Evaluation of in vitro Antioxidant Activities of Ethanol Extracts of Datura innoxia Mill. Leaves and Seeds Harvested in Mali

Aims: The purpose of this study was to evaluate the in vitro antioxidant activities of Datura innoxia Mill. leaves and seeds harvested in Mali. Place and Duration of Study: Collection of plant materials were done at Kolondieba (Mali) in june 2016. Evaluation of polyphenols and flavonoids contents, determination of antioxidant activities were done at Laboratory of Plant and Food Biochemistry and Biotechnology of University of Sciences, Techniques and Technologies of Bamako (Mali) between august 2016 and March 2017. Methodology: The leaves and seeds were collected in Kolondieba, Mali. The phytochemical screening based on the standard methods of tube reactions has been performed with ethanol extracts. The quantitative estimation of total polyphenols was made by the Folin-Ciocalteu method and that of flavonoids by the use of aluminum trichloride. The in vitro antioxidant activities of the ethanol extracts was determined by three methods: the 1,1- diphenyl - 2- picrylhydrazyl (DPPH) test, the ferric reducing ability power (FRAP) test and the Phosphomolybdate test (PPM). Results: The phytochemical screening revealed that leaves and seeds of this plant contains alkaloids, polyphenols, flavonoids, coumarins, tannins, triterpenes and saponin. Phenolic contents of ethanol extracts are 30.97 ± 0.33 mg equivalent gallic acid / g in leaves and 14.02 ± 0.15 mg equivalent gallic acid / g in seeds; those of flavonoids are 15.13 ± 0.2 mg equivalent of quercetin / g in the leaves and 4.93 ± 0.41 mg equivalent of quercetin / g in the seeds. The three tests showed that the leaves have a higher level of antiradical activity in vitro than seeds. Conclusion: The results of this work showed that Datura innoxia Mill. has a good antioxidant activity which would justify its use as a potential source of natural antioxidants.

Objective: The aim of this research program is to evaluate the antioxidant potentialities comprehensively utilizing commonly available cruciferous vegetables of India, viz., cabbage, cauliflower, kohlrabi, and radish.Methods: The plant materials were procured and processed for cold extraction procedure using 70% ethanol. The extracts were primarily assessedfor their phytoconstituents and further for their in vitro antioxidant activity using various qualitative and quantitative estimations. The results of quantitative estimations were expressed in terms of equivalence with respective standards, and the antioxidant potentiality of various scavenging and chelating activities were expressed in terms of 50% effective concentration. Further, correlation studies were made between quantitative and qualitative assays to study the relationship between the effects of different phytoconstituents groups.Results: In this study, the results revealed that all the four cruciferous vegetable extracts possess potential antioxidant activities. Among the extractsunder study kohlrabi has recorded superior antioxidant potential than others. It is clear from the study that the tested cruciferous vegetablesmanifested differential expression of antioxidant capacity due to their phytoconstituents.Conclusion: From the results, it can be concluded that the Cruciferae vegetables have potent antioxidant activity contributing to the use for healthbenefits in addition to their nutritive role as vegetable.Keywords: Cruciferae vegetables, Phytochemicals, In vitro antioxidant activity, Correlation.

Objective: To assess the antioxidant activity from another part of the plant, in this study, leaf extracts in n-hexane were fractionated.Methods: Ten fractions were obtained and tested in vitro for antioxidant activity using two methods, 1,1-diphenyl-2-picrylhydrazyl (DPPH) and ferricreducing antioxidant power (FRAP), to identify the most active fraction.Results: The IC50 of the most active fraction was 36.24 μg/mL using the DPPH method, and the EC50 was 39.54 μg/mL using the FRAP method. Themost active fraction was also shown to contain terpenoids.Conclusion: The most active fraction of an n-hexane extract of the leaves of Gacinia bancana Miq., which was tested by both DPPH and FRAP methodshad antioxidant activities with IC50 and EC50 values of 36.2482 μg/mL and 39.5442 μg/mL, respectively. Phytochemical screening showed that activefraction contains terpenoids.

Honey is a well-known natural sweetener and is rich in natural antioxidants that prevent the occurrence of oxidative stress, which is responsible for many human diseases. Some of the biochemical compounds in honey that contribute to this property are vitamins and phenolic compounds such as phenolic acids and flavonoids. However, the extent to which these molecules contribute towards the antioxidant capacity in vitro is inconsistently reported, especially with the different analytical methods used, as well as other extrinsic factors that influence these molecules’ availability. Therefore, by reviewing recently published works correlating the vitamin, total phenolic, and flavonoid content in honey with its antioxidant activities in vitro, this paper will establish a relationship between these parameters. Based on the literature, vitamins do not contribute to honey’s antioxidant capacity; however, the content of phenolic acids and flavonoids has an impact on honey’s antioxidant activity.

Free radicals, reactive nitrogen species (RNS) and reactive oxygen species (ROS) have been known to contribute several degenerative diseases such as cardiovascular diseases, aging, certain types of cancers, rheumatoid arthritis, neurodegenerative, and diabetes mellitus. In order to overcome the negative effects of these radicals, some scientists have explored some natural antioxidants from plants and it's by-products. The antioxidant can be defined as any substances or samples capable of inhibiting free radical reactions in the oxidation reaction. Due to curcuminoids contained, Curcuma species such as Curcuma longa, Curcuma heyneana, Curcuma mangga, and Curcuma xanthorriza were commonly used for herbal components in some traditional medicine. Several in vitro tests been introduced and used to measure antioxidant activities, namely radical scavenging assay using 2,2’-diphenyl-1-picrylhydrazyl (DPPH) and 2,2′-azinobis-(3-ethylbenzothiazoline-6 -sulfonic acid) (ABTS•+), ferric reducing antioxidant power (FRAP), ferric-thiocyanate, phosphomolybdenum method, cupric ion reducing antioxidant capacity, metal chelating power, beta-carotene bleaching linoleic-ferric-thiocyanate, and thiobarbituric acid methods. This review highlighted the antioxidant activities in vitro of C. longa, C. heyneana, C. mangga, and C. xanthorriza through several tests. To perform this review, several repute databases were analyzed and used. From this review, it can be stated that Curcuma species have powerful antioxidant activities, therefore they could be potential sources of natural antioxidants and can be used as food supplements.

Tinospora cordifolia is known as Giloe and Guduchi, with significant importance in the traditional medicinal systems. It is dioeciously plant. It is mostly used in Ayurved system. It is also known as a ‘Rasayans’ of medicinal system, which develops immune system of the body and protect against infection. The aim of this study is carried out to analyse the phytochemical compounds in leaves and stem extracts of T. cordifolia by using phytochemical screening tests and estimate total flavonoid content (TFC) by using aluminium chloride method in the sample extracts. The leaf and stem extracts of T. cordifolia expressed the presence of several phytochemicals viz., flavonoids, amino acids, diterpines, protein, saponins and carbohydrates. The result of phytochemical screening tests revealed that diterpines and carbohydrates are positive in all extracts of T. cordifolia, but flavonoids and saponins only present in  methanol and ethanol extracts. TFC of T. cordifolia was higher in ethanolic leaves extracts than mathanolic leaves extracts. The studies justify that T. cordifolia use in traditional medicines. The investigation further proposed that the phytochemicals present in stems and leaves of T. cordifolia, which can be use as natural antioxidants in medicinal drugs. Olive pomace (OP) is a valuable food byproduct that contains natural phenolic compounds with health benefits related to their antioxidant activities. Few investigations have been conducted on OP from the United States while many studies on European OP have been reported. OP of Arbequina, the most common cultivar from California, was collected and extracted by water, 70% methanol and 70% ethanol, followed by purification using macroporous absorbing resin. Results showed that the extractable total phenolic content (TPC) was 36–43 mg gallic acid equivalents (GAE)/g in pitted, drum-dried defatted olive pomace (DOP), with major contributions from hydroxytyrosol, oleuropein, rutin, verbascoside, 4-hydroxyphenyl acetic acid, hydroxytyrosol-glucoside and tyrosol-glucoside. Macroporous resin purification increased TPC by 4.6 times the ethanol crude extracts of DOP, while removing 37.33% total sugar. The antioxidant activities increased 3.7 times Trolox equivalents (TrE) by DPPH and 4.7 times TrE by ferric reducing antioxidant power (FRAP) in the resin purified extracts compared to the ethanol crude extracts. This study provided a new understanding of the extraction of the bioactive compounds from OP which could lead to practical applications as natural antioxidants, preservatives and antimicrobials in clean-label foods in the US.

Agrimonia pilosa Ledeb (AL) has received considerable attention as a herbal medicine for its applications in ethnopharmacology with heath benefits. This study aimed to investigate antioxidant activities and A549 growth inhibitory capacity from its root extract (RE) and aerial parts extract (AE). The 50% ethanol extracts were used for the tests. The total polyphenol content and the antioxidant effects comprising ABTS+ and DPPH free radical scavenging activities were evaluated. Phenolic compounds in the extracts were isolated using high performance liquid chromatography (HPLC). Lactate dehydrogenase released in medium was also evaluated. Total phenolic and flavonoid content, and in vitro antioxidant potential of the RE were higher (p<0.01) than that of the AE. Two individual phenolic compounds consisting of 4-hydroxybenzoic acid and p-coumaric acid were firstly identified in both by HPLC. The RE exhibited higher A549 inhibitory capacity compared to the AE and activated the apoptotic proteins of bcl-2, bax, bad, caspase-3 and caspase-9 in A549. In conclusion, the AL extracts were more effective in antioxidant and A549 cells inhibitory capacity.

The objective of the current study was to evaluate the antioxidant activity and enzyme inhibitory effect of different parts of medlar including fruit, leaf and flower bud by using various in vitro methods, and also determination of total phenolic and flavonoid content in the samples. Ethanol extracts of medlar parts were prepared and their antioxidant activities were determined using 1,1-diphenyl-2-picryl-hydrazil (DPPH•) scavenging and β-carotene bleaching methods. The leaf extract showed the strongest antioxidant activity. DPPHradical scavenging activity was in the order of BHA > leaf > bud > fruit. This ordering was the same for β-carotene bleaching activity, tocopherol > leaf > bud > fruit. The highest total phenolic (60.3 ± 1.69 mg GAE g-1 extract) and flavonoid (14.77 ± 1.15 mg QE g-1 extract) content were determined in leaf extract. For possible antidiabetic effects of extracts, α-amylase and α-glucosidase inhibitory activities were investigated, the bud extract showed the highest inhibition activities among the all extracts.

Crossostephium chinensis (L.) (CC) Makino is a common traditional Chinese medicinal plant used to dehumidify and cure rheumatism and arthralgia. The water and methanol extracts of C. chinensis (CCW and CCM) were evaluated for their antioxidant and antiproliferative activities. The antioxidant activities of CC were evaluated by using ABTS radical scavenging, DPPH radical scavenging, nitric oxide scavenging and superoxide scavenging methods. Iron chelating activity, lipid peroxidation, total polyphenol contents, total flavonoid contents and total flavonol contents were also detected. In all the tested models, both CCW and CCM showed their ability to scavenge the free radicals in a does-dependent manner. CCW had higher antioxidant and antiproliferative activities than CCM. In LC-MS-MS analysis, the chromatograms of CCW with good antioxidant activities were established. Rutin might be an important bioactive compound in CCW. The antiproliferative activities of CCW and CCM were also studied in vitro by using human hepatoma HepG2 cells. CCW exhibited good antiproliferative activity. These results indicated that CCW might be used as a potential source of natural antioxidants and as an anti-tumor agent.
