Phyllidiella nigra is an organism that is suspected to have secondary metabolites because their ability to develop its self defense system by camouflage and using chemical compounds derived from their nature diet as deterrent against their predators. The purpose of this study was to isolate symbiotic bacterial derived from P. nigra, extracted and followed by, the antibacterial assays against Escherichia coli and Bacillus megaterium as well as the anti-UV assay. The results showed that the five isolates tested had an antibacterial activity with the highest average inhibition zone against E. coli DSM 498 bacteria, isolate 1 (14.67 mm), isolate 5 (14 mm), and against B. Megaterium DSM 32T bacteria, isolate 3 (13.33 mm). The three isolates which had the highest inhibition zone and P. nigra extract were tested for anti-UV assay using a UV-Vis Spectrophotometer. The results obtained isolate 3 has absorption of UV-A with the UV absorbtion maximum at λ 340 nm and P. nigra extract has absorption on UV-B radiation with UV absorption maximum at λ 290 nm. Key words: Nudibranchia, Bacteria, Anti-bacteial, Anti-UV Phyllidiella nigra merupakan organisme yang diduga memiliki metabolit sekunder karena mampu mengembangkan sistem pertahanan dirinya dengan cara kamuflase dan menggunakan senyawa kimia sebagai racun yang didapat dari makanannya. Tujuan dari penelitian ini yaitu mendapatkan isolat bakteri yang bersimbiosis dengan P. nigra, mendapatkan ekstrak dari baktri simbion, dan menguji antibakteri dan anti-UV ekstrak etil aseta bakteri simbion dengan metode difusi agar terhadap bakteri Escherichia coli dan Bacillus megaterium. Hasil penelitian didapatkan kelima isolat yang diuji memiliki aktivitas antibakteri dengan rerata zona hambat tertinggi terhadap bakteri E. coli DSM 498 yaitu isolat 1 (14,67 mm), isolat 5 (14 mm), dan terhadap baktri B. megaterium DSM 32T yaitu isolat 3 (13,33 mm). Ketiga isolat yang memiliki zona hambat tertinggi dan ekstrak P. nigra diujikan anti-UV menggunakan alat UV-Vis Spektrofotometer. Hasil yang didapat isolat 3 memiliki serapan terhadap radiasi sinar UV-A dengan puncak tertinggi pada λ 340 nm dan ekstrak P. nigra memiliki serapan terhadap radiasi sinar UV-B dengan puncak tertinggi berada pada λ 290 nm. BACKGROUND: Infection is the most common diseases in developing country, including Indonesia. Bacteria that often causes infection is Escherichia coli and Staphylococcus aureus. One of the traditional plants that can be used as an antibacterial is lidah mertua. AIM: The purpose of this study was to find out the profile of chemical compounds by thin layer chromatography method and determine the antibacterial activity of Lidah Mertua leaves by in vitro. METHODS: This research conducted an experimentally using non-polar, semipolar, and polar as solvents to get extract against E. coli and S. aureus as bacterial testing. The antibacterial activity using agar diffusion method to get minimum inhibitory concentration (MIC). RESULTS: The result of the research on thin layer chromatography showed that the compounds contained in the Lidah Mertua leaves were polifenol, steroids and alkaloids. The data obtained were tabulated and analysed descriptively. The antibacterial activity show that n-hexane extract does not provide inhibitory activity. MIC value show that aethyl acetate extract of lidah mertua leaves inhibited the growth of E. coli and S. aureus at concentration 50 mg/mL and 25 mg/mL with diameters of inhibition zone is 8.50 mm and 8.20 mm and methanol extract of lidah mertua leaves inhibited the growth of E. coli and S. aureus at concentration 12.5 mg/mL and 25 mg/mL with diameters of inhibition zone is 8.46 mm and 8.32 mm. CONCLUSION: The profile of chemical compounds by thin layer chromatography method showed that the compounds contained in the Lidah Mertua leaves were polifenol, steroids and alkaloids. The antibacterial activity show that n-hexane extract does not provide inhibitory activity, but aethyl acetate extract of lidah mertua leaves inhibited the growth of E. coli and S. aureus.

Phyllidia varicosa is an organism belongs to the order Nudibranchia, which is commonly known as a sea rabbit. This organism is able to synthesize secondary metabolites from food. The purpose of this study was to obtain P. varicosa extract and symbiotic bacterial extract from P. varicosa, then determine the antibacterial activity of P. varicosa extract and the symbionic bacteria extract against Escherichia coli DSM498, Bacillus megaterium DSM32T and anti-UV activity. P. varicose symbiotic bacterial were isolated and extracted. The results obtained 5 bacterial isolates. The results of antibacterial assay of isolates PhVa 1.1, PhVa 1.3, PhVa 2.1, PhVa 2.3 and PhVa 2.4 shown that these isolates have an antibacterial activity against E. coli DSM498 and B. megaterium DSM32T. Anti-UV assay results shown an absorption at UV-A with the highest value of 1.991 at λ 340 nm. Tujuan dari penelitian ini yaitu untuk mendapatkan ekstrak P. varicosa dan ekstrak bakteri simbion dari P. varicosa, kemudian mengamati aktivitas antibakteri dari ekstrak P. varicosa dan ekstrak bakteri simbionnya terhadap bakteri Escherichia coli DSM498 dan Bacillus megaterium DSM32T serta menguji aktivitas anti-UV. Bakteri yang bersimbion dengan P. varicosa diisolasi dan diekstraksi, lalu diuji bioaktivitas antibakteri dan diuji anti-UV terhadap ekstrak P. varicosa dan ekstrak bakteri simbionnya. Hasil akhir dari penelitian ini yaitu didapatkan 5 isolat bakteri. Hasil uji aktivitas antibakteri yaitu isolat PhVa 1.1, PhVa 1.3, PhVa 2.1, PhVa 2.3 dan PhVa 2.4 memiliki aktivitas antibakteri terhadap bakteri E. coli DSM498 dan B. megaterium DSM32T. Abstract Squid is one of the export commodities in Indonesia. In general, the use of squid meat, while the ink is only as waste. In fact, Squid ink contain bioactive compound that potential as anti-inflammatory, antihypertensive, anti-diabetic,anti-microbial and anti-malaria agents. The purpose of the study is to determine the types of secondary metabolite compounds contained in n-hexane extract of Loligo sp. ink using maceration method to determine its antibacterial activity against Escherichia coli. The results of secondary metabolite compounds obtained from the n-hexane extract of Loligo sp. ink are alkaloid, saponins, glycosides and phenol. The results of antibacterial test against E. coli using the disc method obtained the average of inhibition zone diameter at the concentration of 4% is 6.3 mm (intermediate), concentration of 8% is 7.83 mm (intermediate), concentration of 16% is 14.5 mm (susceptible) and concentration of 32% is 10.83 mm (intermediate). The antibacterial activity in n-hexane extract of Loligo sp. ink is optimal at the concentration of 16% against E. coli bacteria.

ABSTRACTObjective: The purpose of this study was to evaluate the antibacterial activity of chitosan, hydrolyzed coconut oil and their combination againstBacillus cereus and Escherichia coli.Methods: The materials used in this study were powder of chitosan (obtained from prawn shell produced by Laboratory of Research Centre FMIPAUniversity of Sumatera Utara) and virgin coconut oil (VCO) product of Siti Nurbaya-Indonesia. VCO was partially hydrolyzed by Lipozyme TL IM(active at sn-1,3 position) and the result called hydrolyzed virgin coconut oil (HVCO). The bacteria used in this study were B. cereus and E. coli. Theantibacterial activity of chitosan in 1% acetic acid and HVCO in dimethylsulfoxide was tested by Kirby–Bauer agar diffusion method using paper discwith diameter of 6 mm.Results: The results showed that the minimum inhibitory concentration of chitosan against B. cereus and E. coli is at concentration of 0.05% withinhibition zone diameter of 6.86 mm and 7.56 mm, respectively. MIC of HVCO against B. cereus is at concentration of 0.25% with inhibition zonediameter of 6.40 mm, and against E. coli is at a concentration of 0.50% with inhibition zone diameter of 6.20 mm. The inhibition zone diameter ofchitosan 0.05% and HVCO 0.25% in combination against B. cereus is 8.33 mm which is higher than half the sum of chitosan 0.05% and HVCO 0.25%(6.63 mm). The inhibition zone diameter of chitosan 0.05% and 0.5% HVCO in combination against E. coli is 8.53 mm which is higher than half thesum of chitosan 0.05% and HVCO 0.5% (6.53 mm).Conclusion: The findings of this study indicate that chitosan is more antibacterial than HVCO, and the interaction between chitosan and HVCO incombination demonstrated to be synergistic against B. cereus and E. coli.Keywords: Antibacterial, Chitosan, Coconut oil, Combination, Bacillus cereus, Escherichia coli.®

Penelitian ini bertujuan untuk menguji daya hambat dari ekstrak metanol beberapa jenis porifera terhadap pertumbuhan bakteri E. coli dan S. aureus. Pengujian aktivitas antibakteri menggunakan metode Kirby-Bauer, yaitu dilakukan dengan mengukur zona hambat di sekeliling cakram kertas. Ekstrak Haliclona sp dapat menghambat pertumbuhan S. aureus pada konsentrasi ekstrak 30%, 60% dan 90% dengan hasil pengukuran diameter zona hambat 13.50 mm, 20.67 mm dan 27.33 mm; serta menghambat pertumbuhan E. coli dengan diameter zona hambat 10.08 mm, 12.83 mm dan 14.17 mm. Daya hambat ekstrak Agelas sp terhadap S. aureus menunjukkan diameter zona hambat 8.33 mm dan hanya menunjukkan pada konsentrasi ekstrak 90%, sementara daya hambat ekstrak Agelas sp pada konsentrasi 30%, 60% dan 90% terhadap E. coli menunjukkan diameter zona hambat sebesar 7.67 mm, 10.17 mm dan 14.17 mm. Daya hambat Spheciospongia sp terhadap S. aureus dan E. coli hanya terlihat pada konsentrasi ekstrak sebesar 90% dengan diameter zona hambat adalah 8.42 mm dan 8.75 mm. Berdasarkan hasil yang diperoleh, dapat disimpulkan bahwa ekstrak Haliclona sp memiliki potensi aktivitas antibakteri yang dapat digunakan sebagai bahan dasar pembuatan obat antibiotik.This research aimed to test the inhibition capabilities of methanol extract from several kinds of Porifera on Escherichia coli and Staphylococcus aureus growth. The antibacterial activity test using the Kirby-Bauer method, which delivered by measuring the inhibition zone around paper disc. The extract of Haliclona sp can inhibit the S. aureus growth at 30%, 60% and 90% of extracts concentration with the measurement of inhibition zone diameters are 13.50 mm, 20.67 mm and 27.33 mm; also inhibit the E. coli growth with inhibition zone diameters are 10.08 mm, 12.83 mm and 14.17 mm. The inhibition capability of Agelas sp extract on S. aureus shows that the inhibition zone diameters is 8.83 mm and only appear at 90% of extracts concentration, meanwhile the inhibition capability of Agelas sp extract at concentration 30%, 60% and 90% on E. coli shows diameters of inhibition zone are 7.67 mm, 10.17 mm and 14.17 mm. The inhibition capability of Spheciospongia sp on S. aureus and E. coli only occurred at 90% of extracts concentration with inhibition zone diameters 8.42 mm and 8.75 mm. Based on the results, it can be assumed that extracts of Haliclona sp has a potential antibacterial activity that can be used as a basic ingredients for antibiotic medicine.

Abstract: The aim of this study is to investigate the antibacterial effect of endophytic fungi originated from the root of mangrove Rhizophora stylosa growing on Mangrove Plantation Area around DAS Simpang 5 Jl. Piere Tendean Manado. The method of this research was using combination of PDA and NA media. Two species of endophytic fungi were extracted from the root of R. stylosa, black mycelium fungi as isolate A and brown mycelium fungi as isolate B. The activities of both isolates were tested against pathogenic bacteria Staphylococcus aureus and Escherichia coli. The result showed the same inhibition zone of isolate A and B to S. aureus, while, isolate A showed bigger the inhibition zone in comparison to isolate B against E. coli. As a conclusion, the endophytic fungi taken from the root of R. stylosa have antibacterial activity toward S. aureus and E. coli. Piere Tendean Manado. Metode penelitian dilakukan dengan menggunakan kombinasi media Potato Dextrose Agar (PDA) dan Nutrient Agar (NA). Dari akar bakau R. stylosa diperoleh dua  jenis jamur endofit yaitu isolat A dengan karateristik miselium jamur berwarna hitam dan isolat B yang dengan karakteristik miselium berwarna coklat. Kedua isolat ini selanjutnya diujikan aktivitasnya terhadap bakteri Staphylococcus aureus dan Escherichia coli. Hasil penelitian menunjukkan isolat A dan B memberikan  zona hambat yang sama terhadap bakteri Staphylococcus aureus dan pada bakteri Escherichia coli isolat A menunjukkan diameter zona hambat yang lebih besar dibandingkan isolat B. Kesimpulan, Jamur endofit yang diisolasi dari akar bakau R. Stylosa memiliki efek antibakteri terhadap pertumbuhan bakteri S. aureus dan E. coli. The emergence of resistant bacteria strain has become a global health concern. It encourages the exploration of potential antibacterial agents, particularly from natural sources. The aim of this study was to investigate the antibacterial activity of ethanol extract of root, stems, leaves, and flowers of Polygonum pulchrum Blume against Staphylococcus aureus and Escherichia coli, through disc diffusion method using cup-plate method. Inhibition zone against S. aureus from roots, stems, leaves, and flowers ethanol extract were 3.5 mm, 2.5 mm, 2.25 mm, and 2.62 mm, respectively, while the inhibition zone against E. coli were 2.25 mm, 2.12 mm, 1.62 mm, and 1.75 mm, respectively. In conclusion, ethanol extract of root, stem, leaves, and flower of P. pulchrum Bl possessed weak antibacterial activity against S. aureus and E. coli.Keywords: P. pulchrum Bl, antibacterial, E. coli, S. aureus, cup-plate technique

Abstract. Rusdaryanti AF, Amalia U, Suharto S. 2020. Antibacterial activity of CaO from blood cockle shells (Anadara granosa) calcination against Escherichia coli. Biodiversitas 21: 2826-2830. Calcium carbonate (CaCO3) in blood clamp shells (BCS) was able to be converted into calcium oxide (CaO) through a calcination process. Some research stated that CaO can be used in the food industries b, one of which is as an antibacterial agent. The purpose of this study was to determine the most optimal size of BCS's powder during calcination as an antibacterial agent and its effect on the activity of Escherichia coli. Data were analyzed using analysis of variance and Kruskal-Wallis test. The results showed that 200 mesh size of BCS’s powder produced the highest yield of CaO at about 98.68% compared to 120 and 230 mesh size. The best concentration of CaO powder as an antibacterial was 3.5% with a pH of 11.3 ± 0.17. CaO powder had antibacterial activity against E. coli with minimum inhibitory concentration’s value (MIC) of-0.115; a minimum bactericidal concentration’s value (MBC) of 0 CFU/mL; inhibition zone of 3.23 ± 0.2 mm. By the PCR method, DNA degradation has occurred in E. coli cells. The differences of CaO powder concentration had significant effect (P <0.05) on the inhibition zone of E. coli.

Telah dilakukan penelitian yang bertujuan untuk mengidentifikasi senyawa metabolit sekunder  dari ekstrak daun kaf menggunakan tiga jenis pelarut yakni n-heksan, etil asetat, dan metanol. Ekstrak yang diperoleh diuji aktivitas antibakterinya dengan metode difusi yang menggunakan bakteri Staphylococcusaureus dan Escherichia coli. Dari hasil pengujian fitokimia, ketiga ekstrak tersebut menunjukan bahwa sampel mengandung senyawa; alkaloid, saponin, steroid, flavonoid dan tanin. Pengujian aktivitas antibakteri  dengan metode difusi menunjukan hasil yang paling tinggi aktivitas antibakteri adalah  estrak etil asetat dengan zona hambat 35,75 mm terhadap bakteri  Staphylococcus aureus, sedangkan pada ekstrak metanol memiliki zona hambat 23,75 mm terhadap bakteri Escherichia coliResearch has been carried out aimed at identifying secondary metabolites of kaf leaf extract using three types of solvents, namely n-hexane, ethyl acetate, and methanol.The obtained extracts were tested for antibacterial activity by diffusion method using Staphylococcus aureus and Escherichia coli bacteria. From the results of phytochemical testing, the three extracts showed that the sample contained compounds; alkaloids, saponins, steroids, flavonoids and tannins. Antibacterial activity testing with diffusion method showed the highest antibacterial activity was estracethyl ethyl acetate with 35.75 mm inhibition zone against Staphylococcus aureus bacteria, whereas methanol had 23.75 mm inhibitory zone against Escherichia coli bacteria

Circulation of synthetic antibacterial chemicals which is used continuously can cause resistance to bacteria. Alternative synthetic antibiotics research then comes up as an urgent need. Recently, the discovery of new antibacterial agents is looking for Actinomycetes. Actinomycetes are one of the active marine bacteria which produce secondary metabolites that could be used as antibacterial. The objective of this study was to evaluate the antibacterial activities of Actinomycetes symbiont with Halimeda macroloba, Gelidiella acerosa, and Turbinaria ornata in Pangandaran beach, Indonesia. A total of 41 Actinomycetes were isolated from 3 species of seaweed. Rep PCR amplification method was used to characterize and identify the relationship between samples. Primer BOX A1R (5’-CTACGGCAAGGCGACGCTGACGCTGACG-3’), 27F (5'-AGTTTGATCMTGGCTCAG-3') and 1492R (5'-TACGGYTACCTTGTTACGACTT-3') was used in amplification 16s DNA. Six isolates from the representative cluster dendogram were selected for secondary metabolites extraction using ethyl acetate solvent, meanwhile agar diffusion method was used as Antibacterial activity assay using E. coli, Proteus sp., and Enterobacter sp. as the bacterial test. The results showed that the activity of secondary metabolite extracts of Actinomycetes symbiont with Halimeda macroloba presented the highest inhibition zone on 3 bacterial tests. In the future, it could be a potential antibacterial agent against animal pathogenic bacteria.
