Objectives: The aim of this pilot study was to evaluate dynamic thiol-disulfide homeostasis as a novel oxidative stress parameter in multiple sclerosis (MS), neuromyelitis optica spectrum disorders (NMOSD), and myelin oligodendrocyte glycoprotein antibody-associated disease (MOGAD) to better understand the role of thiol homeostasis in neuroimmunological diseases.Methods: A total of 85 participants were included in this study, consisting of 18 healthy controls, 52 patients diagnosed with MS, seven with NMOSD, and eight with MOGAD. We measured total thiol (–SH+-S–S–) and native thiol (–SH) levels in the serum of all the participants, and in a subset of patients (n = 11), these parameters were investigated in paired cerebrospinal fluid (CSF) and serum samples. Dynamic disulfide concentrations were calculated separately. Finally, we determined if there was any relationship between clinical features and dynamic thiol homeostasis.Results: There was a statistically significant difference between serum and CSF levels of biomarkers of thiol homeostasis. Serum total thiol (317.88 ± 66.04) and native thiol (211.61 ± 44.15) levels were significantly lower in relapsed patients compared to those in remission (368.84 ± 150.36 vs. 222.52 ± 70.59, respectively).Conclusions: Oxidative stress plays a crucial role in the physiopathology of neuroimmunological diseases. Thiol homeostasis may be useful for monitoring disease activity.

Diminished testosterone levels conjoined to cardiovascular risk factor mainly myocardial infarction which broadens the risk of cardiovascular mortality referring to age. Ischemic preconditioning (IPC) is one of the interventions to shield such injury. The present study implicated the possible involvement of caveolin and heat shock protein 72 (HSP-72) during stress in orchidectomy (OCD) challenged rats. OCD was performed in male rats and kept for 6 weeks to observe the reduction in the level of testosterone. Isolated perfused heart of normal and OCD group was subjected to ischemic insult as per IPC cycle. Myocardial infarct size, haemodynamic, enzymatic and oxidative stress parameter were assessed for each heart. Diadzein (DDZ) a caveolin inhibitor was administered before the isolation of heart and it significantly decreases myocardial infarct size, release of lactate dehydrogenase, creatinine kinase and oxidative stress marker. DDZ also potentiated the effect IPC-mediated increase in the heart rate and coronary flow. The effect of caveolin inhibitor was remarkably reduced by quercetin administered before 1 h. of the administration DDZ. The findings of this study revealed that protection of myocardium induced by caveolin inhibitor pretreatment has not been lost in OCD rat heart.

A peptic ulcer is an alimentary tract injury that leads to a mucosal defect reaching the submucosa. This work aimed to optimize and maximize ellagic acid (EA) loading in Ca pectinate floating beads to maximize the release for 24 h. Three factors were selected: Ca pectinate concentration (X1, 1–3 w/v %), EA concentration (X2, 1–3 w/v %) and the dropping time (X3, 10–30 min). The factorial design proposed eight formulations. The optimized EA–Ca pectinate formulation was evaluated for the gastric ulcer index and the oxidative stress parameter determination of gastric mucosa. The results indicated that the optimum EA–Ca pectinate formula significantly improved the gastric ulcer index in comparison with raw EA. The protective effect of the optimized EA–Ca pectinate formula was further indicated by the histopathological features of the stomach. The results of the study indicate that an EA formulation in the form of Ca pectinate beads would be effective for protection against gastric ulcers because of Nonsteroidal anti-inflammatory drugs (NSAID) administration.

Obesity induces the formation of reactive oxygen species (ROS) which is produce oxidative stress that is involved in various pathological processes such as malondialdehyde (MDA) which is a highly toxic molecule. In Indonesia, Muslims are fasting for 13-14 hours during Ramadhan. Researchers interested in conducting research on the effect of fasting Ramadhan on oxidative stress (MDA levels) in individuals with obesity. This research is a prospective cohort. The study was held on  March –  May 2019 at Outpatient Clinic Endocrinology of RSUP. H. Adam Malik Medan with the approval of the FK  USU Research Ethics Commission and according to the criteria of inclusion and exclusion. The Data is analyzed by using the SPPS program where p < 0.05 is considered significant. The results of the study obtained a meaningful decline in the level of MDA before and after the fasting of Ramadhan (p < 0.001). The median rate of MDA before Ramadhan was 4.88 nmol/ml (1.68-101,5 nmol/ml) and after Ramadhan fasting the median rate of MDA became 3.96 nmol/ml (0.97-92,20 nmol/ml). There was decreased of oxidative stress parameter MDA levels after Ramadhan fasting in obese patients.

The aim of this study was to investigate the effect of acupuncture on dynamic thiol–disulphide homeostasis and ischemia-modified albumin (IMA) levels as a novel oxidative stress parameter in migraine patients. Acupuncture treatment was applied to the 22 volunteer migraine patients. The acupuncture treatment consists of 5 sessions with 2 sessions per week. Blood samples have been collected before performing acupuncture, after the 1st session and after the 5th session of the acupuncture. And for the control group blood samples were collected only once. A total of 44 participants were included in the study, and 24 of whom were diagnosed with migraine. The total thiol, native thiol and ischemia-modified albumin (IMA) levels in the serum were measured in all patients and healthy individuals. The dynamic disulphide bond and ischemia-modified albumin (IMA) levels were calculated from these values. The total and native thiol levels of migraine patients participating in the study were found to be significantly higher than the total and native thiol levels of healthy individuals. The total thiol, native thiol, and dynamic disulphide bond levels, and ischemia-modified albumin (IMA) were correlated with attack frequency, pain intensity, or migraine type. Thiol-disülfide homeostasis can play roles in the etiology and severity of Migraine.

Background: The continue efforts for long term reducing of intraocular pressure (IOP) in patients with open angle glaucoma, focused the attention on those patients who received different therapies, other that topical drugs for IOP reduction, that can influence this parameter.. The aim of this study was to evaluate the IOP, total oxidative stress and anti-oxidant capacity of plasma before and after nicergoline therapy in patients with ischemic stroke and associated open angle glaucoma. Material and method: a group of 35 patients with ischemic stroke and chronic open angle glaucoma (under topical beta-blockers therapy) was studied regarding the values of IOP, total oxidative stress (TOS) and total antioxidant capacity (TAC) before (T1) and after 6 month (T2) of nicergoline therapy. Results: IOP values for both eyes were significantly reduced when the values of T1 examination were compared with those of T2 assessment. The total oxidative stress parameter was also significantly reduced after nicergoline therapy together with increasing of total antioxidant capacity of plasma. Conclusions: besides its positive effects on neuronal metabolism for ischemic stroke patients, nicergoline is able to influence the IOP in patients with open angle glaucoma and to improve the plasmatic oxidative stress/antioxidant balance. By this mechanism nicergoiline can contribute to a neuroprotection and better visual function preservation for these patients, improving their chances to neuro-motor rehabilitation and their quality of life.

Acrylamide (ACR) exposure leads to neuronal damage in both experimental animals and humans and has been established as a neurotoxin. The primary focus of this study was to assess the potential ameliorative effect of aqueous extract of coriander seeds against ACR-induced neurotoxicity in a Drosophila model. In this co-exposure study, adult male flies (8–10 days) were exposed (7 days) to ACR (5 mM) with or without aqueous extract of coriander seeds (0.005% and 0.01%) in the medium. The incidence of mortality among flies fed with ACR and extract was significantly reduced (40-60%). The ACR fed flies showed improvement in negative geotaxis assay when co-exposed with coriander extract (50-60%). Further, a significant attenuation in the ACR induced oxidative stress parameter such as Reactive Oxygen Species (ROS), and malondialdehyde (MDA) was evident. Although the effect of ACR with or without extract showed a differential effect on the activities of superoxide dismutase (SOD), catalase (CAT) and glutathione –S – transferase (GST), the reduced levels of Thioredoxin Reductase (TRR) activity was robustly enhanced with extract supplement. Further, the cholinergic and dopaminergic function was assessed among all flies. While ACR induced elevation of acetylcholine esterase (AChE) activity remained unaltered with coriander supplement, a significant improvement was evident in the ACR induced reduction of dopamine levels.  While the underlying mechanism of action needs further investigation, the protective effects may be at least in part related to the antioxidant capacity of the phytoconstituents of coriander.

Rheumatoid arthritis (RA) is a chronic inflammatory disease that reduces life quality and requires long-life therapy. Quercetin (Que) is a natural flavonoid with antioxidant and anti-inflammatory effects. (3-(2-(4-Chlorophenyl)-2-oxoethyl)-5-((6-methyl-4-oxo-4H-chromen-3-yl)methylene) thiazolidine-2,4-dione (TZD) is a thiazolidinedione derivative synthesized in our laboratory. This study was designed to investigate the antioxidant effects of the Que and TZD derivative administration in adjuvant-induced arthritic (AIA) rats. AIA was induced in Wistar rats by the intraplantar injection of Freund�s complete adjuvant (FCA), unilaterally in the right hind paw. The control non-arthritic rats and the arthritic rats were treated with Que (30 mg/kg/day) or TZD derivative (12 mg/kg/day) for 21 days. The antioxidant effects of 5-chromen-yl- thiazolidinedione were compared to Que. The serum levels of malondialdehyde (MDA) and protein carbonyl (PC) groups, the superoxide dismutase (SOD) and catalase (CAT) activity were assessed. AIA rats showed significatly increased oxidative stress parameter levels in the blood. The results indicated that the TZD derivative decreased the blood oxidative stress parameters in the treated arthritic rats, compared to Que. The antioxidant effects of 5-chromen-yl-thiazolidinedione in AIA suggest its therapeutic properties for the clinical treatment of RA.
