Medicinal compositions based on natural vegetal raw materials and individual substances for prevention and treatment of urolithiasis: substantiation of composition and biological screening

Kidney and urinary tract diseases are an urgent problem of our time. An increase in the level of kidney and urinary tract diseases occurs with a decrease in the quality of life and ecology. Recently, due to changes in nutrition, sedentary lifestyle, and the impact of various adverse environmental factors, the formation of kidney stones has become increasingly common. Numerous metabolic disorders in urolithiasis require the use of various drugs. At the same time, despite the constant expansion of the range of synthetic drugs, herbal drugs occupy a significant place in the treatment and prevention of kidney pathology. Therefore, the creation of combinations of medicinal plant raw materials and individual chemical substances that are effective in urolithiasis is an urgent issue of modern medicine and pharmacy. Purpose. Establishing optimal ratios within constructed combinations for membrane-stabilizing and antioxidant activity as an indirect measure of specific activity The scientific novelty of the study is the design of compositions effective for dissolving urinary calculus. Materials and methods. We used technological methods for designing compositions based on natural plant raw materials and individual chemical substances — indapamide, ketoprofen, and mexidol. Biological methods: the effect of the developed combinations on daphnia was studied. Biological screening characterizes the presence of membrane-stabilizing and antioxidant effects in the developed formulations and can be a preliminary indicator for determining specific activity. Results. It was found that the most effective compositions for the vital activities of daphnia were: Composition 1: Sum of individual dry extracts + Indapamide Composition 2: Sum of individual dry extracts + Ketoprofen Composition 3: Sum of individual dry extracts + Mexidol Conclusion. We have developed the following compositions: 1. Sum of individual phytoextracts (dyer’s madder rhizomes with roots (extractant — 70 % ethanol), Orthosiphon stamineus leaves (extractant — 40 % ethanol), knot grass (extractant — 70 % ethanol), birch buds (extractant — 70 % ethanol), peppermint leaves (extractant — 96 % ethanol), bay laurel leaves (extractant — 40 % ethanol), common licorice rhizomes with roots (extractant — 0.25 % ammonia solution) + indapamide. 2. Sum of individual phytoextracts (dyer’s madder rhizomes with roots (extractant — 70 % ethanol), Orthosiphon stamineus leaves (extractant — 40 % ethanol), knot grass (extractant — 70 % ethanol), birch buds (extractant — 70 % ethanol), peppermint leaves (extractant — 96 % ethanol), bay laurel leaves (extractant — 40 % ethanol), common licorice rhizomes with roots (extractant — 0.25 % ammonia solution) + ketoprofen. 3. They have membrane-stabilizing and antioxidant activity, which has been tested by biological screening on daphnia. The presence of these two types of activity is the initial screening for a subsequent pharmacological study — the determination of specific activity.

The predominance of synthetic and genetically modified foods in the diet of a modern person often leads to disruption of natural metabolic processes and the accumulation of toxic free radicals in the body, which provokes the development of various diseases. Natural antioxidants from medicinal plants can help maintain health and stop the processes of peroxidation. In our opinion, Аstragalus onobrychis can become a promising source of natural antioxidants. The aim of this study was to establish the antioxidant activity in extracts from the herb Astragalus onobrychis. In order to obtain objective and reliable results during our research, we used two methods: the method of titrimetry and spectrophotometry of free radicals. The titrimetric method is based on the chemical interaction between an oxidizing agent (potassium permanganate) and natural antioxidants present in extracts from plant raw materials, since they exhibit the properties of reducing agents. The spectrophotometric method makes it possible to assess the value of antioxidant activity and the duration of the reaction by assessing the interaction of 2,2-diphenyl-1-picrylhydrazil with plant antioxidants. To establish the relationship between the antioxidant activity and the presence of phenolic compounds in the plant, the content of flavonoids and hydroxycinnamic acids in the herb Astragalus onobrychis was determined. As a result of the studies, the total antioxidant activity of extracts from the herb Astragalus onobrychis was established and it was revealed that its value depends on the type of extractant used, since this factor is associated with the content of various groups of natural antioxidants in the extraction. The data obtained indicate the prospects of using the herb Astragalus onobrychis as a source of natural antioxidants.

The development of targeted beverages is a priority for healthy eating. Plant raw materials containing a wide variety of biologically active substances are used for their production. The presence of these ingredients helps to improve many physiological processes in the body, as well as to increase its immune status. The high efficiency of ultrasonic effects on various technological processes was confirmed by numerous studies. Ultrasonic extraction was carried out at t = 20 ? and an ultrasonic wave frequency of 22 kHz. As a result of the research, it was found that the equilibrium state is achieved within 15 minutes with ultrasound exposure. Natural raw materials, their chemical composition and the influence on the human body were studied in the work to develop new beverages recipes. The selected ingredients in a certain quantitative ratio made it possible to obtain beverages with improved bio-corrective properties, which allows satisfying the daily requirement of the human body for biologically active substances. Ultrasonic exposure has a positive effect on the organoleptic characteristics of the beverage and the aftertaste, as proved by the comparative organoleptic evaluation of the samples. The functional properties of the beverages developed were confirmed experimentally. The following was determined in the beverages: macronutrients content, antioxidant activity, vitamin C content, shelf life duration. The ultrasonic extraction method application in the technology of beverages manufacturing allows: to reduce the preparation time by half, to reduce the amount of raw materials by 15%, to increase the content of macronutrients by 45%, to increase the content of vitamin C by 35%, to increase the antioxidant activity by 45-50%, to increase storage stability by 50-60%, to improve organoleptic characteristics. The results obtained prove the intensifying effect of ultrasonic exposure in the production of functional beverages.

The article analyzes modern technologies for storing fruit and vegetable products and presents possible options for their modernization. From this perspective, the improvement of canning technology using non-traditional methods of processing plant raw materials is an urgent task, the solution of which will allow preserving fruit and vegetable products, reduce the amount of waste and reduce the impact of vegetable processing plants on natural water bodies. Fruits and vegetables are perishable products and require special storage conditions from the moment they are available for sale to purchase by consumers. Keeping them fresh for as long as possible is quite a challenge, as some plants are able to continue maturing even after packaging and produce ethylene, which initiates certain changes in texture and color, tissue degradation. The authors pay special attention to the aspects of innovative technologies in the creation of edible films and coatings, which are currently the only type of biodegradable polymer packaging that does not require individual collection and special conditions of disposal. This is their main difference from traditional biodegradable plastics, which were originally proposed as an alternative to synthetic polymeric materials, and all were convinced that their creation once and for all will solve the problem of waste polymer packaging, which today threatens to become a global environmental disaster. At the same time, they are always more expensive than conventional plastics, in many cases can not be subjected to joint recycling; being in the deep layers of the landfill, they emit greenhouse gas methane. In addition, the raw materials for biodegradable plastics are provided by the same soil and water resources on which food is grown today (except for a very small amount of plastics produced from agricultural waste), and therefore their raw material base will always be limited. Recommendations are formulated on the organization of effective conservation of plant raw materials, intended for further processing or sale in retail and wholesale trade networks.

This article shows the possibility of using a biotechnological method to obtain biologically active compounds based on in vitro cultivation of callus cultures. The callus biomass of Adonis vernalis was obtained in vitro by using the biotechnological method. The extracts based on callus biomass of Adonis vernalis were obtained. The content of flavonoids and phenolic compounds was determined. The antioxidant activity of callus biomass of Adonis vernalis has also been studied. The research results show that the content of biologically active substances and biological activity in callus biomass does not differ from plant raw materials.

The use of the method of ultrasonic extraction in the technology of preparing drinks allows one: to reduce the preparation time by half, to reduce the amount of raw materials by 15 %, to increase the content of macronutrients by 45 %, to increase the content of vitamin C by 35 %, to increase the antioxidant activity by 45-50 %, increase stability during storage by 50 – 60 %, increase organoleptic characteristics. The results obtained prove the intensifying effect of ultrasonic exposure in the production of functional drinks.

Emphasis is placed on the efficiency of production of alternative types of biofuels by rational selection of species and the intensity of biomass formation of plants of the appropriate chemical composition. It is determined that favorable natural and climatic conditions of the South of Ukraine, the presence of built rice irrigation systems make it possible to obtain high yields of high quality rice grain, while forming significant amounts of by-products in the form of straw, husk, flour, which are a source of solid biofuel.As a result of research of bioenergetic potential of Ukrainian rice varieties it is shown that the highest values of energy yield under the use of by-products (straw and husk) are characterized by Viscount and Premium varieties, which studied to determine the impact of agronomic factors on the formation of rice productivity. straw and husks as sources of bioenergy plant raw materials as solid biofuels.

Given the important role of antioxidants in the cellular degeneration process, as well as the increased interest in recent years related to the use of natural antioxidants in therapy, the present study aims to investigate the antioxidant activity of a new pharmaceutical product containing natural antioxidants extracted from plant raw materials. In a first step, the product conventionally named “CILTAG”, containing a mixture in equal proportions of 10% hydroalcoholic extractive solutions of dried plant raw materials from Medicaginis herba, Trifolii pratense flores, Ginkgo bilobae folium, Myrtilli fructus, and Cynosbati fructus, was obtained. In the second stage, the antioxidant activity of the hydroalcoholic extractive solutions included in the pharmaceutical product was tested by chemiluminescence and electrochemical methods and by the superoxide dismutase (SOD) method. The electrochemical determination of the antioxidant capacity of hydroalcoholic extractive solutions was based on a method that provides an indirect evaluation of the presumed antioxidant properties of some compounds using Trolox (6-hydroxy-2,5,7,8-tetramethylchroman-2-carboxylic acid), as a standard measurement. The experimental results indicate a significant antioxidant potential for both the analyzed plant extractive solutions that are part of CILTAG and the final product.

The antioxidant activities of extracts of purple, yellow and orange carrot varieties were evaluated by in vitro methods i.e. DPPH, ABTS, FRAP, and CUPRAC. The total polyphenols concentration was evaluated using Folin-Ciocalteu technique. The content of anthocyanins, chlorophylls and carotenoids was evaluated in fresh and frozen carrots. The anthocyanin concentration was also determined in carrot extracts. The effect of certain parameters of ultrasound-assisted extraction, i.e. extraction time, concentration and acidifying of extractant, as well as the type of raw material (fresh or frozen) was analyzed. The antioxidant potential as well as content of polyphenols and plant pigments were the highest in purple carrot extracts. The most effective extractant seems to be 70% (v/v) ethanol. The extension of the extraction time, in majority of cases, led to an increase of antioxidant activity of extracts, whereas the freezing of raw material generally decreased the above-mentioned activities. The effect of acidification of extractant is ambiguous and depends on the method applied to evaluate antioxidant activity.

The Russian market is oversaturated with grain raw materials in the composition of mixed fodders, which are characterized by a protein deficiency and poor digestibility of nutrients. One of the problems of modern animal husbandry is to increase productivity, and most importantly, to reduce the cost of production due to the efficient use of feed nutrients. Phytobiotics have antiviral, antimicrobial, and immunomodulating effects. Feed consumption is increased and the acid-base environment is normalized when they are included in any diet. Phytobiotics are safe for farm animals and birds. Studies of the amino acid composition and antioxidant activity of the protein feed additive obtained from the leafy mass of red clover plants were carried out in the work. The qualitative and quantitative composition of amino acids and the value of antioxidant activity prove the advisability of introducing a fodder protein supplement into the diet of feeding farm animals and birds. The main advantage of feeding a plant-based supplement is that farm animals and poultry are not exposed to any risks, as well as the possibility of its use as a flavoring and an appetite stimulant. It is assumed that the application of an additive from the vegetative mass of red clover will stimulate growth, reproduction, weight gain and will have antibacterial and immunostimulating effects.
