The hypothesis that a cold plasma jet has the antimicrobial effect against Enterococcus faecalis biofilms was tested in vitro. 27 hydroxyapatite discs were incubated with E. faecalis for six days to form a monoculture biofilm on the disc surface. The prepared substrata were divided into three groups: the negative control, the positive control (5.25% NaOCl solution), and the plasma treatment group. Resultant colony-forming unit counts were associated with observations of bacterial cell morphology changes using scanning electron microscopy (SEM). Treatment of E. faecalis biofilm with the plasma and 5.25% NaOCl for 5 min resulted in 93.1% and 90.0% kill (P<0.0001), respectively. SEM detected that nearly no intact bacteria were discernible for the plasma-exposed HA disc surfaces. The demonstrated bactericidal effect of the plasma with direct surface contact may be due to the enhanced oxidation by the locally produced reactive plasma species.

AIM: To evaluate antimicrobial effect of diode laser when used adjunctively with sodium hypochlorite, methylene blue or toluidine blue in Enterococcus faecalis inoculated root canals. METHODOLOGY: 70 extracted uniradicular premolars were prepared with ProTaper files and sterilized. 5 were kept aside as negative control (G1) and the remaining 65 were inoculated with Enterococcus faecalis and incubated for 7 days. Following this, 5 were kept as positive control (G2) and the other 60 were randomly distributed into six groups: G3, diode laser irradiation (1.5W) with sodium hypochlorite; G4, diode laser (3W) with sodium hypochlorite; G5, diode laser (1.5W) with methylene blue; G6, diode laser (3W) with methylene blue; G7, diode laser (1.5W) with toluidine blue; G8, diode laser (3W) with toluidine blue. Subsequently, turbidity was assessed and CFU count determined following intracanal sampling and plating. RESULTS: G4 exhibited significantly lower mean CFUs/mL (P˂0.001) among the experimental groups. G4 demonstrated the most (98.8%) while G7 exhibited the least antibacterial effect (28.66%). Highest turbidity was observed in G5 (0.1310) while the lowest was noted in G4 (0.0287). CONCLUSION: Diode laser used adjunctively with sodium hypochlorite exhibited superior antibacterial efficacy when compared to its use with photosensitizer dyes like methylene blue and toluidine blue.

Aim: To comparatively evaluate the antimicrobial effect of turmeric and ginger extracts on Streptococcus mutans in in-vitro conditions. Material & Method: An in-vitro experimental study was conducted in a laboratory setting. Ethanolic extract of Ginger and Turmeric was prepared separately by cold masseration technique. The extract of each was then diluted with an inert solvent, Dimethyl Formamide, to obtain 5 different concentrations (2%, 4%, 6%, 8%, and 10%) of each. 0.2% chlorhexidine was used as a positive control and dimethyl formamide was used as negative control. The different extracts, along with controls, were then subjected to microbiological investigation to determine which gave a wider zone of inhibition against streptococcus mutans. The zone of inhibition was measured in millimeters. Results: Turmeric extracts presented the largest zone of inhibition 33mm at the concentration of 8%, while Ginger extract showed a zone of inhibition of 34mm at the concentration of 10%. Conclusion: Ethanolic extract of Turmeric demonstrated antimicrobial activity against Strptococcus Mutans at a lower concentration than that of Ginger. Purpose: To investigate the anti-tuberculosis potential of twelve commercially available pyridone analogues against Mycobacterium tuberculosis H37Rv strain.Methods: Twelve commercially available pyridone-based compounds were screened against M. tuberculosis H37Rv using different susceptibility tests. The most active or lead compound was further evaluated in detail for its anti-tuberculosis (anti-TB) potential. Kill kinetics was used to determine the dynamics of its anti-TB activity in vitro.Results: Compounds d, j and k were potent against M. tuberculosis H37Rv, with minimum inhibitory concentrations (MICs) of 10, 5 and 10 μg/mL, respectively. The standard anti-TB drugs used in this study (positive control drugs) demonstrated MIC of 2.5 μg/mL. The anti-TB effect of compound j was comparable with those of the standard drugs (RIF, LVX, AMK, EMB and INH). The minimum bactericidal concentration (MBC) of compound j was 10 μg/mL. It produced an MIC of 5 μg/mL in agar proportion method (APM). However, its MIC in Middlebrook 7H9 broth supplemented with 10 % fetal bovine serum (FBS) and 4 % bovine serum albumin (BSA) increased 4- and 8-fold, respectively. The bactericidal effect of compound j was time- and concentration-dependent at dilutions above 2x MIC. Combination of compound j with RIF, LVX or AMK exhibited fractional inhibitory concentration index (ΣFIC) of 1, indicative of additive drug-drug interactions. However, combination with INH or EMB produced a ΣFIC of 2. None of the tested drug combinations was antagonistic.Conclusion: Compound j exhibits potent time- and concentration-dependent antimicrobial effect against M. tuberculosis H37Rv. Thus, it may be suitable as an adjunct to current treatment of drug sensitive and drug-resistant TB. The purpose of thisin vitrostudy was to assess the effect of hydrogen peroxide on the antibacterial substantivity of chlorhexidine (CHX). Seventy-five dentine tubes prepared from human maxillary central and lateral incisor teeth were used. After contamination withEnterococcus faecalisfor 14 days, the specimens were divided into five groups as follows: CHX, H2O2, CHX + H2O2, infected dentine tubes (positive control), and sterile dentine tubes (negative control). Dentine chips were collected with round burs into tryptic soy broth, and after culturing, the number of colony-forming units (CFU) was counted. The number of CFU was minimum in the first cultures in all experimental groups, and the results obtained were significantly different from each other at any time period (). At the first culture, the number of CFU in the CHX + H2O2group was lower than other two groups. At the other experimental periods, the CHX group showed the most effective antibacterial action (). Hydrogen peroxide group showed the worst result at all periods. In each group, the number of CFU increased significantly by time lapse (). In conclusion, H2O2had no additive effect on the residual antibacterial activity of CHX.

Recurrent endodontic infections of treated teeth are mainly linked to microbial persistency in the root canal system. Enterococcus faecalis is a bacterium that is often identified in these recurrent infections. Calcium hydroxide (Ca(OH)2) is a widely used root canal medicament and is now being developed to be produced from natural materials. Ca(OH)2, which is synthesized from natural limestone from Palimanan, Indonesia, has a structure similar to Ca(OH)2, which is often used as a root canal medicament. The purpose of this study was to determine the antimicrobial property of Ca(OH)2 paste synthesized from natural limestone compared to conventional Ca(OH)2 paste. This study is an in vitro experimental study consists of 3 groups (n=24, namely, conventional Ca(OH)2 paste, limestone synthesized Ca(OH)2 paste, and saline as the negative control, for 2 intervals of time and evaluated by Total Plate Counting (TPC) method. Samples were mandibular first premolar root canal prepared by root canal preparation and contaminated with E. faecalis (ATCC 29212). The infected root canals of each group were treated by the medicaments, and samples from the root canals were taken on day 7 (n=4) and 14 (n=4). Results showed that both Ca(OH)2 groups had antimicrobial activities against E. faecalis. The limestone Ca(OH)2 paste group had the least amount of bacterial colonies on the 7th day compared to other groups. In contrast, on the 14th day, the conventional Ca(OH)2 paste had the least bacterial colonies. It can be concluded that the Ca(OH)2 paste synthesized from natural limestone has an antimicrobial effect on E. faecalis, the antimicrobial effect was higher on the 7th day but decreases on the 14th day, whereas the conventional Ca(OH)2 had a higher antimicrobial effect on day 14 compared to day 7. Further studies are expected to improve the long-term antimicrobial effectiveness and sustainability of the natural limestone synthesized Ca(OH)2 paste.

Background: Enterococcus faecalis (E. faecalis) is a bacterium commonly detected in the root canals of teeth with post-treatment apical periodontitis or advanced marginal periodontitis. It has the ability to live in an extreme environment and survive as an organism with its virulence factor possibly contributing to the pathogenesis of post-treatment apical and marginal periodontitis. Photodynamic therapy (PDT) is an urgently required alternative method of improving therapy effectiveness. Photodynamic therapy combined with conventional endodontic treatment decreases the number of antibioticresistant bacteria and biofilms. Chlorophyll is one of the photosensitizers added to enhance the absorption of light in photodynamic therapy. Purpose: The purpose of this study was to determine the antimicrobial effect of the combination of photodynamic laser therapy and Alfalfa chlorophyll in E. faecalis. Methods: In vitro study using E. faecalis distributed between negative control (C-) and positive control (C+), treatment groups using various energy doses of a 405 nm diode laser (2.5, 5, 7.5, 10, 12.5, 15, 17.5, 20 J/cm2) with (G1) and without alfalfa chlorophyll as organic photosensitizer (G2). The suspension was inoculated on Tryptocase Soy Agar (TSA) and incubated at 37° C for 24 hours. The number of colonyforming units per milliliter (CFU/ml) was determined. The results were analyzed by ANOVA with p value ≤0.05. Results: A 405 nm irradiating laser with or without a photosensitizer can decrease E. faecalis viability percentage through the administering of various energy doses. The highest decrease (42%) was obtained in the group without a photosensitizer using 20 J/cm2, while 10 J/cm2 in the group with a photosensitizer proved the most effective dose (25%). Conclusion: The results of this study showed a decrease in the viability of E. faecalis exposed to a 405 nm (40 mW) laser. An irradiating process using a 405 nm laser without a photosensitizer (Alfalfa chlorophyll) resulted in the highest percentage decrease (42%) in E. faecalis bacterial viability.

Shallot (Allium ascalonicum L.) has been known as traditional medicine. Shallot which has same genus with garlic(Allium sativum L.) contains allicin that is also found in garlic and has been suspected has fungicidal activity toCandida albicans. It is supported by several researches. Therefore, shallot is suspected has antifungal activity too.The aim of this research was to know antifungal activity of shallot’s water extortion againsts Candida albicans invitro. This was a laboratory experimental research which used completely randomized design, with diffusion method.Shallot’s water extortion was devided into three concentrations, there were 50%, 100% and 200%. Ketoconazole 2%was positive control and aquadest was negative control. The result of this research based on analysis of varians(Anova), there was significant difference between several treatments and was confirmed with Duncan New MultipleRange Test (DNMRT) p<0,05, there was significant difference between 100% shallot’s water extortion with othertreatments, but there was no significant difference between 50% shallot’s water extortion with 200% shallot’s. Theconclusion was shallot’s water extortion had antifungal activity againsts Candida albicans with the best concentration100%, but it was lower than ketoconazole 2%.

Abstract Background The banana or scientifically referred to as Musa sp., is one of the most popular fruits all over the world. Almost all parts of a banana tree, including the fruits, stem juice, and flowers are commonly used as traditional medicine for treating diarrhoea (unripe), menorrhagia, diabetes, dysentery, and antiulcerogenic, hypoglycemic, antilithic, hypolipidemic conditions, plus antioxidant actions, inflammation, pains and even snakebites. The study carried out was to evaluate in vitro anti-urolithiatic activity from different types of Musa pseudo-stems. Methods Observing anti-urolithiathic activity via in vitro nucleation and aggregation assay using a spectrophotometer followed by microscopic observation. A total of 12 methanolic extracts were tested to determine the potential extracts in anti-urolithiasis activities. Cystone was used as a positive control. Results The results manifested an inhibition of nucleation activity (0.11 ± 2.32% to 55.39 ± 1.01%) and an aggregation activity (4.34 ± 0.68% to 58.78 ± 1.81%) at 360 min of incubation time. The highest inhibition percentage in nucleation assay was obtained by the Musa acuminate x balbiciana Colla cv “Awak Legor” methanolic pseudo-stem extract (2D) which was 55.39 ± 1.01%at 60 min of incubation time compared to the cystone at 30.87 ± 0.74%. On the other hand,the Musa acuminate x balbiciana Colla cv “Awak Legor” methanolic bagasse extract (3D) had the highest inhibition percentage in the aggregation assay incubated at 360 min which was obtained at 58.78 ± 1.8%; 5.53% higher than the cystone (53.25%).The microscopic image showed a great reduction in the calcium oxalate (CaOx) crystals formation and the size of crystals in 2D and 3D extracts, respectively, as compared to negative control. Conclusions The results obtained from this study suggest that the extracts are potential sources of alternative medicine for kidney stones disease.

OBJECTIVE: To evaluate the in vitro ionic degradation and slot base corrosion of metallic brackets subjected to brushing with dentifrices, through analysis of chemical composition by Energy Dispersive Spectroscopy (EDS) and qualitative analysis by Scanning Electron Microscopy (SEM). METHODS: Thirty eight brackets were selected and randomly divided into four experimental groups (n = 7). Two groups (n = 5) worked as positive and negative controls. Simulated orthodontic braces were assembled using 0.019 x 0.025-in stainless steel wires and elastomeric rings. The groups were divided according to surface treatment: G1 (Máxima Proteção Anticáries®); G2 (Total 12®); G3 (Sensitive®); G4 (Branqueador®); Positive control (artificial saliva) and Negative control (no treatment). Twenty eight brushing cycles were performed and evaluations were made before (T0) and after (T1) experiment. RESULTS: The Wilcoxon test showed no difference in ionic concentrations of titanium (Ti), chromium (Cr), iron (Fe) and nickel (Ni) between groups. G2 presented significant reduction (p < 0.05) in the concentration of aluminium ion (Al). Groups G3 and G4 presented significant increase (p < 0.05) in the concentration of aluminium ion. The SEM analysis showed increased characteristics indicative of corrosion on groups G2, G3 and G4. CONCLUSION: The EDS analysis revealed that control groups and G1 did not suffer alterations on the chemical composition. G2 presented degradation in the amount of Al ion. G3 and G4 suffered increase in the concentration of Al. The immersion in artificial saliva and the dentifrice Máxima Proteção Anticáries® did not alter the surface polishing. The dentifrices Total 12®, Sensitive® and Branqueador® altered the surface polishing.

The sugarcane aphid, Melanaphis sacchari Zehntner (Hemiptera: Aphididae), is the main pest of sorghum, Sorghum bicolor L. Moench (Poaceae), in Mexico. To control this insect, farmers currently use synthetic chemical insecticides, which are toxic to humans and biodiversity. However, natural products are a promising potential source of safer alternative means to control different agricultural pests. The main objective of this study was to evaluate the insecticidal effect of contact by fumigation of pure molecules of four commercial fatty acids (palmitic, stearic, pentadecanoic and linoleic acids), the phytosterol ß -sitosterol, and the flavonoid rutin. The results showed that fatty acids were the most effective against M. sacchari ; the highest mortality rate (85%) was produced by linoleic acid and the LC 50 was 1,181 ppm, followed by stearic and palmitic acids with mortality percentages of 74 and 63%, respectively, at a concentration of 2,500 ppm at 72 h. The positive control, imidacloprid, had 100% mortality in 24 h and the tween 20 negative control exhibited 4% mortality in 72 h. Our results show that commercial fatty acids are effective against adults of M. sacchari , and can be considered an environmentally friendly alternative to the frequent use of synthetic chemical insecticides.
