Mobile phones from people of different walks of life were examined through culture dependent based approach. Bacteria were isolated from the surface of the mobile phones. Various microbiological and biochemical tests such as Gram staining, citrate utilization test, urease test, TSI test, catalase test, methyl red test and indole test were performed for the isolated bacteria. This study highlights the health risk posed by the microbiota in the mobile phone’s surface

Abstract The aim of this study was to isolate and characterize the Phosphate solubilizing bacteria from the rhizosphere of Zea mays L., Jeneponto Regency. This research was conducted in several stages; i.e, sampling, medium preparation, sample dilution, isolation, characterization in the form of gram staining, biochemical tests, and quantitative tests of phosphate solubility. Soil samples were diluted in 0.9% NaCl and soil containing microbes was isolated on the Picovskaya medium. Three isolates were obtained which could dissolve phosphate, namely J2KN1, J3KR2, and J3TG3 isolates. The isolates were generally round in shape with raised elevations, white, slimy, smooth, shiny surface, milky white, shape like coccus and bacillus, and gram-negative. Some of the isolates had positive motility, indole, voges, methyl red, glucose, and sucrose fermentation in the biochemical test. The quantitative tests of the ability to dissolve phosphate showed that J2KN1 isolate had the highest concentration of 51.1 μM, and the J3KR1 and J3TG3 isolates had a concentration of 45.2 μM and 37.6 μM, respectively.

We evaluated two protocols for isolation of Helicobacter pylori in stool from biopsied and nonbiopsied children. Twenty-three child patients whose presumptive positivity or negativity was diagnosed by endoscopy and a rapid urease test at site were used to compare biopsy-based tests with stool-based tests (H. pylori stool antigen test and stool culture). Their gastric activity and bacterial density were graded by the updated Sydney system. Biopsy and stool specimens were cultured on Campy-blood and Belo horizonte agar plates after enrichment in selective Campy-Thio medium. To compare two stool culture protocols, stools from 20 nonbiopsied children were tested by the HpSA test and cultured either as above or after treatment with cholestyramine. Grown colonies were screened by Gram staining, slide agglutination using anti-H. pylori monoclonal IgG; positive isolates were tested by biochemical tests and polymerase chain reaction for H. pylori-specific ureA gene. Coccoid H. pylori was isolated in stool samples from the biopsied patients whose bacterial density was two to four in histology. Their oxidase was slightly positive but became positive after two subcultures, while additional biochemical tests confirmed the isolation of H. pylori. Similar coccoid but oxidase positive H. pylori was isolated from three nonbiopsied children with the protocol of cholestyramine treatment only. The density of bacteria in the stomach may influence the recovery of H. pylori from stool; inactivation of bile with cholestyramine improves the yield in culture and favors isolation of an enhanced metabolic form of bacteria.

Aim: The aim of this study is to determine the presence of microbial air contaminants in the operating theatre at a teaching hospital. Objective: Airborne microbial level in operation theatre is one of the significant risks in hospital as it can increase the surgical site infection and nosocomial infections. Background: Duo SAS Super 360 Air Sampler was used to collect the airborne samples in triplicate each for nutrient and MacConkey agar at eight operation rooms and two corridors for morning and evening sessions. Sampling was conducted for three months and repeated every two months. Microbiological culture, gram staining and biochemical tests such as catalase test, oxidase test, coagulase test, Triple Sugar Iron Agar test, urease test, citrate test, Sulfide, Indole, Motility test, Methyl Red Voges-Proskauer test, disc diffusion test, and Albert’s stain were performed on the pure isolated culture. Methods: Sampling was conducted for three months and repeated every two months. Microbiological culture, gram staining and biochemical tests such as catalase test, oxidase test, coagulase test, Triple Sugar Iron Agar test, urease test, citrate test, Sulfide, Indole, Motility test, Methyl Red Voges- Proskauer test, disc diffusion test, and Albert’s stain were performed on the pure isolated culture. Bacteria that were present in the operation rooms were Bacillus spp., Micrococcus spp. and Staphylococcus spp. while Pseudomonas aeruginosa and Acinetobacter baumannii were absent. The bacteria identified in the operation theater may also cause surgical site infections and nosocomial infections to the patients, although the microbial contamination in the air of the operation theatre is low. Conclusion: Strengthening surveillance on the hygienic condition of the operation theatre and routine sampling is strongly recommended to control all possible sources and types of infection.


In the year 2018 a report published by TechSci Research entitled “India Chocolate Market Forecast and Opportunities, 2018” reiterated that the chocolate market earnings in India has witnessed the growth rate of 21% from 2013-2018. Indian chocolate market witnessed USD 3.2 billion revenues in 2018. Indian chocolate market is divided into 4 sectors, in which bar chocolates accounts for highest share of 36%. Hence, We had chosen to study the commercially available bar chocolates through culture dependent methods. We had selected 3 commercially available bar chocolates namely Cadbury Dairy milk, Cadbury 5 Star and Cadbury Perk. Bacteria were isolated from these 3 brands of chocolate by streak plate method. Microbiological and Biochemical assays such as Simple staining, Gram staining, Catalase test, Methyl Red test, Oxidase test, Vogus Proskaur test, Indole test, Citrate utilization test and Starch hydrolysis test were performed. The bacterial isolates were found to be Rod shaped and Gram negative. The bacterial isolates were found to be negative for all the above mentioned tests except Methyl Red and Starch hydrolysis test. Thus, these findings highlight the impending health hazard posed by the commercially available bar chocolates.

The purpose of this study was to detect the presence of bacteria Aeromonas sp in tilapia (Oreochromis niloticus). Nine fish with an average body weight of 150-200 g were used in this study as samples. Sampling was conducted at three different locations which were representative of fish culture sites located on Lake Tondano. The number of sample gathered from each location was and 3 fish which were taken randomly. Fish samples were taken by using a scoop, then the samples were taken alive by placing in an oxygen-filled plastic separately and taken direcly to lab Fish Disease Control and Environmental Center, Tateli, Department of Marine and Fisheries of North Sulawesi province, for examination the presence of bacteria. Isolation of bacteria were conducted by taking samples from gill and kidney. This study was carried out from April - July 2013. Identification of the presence of bacteria was done through a series of observations of colony morphology and gram staining of bacteria, followed by a series of biochemical tests: oxidase test , catalase test , test TSIA , H2S production test , indole test , motility test , citrate test, and test O/F. It could be concluded that 22.22 % of farmed nile tilapia in Lake Tondano were infected with the Aeromonas sp with the following percentage from each site: Paleloan village 7.40% , 7.40% Toulimembet village, and 7.40% Eris village. A novel thermotolerant bacterium, designated SgZ-8T, was isolated from a compost sample. Cells were non-motile, endospore-forming, Gram-staining positive, oxidase-negative and catalase-positive. The isolate was able to grow at 20–65 °C (optimum 50 °C) and pH 6.0–9.0 (optimum 6.5–7.0), and tolerate up to 9.0 % NaCl (w/v) under aerobic conditions. Anaerobic growth occurred with anthraquinone-2,6-disulphonate (AQDS), fumarate and NO3 - as electron acceptors. Phylogenetic analysis based on the16S rRNA and gyrB genes grouped strain SgZ-8T into the genus Bacillus , with the highest similarity to Bacillus badius JCM 12228T (96.2 % for 16S rRNA gene sequence and 83.5 % for gyrB gene sequence) among all recognized species in the genus Bacillus . The G+C content of the genomic DNA was 49.3 mol%. The major isoprenoid quinone was menaquinone 7 (MK-7) and the polar lipids consisted of diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine and an unidentified phospholipid. The major cellular fatty acid was iso-C16 : 0. On the basis of its phenotypic and phylogenetic properties, chemotaxonomic analysis and the results of physiological and biochemical tests, strain SgZ-8T ( = CCTCC AB 2012108T = KACC 16706T) was designated the type strain of a novel species of the genus Bacillus , for which the name Bacillus thermotolerans sp. nov. is proposed.

Mobile phones (MPs) are commonly used both in the personal and professional life. We assessed microbiological contamination of MPs from 108 students in healthcare professions (HPs), in relation to their demographic characteristics and MPs handling habits, collected by means of a questionnaire. Cultural and biochemical tests were performed, and statistical analyses were carried out. Staphylococci were present in 85% of MPs, Enterococci in 37%, Coliforms in 6.5%; E. coli was never detected. Staphylococcus epidermidis was the most frequently isolated staphylococcal species (72% of MPs), followed by S. capitis (14%), S. saprophyticus, S. warneri, S. xylosus (6%), and by S. aureus (4%). Heterotrophic Plate Counts (HPC) at 37 °C, ranged from 0 to 1.2 × 104 CFU/dm2 (mean = 362 CFU/dm2). In univariate analysis, the male gender only was significantly associated with higher HPCs and enterococcal contamination. Multiple linear regression models explained only 17% and 16% of the HPC 37 °C and staphylococcal load variability, respectively. Developing specific guidelines for a hygienic use of MPs in clinical settings, for preventing cross-infection risks, is advisable, as well as introducing specific training programs to HP students. MPs decontamination procedures could also be implemented in the community.

Chronic suppurative otitis media (CSOM) is a prevailing and notorious infection in developing countries causing serious local damage and threatening complications. The purpose of the present study was determine the microbiological profile of isolates from discharge in CSOM. This study was conducted at out-patient department of ENT at Rajshahi Medical College Hospital, Rajshahi from January 2014 to December 2014. Samples were taken from 185 patients (both male and female) in all age groups suffering from chronic suppurative otitis media. Their Gram staining, culture, and biochemical tests were carried out to identify the organisms. It showed the predominance by staphylococcus aereus (29.13%), followed by Pseudomonas Aeruginosa (22.83%), Streptococcus Pyogen (14.96%), E.Coli (9.44%), Proteus Mirabilis (6.29%), Klebsiella Pneumonia ( 4.72%). We used broad-range bacterial PCR combined with DNA sequencing to examine prospectively cerebrospinal fluid (CSF) samples from patients with suspected meningitis. Fifty-six CSF samples from 46 patients were studied during the year 1995. Genes coding for bacterial 16S and/or 23S rRNA genes could be amplified from the CSF samples from five patients with a clinical picture consistent with acute bacterial meningitis. For these patients, the sequenced PCR product shared 98.3 to 100% homology with the Neisseria meningitidis sequence. For one patient, the diagnosis was initially made by PCR alone. Of the remaining 51 CSF samples, for 50 (98.0%) samples the negative PCR findings were in accordance with the negative findings by bacterial culture and Gram staining, as well as with the eventual clinical diagnosis for the patient. However, the PCR test failed to detect the bacterial rRNA gene in one CSF sample, the culture of which yielded Listeria monocytogenes. These results invite new research efforts to be focused on the application of PCR with broad-range bacterial primers to improve the etiologic diagnosis of bacterial meningitis. In a clinical setting, Gram staining and bacterial culture still remain the cornerstones of diagnosis.
