Our experiment was conducted at a special dairy farm in Dakahlia Governorate between the periods (March –May 2018), This farm consisted of 400 Holestien cows; from the total of the animals only 210 lactating cows. The established experiment applied on 40 cows suffered from different types of anestrum detected by ultrasonography as follow (15 cows suffer from cystic ovary, 15 cows suffer from smooth inactive ovaries and 10 cows suffer from persist corpus luteum to study the effect of progesterone device insertion in dairy cattle and its effect in fertility. On day 0, cattle at random stage of estrous cycle received controlled internal drug release vaginal insert (CIDR).We left the CIDR in the vagina for seven days as we inject PGF2 on day 6 and remove the CIDR on day 7, blood samples were collected from 25 animals at zero day, 3rd, 7th and 9th day from the tail vein, and then we follow the estrous and detected the estrus cow for AI and apply ultrasonography for pregnancy diagnosis after 30 day from insemination From this study it was concluded that the use of progesterone for 7 days +i.m. injection of PGF2α in the 7th day can applied to dairy cattle to restart ovarian activity and it is an effective treatment for different infertility cases like cystic ovarian disease, persist corpus luteum and smooth in active ovaries. Moreover present study provides evidence for the importance of prior exposure to progesterone for cows to express estrous behavior, increase number of pregnant animals and increase conception rate.

ABSTRACTMelatonin was fed daily at 14.00 h to eight non-lactating and six lactating hinds. Feeding was begun on 27 July 1983 (day 1) and continued until 21 September (day 57). Six non-lactating hinds were used as controls. Blood samples were taken about every 10 days from day 42 until day 73. Plasma progesterone and prolactin were determined by radioimmunoassay. Progesterone values of greater than 0·63 μg/1 were taken to indicate the presence of a corpus luteum and that the animal had ovulated. In the melatonintreated, non-lactating group seven of the eight hinds showed ovarian activity compared with two of the six controls (P < 0·01). None of the lactating hinds treated with melatonin showed evidence of ovarian activity. In both the lactating and non-lactating hinds treated with melatonin, prolactin levels were never greater than the detection limit of the assay (7·4 μg/l), while, in the control hinds mean prolactin levels were elevated on all but the last sampling day. It is, therefore, possible to induce early ovarian activity in non-lactating red deer hinds by feeding melatonin. However, although melatonin treatment similarly depressed plasma prolactin levels in both lactating and non-lactating hinds, it was unable to overcome the inhibitory effect of lactation on reproduction.

An adequate and clean artificial insemination (AI) technique is recommended to maximize reproductive outcomes in dairy cattle. The objective of this study was to evaluate the effectiveness of using protective sheaths (PS; Continental Plastic Inc., Delavan, WI, USA) to minimize contamination of the AI catheter (AIC) on pregnancies per AI (P/AI) in lactating dairy cattle. A previous study reported no improvement on cattle fertility when using PS during first service AI (King et al. 1984 Can. Vet. J. 25, 327). Lactating cows housed in free-stall barns on a commercial dairy farm with a rolling herd milk production average of 10.140 kg were presynchronized with 2 injections of PGF2α (25 mg; Lutalyse, Pharmacia & Upjohn, Kalamazoo, MI, USA) given 14 days apart (starting at 26 ± 3 d postpartum) followed by Ovsynch [OV; GnRH-7 d-PGF2α-56 h-GnRH-16 h-timed-AI(TAI)] 12 days later. Cows presenting signs of standing heat any time during the protocol received AI, whereas the remaining animals were subjected to TAI16 h after second OV GnRH (100 μg; Cystorelin, Merial, Duluth, GA, USA). At the moment of AI (one AI technician), 996 services from lactating dairy cows were randomly assigned to 1 of the 2 groups; with (TRT, n = 487) or without (CON, n = 509) the use of PS. In the TRT group, the AIC protected with a PS was introduced into the vagina; once in the cranial portion of the vagina adjacent to the cervical os, the PS was pulled back and only the AIC was manipulated through the cervix into the uterine body for semen deposition. In the CON group, cows received AI without the PS. Additionally, sterile cotton swab (BD, Franklin Lakes, NJ, USA) samples were taken from AIC (n = 51) after AI from both treatment groups. Pregnancy diagnosis was determined by ultrasonography 42 ± 3 d after AI. Data analyses were performed using GLIMMIX (P/AI) and FREQ (culture) procedures of SAS (SAS Institute Inc., Cary, NC, USA). Cultured swab samples revealed that the use of PS was effective in minimizing contamination of the AIC (bacterial growth on AIC; TRT = 57.7% v. CON = 100%; P < 0.0002). Overall, the proportion of cows pregnant (all services) was greater (P = 0.03) for cows in TRT (42.7 ± 2.2%) compared with CON group (36.1 ± 2.1%). For first services postpartum, P/AI did not differ (P = 0.87) between CON (43.01 ± 4.4%) and TRT (43.8 ± 4.6%) groups. However, P/AI for second or greater services (≥2S) were greater (P = 0.007) in TRT (43.8 ± 2.9%) than in CON cows (32.3 ± 2.6%). Results from this study suggested that the use of PS during AI improved P/AI for ≥2S in lactating dairy cows. Performing a clean AI technique through the use of PS may be a cost-effective strategy to improve reproductive outcomes in dairy cattle. Further investigation is needed under various reproductive management conditions to confirm and determine the underlying mechanisms for these findings. Authors thank Coba/Select Sires Inc. The objective of the present study was to measure serum levels of obestatin by ELISA with the purpose of establishing parameters and being able to use it as an endocrine marker for the selection of reproductive characteristics of milk and beef cattle. In test 1, 15 Creole cows with genetic tendencies of Charolais, Angus, Brangus, and Gelbvieh were sampled in diestrus and 15 in proestrus, determining their structures with an Aloka Prosound ultrasound with a 7.5-MHz linear transducer. In test 2, 16 Holstein cows were sampled in 2 different ovarian stages, diestrus and proestrus. The standardized technique was a bovine obestatin 96-well ELISA kit from My Biosource Inc. The data were analysed by measuring central tendency and linear regression, where the average levels of obestatin were related to the corpus luteum size and size and number of follicles in milk and beef cattle. We found average levels of 556.99 and 453.35 pg for beef and dairy cattle, respectively (P&lt;0.05). We also found a significant difference (P&lt;0.05) in obestatin concentrations depending on the size of the follicles (751.74, 615.33, and 357.39 pg for follicles of 15, 10, and 5mm, respectively). We found a significant difference in concentration of obestatin (644.53, 550.69, and 455.88 pg; P&lt;0.05) for corpus luteum sizes of &gt;20mm, 10–20mm, and &lt;10mm, respectively. In dairy cattle, obestatin levels were also evaluated at different stages of the cycle, and we found concentrations of 414.79 pg in diestrus and 530.48 pg in proestrus (P&lt;0.05). We conclude that obestatin concentration is positively associated with a greater number and size of follicles as well as with larger corpus luteum because its effect on cell proliferation interacting positively with high levels of progesterone in the blood. We found higher concentrations in beef cattle than in dairy cattle.

The objectives of the current research were twofold: (i) to determine the prevalence of multidrug-resistant (MDR) Salmonella in the various classes of dairy cattle and (ii) to determine if comingling of calves from multiple farms at a heifer feedlot serves as a transmission vector for Salmonella back to the dairy farm. Four large commercial dairies in the southwestern United States were sampled in October 2005 and again in March 2006. Fecal samples were collected from hutch calves, 12-and 24-month-old heifers, lactating cows, dry cows, and cattle in the sick-fresh pen and cultured with brilliant green agar supplemented with novobiocin (BGAnov) to estimate the overall Salmonella prevalence, or with tetracycline (BGAtet) to estimate MDR Salmonella. Antimicrobial susceptibility testing was conducted with the National Antibiotic Resistance Monitoring System (NARMS) testing panel, and a portion of the isolates were serotyped. Salmonella prevalence among groups ranged from 0 to 96% positive, with the highest incidence observed in the hutch calves and cattle in the sick-fresh pen. Twenty-eight different serotypes were identified with serotype Reading accounting for the majority of isolates cultured on BGAtet. Nearly all (100 of 103) isolates cultured on BGAnov and screened for antibiotic resistance were pan susceptible, whereas over one-half (64%) of the isolates cultured on BGAtet were MDR. Forty isolates displayed the ACSSuT resistance pattern, and 36 isolates displayed the MDR-AmpC pattern of the 72 isolates examined following culture on BGAtet. The incidence of Salmonella cultured on BGAtet was low (9%) in all heifers and only one MDR isolate was cultured (from a 12-month-old heifer), suggesting the risk of transmission of Salmonella from the heifer feedlot back to the dairy is low. Results of this research suggest the incidence of MDR Salmonella, found primarily in hutch calves and cattle in the sick-fresh pen, is low in comparison to the overall Salmonella prevalence.

A modern dairy farm is a complex biotechnical “man-machine-animal” system, where purposeful human activity concentrates mainly on the control of the “machine” and “animal” subsystems, thus making the whole system ergatic. Increasing the interaction effi ciency of machine subsystems with biological objects (animals) requires an in-depth study of the properties and characteristics of the latter, their behavior, adaptive and refl ex mechanisms that ensure the mutual adaptation of machine and biological subsystems. The paper considers general functionality of the “animal” subsystem, which includes lists of monitored parameters (functions) in pre-weaning, pre-lactation and lactation periods. In a similar way, functionals of the subsystems of the general musculoskeletal development of the animal’s body, respiratory and digestive organs, comfort of the habitat, development and control of the reproductive organs of lactating cows were obtained accompanied with a list of controlled functions and parameters. To carry out a set of research activities in this fi eld, FSAC VIM is planning to carry out a complex project that will increase the levels of automation, digitalization and intellectualization of animal husbandry, provide for comfortable environment, optimal rediced-impact service modes for animals and their extended productive longevity, increased quality of milk and autonomous functioning of individual local biotechnical subsystems.

This study was carried out on clinical cases of retained fetal membranes in crossbred cows presented at College Clinics and College dairy farm and from Villages in and around Mhow. The blood samples were collected from jugular vein just before 12 hr. postpartum and on 7th day postpartum. Haematological profile revealed that the mean values of haemoglobin, neutrophil and monocyte count after 12 hrs and 7th day postpartum were significantly lower and lymphocyte count was significantly higher in RFM cows (n=18) than normally calved cows (n=6). The differences in mean TLC, eosinophil and basophil counts were non-significant at 12 hrs postpartum, however on 7th day postpartum the TLC and eosinophil count were significantly higher and basophil count was non-significantly different in RFM cows than the normally calved cows.

Staphylococcus aureus is one of the most concerning mastitis-causing pathogens in dairy cattle. Using basic microbiological techniques, S. aureus is typically identified by colony characteristics and hemolysis on blood agar where isolates without hemolysis are typically considered to be coagulase-negative staphylococci (CNS) isolates. Herein, we present a decade-long case study where suspected S. aureus isolates from one Georgia dairy farm were further tested to confirm presumptive identification. Presumptive identification of bacterial growth from 222 mammary secretions from bred Holstein heifers and lactating cows was conducted at the time of collection. Presumptive identification of S. aureus on blood agar was based on observation of colony morphology, color, and presence or absence of a broad zone of incomplete hemolysis and a smaller zone of complete hemolysis at 48 h. Those without hemolysis were presumptively characterized as CNS. All isolates were further plated on mannitol salt agar and a coagulase test was performed. A positive for both of these tests together was deemed to be S. aureus. A selection of isolates was tested using API® Staph to biochemically confirm S. aureus identification. Data showed that 63.96% of isolates presumed to be CNS isolates were identified as S. aureus, 9.46% of isolates presumed to be CNS isolates were identified as coagulase-positive staphylococci (CPS) species (but not S. aureus), and 26.58% of samples that were presumed to be CNS isolates were identified correctly.

The objective of this study was to investigate the prevalence of anti-Neospora caninum antibodies in cattle from milk producing farms of the microregion of Batalha, state of Alagoas, Brazil, as well as to identify the risk factors associated with the infection. Blood samples were collected from 1,004 cattle of 17 farms for the serological investigation regarding the presence of anti-N. caninum antibodies by the Indirect Immunofluorescence Reaction Technique (IMRT). From the total amount of samples analyzed, 77/1,004 (7.67%) were positive and 927/1,004 (92.33%) were negative. The logistical regression identified that cattle from farms without consortium breeding have an infection risk 6.33 (p<0.001; C.I. 2.89-13.10) times higher than cattle from farms with that type of breeding. Cattle from farms where the aborted fetuses are not adequately buried have an infection risk 3.04 (p<0.001; C.I. 1.64-5.63) times higher than cattle from farms with adequate destination of these fetuses. Infection by N. caninum occurs in cattle of the investigated region. The factors identified in our study can be used as risk indicators, so that control measures could be implemented to avoid infection by N. caninum in the herds of this region.

Three experiments were conducted to evaluate endocrine profiles and to determine the morphological and functional characteristics of corpora lutea (CL) following the onset of postpartum ovarian activity in beef cows, suckled by a single calf once daily. In exp. 1, blood samples were collected from 12 cows at 6-h intervals beginning 25 days postpartum until ovariectomy which was carried out on each of two cows on days 25, 27, 29, 31, 33, 35 after parturition. Ovarian structures were examined grossly and histologically. In exp. 2, blood samples were collected from eight cows at 6-h intervals for 18 days beginning 25 days postpartum and at less frequent intervals thereafter. Laparotomies were carried out on day 36 after calving, the ovaries were observed, CL were sampled and the residual tissue was marked with charcoal. A second laparotomy was performed on day 50. Luteal tissue samples were processed and examined by light and electron microscopy. Luteal cell types were evaluated, percent of area covered by large cells was determined and average luteal cell diameter was calculated. In exp. 3, seven cows were bled daily from parturition until day 25 postpartum. Serum from all experiments was assayed for progesterone (P4), FSH, LH and a prostaglandin metabolite (PGFM). The data showed that serum PGFM levels declined from a peak at calving to basal levels by 10 days postpartum which was well before the first ovulation. In all instances the observed peaks of serum LH and serum FSH preceded the first rise in P4 which, in eight of nine cases, was due to a functional CL. These CL were functional for periods of time ranging from 3 to 12 days. The regression of short-lived CL appeared abnormal compared to the longer-lived CL in which regression was characterized by a rapid decline in P4 and elevated blood PGFM. These data show clearly that the first increase in P4 is preceded by a typical LH surge, followed by ovulation and CL formation which has a variable life-span. A study was made (1 970 to 1973) of the reproductive performance of Shorthorn and Brahman crossbred cows grazing pasture in the dry tropics of north Queensland. Breed had a variable effect on the conception rates of lactating cows, with Brahman crossbreds having significantly higher (P < 0.05) conception rates in the first year. The two breeds had similar conception rates in the following year. In the third year, overall conception rates of both breeds were reduced due to a prolonged dry season, with Shorthorn cows having significantly higher (P < 0.01) conception rates than Brahman cows. However, during this dry period, twice as many Shorthorn cows required survival feeding as did Brahman cows. Calving rates followed conception rates closely, prenatal loss in each breed being low. In all years, Shorthorns had significantly earlier (P < 0.05) calving dates than Brahmans. The interval from calving to first recorded ovarian activity was significantly longer (P < 0.05) in Brahman cross than in Shorthorn cows in only one of the three years. There was no significant difference between breeds for the interval from first recorded ovarian activity to conception, but this interval decreased with increasing age of the cow. Breed had a variable effect on gestation length. Brahman cows were generally heavier each year in April and in better condition throughout the experimental period than Shorthorn cows.
