Background: LncRNAs have been shown to be implicated in the initiation and progression of gastric cancer (GC). Here, the transcription levels of the AK023391 and FOXO3a genes, as well as their relationship with clinical traits, were examined in GC. Methods: This cross-sectional study was performed on 50 formalin-fixed paraffin-embedded (FFPE) GC tumor and 50 normal tissues. The RNA levels of the FOXO3a and AK023391 genes encoding lncRNAs were evaluated by real-time RT-PCR. Results: A significant difference was observed in the mRNA level of AK023391 between GC tumor and normal tissues (Mean difference = 2.683, P = 0.0371). The transcription level of FOXO3A showed no remarkably significant difference between GC and normal tissues (Mean difference = 0.9177, P = 0.2199). In addition, AK023391 and FOXO3a mRNA expression levels showed no significant associations with the clinicopathologic features of GC. Also, we found no significant correlation between the expressions of AK023391 and FOXO3a in GC tissues (R = 0.014; P = 0.34). Conclusions: Our finding suggested that there might be a significant relationship between the expression level of AK023391 and GC. Our results also showed that there was no correlation between the mRNA levels of the AK023391 and FOXO3a genes in GC tissues.

Abstract Background People are at a high risk of gastric cancer if their first-degree relatives suffered from atrophic gastritis (AG), intestinal metaplasia (IM), intraepithelial neoplasia (IEN), dysplasia (DYS), or gastric cancer (GC). This study was performed to analyse the association between FDR-GC and GC precursors. Methods A cross-sectional study was performed to screen the prevalence of GC precursors from November 2016 to September 2019. A total of 1329 participants with FDR-GC, 193 participants with a family history of non-gastric cancer in FDRs (FDR-nGC), and 860 participants without a family history of cancer in FDRs (FDR-nC) were recruited in this study. The logistic regression model was used in this study. Results The prevalence of normal, Non-AG, AG/IM, IEN/DYS, and GC was 31.91, 44.21, 13.81, 8.73, and 1.34%, respectively. The prevalence of IEN/DYS was higher in people with FDR-GC and FDR-nGC (FDR-GC: odds ratio (OR) = 1.655; 95%CI, 1.153–2.376; FDR-nGC: OR = 1.984; 95%CI, 1.122–3.506) than those with FDR-nC. The younger the age at which FDRs were diagnosed with GC, the more likely the participants were to develop AG/IM (Ptrend = 0.019). The risk of precursors to GC was higher in participants whose FDR-GC was the mother than in those whose FDR-GC was the father or sibling (OR, non-AG: 1.312 vs. 1.007, 1.274; AG/IM: 1.430 vs. 1.296, 1.378; IEN/DYS: 1.988 vs. 1.573, 1.542). There was no statistically significant difference in non-AG (OR = 1.700; 95%CI, 0.940–3.074), AG/IM (OR = 1.291; 95%CI, 0.579–2.877), and IEN/DYS (OR = 1.265; 95%CI, 0.517–3.096) between participants with one or more FDR-GC. Conclusion People with FDR-GC and FDR-nGC are at a high risk of IEN/DYS. When an FDR was diagnosed at a younger age, the risk of AG/IM was higher. The risk of GC precursors was higher in people whose FDR-GC was the mother.

Purpose.The purpose of this study was to investigate the correlation of kinematic variables with quality of trunk control in poststroke patients.Methods.This cross-sectional study included stroke subjects with mild to moderate motor deficit corresponding to Brunnstrom stages 3-4. Trunk functional performance was measured using bed mobility monitor system. All tasks were repeated ten times for both directions in each subject. Outcome measurements included the movement time and displacement of center of pressure (CoP) from supine to side lying and returning.Results.The results revealed that a significant longer turning time was observed when turning from the paretic side toward the nonparetic side compared to the other direction, with an estimated mean difference of 0.427 sec (P=0.005). We found a significant difference in the time of rolling back to supine position between two directions. The displacement of CoP in rolling back from side lying on the nonparetic side was smaller than that from the paretic side with an estimated mean difference of −0.797 cm (P=0.023).Conclusions.The impaired trunk mobility was associated with increased movement time and decreased displacement of CoP in poststroke patients. Trunk rolling performance has potential in assessment of stroke patients.

Abstract Gastric cancer is found at a rate of 2.4 to 3.5 percent in Indonesia, with the majority of cases discovered at an advanced stage. Cyclin D1 is a protein that promotes cancer cell growth. It has been shown to be expressed in a variety of cancers, including stomach cancer. A recent study of cyclin D1 in gastric cancer has been associated with lymph node involvement, metastasis, poor prognosis, and lack of response to platinum chemotherapy. This study aims to determine the relationship between cyclin D1 expression and clinicopathological features in gastric cancer. This cross-sectional study used medical records and paraffin blocks of gastric cancer patients at Cipto Mangunkusumo General Hospital, Jakarta, in 2015–2020. Demographic data, clinical characteristics, radiological findings, histopathological features, and cyclin D1 expression were collected and examined. Data was collected from 39 subjects. Most of the subjects experienced eating disorders (69.23%), weight loss (76.92%), melena (53.85%), and anemia (51.28%). Tumor locations were mostly found in the cardia and corpus of the gaster. Overexpression of cyclin D1 was found in 30.77% of cases. Cyclin D1 expression was greater in subjects with liver metastases (50% vs. 14.8%, p 0.04). Cyclin D1 expression was not associated with tumor location, TNM stage, and histopathological findings.

Abstract Background The aim of this study was to investigate the DNA methylation pattern in CpG islands of NIS and TIMP-3 promoters in thyroid cancers and the matched non-tumoral tissues as well as multinodular goiter (MNG) tissues. This cross-sectional study was performed on total 64 patients including 28 papillary thyroid cancer (PTC), 9 follicular thyroid cancer (FTC) and 27 MNG cases. Method: The bisulfite sequencing PCR technique was used to evaluate the promoter methylation pattern of the NIS and TIMP-3 genes. Results NIS mRNA levels were decreased in both of PTC and FTC tumoral tissues compared to their adjacent normal tissues (P = 0.04 and P = 0.03, respectively). TIMP-3 expression was also reduced in both PTC and FTC tumoral tissues compared to the adjacent non-tumoral tissues (P = 0.02 and P = 0.03, respectively). Moreover, a significant reduction in TIMP-3 expression was observed in FTC tissues compared to the MNG samples (P = 0.05). The methylation on NIS promoter was not a common event in PTC samples, but it was frequent in FTC (P < 0.05). A significant hyper-methylation was shown in TIMP-3 promoter in both PTC and FTC tissues compared to the non-tumoral and MNG samples (P < 0.05). Conclusion Aberrant promoter DNA methylation suggests potential utility to differentiate benign and malignant PTC and FTC tissues for early diagnosis, and contribution for personalized clinical management and surveillance. According to the reversibility of DNA methylation, these events may be potential targets for demethylating treatments.

Background: Some recent reports have noted that copper deficiency can occur in obese patients who have undergone bariatric surgery, such as Roux-en-Y (RY) gastric bypass or biliopancreatic diversion, or in patients who receive enteral nutrition through a jejunostomy. No reports appear to have assessed the serum copper state of patients following gastrectomy with RY reconstruction for gastric cancer. Methods: A cross-sectional study was conducted from June 2013 to December 2014. Serum copper levels (SCLs) in 242 out-clinic patients who underwent curative gastrectomy were obtained. Patients were classified into an RY group (n = 208) and a non-RY group (n = 34). Results: Hypocupremia was identified in 3 patients in the RY group (1.4%), and 2 patients in the non-RY group (5.9%; p = 0.146), but none experienced any symptoms caused by hypocupremia. No significant difference in the mean SCL was seen between the RY group (105.8 ± 21.2 µg/dl) and non-RY group (107.9 ± 22.7 µg/dl; p = 0.499). In the RY group, the mean SCL was significantly lower in younger patients, patients with follow-up period <3 years, and male patients. Conclusion: Some patients developed hypocupremia after gastrectomy with RY reconstruction, but the number is acceptably low, and physical symptoms were unusual.

26 Background: Risaralda (Colombia) has an irregular pattern in the risk of death for gastric cancer; for these reasons, the survival analysis was made with the information of gastric cancer patients treated in Oncólogos del Occidente clinic during 2014. Methods: A cross-sectional study was done with oncologic patients in order to evaluate the replicability of a binary list of signs and symptoms of gastric cancer, also its corresponding principal component factors for the proper identification of their dimensions. A sum of the set of signs and symptoms was done. This total sum of signs and symptoms was used to identify the sensitivity and specificity of each of cut-off related to the patient’s death condition. This evaluation was established by the ROC (Receiver Operating Characteristic) methodology, which is a graphic representation of the sensitivity in front of the specificity for a classifying binary system that may vary the threshold of rejection. The analysis was done in Stata. Results: There were 173 patients, of whom 69.9% were males; the group with the highest incidence was the 60-69 years old group (32%); These patients referred 5 signs and symptoms that conformed the list from which every single item behaved as a binary variable. Cronbach’s alpha was 0.62 and the analysis of principal components factors defined only one dimension. The ROC curve established a sensitivity of 65% and a specificity of 72% for the determination of the risk of death with a score equal or higher to 4 in the sum of the list of signs and symptoms. Adding the gender, the sensitivity increased to 66%. In the ≥ 80 years old group the sensitivity was 70.9%, meanwhile the group of ≥ 40 years old the sensitivity was 93%. Conclusions: The list of signs and symptoms could be a diagnostic technology relevant for unspecific clinical but has an important high frequency in Colombia for gastric cancer patients. The sensitivity was very variable according to the age range, being more sensible and specific for the patients of the ≥ 40 years old group.

AbstractGastric cancer has high malignancy and early metastasis, which lead to poor survival rate. In this study, we assessed the expressions and prognostic values of MS4A family, a newly recently discovered family, by two online dataset, GEPIA and Kaplan Meier-plotter. From these results eight members, MS4A2, MS4A6, MS4A7, MS4A8, MS4A14, MS4A15, TMEM176A and TMEM176B showed positive expression in gastric cancer or normal tissues, and these genes were screened for further analysis of prognostic values. We observed that low mRNA expressions of MS4A2, MS4A7, MS4A14, MS4A15, TMEM176A and TMEM176B were correlated with better overall survival (OS) in all gastric cancer patients, while high mRNA expression of MS4A6 was observed to be associated with good prognosis. MS4A8’s high mRNA level was correlated to better OS in diffuse gastric cancer patients. Further, we estimated prognostic values of MS4A family in gastric cancer patients with different clinic-pathological features, including clinical stages, differentiation level, lymph node status and HER2 status. Our results indicate that these eight MS4A members can estimate prognosis in patients with different pathological groups. In conclusion, MS4A family members are potential biomarkers, and may contribute to tumor progression in gastric cancer.

Abstract Our study aimed to investigate the associations between DEHP exposure and serum thyroid hormone levels in 347 adolescents and young adults. We measured DEHP metabolites including mono(2-ethylhexyl) phthalate (MEHP), mono(2-ethyl-5-oxohexyl) phthalate (MEOHP), mono(2-ethyl-5-hydroxyhexyl) phthalate (MEHHP), mono(2-ethyl-5-carboxypentyl) phthalate (MECPP) and mono(2-carboxmethyl)hexyl phthalate (MCMHP) in their urine. Total thyroxine (TT4), total triiodothyronine, free triiodothyronine, free thyroxine (FT4), thyroid-stimulating hormone and the mRNA levels of thyroid peroxidase (TPO), thyroglobulin (TG), sodium iodide symporter (NIS), thyroid transcription factor 1 (TTF-1) and paired box gene 8 (PAX-8) in serum were measured. The results of statistical analysis showed that urinary DEHP metabolites were generally negatively associated with TT4 levels in serum. In the males, the FT4 levels showed positive associations with urinary MEHP, MECPP, MCMHP, and ∑DEHP. The mRNA level of TG was significantly positively correlated with the levels of MECPP, MCMHP and ∑DEHP, while the level of NIS mRNA was significantly positively correlated with the levels of MEOHP. There was a significant positive correlation between TTF-1 mRNA level and urinary MEOHP, MECPP, MCMHP and ∑DEHP.

Purpose The aims of this study were to characterize hearing symptoms, habits, and sound pressure levels (SPLs) of personal audio system (PAS) used by young adults; estimate the risk of developing hearing loss and assess whether instructions given to users led to behavioral changes; and propose recommendations for PAS users. Method A cross-sectional study was performed in 50 subjects with normal hearing. Procedures included questionnaire and measurement of PAS SPLs (real ear and manikin) through the users' own headphones and devices while they listened to four songs. After 1 year, 30 subjects answered questions about their usage habits. For the statistical analysis, one-way analysis of variance, Tukey's post hoc test, Lin and Spearman coefficients, the chi-square test, and logistic regression were used. Results Most subjects listened to music every day, usually in noisy environments. Sixty percent of the subjects reported hearing symptoms after using a PAS. Substantial variability in the equivalent music listening level (Leq) was noted ( M = 84.7 dBA; min = 65.1 dBA, max = 97.5 dBA). A significant difference was found only in the 4-kHz band when comparing the real-ear and manikin techniques. Based on the Leq, 38% of the individuals exceeded the maximum daily time allowance. Comparison of the subjects according to the maximum allowed daily exposure time revealed a higher number of hearing complaints from people with greater exposure. After 1 year, 43% of the subjects reduced their usage time, and 70% reduced the volume. A volume not exceeding 80% was recommended, and at this volume, the maximum usage time should be 160 min. Conclusions The habit of listening to music at high intensities on a daily basis seems to cause hearing symptoms, even in individuals with normal hearing. The real-ear and manikin techniques produced similar results. Providing instructions on this topic combined with measuring PAS SPLs may be an appropriate strategy for raising the awareness of people who are at risk.
