Fish is in high demand as food, food additives, and supplements as they are a rich source of carbon, proteins, vitamins, and minerals. Fish has been established to possess bacterial populations on or in their skin, gills, digestive tract, etc. with their microbial diversity often reflecting the bacterial populations of the surrounding water which are either allochthonous or allochthonous. This study isolated and enumerated aerobic bacteria flora in the gastrointestinal tract and gills of four culturable freshwater fish (Silver catfish, Tilapia, Clarias, and Heterobranchus). These species of cultural freshwater fish were obtained and each adult specie held in a separate glass containing unchlorinated water and transferred to the laboratory. The quantitative and qualitative estimation of the bacteria flora present in the gill and Gastro-Intestinal Tract (GIT) of fish were investigated. The Mean total plate count on Nutrient Agar (NA), Blood Agar (BA), MacConkey Agar (MCA), Cysteine-Lactose-Electrolyte-Deficient Agar, (CLED Agar) and Salmonella – Shigella Agar (SSA) was found to be 60 and 40 CFU, 150 and 80 CFU, 100 and 90 CFU and 80 and 70 CFU respectively. Most of the isolates were of public significance. The results showed that fish contains a large number of microbiotas which may play a role in nutrition and health.

The soil fertility aspect is characterized by the good biological properties of the soil. One important element of the soil biological properties is the bacterial population present in it. This research was conducted in the laboratory of Microbiology University of Malikussaleh in the May until June 2016. This study aims to determine the number of bacterial populations in soil organic and inorganic so that can be used as an indicator to know the level of soil fertility. Data analysis was done by T-Test that is by comparing the mean of observation parameter to each soil sample. The sampling method used is a composite method, which combines 9 of soil samples taken from 9 sample points on the same plot diagonally both on organic soil and inorganic soil. The results showed the highest bacterial population was found in total organic soil cfu 180500000 and total inorganic soil cfu 62.500.000

SummaryExperiments were done to determine the fate of soluble mucins in the gastrointestinal tract of sheep. Incubation of a soluble mucin with liquor from large intestinal contents resulted in loss of mucin. Some of this loss was due to soluble enzymes. The loss of mucin was less when incubation was with rumen liquor and variable when with ileal liquor.The mean amounts of nitrogen in a soluble mucin fraction which was soluble in trichloroacetic acid (T.C.A.-soluble mucin) were 2·5, 7·3 and 20·0 mg per 100 ml in rumen caecal and faecal liquors respectively. These amounts were only a small proportion of the total soluble nitrogen in these fluids.Amounts of T.C.A.-soluble mucin, measured by sugar content, were greatest in contents from the small intestine. When the amounts of T.C.A.-soluble mucin were compared with the amounts of lignin in the samples, there was an increase between the abomasum and the first part of the small intestine and then a decrease to the caecum. Amounts relative to lignin were low in other parts of the gastro-intestinal tract.The greatest ratio of fucose to rhamnose in T.C.A.-soluble mucin from gastro-intestinal liquors was in ileal liquor. The presence of the two methyl pentoses, fucose and rhamnose, indicates that the T.C.A.-soluble mucin is derived from mucus and bacteria.

Studies to determine the appropriateness of the use of populations of indicator bacteria on poultry carcasses for process verification were undertaken in commercial slaughterhouses. Samples were collected from neck skin by excision or from whole carcass rinses and were examined for a range of presumptive process hygiene indicator bacteria. Coefficients of variation were calculated for each bacterial indicator and were significantly lower in excised samples, indicating more reproducible bacterial recovery by this sampling method. Total viable counts of aerobic bacteria, Enterobacteriaceae, and Pseudomonas in samples collected by excision had the lowest coefficients of variation when compared with other indicators and were therefore used for further study. The uncertainties associated with the quantification of each bacterial indicator were calculated and were lowest overall for total viable counts of aerobic bacteria. In general, uncertainty was higher for lower bacterial numbers. Results of microbiological testing on pooled excised neck skin samples were not significantly different from the mean of individually analyzed samples. Bacterial numbers increased by 1 log unit when cultures were stored under chilled conditions typical of those used for transporting samples to external laboratories, but the increases were not significant for Pseudomonas and aerobic bacteria when storage time was less than 17 h. Weak relationships were identified between bacterial indicator numbers and duration of processing, although cleanliness of the processing environment diminished visibly during this time. In the plants visited for this study, there was a poor relationship between presumptive bacterial indicator numbers and process hygiene. Consequently, bacterial analyses for process verification purposes may be of limited value.

Preservation of probiotics by lyophilization is considered a method of choice for developing stable products. However, both direct consumption and reconstitution of dehydrated probiotic preparations before application “compromise” the survival and functional characteristics of the microorganisms under the stress of the upper gastro-intestinal tract. We evaluated the impact of different food additives on the viability, mucin adhesion, and zeta potential of a freeze-dried putative probiotic, Lactiplantibacillus (Lp.) plantarum HAC03. HAC03-compatible ingredients for the formulation of ten rehydration mixtures could be selected. Elevated efficacy was achieved by the B-active formulation, a mixture of non-protein nitrogen compounds, sugars, and salts. The survival of Lp. plantarum HAC03 increased by 36.36% compared rehydration with distilled water (4.92%) after passing simulated gastro-intestinal stress conditions. Cell viability determined by plate counting was confirmed by flow cytometry. B-active formulation also influenced Lp. plantarum HAC03 functionality by increasing its adherence to a Caco-2 cell-line and by changing the bacterial surface charge, measured as zeta potential.Hydrophobicity, mucin adhesion and immunomodulatory properties of Lp. plantarum HAC03 were not affected by the B-active formulation. The rehydration medium also effectively protected Lp. plantarum ATCC14917, Lp. plantarum 299v, Latilactobacillus sakei (Lt.) HAC11, Lacticaseibacillus (Lc.) paracasei 532, Enterococcus faecium 200, and Lc. rhamnosus BFE5263.

Probiotic products are viewed as an alternative to the use of antibiotics in freshwater fishes farming. Probiotic organisms include bacteria, yeast, and filamentous fungi offering different benefits to fish including growth promotion, inhibition of pathogen colonisation, and improvement of nutrient digestion, water quality, and stress tolerance, as well as enhancement of reproduction. For these reasons, this review aims to identify the main trends in probiotic amendment in freshwater fishes. Strategies to incorporate the probiotic strains in the fish feed or pellets to allow optimal viability of the strains as they reach the fish gastrointestinal tract (GIT) are crucial in probiotic research and commercial applications for freshwater fish.

This article describes thecarrageenan supplement and its effects on the body in in vitro and in vivo experiments. The experimental part is devoted to analysis of the intestinalmicrobiota of laboratory rats with the consumption of the carrageenan dietary supplement in the amount of about 4,4 % of the standard feed.

The spatial distributions of properties of biofilms have been investigated by using three different kinds of biofilms as test materials. Biofilms, cultured by laboratory-scale rotating drum biofilm reactors with synthetic wastewater, were first cut into 10 to 20 µm thick slices using a microtome, and then apportioned into samples representing 3 or 4 layers. The biofilm properties of each layer were investigated by measuring the densities, phospholipid concentrations, and AR18 dye adsorption abilities. The bacterial population distributions and the metabolically active bacterial distributions were studied by plate count methods or a MPN method, and the tetrazolium dye (INT) reduction method, respectively. Based on statistic evaluations, the micro-slicing technique, the procedure for analyzing phospholipid concentrations of biofilms, and the AR18 dye adsorption tests were suitable to be used in biofilm studies. It was found that the densities of biofilms in the bottom layers were 4 to 7 times higher than those in the top layers. For thick biofilms (thickness > 500 µm), the INT active bacteria decreased from 82-89% in the top layers to 5-11% in the bottom layers. The porosities of thick biofilms changed from 83-92% in the top layers to 56-64% in the bottom layers. For thin biofilms (thickness < 500 µm), the porosities of biofilms changed from 72-75% in the top layers to 35-44% in the bottom layers. Highly spatial distributions of bacterial populations, mean pore radius, and specific surface areas were also observed. As a result of these spatial distributions, the ratio of effective diffusivity to diffusivity in the bulk solution also shows a decrease with depth of the biofilm. Assuming biofilm properties are of a uniform distribution may be an over-simplified assumption, valid only in specific cases.

A study was designed to determine populations of aerobic bacteria, coliforms, sorbitol-negative bacteria, and Listeria monocytogenes during display at 4 and 12°C of ground beef patties made with meat from animals fed diets supplemented daily (for 100 days) with 0, 1,000, or 2,000 IU of vitamin E. The patties (113.5 g) were either left uninoculated or were inoculated with Escherichia coli O157:H7 or L. monocytogenes and were tray-overwrapped and stored (at 4 or 12°C for 8 to 10 or 4 to 6 days, respectively) while being continuously exposed to fluorescent light in a display setting. Patties were visually evaluated for overall appearance (based on color and/or discoloration) twice a day and analyzed for microbiological counts at 2-day intervals during display at 4°C and at 0, 1, 2, 3, 4, and 6 days during display at 12°C. Use of beef from animals fed supplemental vitamin E (“high-vitamin E beef”) resulted in ground beef patties which, when stored at 4°C, maintained visually acceptable color longer than did patties made from control beef (from animals not fed supplemental vitamin E), but effects on microbial growth were less pronounced. In general, use of high-vitamin E beef versus control beef in patty manufacture had no major effect on populations of aerobic bacteria, coliforms, sorbitol-negative bacteria, or L. monocytogenes in ground beef patties displayed at 4 or 12°C. Listeria monocytogenes multiplied at 12°C, but growth was similar among ground beef patties made from high-vitamin E beef versus control beef. Overall, changes in bacterial populations were similar in ground beef patties derived from meat from animals with or without added vitamin E in their diets, but control ground beef became visually unacceptable sooner.

Bologna products most frequently are stored and consumed as refrigerated products. Thus bacteria that survive processing or those that contaminate the product subsequent to processing are not destroyed. Ten types of presliced, vacuum-packaged bologna products were purchased from a high-volume retail market and analyzed for total aerobic plate count (APC) and common foodborne pathogens. No Salmonella were isolated. Less than 1% of the 419 samples analyzed contained either Clostridium perfringens or Escherichia coli, Staphylococcus aureus was isolated from 4% of the samples, but only one sample contained more than 1000/g. Just over 5% of the samples contained coliform organisms. The manufacturer appeared to play an important role in bacterial quality of the finished items. An APC < 5 × 106/g is a realistic criterion for bologna products at the time of delivery to retail markets.
