazed plots when aphid numbers were reduced by grazing in 1993, but were not greatly affected by grazing in 1994 when aphid numbers were low. Coccinellid numbers paralleled trends in aphid numbers. Continuous, intensive grazing or the early application of an insecticide at a low rate followed by grazing were effective approaches for suppressing pea aphid in alfalfa.

AbstractDespite the central role of hemocytes in crustacean immunity, the process of hemocyte differentiation and maturation remains unclear. In some decapods, it has been proposed that the two main types of hemocytes, granular cells (GCs) and semigranular cells (SGCs), differentiate along separate lineages. However, our current findings challenge this model. By tracking newly produced hemocytes and transplanted cells, we demonstrate that almost all the circulating hemocytes of crayfish belong to the GC lineage. SGCs and GCs may represent hemocytes of different developmental stages rather than two types of fully differentiated cells. Hemocyte precursors produced by progenitor cells differentiate in the hematopoietic tissue (HPT) for 3 ~ 4 days. Immature hemocytes are released from HPT in the form of SGCs and take 1 ~ 3 months to mature in the circulation. GCs represent the terminal stage of development. They can survive for as long as 2 months. The changes in the expression pattern of marker genes during GC differentiation support our conclusions. Further analysis of hemocyte phagocytosis indicates the existence of functionally different subpopulations. These findings may reshape our understanding of crustacean hematopoiesis and may lead to reconsideration of the roles and relationship of circulating hemocytes.

AbstractIn the current study, we investigated the functional response of Harmonia axyridis adults and larvae foraging on Acyrthosiphon pisum nymphs at temperatures between 15 and 35 °C. Logistic regression and Roger’s random predator models were employed to determine the type and parameters of the functional response. Harmonia axyridis larvae and adults exhibited Type II functional responses to A. pisum, and warming increased both the predation activity and host aphid control mortality. Female and 4th instar H. axyridis consumed the most aphids. For fourth instar larvae and female H. axyridis adults, the successful attack rates were 0.23 ± 0.014 h−1 and 0.25 ± 0.015 h−1; the handling times were 0.13 ± 0.005 h and 0.16 ± 0.004 h; and the estimated maximum predation rates were 181.28 ± 14.54 and 153.85 ± 4.06, respectively. These findings accentuate the high performance of 4th instar and female H. axyridis and the role of temperature in their efficiency. Further, we discussed such temperature-driven shifts in predation and prey mortality concerning prey-predator foraging interactions towards biological control.

The basic biological action of radiation must initially involve particular cells. The sensitivity of various cells differs but some of the differences may be due to a dose-rate effect. When cells are irradiated with doses in the lethal range, inhibition of cell division occurs. A notable feature of mitotic inhibition is that it depends not only on dose but on dose rate. Chromosome breaks produced in cells may be a visible manifestation of an interruption in chemical synthesis produced by the passage of ionizing particles. The effect of oxygen is apparently of primary importance in biological changes produced by irradiation. Almost all are decreased in its absence. The current trend in research on the cellular level is towards determining the more obscure injuries which cannot be seen under the microscope.

Ribosomal proteins (RPs) are indispensable in ribosome biogenesis and protein synthesis, and play a crucial role in diverse developmental processes. In the present study, we carried out a comprehensive analysis of RPs in chordates and examined the expression profiles of the complete set of 92 cytoplasmic RP genes in Nile tilapia. The RP genes were randomly distributed throughout the tilapia genome. Phylogenetic and syntenic analyses revealed the existence of duplicated RP genes from 2R (RPL3, RPL7, RPL22 and RPS27) and 3R (RPL5, RPL19, RPL22, RPL41, RPLP2, RPS17, RPS19 and RPS27) in tilapia and even more from 4R in common carp and Atlantic salmon. The RP genes were found to be expressed in all tissues examined, but their expression levels differed among different tissues. Gonadal transcriptome analysis revealed that almost all RP genes were highly expressed, and their expression levels were highly variable between ovaries and testes at different developmental stages in tilapia. No sex- and stage-specific RP genes were found. Eleven RP genes displayed sexually dimorphic expression with nine higher in XY gonad and two higher in XX gonad at all stages examined, which were proved to be phenotypic sex dependent. Quantitative real-time PCR and immunohistochemistry ofRPL5b and RPL24 were performed to validate the transcriptome data. The genomic resources and expression data obtained in this study will contribute to a better understanding of RPs evolution and functions in chordates.

A survey was conducted in seven districts namely Bogura, Rajshahi, Jashore, Narsingdi, Cumilla, Jamalpur and Gazipur to assess the present status of the usage of pesticides and ripening chemical in major vegetable crops such as tomato, brinjal, country bean and bitter gourd. A total of 280 respondents having 40 respondents from each district were selected randomly for the study. The maximum number of vegetable growers belonged to the age group of 21-40, which is about 50%. About 41% and 25% of farmers accomplished their primary and secondary education in the study areas. Tomato fruit had the highest yield (27.74 tha-1) whereas the highest gross margin was attained from country bean 4,06,832 Tk.ha-1. Almost all of the vegetable growers were used synthetic pesticides (chemical group of Cypermethrin, Emamectin Benzoate, Chlorpyriphos, Carbendazim, Lambda Cyhalothrin, Mencozeb etc.) for protecting their crops from pests and most of them used own hand pump sprayer. Farmers of the study areas applied synthetic pesticides frequently with much higher dosages (8-30 times) than the recommendation. Few farmers practice Integrated Pest Management (IPM) for their crops. Seventy five percent farmers had protective measure during insecticide-pesticide spray and about 40% growers felt uncomfortable after hand spray to the crops. Most of the tomato growers in the study areas (Rajshahi and Jashore) were applied Plant Growth Regulator (PGR)/ripening agents mainly Ethephon @ 2500-8000ppm before 1-3 days of harvest in immature green tomato (1-4 times) for uniform color development to get higher price in the early market. Few traders (10-15%) were applied Ethephon in premature vegetables after harvest. It is strongly recommended to use IPM technology for controlling insects and pest and to create awareness regarding pesticides use practice and safety precautions. Bangladesh J. Agril. The harlequin ladybird, Harmonia axyridis , has emerged as a model species for invasion biology, reflecting its remarkable capacity to outcompete native ladybird species when introduced into new habitats. This ability may be associated with its prominent resistance to pathogens and intraguild predation. We recently showed that the constitutive antibacterial activity present in the haemolymph of H. axyridis beetles can be attributed to the chemical defence compound harmonine. Here, we demonstrate that H. axyridis differs from other insects, including the native ladybird Coccinella septempunctata, by reducing rather than increasing the antimicrobial activity of its haemolymph following the injection of bacteria. However, both species produce new or more abundant proteins in the haemolymph, indicating that bacterial challenge induces innate immune responses associated with the synthesis of immunity-related proteins. Our results suggest that H. axyridis beetles can switch from constitutive chemical defence to inducible innate immune responses, supporting hypothesis that inducible antimicrobial peptides protect host beetles against pathogens that survive constitutive defences. These alternative antimicrobial defence mechanisms may reflect a trade-off resulting from fitness-related costs associated with the simultaneous synthesis of harmonine and antimicrobial peptides/proteins.

Micro-PIXE and inductively coupled plasma mass spectrometry (ICP-MS) were applied to elemental distribution analyses in plant root apex which is composed of various types of tissues and cells in different developmental stages. ICP-MS was so sensitive that a large number of elements including Na, Mg, P, S, K, Ca, Mn, Fe, Cu, Zn, Se, Rb, Sr and Cs could be determined quantitatively. These fourteen elements included almost all the essential elements for plant growth. Only a rough estimation, however, could be obtained by ICP-MS for the elemental distribution at the tissue level, by analyzing sections from the root apex. On the other hand, micro-PIXE was effective for detailed mappings of elemental distributions. The images of elemental distributions were obtained for Na, Mg, P, S, K, Ca, Mn, Fe and Zn, corresponding to the microscopic images of the root structures. The localizations of P, K and Zn in some tissues were observed by the mappings. These results indicated that micro-PIXE and ICP-MS have different, but complementary abilities for the investigation of elemental distributions in plant tissues.
