PHENOTYPIC CHARACTERIZATION OF BENECKEA PARAHAEMOLYTICA: A PRELIMINARY REPORT1

Eighty-six strains which were isolated from cases of gastroenteritis and had the general properties of the genus Beneckea were submitted to an extensive nutritional, physiological, and morphological characterization. The results indicated that this collection of strains, which included the type strain of Beneckea parahaemolytica, was phenotypically homogeneous and distinguishable from the other known species of Beneckea by multiple, unrelated, phenotypic traits. When grown in liquid medium, strains of B. parahaemolytica had single, sheathed, polar flagella; when grown on solid medium, these strains had unsheathed, peritrichous flagella in addition to the sheathed, polar flagellum. Additional traits of use for differentiation of this species from the remaining species of the genus Beneckea were the ability of B. parahaemolytica to grow at 40 C, utilize d-galactose, l-leucine, l-histidine, and putrescine and the inability to utilize sucrose, dl-β-hydroxy-butyrate or give a positive Voges-Proskauer reaction. The validity of some of the traits previously used to identify B. parahaemolytica as well as the possible difficulties encountered in the identification of this organism from marine sources are considered.

Abstract Background: The developmental biology of K. scorpioides is described, based on the phenotype. This species is important for the flora because they are excellent seed disseminators. In addition, basic embryological information is not yet fully clarified, and this research provides unprecedented information on the chelonian embryology of the Amazonian fauna. Methods: Females were monitored throughout the reproductive cycle, by video monitoring, to identify nests and the presence of newly laid eggs. At regular weekly intervals, embryo samples were collected fixed in a 4% paraformol solution and preserved in 70% alcohol. For the embryonic characterization we used a stereomicroscope and the scanning electron microscopy method. Results: We describe 15 embryonic stages for a 15-week (105-day) incubation cycle. Only at 42 days (6th week) was the morphological characterization of a chelonian observed and at the 12th week (Stage XII) the phenotypic characterization of the species kinosternon scorpioides. Conclusions: In view of the evidence, we found that these phases are similar to the other turtles, with structural variations in the appearance and disappearance of structures due to the specific characteristics for the species.

Phytophthora infestans, the causal agent of late blight disease, has been reported in the United States and Canada since the mid-nineteenth century. Due to the lack of or very limited sexual reproduction, the populations of P. infestans in the United States are primarily reproducing asexually and, thus, show a simple genetic structure. The emergence of new clonal lineages of P. infestans (US-22, US-23, and US-24) responsible for the late blight epidemics in the northeastern region of the United States in the summers of 2009 and 2010 stimulated an investigation into phenotypic traits associated with these genotypes. Mating type, differences in sensitivity to mefenoxam, differences in pathogenicity on potato and tomato, and differences in rate of germination were studied for clonal lineages US-8, US-22, US-23, and US-24. Both A1 and A2 mating types were detected. Lineages US-22, US-23, and US-24 were generally sensitive to mefenoxam while US-8 was resistant. US-8 and US-24 were primarily pathogenic on potato while US-22 and US-23 were pathogenic on both potato and tomato. Indirect germination was favored at lower temperatures (5 and 10°C) whereas direct germination, though uncommon, was favored at higher temperatures (20 and 25°C). Sporangia of US-24 released zoospores more rapidly than did sporangia of US-22 and US-23. The association of characteristic phenotypic traits with genotype enables the prediction of phenotypic traits from rapid genotypic analyses for improved disease management.

A preliminary morphological screening and isolation of bacterial colony from latex industrial wastewater was carried out. Bacteria colonies from latex processing wastewater were isolated from a local latex processing industry. It was found that 17 bacterial isolates had been purified grown on nutrient agar under 35˚C. The colonies were then purified and morphologically indicated via Gram staining and motility test. After morphological observation, it was identified that out of 17 isolates, 9 isolates were Gram positive and 8 isolates were Gram negative. There are 11 out of 17 colonies were rod-shaped bacterial colonies, while the other 6 colonies were cocci-shaped bacteria. There were 11 colonies of gliding bacteria, three colonies were non-motile bacteria and the other three colonies were flagellated bacteria. This study is only limited to morphological observation as the main aim of this study was to investigate the potential occurrence of viable growth in treated latex processing wastewater. The bacterial colonies were classified base on their morphological properties shown. This study has classified several genera such as Staphylococcus, Escherichia, Thiobacillus, Arthrobacter and other Genus. The growth curve of 17 isolates studied and the chemical oxygen demand were determined.

<p>Genetic divergence of five mulberry accessions including Embu, Thika, Thailand (<em>M. alba</em>), Kanva-2 and S41 (<em>M. indica</em>) grown in Kenya were examined using twelve phenotypic traits. The assessment of phenotypic traits was done in a field study in two localities, Nairobi and Eldoret. The traits that were significantly different across the mulberry accessions included lamina width and petiole length (P ? 0.01), petiole width and growth height (P ? 0.05), internodes distance and number of branches (P ? 0.001). The Duncan’s Multiple Range Test (DMRT) results were used to generate a dendrogram derived from hierarchical cluster analysis that further partitioned the mulberry accessions into four groups. Embu and Thailand accessions grouped together while S41, Thika and Kanva-2 accessions grouped separately. Embu and Thailand accessions were characterized by fewer numbers of branches than the rest of the accessions. Thika accession had high number of branches and short internode distance. Significant and positive correlations were found between leaf yield traits except in internode distance and number of branches which were significant and negatively correlated. Significant and positive correlations can be utilized since they are rewarding for mulberry leaf yield improvement.</p>

This work was carried out on the phenotypic characteristics of the Kababich sheep exploited in the peri-urban zone of N'Djamena (Chad). This phenotypic characterization of Kababich sheep should be continued on local breeds for genetic improvement and objective optimization of productivity. The objective of this study is the characterization of phenotypically Kababich sheep exploited in the peri-urban area of ​​N'Djamena-Chad. It was conducted in 39 farms and involved 1035 sheep aged less than one year old. Phenotypic traits were measured according to the guideline defined by FAO. A total of 12 variables were measured including five qualitative and seven quantitative. The results obtained show a positively significant correlation (p> 0.001) between the age of the sheep estimated by the breeder and the dentition (0.83). The ears are drooping and measure 17.90 ± 2.77 cm. The color of the brown dress (fawn) and the smooth and short coat are dominant. The females are matte and the rams of brown dress have horns in the forms of the vestiges. The Kababich is large with a height at the withers of 81.50 ± 9.40 cm, a rump height of 83.24 ± 9.42 cm and a chest circumference of 88.29 ± 9.22 cm. The body is 72.82 ± 9.63 cm long, has a basin width of 19.89 ± 2.53 cm and a chest depth of 40.68 ± 5.16 cm. These measurements show that Kababich is a meat breed by excellence. In addition, the sexual dimorphism very accentuated in this race orients towards a valorization of the young males in the fattening workshops.

This paper presents the results obtained from the synthesis and morphological characterization of different magnetite samples:  La0.67-x Prx Ca0.33 MnO3.LaMn1-x Cox O3 and LaMn1-x Nix O3 at 0.13 ≤ 𝑥𝑥 ≤ 0.67 produced by a solid-state reaction mechanism and 𝐿𝐿𝐿𝐿𝑀𝑀𝑀𝑀1−𝑥𝑥(𝐶𝐶𝐶𝐶/𝑁𝑁𝑁𝑁)𝑥𝑥𝑂𝑂3 at 0.0 ≤ 𝑥𝑥 ≤ 0.5 produced by the sol-gel method. These samples were characterized using X-ray diffraction spectroscopy and by measuring electric resistivity and magnetic susceptibility which were carried out as a function of temperature. Notably, the effects of strain and compressive strength on the lattices of magnetite samples were highly dependent on the concentration of 𝑃𝑃𝑟𝑟, 𝐶𝐶𝐶𝐶, and 𝑁𝑁𝑁𝑁. Moreover, the transition temperatures of metal-insulator and ferromagnetic-paramagnetic phases also largely depend on these strength effects, e.g., at higher concentrations of 𝑃𝑃𝑟𝑟, effects of increased strain strength were observed, relocating the shifts of ferromagnetic-paramagnetic transitions to lower temperatures. On the other hand, effects of increased compressive strength were observed at higher concentrations of 𝑁𝑁𝑁𝑁 and 𝐶𝐶𝐶𝐶, relocating the shifts of ferromagnetic-paramagnetic and metal-insulator transitions to higher temperatures.

Legionella-like isolates, strains W03-356T, W03-357 and W03-359, from three independent water samples from the river Elbe, Germany, were analysed by using a polyphasic approach. Morphological and biochemical characterization revealed that they were Gram-negative, aerobic, non-spore-forming bacilli with a cut glass colony appearance that grew only on l-cysteine-supplemented buffered charcoal yeast extract agar. Phylogenetic analysis based on sequence comparisons of the 16S rRNA, macrophage infectivity potentiator (mip), gyrase subunit A (gyrA), ribosomal polymerase B (rpoB) and RNase P (rnpB) genes confirmed that the three isolates were distinct from recognized species of the genus Legionella. Phenotypic characterization of strain W03-356T based on fatty acid profiles confirmed that it was closely related to Legionella rubrilucens ATCC 35304T and Legionella pneumophila ATCC 33152T, but distinct from other species of the genus Legionella. Serotyping of the isolates showed that they were distinct from all recognized species of the genus Legionella. Strains W03-356T, W03-357 and W03-359 are thus considered to represent a novel species of the genus Legionella, for which the name Legionella dresdenensis sp. nov. is proposed. The type strain is W03-356T (=DSM 19488T=NCTC 13409T).

Bacterial strain 5bvl1, isolated from a chromium-contaminated wastewater treatment plant and identified as Ochrobactrum tritici, was resistant to a broad range of antibiotics, to Cr(VI), Ni(II), Co(II), Cd(II), and Zn(II), and was able to grow in the presence of 5% NaCl and within the pH range 4–10. Characterization showed that strain 5bvl1 could be considered a halotolerant and alkalitolerant microorganism resistant to high concentrations of Cr(VI). This strain was able to grow aerobically in up to 10 mmol·L–1 Cr(VI). Cr(VI) resistance was independent of sulphate concentration. Under aerobic conditions strain 5bvl1 was also able to reduce high Cr(VI) concentrations (up to 1.7 mmol·L–1). Increasing concentrations of Cr(VI) in the medium lowered the growth rate of strain 5bv11 but the reduction in growth rate could not be directly correlated with the amount of Cr(VI) reduced. Unlike the type strain, which was only able to reduce Cr(VI), strain 5bvl1 was resistant to Cr(VI) and able to reduce it. Moreover, in strain 5bvl1, the rate and extent of Cr(VI)-reduction were higher than in the other strains of the genus Ochrobactrum. Ochrobactrum strain 5bvl1 resists high Cr(VI) concentrations and has a high Cr(VI)-reducing ability, making it a valuable tool in bioremediation.Key words: Ochrobactrum, Cr(VI) resistance, Cr(VI)-reduction, heavy metal, bioremediation.

Pear stem wart and pear stem canker, which have been considered as two different fungal diseases caused by pathogens belonging to Botryosphaeria spp., commonly occur and cause serious damage in the main pear-producing areas in China. To identify the species of this genus infecting pear in China, 131 Botryosphaeria isolates were recovered from pear samples exhibiting symptoms collected from 20 different provinces and areas. Morphological characterization and phylogenetic analyses of the ribosomal DNA internal transcribed spacer region and the β-tubulin and EF1-α genes revealed that Botryosphaeria dothidea, B. rhodina, B. obtusa, and B. parva were associated with different pear stem wart and stem canker symptoms. Remarkably, all isolates of B. dothidea were obtained from the samples showing either stem wart or stem canker lesions; however, the isolates of the other three species were obtained only from the samples showing stem canker. Pathogenicity tests on the pear shoots showed that B. dothidea isolates could induce stem wart or stem canker lesions but all the isolates of the other three species could only induce stem cankers. However, the isolates of B. parva, B. rhodina, and B. obtusa exhibited higher virulence than that of the B. dothidea isolates on the pear fruit. Our results suggest that B. dothidea is the common causal agent for these two diseases (a pear stem wart and a pear-related stem canker), whereas B. parva, B. rhodina, and B. obtusa only cause pear stem canker diseases. To our knowledge, this study represents the first report for biological and molecular characterization of four Botryosphaeria spp. isolated from pear plants showing stem wart and stem canker in China.

Biodiversitas 21: 5448-5454. Biofuel is an alternative to fossil fuels that are environmentally friendly with low emissions. Biofuel from biomass microalgae, especially Chlorella vulgaris, has an essential role in biofuel production. Increasing biomass microalgae was done by co-culture between microalgae and bacteria. This research aims to determine the potential of bacterial isolates to produce the IAA hormone and identify the highest isolate with the ability to synthesis IAA from four lakes in East Java. This research was conducted by culturing bacterial isolates in the Tryptic Soy Broth (TSB) to add tryptophan media in various periods of incubation. The absorbance was measured with UV-Vis spectrophotometry at a wavelength of 530 nm for determining IAA-production from bacterial isolates. The results showed that the "12" code bacterial isolate from Ranu Grati produced the highest IAA hormone concentration, with an average of 30.23 ppm. The morphological characterization of the highest IAA-producing bacteria showed that isolate included the Enterobacteriaceae group and phenotypic characterization include Enterobacter cloacae complex (ECC).
