Surface browning after harvest is the primary constraint affecting the storage life and market circulation of rambutans. In this study, rambutan fruits were soaked in sodium nitroprusside at different concentrations and stored at 25°C for 8 days to explore the effects on postharvest quality and browning. The weight loss, browning index and superoxide anion radical, hydrogen peroxide and malondialdehyde contents of the treated fruits were reduced compared to those of the control fruits (soaked in distilled water). And fruits treated with sodium nitroprusside had a higher total phenolic content and lower polyphenol oxidase and peroxidase activity. In addition, compared with the control, the treated fruits exhibited higher phenylalanine ammonia lyase, ascorbate peroxidase and superoxide dismutase activities; titratable acidity; and soluble solid, vitamin C and protein contents, indicating high fruit quality. Overall, sodium nitroprusside treatment at 200 μmol L−1 demonstrated the most positive preservation effects. Therefore, sodium nitroprusside treatment, particularly at 200 μmol L−1, can be used as an eco-friendly, safe and convenient method for postharvest quality management and high-efficiency preservation of rambutan fruits.

The improvement of crop yield and quality are currently of prime concern to achieve global food security and sustainability. Plant elicitation with salicylic acid (SA) has been shown to play a key role in regulating plant growth and productivity. Here, the effects of foliar SA application (1, 2, and 4 mM) on antioxidant capacity and phenolic metabolism, among other biochemical parameters, were studied in okra leaves. The results showed that SA treatments markedly increase the levels of endogenous free SA and trigger the accumulation of chlorophylls, soluble proteins, proline, antioxidant capacity, and phenolic compounds, alongside the stimulation of the enzymatic activities of phenylalanine ammonia-lyase and peroxidase, with these changes being paralleled by decreased levels of lipid peroxidation. The beneficial effects of SA treatments were also tested on field-grown okra plants. Results revealed that yield-associated agronomic attributes significantly increased upon SA treatments, especially at 2 mM SA. Altogether, these results point to a connection between SA-induced antioxidant system strengthening and crop growth and productivity. Therefore, foliar application of SA seems to be a potent and workable approach to enhance okra crop quality and yield.

Abstract Fresh-cut Chinese water chestnuts (CWCs) are prone to quality deterioration during storage, which does not meet consumer demand. In this study, the effect of exogenous melatonin (5 mM) on the quality and potential mechanisms in fresh-cut CWC was investigated. The results showed that melatonin treatment alleviated the cut-surface discoloration of CWCs. Not only did this treatment significantly slow down the increase in browning degree and b* as well as the decrease in L*, but also significantly delayed the loss of weight and total soluble solids. Further investigations indicated that melatonin-treated fresh-cut CWCs exhibited significantly lower total phenolics and soluble quinones and suppressed the activities of phenylalanine ammonia-lyase, polyphenol oxidase, and peroxidase. Meanwhile, when fresh-cut CWCs were treated with melatonin, the total flavonoid concentration was significantly decreased compared to the control. Additionally, melatonin significantly inhibited the accumulation of H2O2 and malondialdehyde (MDA) as well as enhanced the activities of superoxide dismutase and catalase by promoting the production of O2 -·. In summary, melatonin treatment may delay the surface discoloration of fresh-cut CWCs by inhibiting phenolic compound metabolism and improving antioxidant capacity, thereby effectively maintaining the quality, and prolonging the shelf life of fresh-cut CWCs.

This research analyzed the phytochemical profile of prickly pear fruits from ‘Orito’ cultivar stored under cold conditions (2 °C, 85–90% RH) and shelf-life conditions at room temperature (stored at 20 °C for three days after cold storage) for 28 days, mimicking the product life cycle. A total of 18 compounds were identified and quantitated through HPLC-DAD-MS/MS (High-Performance Liquid Chromatographic -Diode Array Detector- Mass Spectrometry) analyses. Phenolic acids such as eucomic acid and betalains such as indicaxanthin were the predominant chemical families, and piscidic acid was the most abundant compound. During cold storage, the content of eucomic acid isomer/derivative and syringaresinol increased, and citric acid decreased, which could be caused by the cold activation of the phenylalanine ammonia-lyase (PAL) and polyphenol oxidase (PPO) enzymes. However, no significant differences were found in the content of these compounds during shelf-life storage. These results showed that ‘Orito’ fruit marketability would be possible up to 28 days after harvesting, retaining its profile, which is rich in bioactive compounds.

Abstract Objectives Chilling injury is a prominent physiological disorder in longkong fruit pericarp when stored under 13 °C for a prolonged period. This study aimed to investigate the effects of individual salicylic acid (SA) and ultrasonication (US) treatments and of the combination salicylic acid and ultrasonication (SA-US) on alleviating the chilling injury symptoms in longkong fruit pericarp when in prolonged cold storage. Materials and methods SA (1 mmol/L) and US (40 kHz, 10 min at 90% amplitude, 350 W) were used as individual and combined (SA-US) treatments to control the chilling injury in longkong pericarp. The various quality measures were checked every 2 days in longkong for up to 18 days of cold storage (13 °C, 90% relative humidity). Results The results revealed that the control fruits treated with water exhibited severe chilling injury symptoms followed in rank order by US, SA, and SA-US cases. Treatments such as US and SA alone were more effective in controlling chilling injuries than control, while only minimal significant differences were noticed between them. On the other hand, the longkong pericarp treated with the SA-US combination had significantly increased antioxidant enzyme (superoxide dismutase and catalase) activities and decreased levels of membrane lytic (phospholipase D and lipoxygenase) enzymes and browning-inducing enzymes (phenylalanine ammonia lyase and polyphenol oxidase). Consequently, in the longkong pericarp, the chilling injury index, electrolytic leakage, respiration rate, weight loss, firmness, malondialdehyde content, changes in unsaturated and saturated fatty acid contents, and reactive oxygen species were significantly controlled by this treatment. Conclusions The present study concludes that longkong fruit treatment with a combination of US and SA is an excellent alternative for controlling the chilling injury symptoms and extending the shelf-life.

Phenylalanine ammonia-lyase (PAL) is the first enzyme in the phenylpropanoid pathway and plays a vital role in adoption, growth, and development in plants but in wheat its characterization is still not very clear. Here, we report a genome-wide identification of TaPAL genes and analysis of their transcriptional expression, duplication, and phylogeny in wheat. A total of 37 TaPAL genes that cluster into three subfamilies have been identified based on phylogenetic analysis. These TaPAL genes are distributed on 1A, 1B, 1D, 2A, 2B, 2D, 4A, 5B, 6A, 6B, and 6D chromosomes. Gene structure, conserved domain analysis, and investigation of cis-regulatory elements were systematically carried out. Chromosomal rearrangements and gene loss were observed by evolutionary analysis of the orthologs among Triticum urartu, Aegilops tauschii, and Triticum aestivum during the origin of bread wheat. Gene ontology analysis revealed that PAL genes play a role in plant growth. We also identified 27 putative miRNAs targeting 37 TaPAL genes. The high expression level of PAL genes was detected in roots of drought-tolerant genotypes compared to drought-sensitive genotypes. However, very low expressions of TaPAL10, TaPAL30, TaPAL32, TaPAL3, and TaPAL28 were recorded in all wheat genotypes. Arogenate dehydratase interacts with TaPAL29 and has higher expression in roots. The analysis of all identified genes in RNA-seq data showed that they are expressed in roots and shoots under normal and abiotic stress. Our study offers valuable data on the functioning of PAL genes in wheat.

The soft rot disease caused by Rhizopus stolonifer is an important disease in cherry tomato fruit. In this study, the effect of iturin A on soft rot of cherry tomato and its influence on the storage quality of cherry tomato fruit were investigated. The results showed that 512 μg/mL of iturin A could effectively inhibit the incidence of soft rot of cherry tomato fruit. It was found that iturin A could induce the activity of resistance-related enzymes including phenylalanine ammonia lyase (PAL), polyphenol oxidase (PPO), peroxidase (POD), glucanase (GLU), and chitinase (CHI), and active oxygen-related enzymes including ascorbate peroxidases (APX), superoxide dismutases (SOD), catalases (CAT), and glutathione reductase (GR) of cherry tomato fruit. In addition, iturin A treatment could slow down the weight loss of cherry tomato and soften the fruit. These results indicated that iturin A could retard the decay and improve the quality of cherry tomato fruit by both the inhibition growth of R. stolonifera and the inducing the resistance.
