Monitoring of water quality from the presence of polyaromatic hydrocarbon (PAHs) compounds and its derivates are important for keeping the healthy aquatic environment. Some of those derivates are phenol and several related compounds sharing simmilar structures. This reseach aimed for the detection of those phenol and several similar compounds monitoring due to PAHs degradation. Three identified bacterial isolates of Pseudomonas aeruginosa, Staphylococcus sciuri, and Bacillus amyloliquifaciens were selected based on their phenol degradation characters. On physiological properties all three isolates were observed to degrade several hydrophobic substances such as for naphthalene and anthracene. Yet, genetic analysis indicated that the phenolic degradating oxygenase gene was detected only in the P. aeruginosa and S. sciuri. Applying those isolates for biofilm as biosensor showed a sufficient analytical performance such as their limit of detection between 0.1-0.5 μM.

A new method of phenol measurement based on cyclic voltammetry has been tried using Pseudomonas fluorescens biofilm as a bioreceptor. The research is aimed at developing a new measurement for detecting phenol and related compounds using bacterial biofilm. Therefore, it can be further developed for in situ phenol and related compounds monitoring. The trial evaluated several variables, such as optimum pH value during measurement and biofilm development time. Measurement was conducted at a relatively low concentration (1 ppb – 1 ppm). Based on the obtained data, the optimum measurement was at pH 8 and on the 6th day of developing bacterial biofilm that produced the maximal electrical current. The analytical determination revealed that the sensitivity was at 0.0433 mA/ppm with an R2 value of 0.9208. The apparent limit of detection value was extrapolated around 0.1 ppm; thus, it is suggested that biofilm had potency for measuring phenol at low concentrations. However, this trial needs improvements on biofilm stabilization for broader measured phenol concentrations.

The present study included the collection of 100 samples from various clinical sources for investigating the presence of P. aeruginosa in those sources, the samples have been collected from some hospitals in Baghdad and Hillah city (Al-qassim General Hospital, ,Al-hillah teaching hospital,and Al-hashimya General hospital ) which included wounds, burns, ear and sputum infections. The study was carried out through October 2017 till the end of March 2018. The samples were identified based on the morphological and microscopically characteristics of the colonies when they were culturing or number of culture media as well as biochemical tests, molecular identification were also used as a final diagnostic test for isolates that were positive as they belong to P.aeruginosa bacteria during previous tests based on the OprD gene which has specific sequences for P.aeruginosa bacteria as a detection gene and also consider as virulence factor so it have a synonyms mechanism to antibiotic resistance . The results of the final diagnosis showed that 38 isolates belong to target bacteria were distributed as 18 of burns, 11 isolates of wounds, 6 isolates of ear infection and 3 isolates of sputum, The examination of the sensitivity of all bacterial isolates was done for elected 38 isolation towards the 9 antibiotic by a Bauer - Kirby and the isolates were resistant for a number of antibiotics used such as Ciprofloxacin 65.7%, Norflaxacin 71%, Imipenem 63.1% Meropenem 68.4%, Gentamicin 65.7%, Amikacin 26.3%, Cefepime 68.4%, Ceftazidime 65.7% and Piperacillin 57.8%.Molecular method , All isolates (38) of P. aeruginosa positive for the diagnostic special gene (OprD) genes (100%).

Pseudomonas aeruginosa is one of opportunistic pathogen forming bacterial biofilm. The biofilm sustains the bacterial survival and infections. This study aimed to assess the activity of water extract of papaya leaves on inhibition of cells attachment, growth and degradation of the biofilm using crystal violet (CV) biofilm assay. Research results showed that water extract of papaya leaves contains alkaloids, tanins, flavonoids, and steroids/terpenoids and showed antibacterial activity and antibiofilm against P. aeruginosa. Addition of extract can inhibit the cell attachment and was able to degrade the biofilm of 40.92% and 48.058% respectively at optimum conditions: extract concentration of 25% (v/v), temperature 37.5 °C and contact time 45 minutes. With a concentration of 25% (v/v), temperature of 50 °C and the contact time of 3 days, extract of papaya leaves can inhibit the growth of biofilms of 39.837% v/v.

To determine the resistance pattern of bacterial pathogens involved in infections of the patients aged between 18-64 years, admitted in a ICU from a 1518-bed university-affiliated hospital. A retrospective study of bacterial pathogens was carried out on 351 patients aged between 18-64 years admitted to the ICU, from January to December 2017. In this study there were analysed 469 samples from 351 patients (18-64 years). A total of 566 bacterial isolates were obtained, of which 120 strains of Klebsiella spp. (35.39%%), followed by Nonfermenting Gram negative bacilli, other than Pseudomonas and Acinetobacter (NFB) (75- 22.12%), Acinetobacter spp. (53 - 15.63%), Pseudomonas aeruginosa and Proteus (51 - 15.04%), and Escherichia coli (49 - 14.45%). The most common isolates were from respiratory tract (394 isolates � 69.61%). High rates of MDR were found for Pseudomonas aeruginosa (64.70%), MRSA (62.65%) and Klebsiella spp. (53.33%), while almost all of the isolated NFB strains were MDR (97.33%). There was statistic difference between the drug resistance rate of Klebsiella and E. coli strains to ceftazidime and ceftriaxone (p[0.001), cefuroxime (p[0.01) and to cefepime (p[0.01). The study revealed an alarming pattern of antibiotic resistance in the majority of ICU isolates.

The bacterial resistance development due to the incessant administration of antibiotics has led to difficulty in their treatment. Natural adjuvant compounds can be co-administered to hinder the pathogenesis of resistant bacteria. Sotolon is the prevailing aromatic compound that gives fenugreek its typical smell. In the current work, the anti-virulence activities of sotolon on Pseudomonas aeruginosa have been evaluated. P. aeruginosa has been treated with sotolon at sub-minimum inhibitory concentration (MIC), and production of biofilm and other virulence factors were assessed. Moreover, the anti-quorum sensing (QS) activity of sotolon was in-silico evaluated by evaluating the affinity of sotolon to bind to QS receptors, and the expression of QS genes was measured in the presence of sotolon sub-MIC. Furthermore, the sotolon in-vivo capability to protect mice against P. aeruginosa was assessed. Significantly, sotolon decreased the production of bacterial biofilm and virulence factors, the expression of QS genes, and protected mice from P. aeruginosa. Conclusively, the plant natural substance sotolon attenuated the pathogenicity of P. aeruginosa, locating it as a plausible potential therapeutic agent for the treatment of its infections. Sotolon can be used in the treatment of bacterial infections as an alternative or adjuvant to antibiotics to combat their high resistance to antibiotics.

Abstract We describe here a direct a radioimmunoassay (RIA) for the determination of 6-sulfatoxymelatonin (aMT6s) in plasma, with iodinated aMT6s as tracer. The aMT6s antiserum was raised in rabbit by immunization with a bovine serum albumin conjugate, giving negligible cross-reactivities for related compounds. The low limit of detection (15 pmol/L) allowed a direct assay that required only a 100-microL plasma sample. Dilutions of plasma and of synthetic aMT6s gave the same parallel response in the RIA. A preliminary study showed a circadian variation in healthy volunteers, with mean concentrations ranging from 52 (at 1600-2100 h) to 378 pmol/L (at 0400 h), whereas this rhythm was abolished in pinealomectomized patients. After administration of melatonin orally, or by infusion, the aMT6s concentrations in plasma concorded with previous data on aMT6s production and pharmacokinetics, with aMT6s being cleared from plasma more slowly than melatonin. This assay should have practical application in the development of new pharmaceutical formulations that minimize the hepatic metabolism of melatonin.

The present study was conducted to investigate the water quality of the Trstionica River, Bosnia and Herzegovina. The physicochemical properties (temperature, pH, conductivity, total solids after evaporation at 105 °C), content of metals (calcium (Ca), cadmium (Cd), chromium (Cr), copper (Cu), iron (Fe), magnesium (Mg), manganese (Mn), sodium (Na), nickel (Ni), lead (Pb) and zinc (Zn)), and anions (chloride (Cl–), and phosphate (PO43–)) were determined in water samples collected at seven locations during two sampling periods: unstable weather conditions (precipitation), and stable weather conditions (without precipitation). There was a significant difference in the content of individual parameters in the river water depending on the sampling time. For determination of metals concentrations, which were below the limit of detection, a preconcentration method using an ion-exchange resin was applied. The metals concentrations during the rainy day were in the order Ca > Mg > Na > Fe > Cu > Zn > Pb > Mn with mean values of 343, 6.03, 1.94, 0.18, 0.20, 0.03, 0.02, 0.01 mg dm–3, respectively, and during stable weather conditions: Ca > Mg > Na > Cu > Fe > Mn > Zn with mean values of 288, 7.62, 2.38, 0.11, 0.10, 0.01, 0.01 mg dm–3, respectively. Cd, Cr, and Ni concentrations were below limit of detection in both cases. Obtained values were compared with World Health Organization (WHO) regulations. The results showed that the Trstionica River in the investigated part of the stream meets most of the parameters required by the regulations. The correlation between analysed parameters was assessed, as well. Based on the calculated water quality index values, the water of Trstionica River falls into the category of excellent water.

A fish pond with recommended water quality will produce healthy fishes. Fish ponds with poor water quality will cause fish mortality and outbreak of diseases to fish consumers. Physicochemical analysis was done using standard analytical methods, the total bacterial count was determined by dilution and membrane filtration techniques. Parasitological analysis was done using the centrifugation method. A total of fifteen well waters were sampled during wet season. Results showed that the temperature ranged from 27°C to 29°C, pH, 6.21 to 8.15; dissolved oxygen, 4.28 mg/l to 5.78 mg/l, electrical conductivity, 166.36 µs/cm to 394.00 µs/cm; total dissolved solids, 41 mg/l to 121 mg/l; total suspended solids, 1.00 mg/l to 19.40 mg/l; total solids, 42.00 mg/l to 140.4 mg/l; turbidity values, 7.01 NTU to 10.36 NTU; nitrate, 3.10 mg/l to 28.00 mg/l; total alkalinity, 36 mg/l to 91 mg/l; phosphate, 1.26 mg/l to 13.11 mg/l; sulphate, 0.39 mg/l to 4.37 mg/l; total chloride, 7.08 mg/l to 14.19 mg/l; carbonates, 1.33 mg/l to 2.35 mg/l; bicarbonates, 34.59 mg/l to 89.38 mg/l; total hardness, 25.31 mg/l to 53.04 mg/l; calcium hardness, 23.94 mg/l to 51.96 mg/l; magnesium hardness, 1.08 mg/l to 4.20 mg/l; total acidity, 2 mg/l to 22 mg/l; potassium, 0.04 mg/l to 2.23 mg/l; cadmium, 0.00 mg/l to 0.04 mg/l; lead, 0.01 mg/l - 0.16 mg/l; chromium, 0.00 mg/l - 0.03 mg/l; mercury was not detected, copper, 0.00 mg/l - 0.04 mg/l; arsenic, 0.00 mg/l - 0.02 mg/l; zinc, 0.00 mg/l to 0.02 mg/l; iron, 0.01 mg/l - 1.19 mg/l. The total bacterial counts ranged from 3.60-4.12 log cfu/ml; total coliforms, 14-46 cfu/100ml, Vibrio cholerae, 0-11 cfu/100ml; Vibrio parahaemolyticus, 0-15 cfu/100ml; faecal coliform, 1-9 cfu/100 ml; Acinetobacter calcoaceticus, 0-8 cfu/100 ml; Bacillus subtilis, 0-9 cfu/ml; Staphylococcus aureus, 0-5 cfu/ml; Pseudomonas aeruginosa, 0-12 cfu/100 ml; Pseudomonas fluorescens, 0-12 cfu/100 ml and Clostridium perfringens were not detected in any of the samples. Twelve bacterial species namely Klebsiella pneumoniae, Acinetobacter calcoaceticus, Escherichia coli, Staphylococcus aureus, Vibrio cholerae, Pseudomonas fluorescens, Pseudomonas aeruginosa, Proteus mirabilis, Vibrio parahaemolyticus, Bacillus subtilis, Shigella flexineri and Salmonella typhi were isolated and identified using standard analytical and molecular procedures. Parasites identified were Ichthyobodo species, Diplostomum species, Myxobolus species, Chilodonella species, Bothriocephalus species, Ambiphrya species and Leech species. Salmonella typhi had the highest frequency of isolation (20.63%) while Acinetobacter calcoaceticus and Staphylococcus aureus had the lowest frequency of isolation (2.83%). Ichthyobodo species had the highest frequency of isolation (21.43%) while Leech species had the lowest frequency of isolation (5.71%). Some of the physicochemical, bacteriological and parasitological parameters had values above World Health Organization admissible limits and therefore proper sanitary practices and water treatments must be employed to prevent epidemic among fish consumers.
