Abstract Cyclitols are secondary metabolites with bioactive properties, naturally occurring in plants. Accumulation of such metabolites is directly connected with abiotic stressing factors. This article investigates the influence exercised by a series of abiotic factors including plant treatment with elicitors and cultivability in various regions of Europe, in some cases during two seasons, onto the amounts of cyclitols produced in alfalfa (Medicago sativa L.). The obtained results highlighted that NaCl elicitation acts to increase the quantity of cyclitols, while AgNO3 and Zn(NO3)2 generally decreased the obtained amount of cyclitols. When considering the seasonal impact, samples harvested in August presented a double amount of cyclitols in comparison with those collected in May. Correlation analysis proved that this phenomenon is related mainly to sunshine period versus low humidity. However, our investigation suggests that soil type, salinity level, lack of humidity, the number of sunny days, and plant elicitation play a role on the amount of cyclitols produced in alfalfa plant.

Differences in the assimilation of individual organic compounds (5 mM sugars and L-asparagine) under mixotrophic growth conditions were described for three naturally occurring Haematococcus strains.The effects of assimilation were measured by the growth intensity and size of algal cells, and the effect of colour changes in the cultures was observed. Some compounds caused the cell colouration to change from green to yellow, being the result of chlorophyll disappearance and the accumulation of yellow secondary carotenoids. In the present experiment none of the cultures turned red, thus excluding the intense accumulation of the commercially interesting carotenoid, astaxanthin.

Ecdysteroids (ECs) are steroid hormones originally found in the animal kingdom where they function as insect molting hormones. Interestingly, a relatively high number of these substances can also be formed in plant cells. Moreover, ECs have certain regulatory effects on plant physiology, but their role in plants still requires further study. One of the main aims of the present study was to verify a hypothesis that fenarimol, an inhibitor of the biosynthesis of ECs in the animal kingdom, also affects the content of endogenous ECs in plants using winter wheat Triticum aestivum L. as a model plant. The levels of endogenous ECs in winter wheat, including the estimation of their changes during a course of different temperature treatments, have been determined using a sensitive analytical method based on UHPLC-MS/MS. Under our experimental conditions, four substances of EC character were detected in the tissue of interest in amounts ranging from less than 1 to over 200 pg·g−1 FW: 20-hydroxyecdysone, polypodine B, turkesterone, and isovitexirone. Among them, turkesterone was observed to be the most abundant EC and accumulated mainly in the crowns and leaves of wheat. Importantly, the level of ECs was observed to be dependent on the age of the plants, as well as on growth conditions (especially temperature). Fenarimol, an inhibitor of a cytochrome P450 monooxygenase, was shown to significantly decrease the level of naturally occurring ECs in experimental plants, which may indicate its potential use in studies related to the biosynthesis and physiological function of these substances in plants.

AbstractPolymer composites are attractive for structural applications in the built environment due to their lightweight and high strength properties but suffer from degradation due to environmental factors. While abiotic factors like temperature, moisture, and ultraviolet light are well studied, little is known about the impacts of naturally occurring microbial communities on their structural integrity. Here we apply complementary time-series multi-omics of biofilms growing on polymer composites and materials characterization to elucidate the processes driving their degradation. We measured a reduction in mechanical properties due to biologically driven molecular chain breakage of esters and reconstructed 121 microbial genomes to describe microbial diversity and pathways associated with polymer composite degradation. The polymer composite microbiome is dominated by four bacterial groups including the Candidate Phyla Radiation that possess pathways for breakdown of acrylate, esters, and bisphenol, abundant in composites. We provide a foundation for understanding interactions of next-generation structural materials with their natural environment that can predict their durability and drive future designs.

Sprouts are generally accepted as a pro-healthy food. They are consumed as a source of valuable macronutrients, antioxidants, microelements, and vitamins. Changing growth conditions of sprouts enables modification of their nutritional quality, as well as their safety. Thus, in order to achieve the most desirable composition of the produced sprouts, the conditions for their production are optimized. The aim of this review is to present methods currently used to modify the nutritional quality of plant sprouts. Most scientific works focus on stress conditions inducing the synthesis of secondary metabolites, mainly antioxidants. An increase in their content is achieved after application of physical (e.g., light illumination, temperature) or chemical factors (e.g., salinity stress, phytohormones, metal ions, etc). Though the application of these modifications on a larger scale is problematic. These problems include difficulties in predicting the effect of the stressor and an increased price of the obtained sprouts. However, since it is possible to enrich sprouts with valuable health-promoting substances, these methods are still considered very promising.

Introduction: Galling insects manipulate the development of their host plants. Galls are rich in secondary metabolites, such as phenolic compounds and terpenes. Caryocar brasiliense acts as a host of galling insects. Few studies report the impacts of this plant-insect interaction on the species' morphology, physiology, and secondary metabolism. Objective: The purpose of this study was to verify if there is a correlation between the levels of total phenols and flavonoids in C. brasiliense leaf extracts and environmental variables, such as temperature and precipitation, throughout the year, and to evaluate how galling insects can interfere in this seasonality. Methods: Ethanol extracts from leaves were obtained by soxhlet extraction. The main classes of secondary metabolites were detected by phytochemical prospecting. The levels of total phenols and flavonoids were quantified by the Folin-Dennis tests and complexation reaction with aluminum chloride, respectively. Pearson 's correlation coefficient shows that the levels of phenols and flavonoids were correlated with the average monthly maximum temperatures and precipitation accumulation, respectively, recorded in Gurupi - Tocantins. Results and Discussion: Phytochemical prospecting indicated flavonoids, saponins, tannins, triterpenes, steroids, and alkaloids in both extracts. In C. brasiliense, the samples of healthy leaves suffered a greater variation in the levels of phenols (68.39±2.3 to 279.34±3.1 mg GAE g-1 extract) and flavonoids (164.06± 3.0 at 269.03±3.6 mg QE g-1 extract) compared to leaf samples with galls. A strong positive correlation was found between the contents of total phenols and maximum temperatures and a strong negative correlation with precipitation through Pearson's correlation. Conclusions: The variations observed in the levels of phenols and flavonoids in the extracts of C. brasiliense seem to be related to abiotic factors and the herbivory promoted by the galling insect.

Fungal improvement of the nutritive content of low-quality forages can be affected by several factors among which loss of water-soluble content (WSC) plays a major role. To achieve this aim, two growth conditions i.e. forage-liquid ratios (1:3 and 1:5) and two inoculation times (14 and 28 days) were used to cultivate the selected fungi i.e. Pleurotus ostreatus (PO) at 30°C and Ceriporiopsis rivulosus (CR) at 20°C on Brachiaria decumbens (BD), Andropogon gayanus (AG), Triticum aestivum (TA) straw, Lolium perenne (LP), respectively with the view of selecting the optimal conditions that facilitate the release of WSC. The impact that losses of WSC have on the ability of fungi to improve the nutritive content (i.e. proximate, fibre, secondary metabolites and total antioxidant content) of the forages were then measured using 2 filtering methods i.e. light pump filtering method (LFM) and free flow filtering method (FFM). The optimal conditions that supported increased th soluble was identified as 28th day for both fungi; forage-liquid ratio of 1:5 for both fungi in most of the forages except BD (CR) and BD & AG (PO). The LFM led to lesser or no improvement in the nutritive quality of the upgraded forages while the opposite was recorded with the LFM. The LFM as against the FFM produced upgraded forages with lesser reduction in NDF; similar or higher ADF and lignin contents; and similar reduction in secondary metabolites and TAC when compared with un-improved forages. It can be concluded that the fungal improvement of the nutritive content of low-quality forages was negatively affected by the loss of WSC. It is therefore recommended that fungal improvement of the nutritive quality of low quality forages should be carried out with methods or techniques that facilitates little or no WSC loss in the upgraded forages.

The number of ecometabolomic studies, which use metabolomic analyses to disentangle organisms’ metabolic responses and acclimation to a changing environment, has grown exponentially in recent years. Here, we review the results and conclusions of ecometabolomic studies on the impacts of four main drivers of global change (increasing frequencies of drought episodes, heat stress, increasing atmospheric carbon dioxide (CO2) concentrations and increasing nitrogen (N) loads) on plant metabolism. Ecometabolomic studies of drought effects confirmed findings of previous target studies, in which most changes in metabolism are characterized by increased concentrations of soluble sugars and carbohydrate derivatives and frequently also by elevated concentrations of free amino acids. Secondary metabolites, especially flavonoids and terpenes, also commonly exhibited increased concentrations when drought intensified. Under heat and increasing N loads, soluble amino acids derived from glutamate and glutamine were the most responsive metabolites. Foliar metabolic responses to elevated atmospheric CO2 concentrations were dominated by greater production of monosaccharides and associated synthesis of secondary metabolites, such as terpenes, rather than secondary metabolites synthesized along longer sugar pathways involving N-rich precursor molecules, such as those formed from cyclic amino acids and along the shikimate pathway. We suggest that breeding for crop genotypes tolerant to drought and heat stress should be based on their capacity to increase the concentrations of C-rich compounds more than the concentrations of smaller N-rich molecules, such as amino acids. This could facilitate rapid and efficient stress response by reducing protein catabolism without compromising enzymatic capacity or increasing the requirement for re-transcription and de novo biosynthesis of proteins.
