Due to the large number of people with lactose maldigestion, the dairy industries have increased production and diversity of low lactose and lactose-free foods. Consequently, the need to control the lactose hydrolysis process has also risen. This study aimed to correlate freezing point depression (cryoscopy) and lactose concentration, quantified by high-performance liquid chromatography (HPLC), in UHT milk. To accomplish this, UHT milk samples were subjected to seven lactose hydrolysis treatments, using lactase enzyme, resulting in different lactose concentrations. All samples were subjected to HPLC analysis and freezing point measurement, using a cryoscope. The results were plotted on a graph and a linear regression was performed. There was a strong correlation between lactose concentration and freezing point (R = 0,9973) and the coefficient of determination (R2) was 0,9946, which means that 99,46% of the variability of the response data is explained by the linear regression model. Therefore, the results point to the feasibility of estimating the lactose concentration in milk during the hydrolysis process for the production of low lactose milk, by cryoscopy, a quick analysis, with lower cost compared to HPLC and that is already among the analyses commonly performed in dairy industries.

This article introduces the project NIR4Dairy and presents the studies currently undertaken. Based on an industry partner’s interest, one of the research objectives is to quantify the lactose content of milk protein concentrate in a very low lactose concentration range (e.g. 0.07–0.3%), using a Near Infrared (NIR) transflectance probe connected to a Fourier Transform-Near Infrared (FT-NIR) instrument. An approach to acquire high-quality and repeatable spectra of milk protein concentrate is reported.

The present study aims to investigate the composition including concentrations of IGF-1, IgG and Vit A in colostrum and their effects by litter size and goat parity in 3 groups of goats; Black Bengal (BB), Saanen (SA) and their crossbred (BBSA). Thirty-eight goats were used (23 BB, 7 BBSA and 8 SA). The composition (fat, protein, lactose and total solid; TS) in colostrum (Day 0; D0) and milk (Day 4; D4 and Day 7; D7) were measured. The IGF-1, IgG concentrations were analysed in some samples collected at D0, D4 and D7 while Vit A was analysed only in colostrum. The results showed that colostrum components were similar among experimental groups. However, the colostral IGF-1 concentration of BBSA (983.0 ± 163.6 ng/mL) was higher than that of BB (340.7 ± 85.5 ng/mL, p < 0.01) and SA (417.1 ± 93.9 ng/mL, p < 0.01). The colostral IgG concentration of BB (8.2 ± 0.9 mg/mL) was lower than that of BBSA (12.9 ± 1.7 mg/mL, p < 0.05) and SA (12.9 ± 1.0 mg/mL, p < 0.01). Colostral Vit A concentration in BBSA (787.2 ± 152.6 µg/100 gm) was higher than that in BB (388.9 ± 84.3 µg/100 gm, p < 0.05) but was not different from SA (522.8 ± 96.9 µg/100 gm). Colostrum from all groups contained higher protein and TS but was lower in lactose concentration than milk. The IGF-1 and IgG concentrations in colostrum were much higher than in milk both D4 and D7 (p < 0.001). Additionally, litter size had no effects on colostrum contents but colostrum from goats with a higher parity number had higher IgG concentration. It is concluded that colostrum from BBSA may be superior when fed to BB newborn goats in terms of higher IGF-1, IgG and Vit A contents. Moreover, colostrum from goats with a high parity number contained more IgG content.

The aim of our study was to determine how the ease of calving of cows may influence changes in lactose concentration and other milk components and whether these two factors correlate with each other. To achieve this, we compared data of calving ease scores and average percentage of in-line registered milk lactose and other milk components. A total of 4723 dairy cows from nine dairy farms were studied. The cows were from the second to the fourth lactation. All cows were classified according to the calving ease: group 1 (score 1)—no problems; group 2 (score 2)—slight problems; group 3 (score 3)—needed assistance; group 4 (score 4)—considerable force or extreme difficulty. Based on the data from the milking robots, during complete lactation we recorded milk indicators: milk yield MY (kg/day), milk fat (MF), milk protein (MP), lactose (ML), milk fat/lactose ratio (MF/ML), milk protein/lactose ratio (MP/ML), milk urea (MU), and milk electrical conductivity (EC) of all quarters of the udder. According to the results, we found that cows that had no calving difficulties, also had higher milk lactose concentration. ML > 4.7% was found in 58.8% of cows without calving problems. Cows with more severe calving problems had higher risk of mastitis (SCC and EC). Our data indicates that more productive cows have more calving problems compared to less productive ones.

Milk allergy is among the most common food-related allergies. Milk-based products are recognized as plant protection products (PPPs) in several countries as alternatives to synthetic pesticides. The potential health risk for allergic workers, as well as the general population, is yet to be assessed. An investigation was conducted in the Vaud Canton of Switzerland, where milk-based products are sprayed by helicopter over vineyards. Air lactose concentration was measured at 14 locations via 25 mm IOM Multidust samplers. Residual lactose concentration was measured on the surface of leaves over 7 days following spraying. Surface contamination downwind from the treated area was estimated through computer-based modeling using AgDRIFT® software. The average milk protein concentration inside and outside the vineyard was 0.47 and 0.16 µg/m3, respectively. Milk residues persisted on the leaf surface for an average of three days. Modelling results revealed an estimated order of magnitude of 0.1–0.5 µg/m3 in milk proteins within one hour after the treatment in the close vicinity of the treated area. Our results reveal that the potential exposure to milk proteins in and around helicopter-treated vineyards is not negligible and that prevention messages targeted to individuals with severe allergies should be considered.

Holder pasteurization (HoP; 62.5 °C, 30 min) is commonly used to ensure the microbiological safety of donor human milk (DHM) but diminishes its nutritional properties. A high-temperature short-time (HTST) system was designed as an alternative for human milk banks. The objective of this study was to evaluate the effect of this HTST system on different nutrients and the bile salt stimulated lipase (BSSL) activity of DHM. DHM was processed in the HTST system and by standard HoP. Macronutrients were measured with a mid-infrared analyzer. Lactose, glucose, myo-inositol, vitamins and lipids were assayed using chromatographic techniques. BSSL activity was determined using a kit. The duration of HTST treatment had a greater influence on the nutrient composition of DHM than did the tested temperature. The lactose concentration and the percentage of phospholipids and PUFAs were higher in HTST-treated than in raw DHM, while the fat concentration and the percentage of monoacylglycerides and SFAs were lower. Other nutrients did not change after HTST processing. The retained BSSL activity was higher after short HTST treatment than that following HoP. Overall, HTST treatment resulted in better preservation of the nutritional quality of DHM than HoP because relevant thermosensitive components (phospholipids, PUFAs, and BSSL) were less affected.

In this study, cow reticulorumen content pH and temperature together with the activity were registered using smaXtec boluses, specially designed for animal care. Body weight, rumination time, milk fat/protein ratio, milk yield, milk lactose, milk electrical conductivity, milk somatic cell count and consumption of concentrated feed were registered by Lely Astronaut® A3 milking robots. The cows in this study were assigned into two groups according to the milk lactose concentration: group 1—milk lactose < 4.70% (n = 20), group 2—milk lactose ≥ 4.70% (n = 15). The following cows were further classified according to milk fat and protein ratio: F/P < 1.2 (class 1), F/P = 1.2 (class 2) and F/P > 1.2 (class 3). According to our results, we can conclude that inline registered milk lactose concentration can be used to indicate the health status and reproductive success of fresh dairy cows. Cows with an increased lactose concentration (≥4.70%) showed more activity (54.47%) and had less risk of mastitis (determined by lower milk electrical conductivity (EC) and somatic cell counts (SCC)) and metabolic disorders, determined by milk F/P. A higher glucose concentration was also apparent in the cows with higher lactose concentration. Registered lower levels of milk lactose can be used for early identification of metabolic disorders and mastitis (set at milk SCC ≥ 100 thousand/mL). Lactose levels in cows’ milk were positively associated with their reproductive success.

Abstract Background: The degradation of cellulose and hemicellulose molecules into simpler sugars such as glucose is part of the second generation biofuel production process. Hydrolysis of lignocellulosic substrates is usually performed by enzymes produced and secreted by the fungus Trichoderma reesei. Studies identifying transcription factors involved in the regulation of cellulase production have been conducted but no overview of the whole regulation network is available. A transcriptomic approach with mixtures of glucose and lactose, used as a substrate for cellulase induction, was used to help us decipher missing parts in the network of T. reesei Rut-C30. Results: Experimental results on the Rut-C30 hyperproducing strain confirmed the impact of sugar mixtures on the enzymatic cocktail composition. The transcriptomic study shows a temporal regulation of the main transcription factors and a lactose concentration impact on the transcriptional profile. A gene regulatory network built using BRANE Cut software reveals three sub-networks related to i) a positive correlation between lactose concentration and cellulase production, ii) a particular dependence of the lactose onto the β-glucosidase regulation and iii) a negative regulation of the development process and growth. Conclusions: This work is the first investigating a transcriptomic study regarding the effects of pure and mixed carbon sources in a fed-batch mode. Our study expose a co-orchestration of xyr1, clr2 and ace3 for cellulase and hemicellulase induction and production, a fine regulation of the β-glucosidase and a decrease of growth in favor of cellulase production. These conclusions provide us with potential targets for further genetic engineering leading to better cellulase-producing strains in industry-like conditions.
