This paper analyses the latest techniques for treating wastewater to make it suitable for agricultural applications in regions where irrigation water is scarce. Micro-filtration (MF) techniques yield a significant reduction in Chemical Oxygen Demand (COD), Biological Oxygen Demand (BOD), Total Suspended Solids (TSS), and Total Bacterial Count (TBC) of wastewater, which makes it suitable to be used for irrigational purposes. Microbial Fuel Cell (MFC) technology is a viable solution for treating wastewater discharged from many industrial sectors, such as the food processing industry, for reclaiming water for agro-applications. Such industrial water may seal soil pores if directed untreated to agricultural fields. Concerning the treatment of microbial contamination of wastewater, the removal rate of pressurized membrane bio-booster (MBR) is significantly large for coliform and metals such as lead, copper, chromium, and arsenic. Both electrocoagulation and chemical coagulation are applied in the removal of oxidable chemicals from wastewater. However, the electrocoagulation process shows a higher efficiency in terms of removing COD. Contamination of agricultural fields with heavy metals is considered an adverse impact on the human and animal safety of discharging wastewater into agro-fields. Thus, removing such contaminants should be given the utmost priority in wastewater treatment, especially from industrial discharge, before they are directed to agricultural usage. Factors that govern the sustainability of a given method in a water-scarce region are also discussed.

Companion animals can both protect against, and increase risk for, coercive control and abuse, yet have not been considered in existing UK COVID-19 reports of domestic abuse. This study aimed to explore the nature and frequency of animal-related calls received by UK domestic abuse helpline staff during the COVID-19 pandemic, examine any lockdown-related changes, identify potential commonalities across helpline organisations, and explore perspectives about ongoing animal-related issues in the context of domestic abuse. Semi-structured virtual interviews were conducted with 11 domestic abuse helpline staff workers during the COVID-19 pandemic. Data were subjected to thematic analysis. The analysis revealed four overarching themes. Theme 1) Lockdown-related changes in the frequency and nature of animal-related calls received. Theme 2) Animals as tools for abuse during lockdown, with subthemes a) Manipulating the family-animal bond, and b) Fears over animal safety. Theme 3) Animals as barriers to refuge during lockdown, with subthemes, a) Lack of animal-friendly accommodation, b) Lack of social support systems, and c) Animals as coping mechanisms. Theme 4) Helpline staffs’ awareness of and links to animal friendly accommodation and fostering services. The findings can inform decision making regarding appropriate long-term support needs for multi-species families with complex needs, both during and post-pandemic.

Abstract Background The ability of 3D printing using plastics and resins that are magnetic resonance imaging (MRI) compatible provides opportunities to tailor design features to specific imaging needs. In this study an MRI compatible cradle was designed to fit the need for repeatable serial images of mice within a mouse specific low field MRI. Methods Several designs were reviewed which resulted in an open style stereotaxic cradle to fit within specific bore tolerances and allow maximum flexibility with interchangeable radiofrequency (RF) coils. CAD drawings were generated, cradle was printed and tested with phantom material and animals. Images were analyzed for quality and optimized using the new cradle. Testing with multiple phantoms was done to affirm that material choice did not create unwanted image artifact and to optimize imaging parameters. Once phantom testing was satisfied, mouse imaging began. Results The 3D printed cradle fit instrument tolerances, accommodated multiple coil configurations and physiological monitoring equipment, and allowed for improved image quality and reproducibility while also reducing overall imaging time and animal safety. Conclusions The generation of a 3D printed stereotaxic cradle was a low-cost option which functioned well for our laboratory.

Artemisia annua (AAH) is traditionally used as an anti-malarial, expectorant and antipyretic Chinese medicine. The aim of this study was to explore the therapeutic effect of Qinghao Powder (QHP) on chicken coccidiosis, evaluate the safe dosage of QHP, and provide test basis for clinical medication. High-performance liquid chromatography (HPLC) and thin-layer chromatography (TLC) were used to detect artemisinin in Qinghao Powder (QHP) for quality control. The level of artemisinin in QHP was 81.03 mg/g. A total of 210 chicks (14 days of age) were divided randomly into seven groups: three QHP treatments (0.15, 0.30, and 0.60 g/kg), a toltrazuril control (1.00 mL/L), a sulfachloropyrazine sodium control (SSC, 0.30 g/L), an E. tenella-infected control, and a healthy control group. All the groups were inoculated orally with 7 × 104E. tenella oocysts except for the healthy control group. After seven days of administration, compared with the infected control group, chicks which were administered QHP, SS, and toltrazuril showed less bloody feces, oocyst output, and cecal lesions, and the protection rates were improved. The maximum rBWG and ACI were found in the SS-medicated group, followed by the groups medicated with 0.60 and 0.30 g/kg QHP. Therefore, a 0.30 g/kg dose level of QHP in the feed was selected as the recommend dose (RD) in the target animal safety test, in which 80 broiler chicks (14 days of age) were randomly divided into four major groups (I-healthy control group; II-1× RD; III-3× RD; IV-6× RD), with each group subdivided into two subgroups (A and B) consisting of 10 chicks each. After 7-day (for sub-group A) or 14-day (for sub-group B) administration, compared with the healthy control, treatment-related changes in BWG, feed conversion ratio (FCR), relative organ weight (ROW) of the liver, WBC counts, and levels of RBC, HGB, ALT, AST, and TBIL were detected in the 3× and 6× RD groups. No differences were noted in necropsy for all doses, and histopathological examinations exhibited no QHP-associated signs of toxicity or abnormalities in the liver or kidney. The findings suggest that QHP at a dose of 0.30 g/kg feed would be appropriate for therapy and intermittent treatment of E. tenella-infected chicks, the dosage in clinical applications should be set according to the recommended dose to ensure animal safety.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) variants of concern (VOCs) continue to wreak havoc across the globe. Higher transmissibility and immunologic resistance of VOCs bring unprecedented challenges to epidemic extinguishment. Here we describe a monoclonal antibody, 2G1, that neutralizes all current VOCs and has surprising tolerance to mutations adjacent to or within its interaction epitope. Cryo-electron microscopy structure showed that 2G1 bound to the tip of receptor binding domain (RBD) of spike protein with small contact interface but strong hydrophobic effect, which resulted in nanomolar to sub-nanomolar affinities to spike proteins. The epitope of 2G1 on RBD partially overlaps with ACE2 interface, which gives 2G1 ability to block interaction between RBD and ACE2. The narrow binding epitope but high affinity bestow outstanding therapeutic efficacy upon 2G1 that neutralized VOCs with sub-nanomolar IC50 in vitro. In SARS-CoV-2 and Beta- and Delta- variant-challenged transgenic mice and rhesus macaque models, 2G1 protected animals from clinical illness and eliminated viral burden, without serious impact to animal safety. Mutagenesis experiments suggest that 2G1 could be potentially capable of dealing with emerging SARS-CoV-2 variants in future. This report characterized the therapeutic antibodies specific to the tip of spike against SARS-CoV-2 variants and highlights the potential clinical applications as well as for developing vaccine and cocktail therapy.

Animal use for testing chemicals under REACH continues to increase, despite advances in non-animal safety science during the past 15 years. The application of modern science and technology, and the use of ‘next generation’ weight-of-evidence assessment approaches, are embedded in EU guidance for establishing the safety of cosmetics and foods – and of the ingredients used in these products. However, this is still not the case for the regulation of chemicals. Under the new Chemicals Strategy for Sustainability, thought leaders in human health and environmental protection are calling on the European Commission to quickly embrace the benefits of modern and innovative non-animal safety science, in place of outdated animal testing, if the EU is to be a leader in safe and sustainable innovation under the European Green Deal transformational change ambitions. The European Commission also needs to enable companies to meet their legal obligation to only conduct animal testing as a last resort, by providing a more flexible, science-based and consistent regulatory framework for assuring chemical safety, which supports the integration of data from different sources. We are at a tipping point for closing the gap between regulatory chemicals testing and modern safety science. It is time to join forces, across policy makers, scientists, regulators and lawyers, to lead the paradigm shift needed to deliver what EU citizens want – namely, chemicals and products that are safe and sustainable, without resorting to animal testing.

Abstract Background Daxocox® [Ecuphar/Animalcare Group] contains the selective COX-2 inhibitor enflicoxib, approved in the EU for the treatment of pain and inflammation associated with osteoarthritis in dogs. The safety of Daxocox® was evaluated in a target animal safety study: Groups of 4 dogs per sex each were treated once weekly with placebo or Daxocox tablets at 1-, 3- and 5-times (1X, 3X and 5X) the maximum recommended therapeutic dose of enflicoxib (0, 4, 12 or 20 mg/kg, respectively). After an initial loading dose, dogs in the placebo control, 1X and 3X groups were administered for 32 weeks, and those in the 5X group were administered for 13 weeks. Dogs were subjected to daily food consumption measurements and clinical and dose observations. Body weight measurements, physical examinations, clinical pathology, urinalysis, faecal occult blood (FOB) and electrocardiographic (ECG) and blood pressure measurements, buccal mucosal bleeding time (BMBT), ophthalmology and gastroduodenal endoscopy examinations were conducted throughout the study. At study completion, all dogs were subjected to gross necropsy. Histopathology was performed on selected tissues from all animals in all groups. Results No clinical signs were noted, and no toxicologically relevant dose-associated effects were observed. Conclusions Results show that Daxocox® is well-tolerated and has a broad safety margin when administered as directed in dogs.

The confirmation that Epichloë endophytes are important for pest resistance in New Zealand pastures led to the development of a range of novel endophyte strain × host plant combinations that enhance the persistence of the grass, while mitigating adverse effects on grazing animals. Successfully delivering these endophytes to the pastoral industry has required the development of a range of scientific and commercial capabilities. In March 2012, the New Zealand proprietary seed industry established the Endophyte Technical Committee. This is a cooperative forum to ensure that endophyte strains in commerce or under development are tested uniformly, and to publish industry-agreed descriptions of the animal safety and insect control of commercial endophyte strains each year.

There is a strong industry demand for technically simple and highly efficacious alternatives to heat cautery disbudding in goat kids that can be performed as a stand-alone procedure without adjunct anesthesia, and that result in improved overall welfare through reduced acute pain, reduced tissues healing interval, and a consistent safety record. The objective of this study was to consider the net effect of disbudding techniques on goat welfare by examining vocalization frequency, long-term efficacy and animal safety associated with four alternative caprine disbudding methods against sham-disbudded and heat-cautery controls. Sixty-five commercial male dairy kids were disbudded at 3–10 days of age with one of six disbudding treatments (clove oil injection, caustic paste, two cryosurgical methods, heat-cautery, and sham procedure). Heat cautery was 91% effective, caustic paste was 55% effective, and the other treatments were ineffective. Heat cautery and sham procedures resulted in similar vocalization efforts; freezing with a liquid-nitrogen cooled iron resulted in significantly greater vocalization numbers. No unintended paste transfer injuries were observed with short-term application of the caustic paste. Heat cautery resulted in numerous superficial infections but no permanent injury. Clove oil injection was associated with several unexpected and severe complications including unintended tissue necrosis, temporary paresis, skull defects, meningitis, and death. Collectively, we did not find that any of the alternative methods of disbudding provided a feasible option over heat cautery to improve welfare.
