Purpose: To investigate the gastroprotective effect of Alpinia conchigera rhizome in an ulcer rat model. Method: Sprague-Dawley rats were treated with ethanol extract of Alpinia conchigera at doses of 75 and 150 mg/kg body weight. After 28 days, the rats were sacrificed. Ulcer index, total acidity, pH, hemoglobin levels, and hemorrhagic lesions were determined through macroscopic assessment and histopathological examination. The levels of lipid peroxidation (LPO), superoxide dismutase (SOD), and catalase (CAT), and inflammatory markers (TNF-α, IL-1β, and IL-6) were also determined. Results: Alpinia conchigera significantly reduced ulcer index, total acidity, pH and hemoglobin levels (p < 0.05). Results from histology revealed that the extract protected the mucosa from lesions. Moreover, the extract significantly enhanced gastric mucosal activities of SOD and CAT, and decreased LPO levels and plasma concentrations of TNF-α, IL-1β, and IL-6 (p < 0.05). Conclusion: These results validate the traditional claims on the gastroprotective effects of Alpinia conchigera as revealed through its antioxidant and ulcer-healing properties by stimulation of important cellular mechanisms.

Background: Aging is a process that every living creature will pass through and involves various factors. Increased inflammatory factors can be caused by hypoxia which forms Reactive Oxygen Species (ROS) and damages macromolecules and causes the acceleration of the aging process. Maja plants, empirically have long been used as medicinal plants, research on Maja leaves shows anti-inflammatory and antioxidant potential.Objective: The purpose of this study was to find out and understand the potential of Maja fruit in the presence of inflammatory markers of IL-6 and TNF-α which are involved in the aging process.Methods: This research was in vivo experimental, using male experimental animals Sprague Dawley rats divided into 8 groups (n = 4), and divided into 2 groups (fed with Maja ethanol extract (400 mg/ kg/day, 14 days) and not force-fed). Each group was divided into 4 subgroups (normoxia, hypoxia (O2 8%, N2 92%) for 3, 7 and 14 days). At the end of the trial period, animal blood was examined for IL-6 and TNF-α concentrations using the ELISA method.Results: The results showed an increase in levels of IL-6 and TNF-α both in the group fed and not fed in line with the duration of hypoxia when compared with controls. However, groups that are not force-fed show a higher pattern compared to those who are force-fed.Conclusion : The ethanol extract of Maja fruit can help slow down the aging process.

BACKGROUND <br />Guava is an herbal with proven antioxidant and anti-inflammatory properties. The aim of this study was to investigate the anti-arthritic activity of the ethanol extract of Psidium gujava leaves (EEPG) against complete Freund’s adjuvant (CFA) induced arthritis in rats. <br /><br />METHODS<br />An experimental study was conducted on 40 male Wistar Sprague Dawley rats, which were divided into 5 groups. Each group was induced with 0.2 mL CFA (1 mg/mL) on day 1 and 0.1 CFA mL booster injection on day 5. Group I served as an arthritic control, group II received dexamethasone (6.75 mg.kg-1 orally), group III, IV and V received EEPG at oral doses of 250, 500, and 750 mg/kg BW, respectively, on days 14 to 28. Anti-arthritic activity was observed from the arthritis score, the paw circumference was measured on days 0, 1, 4, 8, 12, 16, 20, 24, and 28, the mobility score was determined on days 12 and 28, and the histolopathology of the knee joint was examined on day 29. <br /><br />RESULTS<br />Ethanol extract of Psidium guajava leaves significantly suppressed the swelling of the paws in chronic phase based on increasing of edema (%), while starting on day 20. EEPG at 250 mg/kg was most effective in significantly reducing arthritis scores (p&lt;0.05). Histopathological examination showed repair of the knee joint synovial membrane and cartilage.<br /> <br />CONCLUSIONS<br />Psidium guajava leaf extract is effective in decreasing the inflammatory response and arthritic symptoms in rats with adjuvant-induced arthritis. Psidium guajava leaves can be developed into an alternative anti-arthritis treatment.

The current study aimed to determine the protective effect of P. reticulatus on ethanol-induced gastric ulcer. For this purpose, thirty-six Sprague-Dawley rats were divided into six groups. The first group served as normal control, while, in other five groups, absolute ethanol was used to induce gastric ulcer. Group II served as a diseased group, while groups III, IV, and V were treated with methanol extract, ethyl acetate fraction, and n-hexane fraction, respectively, in a dose of 400 mg/kg bodyweight. Group VI was given omeprazole in a dose 20 mg/kg bodyweight. The stomachs were removed, ulcer score was evaluated, and histopathological examination of gastric lumen was conducted. Total acidity and pH values were determined in gastric juice. TNF-α and IL-8 mRNA expressions levels were determined using the reverse transcription real-time PCR method. The data indicated that P. reticulatus protected against gastric ulcer, which was evident by attenuation of ulcer score. The pretreatment with P. reticulatus raised the gastric pH and improved all evaluated histopathological parameters such as ulcer score, erosion score, hemorrhage score, fibrinoid necrosis score, inflammatory infiltrate score, and edema score. P. reticulatus significantly reduced mRNA expression levels of TNF-α and IL-8. In conclusion, P. reticulatus possess antiulcer property which might be attributed to downregulation of TNF-α and IL-8 expression levels.

The aim of present investigation was to study the Ulcerative colitis effect of extracts of Caesalpinia crista in acetic acid induced experimental colitis in Sprague Dawley rats. Sprague Dawley rats were divided into nine groups (n=6). The rats were received 7 days pretreatment with chloroform, ethyl acetate, ethanolic extracts of C. crista 200 mg/kg and 400 mg/kg. Ulcerative colitis was induced by intrarectal administration of 1ml of 4% acetic acid solution on 8th day. Prednisolone (2 mg/kg) was used as standard drug administered orally for 3 days. After 48 hrs of colitis induction animals were sacrificed by cervical dislocation to remove colon and distal 5 cm of the colon was dissected. Macroscopical study, Ulcer index of the colon, colonic myeloperoxidase (MPO) and malondialdehyde (MDA) level in colon tissue and blood were studied. Intrarectal instillation of acetic acid caused enhanced ulcer index, myeloperoxidase and malondialdehyde. Ethanol extract of C.crista showed significant effect in lowering ulcer index as well as neutrophil infiltration at a dose of 400 mg/kg in acetic acid induced colitis. The present investigation demonstrates that the ethanol extract of C.crista is of potent therapeutic value in the amelioration of experimental colitis in rat by inhibiting the inflammatory mediator.

AbstractTo investigate the effect of simulated weightlessness on the pharmacokinetics of orally administered moxifloxacin and the antacid Maalox or the antidiarrheal Pepto-Bismol using a tail-suspended (TS) rat model of microgravity. Fasted control and TS, jugular-vein-cannulated, male Sprague-Dawley rats received either a single 5 mg/kg intravenous dose or a single 10 mg/kg oral dose of moxifloxacin alone or with a 0.625 mL/kg oral dose of Maalox or a 1.43 mL/kg oral dose of Pepto-Bismol. Plasma concentrations of moxifloxacin were measured by HPLC. Pharmacokinetic data were analyzed using WinNonlin. Simulated weightlessness had no effect on moxifloxacin disposition after intravenous administration but significantly decreased the extent of moxifloxacin oral absorption. The coadministration of moxifloxacin with Maalox to either control or TS rats caused significant reductions in the rate and extent of moxifloxacin absorption. In contrast, the coadministration of moxifloxacin with Pepto-Bismol to TS rats had no significant effect on either the rate or the extent of moxifloxacin absorption. These interactions showed dose staggering when oral administrations of Pepto-Bismol and moxifloxacin were separated by 60 min in control rats but not in TS rats. Dose staggering was more apparent after the coadministration of Maalox and moxifloxacin in TS rats.

ABSTRACTObjective: To evaluate the antioxidant and chemotherapeutic potential of Curcuma amada Rhizome extract on benzo(a)pyrene (BaP) induced cervicalcarcinoma in Sprague Dawley rats.Methods: A total of 30 female Sprague Dawley rats were selected to establish cervical cancer model and then divided into 5 groups at random withsix mice in each group. Group 1 control, Group 2 BaP (oral), Group 3 BaP for 8 weeks and post-treated with cisplatin (intravenous administration),Group 4 BaP for 8 weeks and post-treated with 250 mg of ethanol extract of C. amada (oral), Group 5 BaP for 8 weeks and post-treated with 500 mgof ethanol extract of C. amada (oral). 4 weeks after the treatment, the animals were sacrificed, serum separated, and cervical tissues were dissected.Antioxidants and the markers carcinoembryonic antigen (CEA), cancer antigens (CAs) 125, gamma glutamyltransferase (GTT) were assayed in serumand the tissue was used for analyzing tumor burden and sectioned for histopathological assays.10% tissue homogenate was estimated for antioxidantsand membrane-bound enzymes.Results: BaP treated group showed significant (p<0.001) incidence of tumor burden, decreased activities of antioxidants, elevated lipid peroxidation,Na+/K+ adenosine triphosphatase (Na+K+ATPase), Calcium adenosine triphosphatase (Ca2+ATPase), Magnesium adenosine triphosphatase (Mg2+ ATPase),CEA, CA 125, GTT. Treatment with C. amada rhizome extract and standard drug cisplatin reverted the antioxidants, serum markers and tissue enzymes.Conclusion: From the results, it can be concluded that C. amada Rhizome extract ameliorated BaP induced oxidative stress in the cervicalcarcinogenicity of rats.Keywords: Curcuma amada, In vivo antioxidant, Chemotherapy, Benzo(a)pyrene, Cervical carcinoma, Tumor markers.

Objective: This study aims to formulate and characterize a transfersome apple peel extract, formulate it into a gel, and compare it with a control gelmade without transfersome.Methods: Both gels were evaluated, stability tested, and penetration tested using Franz diffusion cells on the skin of female Sprague-Dawley rats. Thetransfersome preparations were formulated with different concentrations of the active substance, quercetin: 0.5% (F1); 0.7% (F2), and 1.0% (F3).Results: Based on the characterization results, F1 was selected as the optimum gel formulation because it had spherical morphology, a Dmean volume of106.44±2.70 nm, a polydispersity index of 0.078±0.01, a zeta potential of −49.96±2.05 mV, and a drug efficiency entrapment percentage of 78.78±0.46%.The cumulative amount of quercetin that was penetrated with the transfersome gel was 1514.41±26.31 μg/cm2, whereas the penetration with thecontrol gel extract was 1133.62±18.96 μg/cm2. The cumulative percentages of the penetrated gel transfersome and gel extract were 78.40±1.89%and 49.89±0.88%, respectively. The fluxes of transfersome gel and control gel extract were 52.33±0.11 μg/cm²/hrs and 40.89±0.68 μg/cm²/hrs,respectively.Conclusions: Based on these results, it can be concluded that the gel with transfersome exhibited better penetration than the gel extract alone.

Objective: The central motive of this study is to explore the therapeutic impact of Diosgenin encapsulated Chitosan nanoparticles (DG@CS-NP) on mammary carcinogenesis in female Sprague Dawley rats via modulating hormonal status, cell proliferation, inflammatory responses, and Apoptosis. Methods: 7,12-dimethylbenz(a)anthracene (DMBA) was administered subcutaneously near the mammary gland (25 mg/kg b. wt) to provoke mammary tumor in female Sprague Dawley rats. Following the progress of a tumor, DMBA-induced tumor-bearing rats were medicated orally with 5 mg/kg b. wt of DG@CS-NP. Consequently, the expression of ER, PR, PCNA, Cyclin D1, NF-κB, TNF-α, Bcl-2, Caspases-3, and p53 in experimental rats were revealed via architectural immunohistochemistry. Further, Diosgenin interactions with these proteins were evidently confirmed by molecular docking analysis. Results: As a result, we noticed diminished levels of ER, PR, PCNA, Cyclin D1, NF-κB, TNF-α, and Bcl-2 expressions in DG@CS-NP medicated rats as well as with elevated levels of Caspases-3 and p53 expressions. In DMBA rats, the expressions were vice versa. Additionally, molecular docking analyses support these outcomes by highlighting the strong interaction between Diosgenin and breast cancer targets. Conclusion: These reports prove that DG@CS-NP imposes its therapeutic impact by hormonal adjustments, downregulating proteins involved in inflammation and cellular proliferation, and thereby promotes apoptosis by impeding apoptotic inhibitors.
