Storage and hydro-priming treatments to improve the seed quality of two traditional rice varieties; Batapola-el and Suwendal, from Sri Lanka

Although the demand for traditional rice varieties has increased, low initial germination and poor storability of these varieties impede its popularization. Thus, the aim of the study was to improve the seed quality using priming treatments and to suggest an effective storage practice of two of the traditional rice varieties; Suwendal and Batapola-el, in Sri Lanka. Seeds were subjected to hydro-priming under various time periods (0, 24, 48 and 72 hours). Seeds were stored in gunny, polysac and polythene bags at room temperature, 25 and 8 °C. Seed quality of primed and stored seeds was evaluated using seed germination, seed vigour (seedling growth and conductivity) and seedling emergence. Hydro-priming for 72 hours has increased the seed germination of the two varieties significantly compared to other priming methods. Further, seeds primed for 72 hours had the highest vigour and highest seedling emergence in both varieties. Storing Suwendal and Batapola-el seeds in polythene bags at 8 °C retained the highest viability and highest vigour for more than six months. Thus, hydro-priming for 72 hours could be recommended to enhancethe seed quality of these two tested varieties and storage at 8 °C in sealed polythene bags could be recommended as an effective storage practice to maintain the seed viability till the next sowing season.

An experiment was carried out at Germplasm Centre and Plant Biotechnology Lab, Department of Horticulture, PSTU to evaluate the growth, seed yield and quality of hybrid F1 chilli (cv. Sonic) crossing male line (LTSL-004-M) and female line (LTSL-004-F) along the viability of productive seeds influenced by NPK fertilizers during the period from November 2015 to October 2016. Ten different treatments of NPK fertilizers were used. Result showed that all the characteristics except primary branches and1000 seed weight were influenced significantly due to the application of NPK fertilizers. The tallest plant (69.88 cm), more leaves plant–1 (2491.33), highest number of secondary branches plant-1, tertiary branches plant-1 (5.00 and 88.00, respectively), number of flowers and hybrid fruits plant–1 (1533.00 and 12.33, respectively) and weight of seeds (4.03 g plant–1) along with required more time (54.67 days) for flowering were obtained from the application of 145 kg N ha–1, 175 kg P ha–1 and 96 kg K ha–1 (T4) which produced the highest final seed yield of chilli (4.03 kg ha–1). Application of T5 and T9 showed the highest number of primary branches (2.33). In seed viability characteristics T9 treated seeds showed the highest germination (100, 97.33 and 94.97%) at 1st, (1 MAH - Month After Harvest), 2nd (2 MAH) and 3rd (4 MAH) observations, respectively. The T3 treated seeds performed well in respect of Seed Vigour Index (SVI) at 1st and 2nd observations (8.00 and 7.48, respectively) but T9 treated seeds showed the highest SVI (6.32) at 3rd observation. Above indicating all characteristics were lowest under only recommended doses of NPK T1 as control treatment except days to first flowering. The observations suggested that production of hybrid chilli seeds could be enhanced by applying T4. While seeds of chilli treated by T9 in field level showed long time viable. Application of 145 kg N ha–1, 175 kg P ha–1 and 96 kg K ha–1 may be suggested for seed production of chilli and 110 kg N ha–1, + 175 kg P ha–1 + 115 kg K ha–1 for keeping the long time seed viability. Storage is an important step in the production of soybean seeds, mainly in tropical regions, where high temperatures can drastically reduce seed quality. The storage at cooler temperatures may be a feasible alternative to preserve seed quality during storage. The objective of this study was to evaluate the physiological quality of soybean seeds artificially cooled by a dynamic process and stored in an environment, with non controlled temperature and relativity humidity. Two seed lots of cultivars Monsoy 8757, TMG 115 RR and BRS Valiosa RR were used. After processing and during packaging the seeds were cooled to 18 ºC by a dynamic process. Seed samples were collected after 0, 60 and 120 days of storage, using the following parameters germination, accelerated aging, seedling emergence in sand, speed of emergence index, electrical conductivity, tetrazolium and seed health. Although soybean seeds artificially cooled by the dynamic method may have slightly superior performance in maintaining physiological quality during storage in non-refrigerated warehouse, for lots of high physiological quality, these advantages are not observed.

ABSTRACT To obtain seeds of high physiological quality, it is of paramount importance to define the ideal harvest moment, which oftentimes corresponds to the period when the physiological maturity is reached. This experiment aimed to study the maturation process and determine the best harvest season of maroon cucumber fruits (Liso Gibão cultivar), in order to reach the maximum physiological seed quality. The flowers were labeled during the anthesis and the fruits harvested at 14, 21, 28, 35, 42, 49 and 56 days after the anthesis (DAA). The fruits had their visual aspects registered by photography analysis and, later, the seeds were extracted for physical (water and dry matter contents) and physiological (germination, seedling emergence, emergence speed index, accelerated aging and electrical conductivity) analyses. It was observed that the seeds reach the maximum dry mass (8.08 mg seed-1) between 47 and 49 DAA. However, the seed physiological maturity, characterized by a maximum germination (70 %) and vigor, occurs at 56 DDA. The best season to harvest seeds with a higher physiological quality takes place from 49 to 56 DDA, when the fruits present a yellowish color.

ABSTRACT The season of harvest is one of the most important factors influencing seed quality. This study evaluated the effect of different harvesting times on the physiological quality of Coriandrum sativum L. seeds, cv. Palmeira. The experimental design was completely randomized, with six treatments (harvest times) and four replications. The experiment was conducted during the months of April to October 2010. The first moment to harvest seeds was carried out 15 days after full bloom, when approximately 50% of plants were in flowering phase. Other harvests (22, 29, 36, 43 and, 50 days after bloom) were carried out at 7 days intervals until the seeds reached 16.0% of moisture, which occurred in the 6th harvest. Immediately after each harvest, the seeds were analysed for water content, germination and vigor (first count, seedling emergence and emergence rate index). The harvest season most suitable for C. sativum seeds cv. Palmeira occurred between 46 and 50 days after flowering, when the seeds have the highest physiological quality and moisture content between 20 and 16%.

Rice false smut (RFSm) is a disease of interest globally and in Bangladesh. Its occurrence and spread have drawn attention since 2010 in the country. While many reports are available on the yield loss from the disease, little is known of its effect on grain quality. Besides, there has been no information on how RFSm affects the seed quality with respect to germination. This study attempted evaluation of seed germination and grain quality of rice as affected by false smut disease. Three years’ study provided strong indication of inferior milling outturn and head rice yield in RFSm contaminated seeds. Results further revealed reduced amylose content and increased protein content in the contaminated seeds. Tendency in reduced seed germination, and less cooking time was observed in RFSm contaminated seeds. Future research is suggested to fully understand those causations. Extraction of DNA is very important nowadays in bio-molecular researches. Extracted DNA should be purified and the quality of DNA should also be very high. The objective of the study was to develop a simple efficient method to isolate DNA from the rice varieties in an open laboratory environment, and to eliminate the usage of expensive chemicals and tools. The DNA extraction methods developed by the DNeasy plant kit method supplied by QIAGEN, Cheng et al., Doyle et al. and Michiels et al. were applied to five different rice varieties grown in different parts of Sri Lanka. Based on the quantity and quality of the extracted DNA tested by measuring the absorbance of DNA at 260 nm using Nanodrop® ND-1000 spectrophotometer and measuring the ratio of A260 / A280 and gel running on agarose, the efficiency of the extraction method chosen varied among rice varieties. Among the methods used, the methods introduced by DNeasy plant kit method supplied by QIAGEN and Cheng et al, yielded good and amplifiable quality DNA with satisfactory concentration for all the rice varieties tested. Therefore the modified method of Cheng et al, 1987 could be used to extract DNA from rice varieties instead of the commercially available expensive and hazardous DNeasy plant kit method supplied by QIAGEN.Int J Appl Sci Biotechnol, Vol 3(3): 398-401

Selecting genotypes that combine high productivity with high seed quality is a challenge. The exploration of intracultivar genetic variation is an alternative to significantly increase the annual genetic gain and maximize the selection efficiency for both characteristics. The present study aimed to identify intracultivar variation to improve the seed quality of soybean genotypes derived from the commercial cultivar BRS 284, selected using the HSD (Honeycomb Selection Designs) model. Soybean genotypes selected for two years from two environments with contrasting edaphoclimatic characteristics, according to the principles of the HSD selection model, were cultivated under competition by using the experimental model in randomized blocks with four replicates and evaluated regarding the productivity and physiological quality of seeds. The results showed that genotype 284-3 presented a greater mass of 100 seeds, germination, vigour after accelerated ageing test, seedling emergence and emergence speed index than the other genotypes in both environments, with no significant difference in the standard cultivar regarding seed yield. The HSD method was an efficient selection method to identify intracultivar variation to improve cultivar performance.

Abstract: Harvesting and drying are essential procedures to obtain high quality seeds as well as for their conservation during storage. The goal of this research was to identify the physiological maturity stage and to verify the effect of drying speeds on the quality of zucchini seeds. A completely randomized design in a 3x2x2 factor scheme was used, involving three fruit maturation stages (49, 56 and 63 days after anthesis - DAA), two drying speeds (slow and quick) and two storage periods (0 and 6 months). Seed quality was evaluated by germination, first count, accelerated aging, electrical conductivity, tetrazolium, seedling emergence, emergence speed index and seed health test. Moisture content, thousand seed mass and percentage full seeds were also evaluated. Zucchini seeds reach maximum quality at 49 DAA. Slow or quick drying and storage do not affect the physiological quality of seeds that were harvested at 49 DAA. The delayed harvest of fruits reduces the quality of seeds. Quick drying reduces the incidence of the fungi Alternaria, Cladosporium, Phoma and Fusarium, and increases the occurrence of Aspergillus and Penicillium.
