Natural dibenzo-α-pyrones (DAPs) can be viewed from two opposite angles. From one angle, the gastrointestinal metabolites urolithins are regarded as beneficial, while from the other, the emerging mycotoxin alternariol and related fungal metabolites are evaluated critically with regards to potential hazardous effects. Thus, the important question is: can the structural characteristics of DAP subgroups be held responsible for distinct bioactivity patterns? If not, certain toxicological and/or pharmacological aspects of natural DAPs might yet await elucidation. Thus, this review focuses on comparing published data on the two groups of natural DAPs regarding both adverse and beneficial effects on human health. Literature on genotoxic, estrogenic, endocrine-disruptive effects, as well as on the induction of the cellular anti-oxidative defense system, anti-inflammatory properties, the inhibition of kinases, the activation of mitophagy and the induction of autophagy, is gathered and critically reviewed. Indeed, comparing published data suggests similar bioactivity profiles of alternariol and urolithin A. Thus, the current stratification into hazardous Alternaria toxins and healthy urolithins seems debatable. An extrapolation of bioactivities to the other DAP sub-class could serve as a promising base for further research. Conclusively, urolithins should be further evaluated toward high-dose toxicity, while alternariol derivatives could be promising chemicals for the development of therapeutics.

Choline chloride (CC) application enhanced the tolerance of cluster bean ( Cyamopsis tetragonoloba L.) against salinity stress. The aim of the study was to determine the protective role of CC on plant growth, photosynthesis, and biochemical indicators of oxidative stress. The seeds of BR-99 (tolerant) and BR-2017 (sensitive) were surface sterilized and sown in plastic pots containing river sandy soil. The design of the experiments was completely randomized with 4 replicates per treatment. Three weeks after germination, salinity (150 mM) was imposed. Then plants were sprayed with different concentrations of CC (3, 5, and 10 mM), while normal plants were sprayed with distilled water. Salinity decreased growth attributes, relative water contents, photosynthetic attributes, total soluble proteins, total free amino acids, phenolic, flavonoids, ascorbic acid, proline, and glycine betaine and increased the levels of oxidative stress indicators. However, the application of CC (particularly 5 mM) improved growth attributes, photosynthetic pigments, and activities of antioxidant compounds by reducing the levels of H2O2, malondialdehyde in salt-stressed plants in both cluster bean varieties. BR-99 variety showed more tolerance to salinity stress than that of BR-2017 in the form of greater oxidative defense and osmotic adjustment and clear from greater plant dry masses. Thus, our results showed that the application of CC (5 mM) is an efficient strategy for field use in the areas, where salt stress soils limit agriculture production.

Hip fractures are associated with the highest degree of morbidity and mortality of all fractures in elderly patients and pose a major risk for subsequent fractures. Patients with hip fractures also present accelerated bone turnover despite early stable fracture fixation and early mobilization. We aimed to evaluate oxidative stress in two groups of patients (25 patients each, matched for age, side, and BMI) who underwent internal fixation of hip fractures and total hip arthroplasty for hip osteoarthritis. Blood samples were taken from all patients during admission, the day of surgery, the 4th postoperative day, and the 15th postoperative day. Reduced (GSH) and oxidized (GSSG) glutathione, GSH/GSSG, catalase (CAT), thiobarbituric acid reactive substances (TBARS), protein carbonyls (PC), and total antioxidant capacity (TAC) as a widely used battery of redox biomarkers were recorded from blood samples. Patients with hip fractures who undergo fixation surgery, compared to those with hip osteoarthritis, suffer significant oxidative stress with an active but insufficient first line of oxidative defense, an intensive first line reaction, a very active second line of oxidative defense, and a low plasma antioxidant capacity. Surgery worsened already present lipid- and protein-related tissue damage. The severe oxidative stress observed may explain high morbidity and mortality rates and high bone turnover status, as well as the high incidence of refractures. Furthermore, the question of whether antioxidant therapy measures should be introduced in the management of hip fracture patients is raised.

Pharmaceutical wastes are environmental micro pollutant and potential risk for the ecosystem. Therefore, the present study was planned to find out the effects of different pharmaceutical effluent (PE) regimes on growth, secondary metabolism, and oxidative defense in 2 carrot lines. The seeds of 2 carrot lines (DC-3 and T-29) were spread in plastic pots containing sandy loam soil. The design of experiment was completely randomized with 3 replicates per treatment. At vegetative stage, plants were irrigated with 5 different doses (control), 25%, 50%, 75% and 100%) of PE on every 3-day interval, while control plants were irrigated with canal water. The carrot roots were harvested after 25 days’ application of the treatments to determine various attributes. High concentration of PE caused a substantial decline in growth, beta carotenoids, anthocyanin, total soluble protein, free amino acids, total soluble sugar, phenolic and flavonoid contents and an increase in proline, levels of H2O2 and MDA, activities of antioxidant enzymes such as peroxidase (POD), superoxide dismutase (SOD), and catalase (CAT) in both lines. Moreover, PE caused significant reduction in the levels of essential nutrients (K+, Ca2+) and increased in Na+ content. However, T-29 line was found to be more PE tolerant because it had less H2O2, MDA and ascorbic acid contents. Thus, our findings showed that diluted PE (25%) could not be used for irrigation to increase the growth of plants in nutrients deprived environments without using bio filtration and biocarbon sorption technologies for treatments.
