Abstract This study proposes a novel exploration of perceived prosodic highlights in continuous speech, focusing on the alternative function of indexing and projecting information content deployment in the speech context. Given the assumption that prosodic highlight allocation directly reflects the interlocutors’ information content deployment, this study foregrounds perception-based prominences for indexing both the key information (KEY) and the projector (PJR) that projects the deployment of key/focal information. Two information content planning units (PJR plus its respective projection PJN, and KEY) prompted by prosodic highlights were established, based on quantitative analyses and discriminative acoustic features. Additional analyses confirm a general heavy-to-light information distribution across both units, showcasing that the relative projection trajectory size in the PJR-PJN unit is positively correlated to its position within discourse-prosodic units. Current results, therefore, directly substantiate the cognitive explanation of prosodic projection in speech, as evidence beyond syntactic relationships are drawn and prosodic projection is shown to involve perceived prosodic highlight allocation and information deployment in a fixed pattern. Explorations of prosody-prompted projection shed light on a more comprehensive account of the mechanism behind information planning, hence facilitating a deeper understanding of the composition of context prosody and the derivation of linguistic invariants from speech.

Histones, ubiquitous in eukaryotes as DNA-packing proteins, find their evolutionary origins in archaea. Unlike the characterized histone proteins of a number of methanogenic and themophilic archaea, previous research indicated that HpyA, the sole histone encoded in the model halophile Halobacterium salinarum, is not involved in DNA packaging. Instead, it was found to have widespread but subtle effects on gene expression and to maintain wild type cell morphology; however, its precise function remains unclear. Here we use quantitative phenotyping, genetics, and functional genomic to investigate HpyA function. These experiments revealed that HpyA is important for growth and rod-shaped morphology in reduced salinity. HpyA preferentially binds DNA at discrete genomic sites under low salt to regulate expression of ion uptake, particularly iron. HpyA also globally but indirectly activates other ion uptake and nucleotide biosynthesis pathways in a salt-dependent manner. Taken together, these results demonstrate an alternative function for an archaeal histone-like protein as a transcriptional regulator, with its function tuned to the physiological stressors of the hypersaline environment.

AbstractHeading date is an important agronomic trait of rice (Oryza sativa L.) and is regulated by numerous genes, some of which exhibit functional divergence in a genetic background-dependent manner. Here, we identified a late heading date 7 (lhd7) mutant that flowered later than wild-type Zhonghua 11 (ZH11) under natural long-day (NLD) conditions. Map-based cloning facilitated by the MutMap strategy revealed that LHD7 was on the same locus as OsPRR37 but exhibited a novel function as a promoter of heading date. A single-nucleotide mutation of G-to-A in the coding region caused a substitution of aspartic acid for glycine at site 159 within the pseudo-receiver (PR) domain of OsPRR37. Transcriptional analysis revealed that OsPRR37 suppressed Ghd7 expression in both ZH11 background under NLD conditions and the Zhenshan 97 background under natural short-day conditions. Consistently, the expression of Ehd1, Hd3a and RFT1 was enhanced by OsPRR37 in the ZH11 background. Genetic analysis indicated that the promotion of heading date and reduction in grain yield by OsPRR37 were partially dependent on Ghd7. Further investigation showed that the alternative function of OsPRR37 required an intact Ghd7-related regulatory pathway involving not only its upstream regulators OsGI and PhyB but also its interacting partner Hd1. Our study revealed the distinct role of OsPRR37 in the ZH11 background, which provides a more comprehensive understanding of OsPRR37 function and enriches the theoretical bases for improvement of rice heading date in the future.

Government stability in the German Bundestag is traditionally tied to a parliamentary majority and an opposition minority . Nonetheless, minority governments in other Western democracies show that, despite the lack of a parliamentary majority, they govern stable and effectively together with the opposition . In this article, on the Swedish case, we examine how opposition parties in parliament are involved in the legislative process in a minority government and what patterns they follow in order to maintain governmental stability without neglecting their alternative function . The paper combines theoretical and concep­tual considerations on the adequate understanding of the opposition in the Federal Repub­lic of Germany with empirical findings on cooperation and conflicts between opposition party groups and minority governments . The results show that opposition parties strategi­cally switch between confrontational (Westminster-style) and consensual patterns of behav­ior (republican) . Through this flexible majority finding, opposition parties in parliament can alternately present themselves as policymakers or as an alternative counterpart to the government . This opposition behavior is functionally adequate under the conditions of a pluralized and fragmented party system and the resulting difficulties in forming a stable government majority .

Abstract During mRNA translation, tRNAs are charged by aminoacyl-tRNA synthetases and subsequently used by ribosomes. A multi-enzyme aminoacyl-tRNA synthetase complex (MSC) has been proposed to increase protein synthesis efficiency by passing charged tRNAs to ribosomes. An alternative function is that the MSC repurposes specific synthetases that are released from the MSC upon cues for functions independent of translation. To explore this, we generated mammalian cells in which arginyl-tRNA synthetase and/or glutaminyl-tRNA synthetase were absent from the MSC. Protein synthesis, under a variety of stress conditions, was unchanged. Most strikingly, levels of charged tRNAArg and tRNAGln remained unchanged and no ribosome pausing was observed at codons for arginine and glutamine. Thus, increasing or regulating protein synthesis efficiency is not dependent on arginyl-tRNA synthetase and glutaminyl-tRNA synthetase in the MSC. Alternatively, and consistent with previously reported ex-translational roles requiring changes in synthetase cellular localizations, our manipulations of the MSC visibly changed localization.

l-cysteine is the source of all bacterial sulfurous biomolecules. However, the cytoplasmic level ofl-cysteine must be tightly regulated due to its propensity to reduce iron and drive damaging Fenton chemistry. It has been proposed that inEscherichia colithe component of cytochromebd-I terminal oxidase, the CydDC complex, shuttles excessivel-cysteine from the cytoplasm to the periplasm, thereby maintaining redox homeostasis. Here, we provide evidence for an alternative function of CydDC by demonstrating that thecydDphenotype, unlike that of the bona fidel-cysteine exportereamA, parallels that of thel-cystine importertcyP.Chromosomal induction ofeamA, but not ofcydDC, from a strong pLtetO-1 promoter (Ptet) leads to the increased level of extracellularl-cysteine, whereas induction ofcydDCortcyPcauses the accumulation of cytoplasmicl-cysteine. Congruently, inactivation ofcydDrenders cells resistant to hydrogen peroxide and to aminoglycoside antibiotics. In contrast, induction ofcydDCsensitizes cells to oxidative stress and aminoglycosides, which can be suppressed byeamAoverexpression. Furthermore, inactivation of the ferric uptake regulator (fur)in Ptet-cydDCor Ptet-tcyPcells results in dramatic loss of survival, whereas catalase (katG) overexpression suppresses the hypersensitivity of both strains to H2O2. These results establish CydDC as a reducer of cytoplasmic cystine, as opposed to anl-cysteine exporter, and further elucidate a link between oxidative stress, antibiotic resistance, and sulfur metabolism.

AbstractSocial insect colonies use negative as well as positive feedback signals to regulate foraging behaviour. In ants and bees individual foragers have been observed to use negative pheromones or mechano-auditory signals to indicate that forage sources are not ideal, for example being unrewarded, crowded, or dangerous. Here we propose an alternative function for negative feedback signals during foraging, variance reduction. We show that while on average populations will converge to desired distributions over forage patches both with and without negative feedback signals, in small populations negative feedback reduces variation around the target distribution compared to the use of positive feedback alone. Our results are independent of the nature of the target distribution, providing it can be achieved by foragers collecting only local information. Since robustness is a key aim for biological systems, and deviation from target foraging distributions may be costly, we argue that this could be a further important and hitherto overlooked reason that negative feedback signals are used by foraging social insects.

ABSTRACTDysfunction of mitochondrial DNA replication machinery is a common cause of mitochondrial diseases. The minimal mammalian replisome is made up of DNA polymerase gamma, replicative helicase Twinkle, and single-stranded DNA binding protein. The replisome is localized to the inner mitochondrial membrane and serves as the site of mitochondrial DNA replication and mitochondrial fission. Recently, a sequence homolog of Twinkle was uncovered in the nematode Caenorhabditis elegans. Here, we characterized this homolog, twnk-1, and report that twnk-1 does not function as the primary mitochondrial DNA replicative helicase in this species, as loss of twnk-1 does not result in reduce mitochondrial DNA levels, or result in other expected mitochondrial dysfunctions such as reduced oxygen consumption rates, increased sensitivity to metabolic perturbations, or reduced muscle function. Instead, twnk-1 mutants have increased mitochondrial DNA as they age, and exhibit phenotypes associated with mitochondrial stress, including reduced fecundity, an activation of the mitochondrial unfolded protein response, and mitochondrial fragmentation. Our results suggest in Caenorhabditis elegans, twnk-1 does not function as the mitochondrial DNA replicative helicase, but has an alternative function in regulating mitochondrial function.

AbstractMany protein families harbor pseudoenzymes that have lost the catalytic function of their enzymatically active counterparts. Assigning alternative function and importance to these proteins is challenging [1]. Because the evolution towards pseudoenzymes is driven by gene duplication, they often accumulate in multigene families. Plant cell wall-degrading enzymes (PCWDEs) are prominent examples of expanded gene families. The pectolytic glycoside hydrolase family 28 (GH28) allows herbivorous insects to break down the PCW polysaccharide pectin. GH28 in the Phytophaga clade of beetles contains many active enzymes but also many inactive counterparts. Using functional characterization, gene silencing, global transcriptome analyses and recordings of life history traits, we found that not only catalytically active but also inactive GH28 proteins are part of the same pectin-digesting pathway. The robustness and plasticity of this pathway and thus its importance for the beetle is supported by extremely high steady-state expression levels and counter-regulatory mechanisms. Unexpectedly, the impact of pseudoenzymes on the pectin-digesting pathway in Phytophaga beetles exceeds even the influence of their active counterparts, such as a lowered efficiency of food-to-energy conversion and a prolongation of the developmental period.
