Tripartite motif protein 21 (TRIM21) is an interferon-inducible E3 ligase, containing one RING finger domain, one B-box motif, one coiled-coil domain at the N-terminal, as well as one PRY domain and one SPRY domain at the C-terminal. TRIM21 is expressed in many tissues and plays an important role in systemic autoimmunity. However, TRIM21 plays different roles in different virus infections. In this study, we evaluate the relationship between porcine TRIM21 and PCV2 infection as well as host immune responses. We found that PCV2 infection modulated the expression of porcine TRIM21. TRIM21 can enhance interferons and proinflammatory factors and decrease cellular apoptosis in PCV2-infected cells. These results indicate that porcine TRIM21 plays a critical role in enhancing PCV2 infection, which is a promising target for controlling and developing the treatment of PCV2 infection.

Drought and salinity are major important factors that restrain growth and productivity of rice. In plants, many really interesting new gene (RING) finger proteins have been reported to enhance drought and salt tolerance. However, their mode of action and interacting substrates are largely unknown. Here, we identified a new small RING-H2 type E3 ligase OsRF1, which is involved in the ABA and stress responses of rice. OsRF1 transcripts were highly induced by ABA, salt, or drought treatment. Upregulation of OsRF1 in transgenic rice conferred drought and salt tolerance and increased endogenous ABA levels. Consistent with this, faster transcriptional activation of key ABA biosynthetic genes, ZEP, NCED3, and ABA4, was observed in OsRF1-OE plants compared with wild type in response to drought stress. Yeast two-hybrid assay, BiFC, and co-immunoprecipitation analysis identified clade A PP2C proteins as direct interacting partners with OsRF1. In vitro ubiquitination assay indicated that OsRF1 exhibited E3 ligase activity, and that it targeted OsPP2C09 protein for ubiquitination and degradation. Cell-free degradation assay further showed that the OsPP2C09 protein is more rapidly degraded by ABA in the OsRF1-OE rice than in the wild type. The combined results suggested that OsRF1 is a positive player of stress responses by modulating protein stability of clade A PP2C proteins, negative regulators of ABA signaling.

Developing methods for increasing the biomass and improving the plant architecture is important for crop improvement. We herein describe a gene belonging to the RING_Ubox (RING (Really Interesting New Gene) finger domain and U-box domain) superfamily, PLANT ARCHITECTURE and GRAIN NUMBER 1 (PAGN1), which regulates the number of grains per panicle, the plant height, and the number of tillers. We used the CRISPR/Cas9 system to introduce loss-of-function mutations to OsPAGN1. Compared with the control plants, the resulting pagn1 mutant plants had a higher grain yield because of increases in the plant height and in the number of tillers and grains per panicle. Thus, OsPAGN1 may be useful for the genetic improvement of plant architecture and yield. An examination of evolutionary relationships revealed that OsPAGN1 is highly conserved in rice. We demonstrated that OsPAGN1 can interact directly with OsCNR10 (CELL NUMBER REGULATOR10), which negatively regulates the number of rice grains per panicle. A transcriptome analysis indicated that silencing OsPAGN1 affects the levels of active cytokinins in rice. Therefore, our findings have clarified the OsPAGN1 functions related to rice growth and grain development.

Abstract Background The genetic mechanism of glucose metabolism has not been elucidated in nonalcoholic steatohepatitis (NASH), and many genes are took part in glucose metabolism of NASH. In this study, we used the weighted gene co-expression network analysis (WGCNA) to find the key genes associated with glucose metabolism; Methods Data sets GSE96971 and GSE89632 from Gene Expression Omnibus (GEO) were analyzed by WGCNA. We screened the Hub gene from the GSE96971 dataset, and the selected Hub genes were verified by GSE89632 dataset. We then analyzed the dataset using the Gene Ontology (GO) term enrichment and the Kyoto Encyclopedia of Genome (KEGG) path analysis. Expression levels of the hub genes are assessed by qPCR analysis. The function of hub genes was verified by Nile Red staining and relative glucose consumption detection; Results The hub genes are mannosidase beta like (MANBAL), myc proto-oncogene protein (MYC), caspase 4 (CASP4), CDK5 regulatory subunit associated protein 3 (CDK5RAP3) and ZFP36 ring finger protein (ZFP36) in the datasets GSE96971 and the GSE89632. Further, these genes are mainly involved in the integral component of membrane and plasma membrane, the PI3K-AKT signaling pathway and the olfactory transduction according to the GO and KEGG results. These hub genes were significantly up-regulated in the palmitic acid (PA) cell model and methionine-choline-deficient medium (MCD) cell model. After knocking out the hub genes in PA model and the MCD model of NASH, relative glucose consumption was increased and lipid deposition was reduced compared with the control group; Conclusions MANBAL, MYC, CASP4, CDK5RAP3 and ZFP36 are elevated and involved in the pathogenesis of NASH. Further research on these genes are warranted.

Background: Variants in the ring finger protein 213 ( RNF213 ) gene are known to be associated with increased predisposition to cerebrovascular diseases development. Genomic studies have identified RNF213 as a major risk factor of Moyamoya disease in East Asian descendants. However, little is known about the RNF213 (ring finger protein 213) biological functions or its associated pathogenic mechanisms underlying Moyamoya disease. Methods: To investigate RNF213 loss-of-function effect in endothelial cell, stable RNF213-deficient human cerebral endothelial cells were generated using the CRISPR-Cas9 genome editing technology. Results: In vitro assays, using RNF213 knockout brain endothelial cells, showed clear morphological changes and increased blood-brain barrier permeability. Downregulation and delocalization of essential interendothelial junction proteins involved in the blood-brain barrier maintenance, such as PECAM-1 (platelet endothelial cell adhesion molecule-1), was also observed. Brain endothelial RNF213-deficient cells also showed an abnormal potential to transmigration of leukocytes and secreted high amounts of proinflammatory cytokines. Conclusions: Taken together, these results indicate that RNF213 could be a key regulator of cerebral endothelium integrity, whose disruption could be an early pathological mechanism leading to Moyamoya disease. This study also further reinforces the importance of blood-brain barrier integrity in the development of Moyamoya disease and other RNF213-associated diseases.

Abstract Background Fifth carpometacarpal joint (CMCJ) fracture dislocation is a relatively rare injury and most will require operative treatment because of its unstable nature. Improper reduction and fixation lead to joint surface destruction, pain, and reduced grasping power. Intra-articular fragment reduction is often obscured by dorsally displaced ulnar fragment. Therefore, fifth CMCJ arthroscopy can be advantageous in assisting intra-articular fragment reduction. However, there is no detailed description of the portal landmarks or portals' relationship with adjacent important structures in the literature. Purposes To explore the feasibility and safety of fifth CMCJ arthroscopy, locations of the portals are examined in cadaveric hand specimens. Their proximity to important anatomical structures such as dorsal cutaneous branch of ulnar nerve (DCBUN), ring finger and little finger extensor digitorum communis (EDC), and extensor digiti minimi (EDM) is measured. Methods Fifth CMCJ arthroscopy is performed on 11 cadaveric hand specimens by specialist-level surgeon. The portals are marked and portal positions are further confirmed under the fluoroscopy. Then the cadaveric specimens were undergone anatomical dissection by specialist-level surgeon. During dissection, the spatial relationship between the portal positions and DCBUN, EDC to ring finger and little finger, and EDM is identified. The distance between the portals and the above important structures was measured in millimeters. Results DCBUN was consistently found between fourth metacarpohamate (4-MH) and fifth metacarpohamate (5-MH) portals, with it being closer to the latter (mean distance, 2.03 mm; range, 0–4.43 mm; standard deviation [SD], 1.09 mm). The closest tendon for 4-MH portal is ring finger EDC (mean distance, 2.65 mm; range, 0–5.89 mm; SD, 1.78 mm), while 5-MH portal and accessory portal were closest to EDC (mean distance, 1.88 mm; range, 0–3.69 mm; SD, 1.25 mm) and EDM (mean distance, 7.79 mm; range, 6.63–10.72 mm; SD, 1.49 mm), respectively. During the process of specimen dissection, we found no damage to the above structures after portal introduction. Conclusion The above findings support the use of fifth CMCJ arthroscopy, which can be used for assisted reduction in fifth metacarpal base fracture dislocation and hamate body fracture. Gentle soft tissue spreading technique during portal creation prevents injury to the important structure surrounding the portals. Level of evidence This is a Level V study.

Introduction. The group of jewelry with Christian symbols that existed in the Crimea in the early Middle Ages includes cast bronze rings, on a flat shield of which the image of a holy rider with a cross in his hands is engraved. The rings were found in the South-West Crimea in the burial grounds near the village of Luchistoe, Skalistoe and EskiKermen, as well as in Kerch at the early medieval necropolis of the Bosporus, in the burials of the 7th century. Methods. For the attribution of published products, a circle of analogies is identified, findings from the territory of the Eastern Roman Empire are analyzed. The iconography of the holy rider is compared on various subjects. In the Crimea, three of the published finger rings were found in in situ burials, as part of a closed complex, together with tools with a narrow dating, which makes it possible to clarify the time of existence of this type of products in the region and to determine the method of wearing it. Analysis. The plot depicted on the finger rings has an undeniable resemblance to the scene of the solemn entry of Jesus Christ into Jerusalem, described in all four Gospels and well known from the numerous pictorial monuments of the early medieval period. The iconography of that scene originated in the era of Constantine the Great, under the influence of imperial art and in many ways corresponded to the Triumphal entry of the emperor to Rome or any large city of the empire. The image of Christ the rider on the Crimean finger rings belongs to the iconographic type, which became widespread in the 6th–7th centuries, mainly in Egypt, the Syro-Palestinian region and Asia Minor. The quality of execution of the published rings from the Crimea allows us to speak about their local production. Byzantine products that came to the peninsula thanks to stable trade relations with the empire or were brought by pilgrims from holy places served as models for the Crimean artisans. In the minds of ancient Christians, the image of Christ the rider had a powerful protective power. Placed on the shield of the ring, it endowed the decoration with the properties of an amulet, protecting the person wearing it from all troubles. The rings originating from Crimea belonged to a teenage girl and young women who wore them on their hands – on the right index or ring finger, or in special belt bags, in which, in addition to utilitarian items, various amulets were also put. Results. The study of rings makes it possible not only to expand knowledge about jewelry that existed in the early medieval times, but also to replenish our information about the daily life of the Christian population of the early medieval Crimea.

The design of this IoT (Internet of Things)-based post-stroke patient monitoring tool was built to monitor patient progress remotely. Flex sensor hardware was used to retrieve data from every movement of the patient's finger muscle strength, then processed by the NodeMCU ESP32 after that the data will be sent to the web server via the IoT platform using a local wireless network. The results of research on the development of post-stroke patients can be displayed to a web server in the form of a table plot of data values for the index finger, middle finger, and ring finger. The post-stroke patient monitoring system can be accessed on PC and Android devices. The results of the sample test from the device obtained the highest value of 3.26 MMT and the lowest value of 0.2 MMT so that the patient's finger muscle strength was declared unstable and frequent changes in muscle strength.
