Impact of exogenous carbohydrases and phytase on growth performance and nutrient digestibility in broilers

The effect of supplementing corn–soybean-based diets with Rovabio®Max AP, an exogenous enzyme mix with carbohydrase (xylanase and β-glucanase) and phytase activities to broiler chickens was evaluated. Male Ross 708 broilers were assigned to four diets: positive control (PC), negative control (NC), deficient in metabolizable energy (ME), crude protein (CP), calcium (Ca) and non-phytate phosphorus (nPP), relative to the PC, by 100 kcal kg−1, 5 g kg−1, 1.1 g kg−1and 1.6 g kg−1, respectively, in the starter and grower diets; and NC+Rovabio®at 0.05 or 0.075 g kg−1diet. Starter diets were fed from day 0 to day 21 and grower diets from day 22 to day 42. Body weight (BW) and body weight gain on days 22 and 42 were increased (P<0.05) by enzyme supplementation. Enzyme addition increased (P<0.05) feed intake from day 22 to day 42 and overall (day 0 to day 42). Gain to feed ratio (G:F) was significantly increased (P<0.05) with enzyme supplementation during both starter and grower phases. Enzyme addition increased (P<0.05) serum phosphorus (P) concentration on day 21. On day 42 enzyme supplementation increased (P<0.05) the apparent ileal digestibility of P, dry matter (DM), energy, and N. Addition of a combination of carbohydrases (xylanase and β-glucanase) and phytase to corn–soybean-based diets deficient in ME, CP, Ca and P resulted in a significant increase in growth performance and utilization of P, DM, energy and N in broiler chickens.

Effect of carbohydrases and phytase supplementation on growth performance, nutrient utilization and gut health of nursery pigs was evaluated. Pigs were blocked by body weight (BW) and sex and allocated to four treatments. Treatments were a positive control (PC), a negative control (NC) deficient in metabolizable energy (ME), crude protein (CP), Ca, and non-phytate P (nPP), NC plus Rovabio® Max AP enzyme mix, at 0.05 and 0.075 g kg−1. Apparent total tract digestibility (ATTD) was determined in faecal samples. Apparent ileal digestibility (AID) was determined in ileal digesta samples collected after euthanasia. Lower final BW and average daily gain (ADG) (P < 0.05) were observed in NC compared with PC. Enzyme at 0.05 g kg−1increased (P < 0.05) BW on d 14 and d 41, respectively, and also increased ADG. Enzyme at 0.075 g kg−1increased BW on d 14 and ADG on d 0 to d 14 (P < 0.05). Feed efficiency [gain to feed ratio (G:F)] was greater (P < 0.05) in PC than NC from d 15 to d 41 and from d 0 to d 41. No difference in G:F was observed with enzyme supplementation. Higher (P < 0.05) serum Ca and bone ash were observed in PC than NC. Enzyme increased the ATTD of Ca and P (P < 0.05) compared with NC.

A total of 600 one-day-old male Ross 308 broiler chickens with an average body weight of 43.00 ± 0.42 g were used in a 35 d growth assay to investigate the supplemental effects of three levels (600, 1200, and 1800 β-glucanase units kg−1) of β-glucanase on growth performance, nutrient digestibility, blood parameters, and meat quality in wheat–barley–soybean-meal-based broiler diet. During days 19–35 and overall period, chickens’ body weight gain and feed conversion ratio were significantly (P < 0.05) improved linearly following the levels of β-glucanase supplementation. The digestibility of crude protein and gross energy were significantly (P < 0.05) increased linearly with the levels of β-glucanase supplementation. Significant linear increase (P < 0.05) due to increasing β-glucanase supplementation was observed for blood urea nitrogen (BUN) concentration. Increasing β-glucanase supplementation did not influence (P > 0.05) the meat quality. In conclusion, broiler diets supplementation with increasing β-glucanase could improve the growth performance, enhance the digestibility, and modulate the concentrations of BUN linearly. Meat quality was not influenced.

A series of experiments was conducted to determine the nutritive value of wheat screenings, bakery by-products and wheat mill run used by the feed industry in Canada and to investigate the effect of enzyme supplementation on available energy content, nutrient digestibility and broiler chicken growth performance. Broiler chicken performance (2-wk growth trial) was compared using a wheat/soybean meal-based control diet and diets substituted with wheat by-products (wheat screenings, 200 g kg-1; bakery by-products or mill run, 100 g kg-1). In addition, diets containing one sample of each of the by-products were fed to broiler chickens with enzyme supplementation (Superzyme W®). Chicken performance was not affected by dietary substitution of wheat by-products for wheat as values for control, wheat screenings (three samples), bakery by-products (two samples) and mill run (one sample) were similar for body weight gain (514, 520, 515, 500 g bird-1) and for feed to gain ratio (1.52, 1.52, 1.52, 1.50), respectively. Response to enzyme supplementation was highest for wheat screenings showing a statistically significant (P ≤ 0.05) improvement in body weight gain (523 vs. 548 g bird-1) and feed to gain ratio (1.54 vs. 1.47). To further substantiate the importance of enzyme supplementation, a long-term production trial with broiler chickens was conducted. Five replicate pens of 60 birds per treatment were used in the starter (21 d) and grower (16 d) phases of the experiment consisting of three treatments: positive control (PC) (wheat/soybean meal/fish meal), negative control (NC) (wheat/wheat screenings/barley/soybean/ canola meal) and NC + Enzyme (Superzyme W®). In comparison to PC, the NC diet contained 5% less metabolizable energy, 8–10% less crude protein, amino acids and calcium and 25% less available phosphorus. Body weight gain and feed conversion ratio averaged 2.14, 2.10, 2.17 kg bird-1 and 1.68, 1.76, 1.66 for PC, NC and NC + Enzyme, respectively. A significant (P < 0.05) improvement in broiler chicken performance with enzyme supplementation was further substantiated by the same magnitude of difference in dry matter (71.2, 68.7, 75.1%), starch (92.5, 90.3, 95.6%), non-starch poly saccharides (NSP) (4.8, 15.0, 36.1%), energy (AME) (12.37, 11.83, 12.84 MJ kg-1) and phytate (44.3, 37.5, 69.5%) digestibilities. It is evident from this study that the use of wheat by-products in concert with an effective enzyme supplement in broiler chicken diets will allow for optimum growth performance. Abstract Two experiments were conducted to investigate the effects of a combined α-galactosidase and xylanase preparation on nutrients digestibility and growth performance in broiler chickens. Experiment 1 had 240 broilers allocated to 3 treatments with the dietary supplementation of 0, 300 and 500 g/t of the enzyme combination. Diet and amino acid (AA) digestibility were assessed. Experiment 2 was a 2 × 3 (enzyme × diet) factorial arrangement with 10 replicates of 12 male broilers per replicate. Diets were based on corn-Soybean meal (SBM) diet and had 3 nutritional levels (normal, 2% apparent metabolizable energy (AME) and crude protein (CP) reduction, and 4% AME and CP reduction). Each of these diets was fed with or without enzyme supplementation. Growth performance, chyme viscosity, nutrients digestibility, and endogenous enzymes activity were assessed. In experiment 1, enzyme supplementation improved the digestibility of Ca (P = 0.025) and ileal digestibility of total AA, Pro, Alu, Ile, Lys, His, Thr, Glu, Val, Leu, Tyr and Phe (P &lt; 0.05), and also tended to increase the AME of diets (P &lt; 0.10). In experiment 2, broilers fed the corn-SBM diet with 4% nutrient reduction had better growth performance (P &lt; 0.05), jejunal digesta viscosity at 42 days (P &lt; 0.01), and lower digestibility of gross energy (GE) (P &lt; 0.05) when compared to those fed the normal nutrient diet. Enzyme inclusion increased digestibility of CP (P = 0.044), GE (P = 0.009), raffinose (P &lt; 0.001) and stachyose (P &lt; 0.001), improved average daily gain (P = 0.031), and reduced jejunal digesta viscosity at 42 days (P = 0.011). Besides, similar improvements trend in amylase, trypsin, sucrase, and maltase activity with enzyme inclusion were observed as with energy. These data support that the enzyme supplementation increased nutrients and ileal amino acid digestibility, improved performance and endogenous enzymes activity.

Ross 308 broiler chicks (n = 240) aged 1 day were assigned to five groups for eight replicates (six chicks for each) (3♂ and 3♀). Basal dietary groups were supplemented by 2000, 4000, and 6000 mg/kg cinnamon (CN) for 21 days. Basal diet alone was used as a negative control, and basal antibiotic diet (Colimox) was used as a positive control. At 10, 14, and 21 days of age, chicks that received 2000 mg CN and Colimox had a higher body weight, resulting in an increase in body weight gain. CN also resulted in the maximum improvement in the feed conversion ratio and feed efficiency over 1–21 days at the level of 2000 mg/kg. At days 10, the maximum relative breast weight was 2000 mg/kg of CN. Mean serum albumin concentrations, duodenal villus height, and goblet cell density increased (p < 0.05) by 2000 mg/kg of CN, and mean serum globulin and total protein concentrations and crypt depth increased (p < 0.05) by 6000 mg/kg of CN compared with control. Increased cecal Escherichia coli number was CN dose-dependent. In conclusion, dietary inclusion of 2000 mg/kg CN can be applied as an alternative to in-feed antibiotics for broiler starter diet.

The present study was conducted to evaluate the effect of different levels of methionine supplementation on feed intake, nutrient digestibility and growth performance of growing rabbit. Sixteen weaned crossbred New Zealand White (NZW) growing rabbits (30-35 d) were distributed into four treatment groups having four replications in each group using a Completely Randomized Design (CRD). Basal diet composed of green grass (dhal grass) and concentrate mixture which was offered ad libitum basis for 56 days period. Four levels of methionine such as 0.0% (control), 0.15%, 0.25%, and 0.35% were supplemented randomly to rabbits. Results showed that supplementation of methionine did not affect green grass intake. Cumulative as well as daily concentrate and DM intake were significantly (p<0.05) higher for all methionine groups than control group. Final body weight gain as well as daily, weekly and cumulative body weight gains were improved significantly with increasing level of methionine. It was found that methionine had significant (p<0.01) effect on digestibility of DM, CP, NFE and EE but CF digestibility did not differ significantly. Digestibility was improved with increasing the level of methionine. Feed conversion ratio also decreased significantly with methionine supplementation, and 0.25% methionine group showed the best performance among the four treatments. One of the widespread spices is garlic which consumed by people of different parts of the world but its valuable growth promoting effect is not sufficiently studied in poultry. Growth promoting effect of garlic in broiler chickens was studied together with its carcass characteristics and blood parameters. Broilers were grouped into four: A (1%), B (2%), C (positive control) and D (negative control).  Aqueous extract of garlic (1% and 2%) was prepared by extraction and were administered with drinking water. A group (1%) shows best performance with regards to body weight gain rather than B (2%), positive and negative control. At the end of experiment (35th day), in A group, the live body weight, dressed weight, feed conversion ratio, liver, gizzard, heart, spleen, and pancreas were 1833.2±2.1g (P<0.05), 1229.7±1.9g (P<0.05), 1.63(P<0.05), 47.13±0.4g, 26.15±0.7g, 9.71±1.8g, 2.21±0.2g, 2.27±0.1g respectively. The mean Total Erythrocyte Count (TEC), Haemoglobin concentration (Hb) and Packed Cell volume (PCV) were 2.9± 0.1million/cm3,7.5g ± 0.2g, and 25.8 ± 0.6%, respectively. It is recommended that 1% aqueous extraction of garlic shows better growth and improved carcass qualities in broilers.

A total of 480 one-day-old male Ross 308 broilers with an average initial body weight (BW) of 42 ± 1 g was randomly divided into four treatments (8 replicates/treatment, 15 birds/replicate) by their BW to evaluate the influence of a cocktail of protease and xylanase in different energy density of corn- and soybean-meal-based diet on broilers. This experiment lasted for 35 d and there were two phases; starter (1–18 d) and finisher (19–35 d). The experiment was a 2 × 2 factorial arrangement with two levels of metabolizable energy and the cocktail enzyme with 200 mg of protease kg−1 diet and 100 FXU xylanase kg−1 diet. Both high-nutrient density and cocktail enzyme supplementation increased (P < 0.05) the body weight gain (BWG) during day 1–18 and day 1–35 as well as dry matter retention on day 35. Broilers fed the high-nutrient-density diets had higher (P < 0.05) energy retention on day 35 and relative breast muscle weight. Broilers fed the cocktail enzyme diets significantly increased nitrogen (N) retention and decreased the levels of ammonia (NH3) and hydrogen sulphide (H2S) emission. In conclusion, dietary supplementation of the cocktail enzyme component improved BWG, N retention, and reduced excreta noxious gas emission in broilers.

The aim of this study was to determine the calcium requirements of broiler chickens in starter phase with the addition of phytase enzyme. This experiment used 1,200 broiler chickens (mixed sex) for 21 days rearing period in a closed house. The basal diets was corn and soybean meal which contained 0.097% Ca and 0.123% P-av. The treatments consisted of 6 diets treatment: P1 (basal diet with the addition of 0.42% non-phytate phosphorus (NPP), and then P2 to P6 were basal diet with the addition of 0.22% NPP and 1,000 FTU/kg phytase and the addition of calcium (Ca) at different levels: 0.82% (P2); 0.74% (P3); 0.66% (P4); 0.58% (P5) and 0.50% (P6). The growth performance, feed efficiency, and bone mineralization parameters were studied using Oneway ANOVA in a Completely Randomized Design. Duncan's new Multiple Ranges Test was used to separate means with significant differences. Results showed that 0.90% Ca supplementation without phytase and the reduction levels of Ca from 0.82% to 0.50% increased the amount of feed consumed by birds in all growth phases (P<0.05). The addition of phytase had beneficial effects on increasing body weight (P<0.05) and average of body weight gain (P<0.05), and decreased the value of FCR in 11 - 21 days and 1 - 21 days. The addition of phytase increased protein and energy consumption, followed by increased in the value of PER and  EER (P<0.05). When phytase was added in the diets, reduction levels of Ca in the diets up to 0.50% did not give any adverse effect on the tibia bone ash. It can be concluded that feeding with phytase can sustain growth performance, feed efficiency, and bone mineralization.
